The green water technique has been widely shown to improve fish larvae growth, survival and feed ingestion. Therefore, fish larvae (Sparus aurata L. and Solea senegalensis Kaup) feeding behaviour was studied through gut content analysis, when using different species of microalgae, as the ‘green water’ technique. Six treatments were used: Stain – food green stain; Tetra – microalgae Tetraselmis chuii; Iso – microalgae Isochrysis galbana; Tetra Sup –T. chuii supernatant (obtained from centrifugation); Phyto – a microalgae paste, Nannochloropsis oculata, (Phytobloom®); and C water – clear water, as control. At 9, 16 and 23 days after hatching (DAH) for S. aurata, and 4, 9 and 14 DAH for S. senegalensis, 40 unfed fish larvae were transferred to 3 L experimental tanks, filled with the different ‘green water’ technique. Fish larvae were sampled 2 h after being fed with live prey, anaesthetized and fixed in buffered formaldehyde for posterior gut content determination. Feeding was evaluated by the feeding rate, percentage of larvae with prey items in the digestive tract and feeding intensity, number of prey in each larva digestive tract. Fish larvae feeding ability was influenced by the interaction between light conditions and substances provided by the presence of microalgae during fish larvae development. Sparus aurata was more dependent on microalgae addition than S. senegalensis larvae, which may be related to the type of prey, larval behaviour, ontogeny and physiology. The presence of microalgae influenced the selection of larger prey (Artemia over rotifers) by S. aurata aged 23 DAH.  相似文献

Objective – To compare the histopathologic diagnosis in dogs with spontaneous hemoperitoneum when abdominal ultrasonographic examination detects a solitary versus multiple lesions.
Design – Retrospective cross-sectional study.
Setting – Private veterinary hospital.
Animals – Client-owned dogs presented with spontaneous hemoperitoneum between March 1, 2003 and June 1, 2008.
Interventions – Dogs were divided into 2 groups based on presence of a solitary or multiple abdominal ultrasonographic lesions. Prevalences were compared between groups for malignancy and specifically hemangiosarcoma.
Measurements and Main Results – Ten of 31 (32%) dogs had a solitary abdominal ultrasonographic lesion and 21 of 31 (68%) had more than 1 lesion. The bleeding tissue was characterized as malignant in 8 of 10 (80%) dogs with solitary lesions and 17 of 21 (81%) dogs with multiple lesions; there was no significant difference ( P =1.0) between groups. In this study no association ( P =0.26) was found between the number of abdominal ultrasonographic lesions observed and subsequent diagnosis of hemangiosarcoma.
Conclusions – Solitary abdominal ultrasonographic lesions in dogs with spontaneous hemoperitoneum do not necessarily indicate a lower prevalence of malignancy.  相似文献

The prevalence and contamination levels of Listeria monocytogenes were investigated in live bivalve molluscs for human consumption, collected in various steps of the commercial and sanitary circuits in the North of Portugal. Samples of different species were collected per lot before and after depuration treatment in two depuration units and further, when placed in retail markets. Listeria monocytogenes was isolated from 4% of the samples although with very low contamination levels (less than 100 MPN g?1). None of the environmental (non‐depurated) samples were contaminated with the food‐borne pathogen. The positive samples involved depurated and commercialized samples from two lots, one in each circuit. Two serotypes (1/2a and 3b) were detected among the isolated strains. This study demonstrated the possibility of occurrence of L. monocytogenes contamination of live bivalve molluscs along the sanitary control circuit, including at the retail market level. As L. monocytogenes grows well at refrigerated temperatures and in high salt matrixes such as seafood its presence in these products should not be overlooked.  相似文献

Juniperus thurifera L. is an endemic conifer of the western Mediterranean Basin where it is subjected to a severe climatic stress characterized by low winter temperatures and summer drought. Given the trend of increased warming-induced drought stress in this area and the climatic sensitivity of this species, we expect a negative impact of climate change on growth and ecophysiological performance of J. thurifera in the harsh environments where it dominates. To evaluate this, we measured long- and short-term radial growth using dendrochronology, photosynthesis and water-use efficiency in males, females and juveniles in three sites in Central Spain. Climate was monitored and completed with historical records. Mean annual temperature has increased +0.2?°C per decade in the study area, and the main warming trends corresponded to spring (+0.2?°C per decade) and summer (+0.3?°C per decade). Radial growth and maximum photosynthesis peaked in spring and autumn. Positive photosynthetic rates were maintained all year long, albeit at reduced rates in winter and summer. Radial growth was enhanced by wet conditions in the previous autumn and by warm springs and high precipitation in summer of the year of tree-ring formation. Cloud cover during the summer increased growth, while cloudy winters led to impaired carbon gain and reduced growth in the long term. We argue that maintenance of carbon gain under harsh conditions (low winter temperatures and dry summer months) and plastic xylogenesis underlie J. thurifera's ability to profit from changing climatic conditions such as earlier spring onset and erratic summer rainfall. Our results highlight that not only the magnitude but also the sign of the impact of climate change on growth and persistence of Mediterranean trees is species specific.  相似文献

Context

Forecasting the expansion of forest into Alaska tundra is critical to predicting regional ecosystem services, including climate feedbacks such as carbon storage. Controls over seedling establishment govern forest development and migration potential. Ectomycorrhizal fungi (EMF), obligate symbionts of all Alaskan tree species, are particularly important to seedling establishment, yet their significance to landscape vegetation change is largely unknown.

Objective

We used ALFRESCO, a landscape model of wildfire and vegetation dynamics, to explore whether EMF inoculum potential influences patterns of tundra afforestation and associated flammability.

