The vitellogenesis of Paraechinophallus japonicus (Yamaguti, 1934), the first pseudophyllidean tapeworm of the family Echinophallidae studied using transmission electron microscope, is described on the basis of ultrastructural observations of specimens from the benthopelagic fish Psenopsis anomala (Temminck et Schlegel, 1844) (Perciformes: Centrolophidae). The process of vitellogenesis in P. japonicus follows the same general pattern observed in other tapeworms. Five stages of vitellocyte development have been distinguished. The first stage corresponds to immature cells containing ribosomes and mitochondria. The second stage of development is characterized by the appearance of granular endoplasmic reticulum and Golgi complexes, formation of shell globules and lipid droplets at the periphery of the cell cytoplasm. Vitellocyte of the third stage presents accumulation of shell globules and lipid droplets. During the fourth stage, shell globule clusters are formed, and lipid droplets and rosettes of alpha-glycogen are accumulated. Mature vitelline cells are characterized by a great number of lipid droplets with glycogen in the centre of the cytoplasm, whereas shell globule clusters are situated more peripherally. The interstitial tissue of vitelline follicles of P. japonicus is syncytial with long cytoplasmic projections extending between vitelline cells. The presence of a large amount of lipid droplets in the vitelline cytoplasm within the eggs of P. japonicus may be related to egg accumulation in the uterine sac.  相似文献

Potassium channels are K+-selective protein pores in cell membrane. The selectivity filter is the functional unit that allows K+ channels to distinguish potassium (K+) and sodium (Na+) ions. The filter's structure depends on whether K+ or Na+ ions are bound inside it. We synthesized a K+ channel containing the d-enantiomer of alanine in place of a conserved glycine and found by x-ray crystallography that its filter maintains the K+ (conductive) structure in the presence of Na+ and very low concentrations of K+. This channel conducts Na+ in the absence of K+ but not in the presence of K+. These findings demonstrate that the ability of the channel to adapt its structure differently to K+ and Na+ is a fundamental aspect of ion selectivity, as is the ability of multiple K+ ions to compete effectively with Na+ for the conductive filter.  相似文献

Two polymorphisms of the porcine prolactin receptor (PRLR) gene were previously related to litter size by several authors; however, the magnitude and direction of such effects varied depending on the population analyzed. We have sequenced the complete coding region of the porcine PRLR gene and found 6 nonconservative SNP: C1217T (Leu/Pro406), C1283A (Asp/Ala428), G1439A (Lys/Arg480), T1528A (Met/Leu510), G1600A (Gly/Ser534), and G1789A (Gly/Ser597), within exon 10 of the gene, which encodes the entire intracytoplasmic domain of the protein. Eight haplotypes were found and were segregating at different frequencies in 6 porcine breeds. The effects of each individual SNP and haplotype were evaluated in an Iberian x Meishan F2 population using a univariate mixed-inheritance animal model. Significant effects on the number of corpora lutea were found for PRLR haplotypes (P < 0.012), confirming the previously reported associations of PRLR in this process and highlighting the importance of performing analysis of haplotypes rather than of individual SNP. Suggestive effects or tendencies were found for heart rate at birth (P < 0.10), rectal temperature (P < 0.05), and oxygen saturation (P < 0.10) 1 h after birth, and time to first suckle (P < 0.10). We found greater than expected levels of amino acid variability within the intracellular domain of the porcine PRLR, which have been associated with differences in the number of corpus lutea of sows and the preweaning survivability of piglets.  相似文献

Incubation of eggs by birds and lactation in mammals are regulated by pituitary prolactin (PRL) and associated with an increase in pituitary PRL-producing cells or lactotrophs. However, the mechanisms controlling this increase in lactotroph numbers are not known. PRL secretion in birds is regulated by vasoactive intestinal polypeptide (VIP). This study was designed to determine whether VIP treatment could modulate lactotroph abundance in culture. Anterior pituitary cells were isolated from laying Japanese White Silkie hens and cultured for 2 or 6 days in the absence or presence of VIP. PRL-secreting cells were identified by reverse hemolytic plaque assay. Treatment with VIP for 6 days substantially increased the abundance of PRL-secreting cells from 47.5% under basal conditions to 70.6% of all pituitary cells following VIP stimulation. However, 2-day VIP treatment had no effect. Furthermore, the extent to which the hens were allowed to accumulate eggs in a clutch prior to isolation of the pituitaries did not affect the lactotroph response to VIP in vitro. These findings indicate that chronic VIP stimulation may be responsible for the increased abundance of lactotrophs found in the pituitary glands of incubating hens.  相似文献

Mutations in the nuclear structural protein lamin A cause the premature aging syndrome Hutchinson-Gilford progeria (HGPS). Whether lamin A plays any role in normal aging is unknown. We show that the same molecular mechanism responsible for HGPS is active in healthy cells. Cell nuclei from old individuals acquire defects similar to those of HGPS patient cells, including changes in histone modifications and increased DNA damage. Age-related nuclear defects are caused by sporadic use, in healthy individuals, of the same cryptic splice site in lamin A whose constitutive activation causes HGPS. Inhibition of this splice site reverses the nuclear defects associated with aging. These observations implicate lamin A in physiological aging.  相似文献

Leaf meals are potential alternatives to soybean meal (SBM) in fish diets in developing countries because they are cheaper. Moringa oleifera (MOR) and Leucaena leucocephala (LEU) reduced nutrient digestibility of diets compared with SBM in an earlier study. However, fish raised outdoors consume natural foods, which might offset the negative effects of leaf‐meal diets. We conducted a feeding trial using mixed‐sex Nile tilapia (5.2 g) to assess performance of fish fed 36% protein diets with different concentrations of MOR and LEU leaf meals in place of SBM. Fish in static pools were fed daily to apparent satiation for 59 d. Individual weight gain (30.4–34.7 g), survival (91.8–97.3%), feed conversion ratio (FCR, 1.6–1.9), proximate composition, plasma lysozyme activity, and intestinal proteolytic enzyme activity were similar among diets. However, fish fed diets containing leaf meals had higher concentrations of n‐3 fatty acids than those fed the SBM control. Despite lower nutrient availability of the leaf‐meal diets compared with the soy diet, MOR or LEU could replace up to 30% of the SBM protein without reducing fish performance. Inclusion of poultry meal in the diets and probable nutrient supplementation from natural foods appeared to compensate for the lower nutritional value of the leaf meals compared with SBM.  相似文献

To determine the potential for improving the conversion and deposition of the omega‐3 fatty acids docosahexaenoic acid (DHA; 22:6n‐3) and eicosapentaenoic acid (EPA; 20:5n‐3) in fish, 44 families of rainbow trout were fed a diet low in these components and then evaluated for their ability to convert and store plant oils that did not contain DHA and EPA in their muscle tissue. The range for EPA and DHA detected in the tissues of different families varied between 1.2 and 2.9%, and 3.8 and 7.1% total fatty acid, respectively. After adjusting for covariates, an averaged heritability of 0.78 ± 0.11 for DHA and 0.61 ± 0.17 for EPA was calculated. Expression analysis of genes related to the elongation and conversion of lipids were also analyzed, and significant differences were found in the expression of some genes between groups of families that were delineated as having relative high, medium, and low capabilities of depositing EPA and DHA in their muscle tissue after being reared on the mainly soy/flax oil containing diet. However, none of these genes showed a positive correlation with the high conversion/deposition group .  相似文献
