ABSTRACT: Tyrosinase inhibitor-producing fungus of Trichoderma sp. strain H1-7, which was isolated from marine sediment, was investigated by phylogenetic analysis and physiological characteristics. Phylogenetic analyses of the strain were similar to Trichoderma atroviride or T. viride . Physiological characteristics of the strain were similar to T. viride , and based on these results, it was identified as T. viride . Characterization of tyrosinase inhibitory activity of a culture supernatant of the strain was investigated. The inhibitory activity of the supernatant of the strain decreased after cultivation for more than 3 days. Furthermore, sea water was not essential for the production of the tyrosinase inhibitor (TI). When TI production of the strain was compared to T. viride , the strain showed higher activity than T. viride . From this result, it seemed that the strain had characteristic features comparable to T. viride , which was isolated from the terrestrial environment. As TI production of the strain showed higher potential than that of T. viride , it is necessary to elucidate the chemical structure of TI exemplified in the present study. 相似文献

The acute toxicities of copper were examined for Japanese flounder, Paralichthys olivaceus, and red sea bream, Pagrus major, in terms of fish size and water temperature. Artificial seawater of low pH of 5.4–6.7 was used as testing water to keep dissolved copper concentration at 0.04–41 mg Cu/L. Japanese flounder of 0.3–17 g and red sea bream of 0.5–13 g were exposed to different concentrations of copper for 96 h at 20 C under semistatic condition. Median‐lethal concentration for 96 h of Japanese flounder and red sea bream were 8.7–12.2 and 2.0–5.2 mg Cu/L, respectively. No significant relationships were observed between median‐lethal concentrations for 96 h and fish size for Japanese flounder, while the value decreased significantly with increasing fish size for red sea bream. Effect of water temperature on the acute toxicity was examined for Japanese flounder of 0.3 and 0.4 g at 10, 15, 20, and 25 C and red sea bream of 0.5 and 1.0 g at 12, 15, 20, and 25 C. Ninety‐six‐hour median‐lethal concentrations for Japanese flounder and red sea bream were 5.1–11.2 and 1.0–5.3 mg Cu/L, respectively. No significant relationships were observed between median‐lethal concentrations for 96 h and water temperature for both fish species. 相似文献

A feeding experiment was conducted to examine the effect of a supplemental ethanol extract from a defatted soybean meal on the biliary bile status and intestinal conditions of rainbow trout Oncorhynchus mykiss . A semi-purified control diet based on casein (Cont) and three diets supplemented with the extract (ES), bovine bile salts (BS) and their combination (ESBS) were fed to trout (10 g) for 6 weeks. The growth, feed efficiency ratio, fat digestibility and gallbladder- and hepato-somatic indices of fish fed diet ES decreased. Compared with this group, these parameters were improved in fish fed diet ESBS. The total biliary bile salt content was the lowest in fish fed diet ES and this group had a high proportion of chenodeoxycholyltaurine. The inclusion of the bile salts to diet ES (diet ESBS) increased the total biliary bile salt content. A similar trend was observed in the bile salt concentration of intestinal digesta. Although morphological changes occurred in epithelial cells of the distal intestine of fish fed diet ES, the histological features of fish fed diet ESBS were similar to those of fish fed diet Cont. These results indicate that alcohol-soluble substances of soybean meal cause physiological changes in the biliary bile and intestine of rainbow trout. 相似文献

Unbleached and oxygen-bleached hardwood kraft pulp (UKP and OKP), respectively, were bleached with a culture filtrate containing manganese peroxidase (MnP) and lignin peroxidase (LiP) fromPhanerochaete chrysosporium Burds. The brightness increases of UKP upon biobleaching with the culture filtrate with and without MnSO4 were the same. The brightness increase of OKP with MnSO4 decreased to about half that seen without MnSO4. Changes in the brightness of UKP and OKP by treatment with the culture filtrate were determined. The brightness increased sharply by about eight points during the first 3h. The 3-h treatment was repeated seven times. The brightness increased linearly with the bleaching of UKP. Morphological changes of ramie fibers due to swelling in alkali solution were investigated. The observed twisting and changes in the cross-sectional shape of the fibers could be explained as a geometrical alteration caused by lateral expansion of coiling fibrils, as the unmercerized ramie fiber showed an S-helix structure. The fibril angle of ramie fibers was estimated to be 3°. Although the swollen fiber untwisted to some extent on washing with water, the shape of the twisted ribbons as observed by scanning electron microscopy was retained even after drying. Concentrated 8N NaOH produced only a moderate change in morphology, whereas swelling with 3.5N NaOH at 0°C expanded the cross sections about 30 times over the initial ones accompanied by a drastic change in the shape of the fiber.Part of this paper was presented at the annual meeting of the Wood Research Society of Japan in Kumamoto, April 1996 相似文献

