Summary Seedling mortality caused by excessive salinity is common in establishing furrow-irrigated crops. This study was conducted to evaluate the processes involved and salinity levels leading to seedling mortality in guayule (Parthenium argentatum Gray cv. 593), carrot (Daucus carota L. cv. Imperator-58), chile pepper (Capsicum annuum L. cv. New Mex. 6–4), and tomato (Lycopersicon esculentum Mills cv. Rutgers). Salt accumulation patterns were also evaluated in soil columns subirrigated with waters of 0.8 and 3.9 dS m–1. Seedlings were first grown for 10 to 16 days in greenhouse pots with water of 0.8 dS m–1. Upon emergence of the first true leaf, seedling roots, leaves and stems were independently exposed to different levels of salinity (0.8 to 59 dS m–1) under two diurnal temperature regimes (22–32°C and 24–40°C). When seedling roots were exposed to the saline solutions, mortality was sub stantially greater under the high temperature, and increased greatly at salinity levels of soil solutions exceeding about 5 dS m–1 in guayule and carrot, and 15 dS m–1 in tomato and pepper. Mortality caused by leaf exposures to saline spray was greater under the low temperature with higher relative air humidities, and increased greatly when salinity levels of spray solutions exceeded ap 5, 10, 15 and 20 dS m–1 in guayule, carrot, tomato and pepper, respectively. Physical abrasion of seedling leaves prior to saline water spraying significantly increased mortality. Stem exposure to a thin layer of salted sand having the saturation extract salinity of up to 58 dS m–1 caused no significant increase in mortality. Soluble salts were accumulated mostly in a soil depth of 0 to 0.5 cm at a rate of 35 dS m–1 in 3 weeks when subirrigated with water of 3.9 dS m–1. Under furrow-irrigated conditions, seedling mortality may be induced mainly through leaf and/or root, but not stem, exposure to the salts accumulated at soil surfaces. Leaf-induced mortality can be the most significant process when wind-damaged seedlings are exposed to saline splatters during light showers common to the semi-arid region.Contribution from Texas Agr. Myofibrillar proteins of vertebrate skeletal muscles are insoluble in solutions of ionic strength that approximate physiological conditions. We established a method to solubilize more than 80% of chicken breast muscle myofibrillar proteins in water for the use of meat as a source of food protein. SDS‐polyacrylamide gel electrophoretic patterns of water‐soluble myofibrillar proteins demonstrated that all identified myofibrillar proteins except connectin/titin were soluble in water. A part of α‐actinin was released from myofibrils by repeated washing with 2.5 mmol/L NaCl and 5 mmol/L L‐histidine solution, and subsequent destruction of connectin/titin in washed myofibrils by ultrasonication resulted in solubilization of a large fraction of chicken breast muscle myofibrillar proteins in water. Myofibrillar proteins of chicken leg, pork loin, beef shoulder loin, and lamb were also solubilized in water using this procedure.  相似文献

To clarify the immunologic effects of vaccination in dogs, we monitored total leukocyte and lymphocyte counts, humoral antibody responses, blastogenic responses of lymphocyte, and delayed type hypersensitivity (DTH) responses after vaccination. Mixed vaccines were administered on day 0 except for canine parvovirus (CPV) vaccine which was readministered on day 21. The puppy and adult dogs had a significant decrease in leukocyte and lymphocyte counts on day 7. The puppies showed a significant increase in the blastogenesis of lymphocytes after each vaccination, whereas the adult dogs had no significant changes. However, the adult dogs were divided into two groups, high responders and low responders in blastogenesis of lymphocytes. The dogs with higher or lower response in SI values on day 0 tended to show decrease or increase after the first vaccination, respectively. Since almost all dogs developed high titers of humoral antibody, it is considered that vaccination acts in an immunomodulative fashion. DTH responses to phytohemagglutinin (PHA) and CPV vaccine monitored at 0, 3, and 8 weeks after the first vaccination produced strong reactions, in particular those to CPV vaccine rose significantly after vaccination and maintained the higher responses for at least 2 months. These results suggest that DTH responses to PHA and CPV vaccine are helpful to monitoring non-specific and specific immune functions in vivo, therefore, DTH could be used as simple and rapid immunologic tests in canine practice.  相似文献