Methods

Using two downscaled CMIP3 general circulation models (ECHAM5 and CCCMA) and a mid-range emissions scenario (A1B) at a 1 km2 resolution, we compared simulated tundra afforestation rates and flammability from four parameterizations of EMF effects on seedling establishment and growth from 2000 to 2100.

Results

Modeling predicted an 8.8–18.2 % increase in forest cover from 2000 to 2100. Simulations that explicitly represented landscape variability in EMF inoculum potential showed a reduced percent change afforestation of up to a 2.8 % due to low inoculum potential limiting seedling growth. This reduction limited fuel availability and thus, cumulative area burned. Regardless of inclusion of EMF effects in simulations, landscape flammability was lower for simulations driven by the wetter and cooler CCCMA model than the warmer and drier ECHAM5 model, while tundra afforestation was greater.

Conclusions

Results suggest abiotic factors are the primary driver of tree migration. Simulations including EMF effects, a biotic factor, yielded more conservative estimates of land cover change across Alaska that better-matched empirical estimates from the previous century.

A quantitative real-time polymerase chain reaction method (qPCR) was developed and tested for the detection of Taylorella equigenitalis. It was shown to have an analytical sensitivity of 5 colony-forming units (CFU) of T. equigenitalis when applied to the testing of culture swabs that mimicked field samples, and a high analytical specificity in not reacting to 8 other commensal bacterial species associated with horses. As designed, it could also differentiate specifically between T. equigenitalis and T. asinigenitalis. The qPCR was compared to standard culture in a study that included 45 swab samples from 6 horses (1 stallion, 5 mares) naturally infected with T. equigenitalis in Canada, 39 swab samples from 5 naturally infected stallions in Germany, and 311 swab samples from 87 culture negative horses in Canada. When the comparison was conducted on an individual sample swab basis, the qPCR had a statistical sensitivity and specificity of 100% and 96.4%, respectively, and 100% and 99.1% when the comparison was conducted on a sample set basis. A comparison was also made on 203 sample swabs from the 5 German stallions taken over a span of 4 to 9 mo following antibiotic treatment. The qPCR was found to be highly sensitive and at least as good as culture in detecting the presence of T. equigenitalis in post-treatment samples. The work demonstrates that the qPCR assay described here can potentially be used to detect the presence of T. equigenitalis directly from submitted sample swabs taken from infected horses and also for determining T. equigenitalis freedom following treatment.  相似文献

An indirect enzyme-linked immunosorbent assay (ELISA) has been developed for the species identification of grouper (Epinephelus guaza), wreck fish (Polyprion americanus), and Nile perch (Lates niloticus) fillets. The assay was performed in two different formats, microtiter plates and immunostick tubes, and uses polyclonal antibodies raised in rabbits against muscle-soluble proteins of grouper (anti-GSP), wreck fish (anti-WSP), and Nile perch (anti-PSP). The antibodies were made species-specific by blocking them with the heterologous soluble muscle proteins. Immunorecognition of polyclonal antibodies adsorbed to their specific fish samples was made with swine antirabbit immunoglobulins conjugated to the enzyme horseradish peroxidase. Subsequent enzymatic conversion of the substrate allowed unequivocal identification of the species studied.  相似文献

It is surmised that domestication has reduced geneticvariability in the common bean (Phaseolusvulgaris L.). However, little is known about leaf anatomy responses to water stress in wild or domesticated forms ofP. vulgaris. In this study,anatomical variation was evaluated in watered and water stressed plants of wild and domesticated genotypes of P.vulgaris from Mexico, growing under greenhouse conditions. Five terminal leaflets of completely expanded leaves wereused to evaluate leaf area (LA) and theirmiddle-regions to evaluate leaf thickness (LT), and various anatomical features. Domesticated genotypes showed bigger LAthan that of the wild ones; there was an effect due to the origin of materials, LA being bigger in genotypes from warm and humid lands that those from dry and temperate ones. No differences were detected in LT among materials; however, in domesticated plants the palisadeparenchyma predominated over the spongy one, contrasting with thewild genotypes. Under drought conditions, stomatal abortion was observed in wild and domesticated genotypes, especially on theabaxial surface. We conclude that, in fact, there are contrasting differences in leaf anatomy among wild and domesticated forms of Mexican Phaseolus vulgaris. Results suggest that domestication not only favours the enlargement of the LA, but also an increase in palisade parenchyma, suggesting a divergence toward assimilation in domesticated forms, in contrast with a better internal gas diffusion in wild forms. Domestication seems to have accentuated stomatal abortion as means of a transpirational control mechanism to counteract the effects of drought.  相似文献

Three commonly used flavor industry solvents (propylene glycol, triacetin, and triethyl citrate) were tested for their capacity to interfere with the ability of alpha-, beta-, and gamma-cyclodextrin to form molecular inclusion complexes with flavors. Six flavor compounds (ethyl butyrate, ethyl heptanoate, l-menthol, methyl anthranilate, neral, and geranial) were measured by headspace gas chromatography above 2:1 water/ethanol containing appropriate additions of cyclodextrin and flavor solvent. The smallest and most polar solvent molecule represented by propylene glycol had the least effect on cyclodextrin/flavorant complex formation. In contrast, triacetin, intermediate in size among the three flavor diluents studied, had the greatest effect, even though, based on at least some computed molecular parameters, it appears to be more polar than triethyl citrate. The explanation for this apparent anomaly may lie in differences in the extent to which triacetin and triethyl citrate are able to interact with cyclodextrins by means of partial interaction with the hydrophobic cavities of the latter.  相似文献
