Three haplotypes of the recently discovered bacterium species “Candidatus Liberibacter solanacearum” are described and related to geographic ranges. The first two are associated with Zebra Chip/Psyllid
Yellows of potatoes and other solanaceous plants, vectored by the tomato/potato psyllid Bactericera cockerelli in North and Central America and New Zealand. The third is associated with diseased carrots in Finland and vectored by the
carrot psyllid Trioza apicalis. The haplotypes are described by SNPs on the 16s rRNA, 16s/23s ISR and 50s rplJ and rplL ribosomal protein genes. These SNPs
are inherited as a package across the three genes. Haplotype “a” has been found primarily from Honduras and Guatemala through
western Mexico to Arizona and California, and in New Zealand. Haplotype “b” is currently known from eastern Mexico and northwards
through Texas to south central Washington. These haplotypes show some range overlap in Texas, Kansas and Nebraska. The haplotypes
are not yet known to elicit biological differences in the plant or insect hosts. These apparently stable haplotypes suggest

BACKGROUND: The spotted wing Drosophila, Drosophila suzukii (Matsumura, 1931) (Diptera: Drosophilidae), is an invasive pest of small‐fruit crops. Unlike most other Drosophila, this insect is able to oviposit into and damage ripe and ripening fruit, making it unmarketable. Because this is a new pest in the United States, it is necessary to identify registered insecticides to manage this insect effectively in conventional and organic production systems. RESULTS: The present laboratory bioassays and field trials identified a number of insecticides representing various modes of action that are effective in controlling D. suzukii. Products that performed well in the laboratory bioassay also performed well in the field, indicating that screening of new chemistries in the laboratory is a worthy exercise. Field application of pyrethoids, organophosphates or spinosyns provided 5–14 days of residual control of D. suzukii. The efficacy of the neonicotinoids as adulticides was not satisfactory compared with the other contact‐mode‐of‐action chemistries. Based on the zero tolerance by the small‐fruit industry and the individual effects mentioned above, neonicotinoids are not currently recommended for D. suzukii management. BACKGROUND: RNA interference (RNAi) is a breakthrough technology for conducting functional genomics studies and also as a potential tool for crop protection against insect pests. The major challenge for efficient pest control using RNAi in the field is the development of efficient and reliable methods for production and delivery of double‐stranded RNA (dsRNA). In this paper, the potential of feeding dsRNA expressed in bacteria or synthesized in vitro to manage populations of Colorado potato beetle, Leptinotarsa decemlineata (Say) (CPB), was investigated. RESULTS: Feeding RNAi successfully triggered the silencing of all five target genes tested and caused significant mortality and reduced body weight gain in the treated beetles. This study provides the first example of an effective RNAi response in insects after feeding dsRNA produced in bacteria. CONCLUSION: These results suggest that the efficient induction of RNAi using bacteria to deliver dsRNA is a possible method for management of CPB. This could be also a promising bioassay approach for genome‐wide screens to identify effective target genes for use as novel RNAi‐based insecticides. Chia, Salvia hispanica L., was a staple crop in pre-Columbian Mesoamerica. Despite the great potential of the species as an oilseed crop, little research related to domesticated and wild varieties exists. A study was undertaken to assess genetic diversity among 38 wild and domesticated accessions of S. hispanica collected throughout Mesoamerica by using RAPD markers. Genetic diversity was higher among wild varieties (H
G= 0.15) than all domesticated varieties (H
G= 0.10) and modern commercial domesticated varieties (H

Declines of amphibian populations have been well documented in the US, Canada, and Central America, but little is known regarding the status of amphibian populations in Mexico. In 2000, we surveyed 43 transects from 3 upland regions in Guerrero and Oaxaca, Mexico. We found 161 adult amphibians belonging to 39 species, representing only 19-48% of the anuran fauna known from these regions. We found one dead (Eleutherodactylus saltator) and one dying frog (Ptychohyla erythromma) from two different streams near Chilpancingo, Guerrero. Both frogs were infected with Batrachochytridium dendrobatidis, a pathogenic fungus involved in other declines of amphibian populations. We collected 368 tadpoles; 60 (19%) tadpoles from 9 different streams among the three regions were missing mouthparts, which is indicative of infection by chytrid fungus. We report additional data from the state of Chiapas, Mexico, that document declines, disease, and apparent extirpations from that region as well. Overall, we report 31 populations (representing 24 species) that appear to have been extirpated, including populations of as many as 11 endemic species that have been missing for 16-40 years and may be extinct. 相似文献