Chinese raw tall oil samples, which were obtained from pulp and papermaking factories in Qingzhou and Jiamusi, were first divided into acidic and neutral parts with a diethylaminoethyl-Sephadex (DEAE-Sephadex) gel column. The acidic components were then analyzed qualitatively and quantitatively by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). The results obtained are as follows: Most of the compounds that comprised 0.1% or more of the components were identified. Secodehydroabietic, 8,15-pimaradienoic, 8,15-isopimaradienoic, 7,13,15-abietatrien-18-oic acid, and others were found for the first time in Chinese tall oil; they have not been found in Chinese fatty rosin. The mode of action of soluble silicon against strawberry powdery mildew (Sphaerotheca aphanis var. aphanis) was investigated in four experiments. First, silicon-treated leaves from plants grown with silicate (Si+) and control leaves
were excised, inoculated with conidia, and subsequent germination and formation of appressoria in a petri dish was assessed
after 24 h. The germination rate was 49.7% on Si+ leaves, and was 67.2% on control leaves (t-test, P < 0.01). Second, we soaked cellulose membranes in various solvents and then placed the membranes on 4% water agar, dusted
the membranes with conidia, and examined after 12 h. No difference was apparent between any treatment and the control (distilled
water). Third, strawberries growing hydroponically with additional silicon in the medium were inoculated with conidia, and
leaves were observed with a scanning electron microscope 1–2 days after inoculation. Germ tubes and secondary hyphae were
shorter and had fewer branches on Si+ leaves than on the control. Moreover, penetration appeared to be inhibited. Fourth,
the cuticle was separated from leaves from plants grown as in the third experiment, placed on water agar, and dusted with
conidia. Germination of conidia, observed with a light microscope, on Si+ leaves was suppressed markedly to 40%–60% of that
of the control. These results suggested that soluble silicon induced physiological changes in the cuticle layer after absorption
by the plant. In addition, soluble silicate reduced germination of conidia, formation of appressoria, and possibly the penetration

Isolates of an unidentified Rhizoctonia sp. (UR isolates) were obtained from creeping bentgrass and Kentucky bluegrass with reddish brown sheath and foliar rots.
Because the UR isolates anastomosed with isolates of three varieties of Waitea circinata (var. oryzae, var. zeae, and var. circinata), colony morphology, hyphal growth rate at different temperatures, pathogenicity, sequence analysis of the internal transcribed
spacers (ITS) region of ribosomal RNA genes (rDNA) were compared. The colony color of mature UR isolates was distinct from
isolates of the other three varieties of W. circinata. In pathogenicity tests on creeping bentgrass, the severity of the disease caused by UR isolates was significantly higher
than that caused by the three varieties of W. circinata. Sequence similarities of the rDNA-ITS region between UR isolates and between isolates within each variety were high (97–100%),
but they were lower among isolates from UR and the varieties of W. circinata (88–94%). In a phylogenetic tree based on the rDNA-ITS sequences, UR isolates formed a cluster separate from each of the
clusters formed by the three varieties of W. circinata. These results indicate that the UR isolates clearly differ from the three varieties of W. circinata. We therefore propose that the UR isolates be classified as new Rhizoctonia sp. that are closely related to W.
circinata and that the disease on creeping bentgrass should be called Waitea reddish-brown patch disease (Sekikasshoku-hagusare-byo

Certain cellular responses to stresses and stimuli are regulated by a G-protein-mediated signaling pathway. A rice dwarf
mutant that is defective in the α subunit of the heterotrimeric G-protein was found to be fully protected from blast fungus
by the plant activator probenazole (PBZ) despite the 1-day delay in induction of the PR-10 gene PBZ1 by PBZ. These results suggest that the signaling pathway for protection by PBZ is not via the G-protein, although G-protein
is involved in the induction of PBZ1 by PBZ.