In constructed wetlands (CWs) with horizontal sub-surface flow, nutrient removal, especially phosphorus, is limited because the root biomass fills the pore spaces of the substrate (usually gravel), directing wastewater flow to deeper wetland media; plants are not regularly harvested; the litter formed by decomposing vegetation remains on the surface of the substrate and thus does not interact with the wastewater; and the substrate media often used provide only limited adsorption. Effective nutrient removal including rootzone oxidation, adsorption and plant uptake therefore requires sufficient interaction of wastewater with the treatment media. We assessed the feasibility of biological nutrient removal from wastewater using substrate-free CWs with horizontal flow, planted with two tropical macrophytes namely, Cyperus papyrus and Miscanthidium violaceum. The objectives were to evaluate the system treatment efficiency under semi-natural conditions, and to assess microbial and plant biomass contributions to nutrient removal in the CWs. Results showed high removal efficiencies for biochemical oxygen demand, ammonium-nitrogen (NH4–N) and phosphorus (P) fractions in papyrus-based CWs (68.6–86.5%) compared to Miscanthidium (46.7–61.1%) and unplanted controls (31.6–54.3%). Ammonium oxidizing bacteria in CW root–mats (108–109 cells/gram dry weight) and residual nitrite and nitrate concentrations in the water phase indicated active system nitrification. Papyrus showed higher biomass production and nutrient uptake, contributing 28.5% and 11.2%, respectively, of the total N and P removed by the system compared to 15% N and 9.3% P removed by Miscanthidium plants. Compared to literature values, nitrification, plant uptake and the overall system treatment efficiency were high, indicating a high potential of this system for biological nutrient removal from wastewaters in the tropics. 相似文献

Seeds of cabbage, lettuce, millet, radish, turnip, and wheat were treated with solutions containing Be, Ni, Tl, or V, and subsequent effects on seed germination and radicle elongation were measured after three days. Treatment with low concentrations of Be, Ni, or V stimulated root elongation in most species. Higher concentrations of these elements and all treatment with Tl caused reductions in root elongation. In general, turnip and lettuce were the most sensitive of the plants studied to the metals tested, while wheat and millet were the least sensitive. 相似文献

Commercial pectolytic enzymes were investigated for their influence on phenolics and antioxidant activities of asparagus juice. The antioxidant activity of asparagus juice was analyzed according to 2,2'-diphenyl-l-picrylhydrazyl and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) methods. The enzymes, with the exception of pectinase from Rhizopus sp., contained rutinase, which hydrolyzed rutin to quercetin. Asparagus juice treated with Viscozyme had the highest quercetin content without exhibiting a significant increase in the antioxidant activity. For a pectinase from Aspergillus niger, the antioxidant activity of asparagus juice was markedly reduced. Caution should be paid in the selection of pectolytic enzyme preparations for production of antioxidant activity-rich juice. 相似文献

Brazil nuts have been classified as the foodstuffs that contain the highest level of unadulterated selenium, an essential trace element that appears to prevent cancer. To date, characterization of the selenium species in brazil nuts has not yet been investigated. In this work, various sample preparation approaches, including microwave extractions and enzymatic treatments, are examined with the goal of species preservation and subsequent selenium speciation; of these approaches, an enzymatic treatment with Proteinase K proved most effective. High-performance liquid chromatography (HPLC) separation strategies and inductively coupled plasma mass spectrometry (ICP-MS) detection schemes will also be presented. Extracts are evaluated against available standards for the commercially obtainable seleno-amino acids, selenomethionine (SeMet), selenoethionine (SeEt), and selenocystine (SeCys); selenomethionine was demonstrated to be the most abundant of these seleno-amino acids. Further characterization of unidentified selenium-containing peaks is attempted by the employment of several procedures, including electrospray-mass spectrometry (ES-MS). A peptide structure was identified; however, this was considered a tentative proposal due to the large background produced by the extremely complicated brazil nut matrix. 相似文献

Preparation of soybean oil-based greases: effect of composition and structure on physical properties
Vegetable oils have significant potential as a base fluid and a substitute for mineral oil in grease formulation. Preparation of soybean oil-based lithium greases using a variety of fatty acids in the soap structure is discussed in this paper. Soy greases with lithium-fatty acid soap having C12-C18 chain lengths and different metal to fatty acid ratios were synthesized. Grease hardness was determined using a standard test method, and their oxidative stabilities were measured using pressurized differential scanning calorimetry. Results indicate that lithium soap composition, fatty acid types, and base oil content significantly affect grease hardness and oxidative stability. Lithium soaps prepared with short-chain fatty acids resulted in softer grease. Oxidative stability and other performance properties will deteriorate if oil is released from the grease matrix due to overloading of soap with base oil. Performance characteristics are largely dependent on the hardness and oxidative stability of grease used as industrial and automotive lubricant. Therefore, this paper discusses the preparation methods, optimization of soap components, and antioxidant additive for making soy-based grease. 相似文献