Summary Because of the strategic and industrial importance of natural rubber, there has been renewed interest in cultivating guayule (Parthenium argentatum). This study was performed for assessing feasibility of guayule cultivation with waters high in dissolved salts. The test materials included six USDA selections (11591, 11605, 11619, 11646, 12229 and N576), one cultivar (593) and one hybrid (4265XF). Seedlings were grown for l0 weeks in a greenhouse and transplanted in the spring and in the summer into lysimeters (unit surface dimension of 6 x 7 m) containing loamy sand or silt loam. They were grown with simulated irrigation waters having four levels of salinity (0.8, 2.4, 4.6 and 7.2 dSm–1 with SAR < 13) and an additional water containing 5 mmol L–1 of Mg at 2.4 dSm–1. In the spring planting, over 90% of the transplants survived when furrow irrigated weekly with waters of 4.6 dSm–1 or less. However, transplant growth for the first several months was reduced by half at irrigation water salinity of 4.6 dSm–1. In the summer planting, several fold increases in mortality and growth reduction occurred. Dry top Shrub yields after the two growing seasons following the spring planting averaged 10 Mg ha–1 at 0.8 dSm–1 and declined on the average 15 and 51 % when irrigated with waters of 4.6 and 7.2 dSm–1 respectively. The amount of water used to produce one ton of dry shrub was 1,600 m3 with waters of 0.8 and 4.6 dSm–1, and 1,900 m3 with water of 7.2 dSm–1. The contents of rubber in the shrubs averaged 61 g kg–1 at 0.8 dSm–1 and increased to 70 g kg–1 at 7.2 dSm–1. whereas resin contents were not affected by the salt treatments. Resulting rubber yields were reduced on the average by 8.5 and 44% at 4.6 and 7.2 dSm–1. respectively, because of the reduction in shrub yields. Selection N576 produced the largest rubber yields with the highest rubber content at all levels of salinity. Increasing Mg concentrations from 0.5 to 5 mmol L–1 in the irrigation waters reduced neither yields nor transplant survival. Two experiments were conducted to compare the effect of estrus induction by controlled internal drug release (CIDR) and intravaginal cream containing 500 mg progesterone (P cream) in ewes during the non-breeding season. In the first experiment, twenty-four ewes were randomly grouped for two treatments with the different intravaginal devices for 12 days: Group A was the CIDR group and Group B was the P cream group. Blood was collected from all treated ewes, and progesterone (P(4)), estradiol 17-beta (E(2)) and luteinizing hormone (LH) concentrations were measured by enzyme immunoassay. In the second experiment, the conception rates from natural mating, estrus-detected AI (inseminated 12 h after estrus detection), or fixed-time AI (inseminated 42 h after removal of an intravaginal device) in 127 ewes treated with CIDR or P cream were compared. In Experiment 1, the rate of estrus induction and the time of estrus onset after device removal were 91.7% and 36.3 +/- 15.7 h in Group A, and 100% and 35.0 +/- 12.6 h in Group B, respectively. There were no significant differences between the devices. The mean plasma P(4) concentration in Group B was significantly (P < 0.01) lower than Group A between day -9 and day -1 (Day 0: the day of device removal). However, no significant differences were found in the mean E(2) concentrations of the two groups after treatment. The mean time of estrus onset in ewes with an observed LH surge and the time of LH surge after treatment were 23.3 +/- 8.7 h and 30.3 +/- 5.0 h for Group A and 27.6 +/- 6.5 and 26.3 +/- 8.0 h for Group B, respectively, and there were no significant differences. However, a significant difference (P < 0.05) was found in the mean time from the time of estrus onset to LH surge between Group A (6.4 +/- 6.7 h) and Group B (-1.3 +/- 4.1 h). In Experiment 2, the conception rates for natural mating, estrus-detected AI, and fixed-time AI were 55.0, 29.4, and 25.0% for Group A and 40.7, 25.0, and 42.1% for Group B, respectively, and there were no significant differences. These results suggest that the effect of induction of estrus and ovulation and the rate of conception after treatment were comparable to CIDR even though the plasma P(4) concentration of the P cream method tended to be low during the insertion period.  相似文献

N-acetylglucosaminyltransferase I (GnT I; EC 2.4.1.101), which catalyzes the first step in the conversion of oligomannose to complex or hybrid N-glycans of glycoproteins, was found in media cultured with bovine oviduct epithelial cells (BOEC) obtained from non-pregnant cows during the follicular phase. Combined treatment with specific hormones increased GnT I release from BOEC. Luteinizing hormone (LH; 10 ng/ml) alone slightly, but together with 17beta-estradiol (E2; 1 ng/ml), synergistically increased GnT I activity. Vascular endothelial growth factor (VEGF) and tumor necrosis factor (TNF) alpha, which have been shown to have their highest activities in the bovine oviduct during the periovulatory period, also increased in GnT I activity. This study provides the first evidence of an increase of GnT I release from BOEC in vitro, and shows that endocrine as well as local factors such as LH, VEGF and TNFalpha increase this activity. The results suggest that GnT I activity in the bovine oviduct may contribute to the induction of glycosylation and thereby contributing to the provision of the optimal microenvironment for fertilization and early development of the embryos.  相似文献

The present study was conducted to obtain new information on relationships among serum testosterone (T), estradiol-17 beta (E(2)), follicle-stimulating hormone (FSH), and luteinizing hormone (LH) concentrations and histology of seminiferous tubules in captured common minke and Bryde's whales during the feeding season. Blood samples and testes were collected from common minke (n=39 for blood samples, n=15 for testes) and Bryde's (n=14 for blood samples, n=7 for testes) whales captured from May 2001 to August 2001 in the Western North Pacific. Serum T concentrations, in 35.9% of the common minke and 57.1% of Bryde's whales, were below the detection limit (< 2.5 pg/ml). There were no significant differences in the serum concentrations of E(2), FSH, and LH among immature, mature common minke and Bryde's whales except that LH levels of immature Bryde's whales was higher than those of common minke whales. In most seminiferous tubules of mature whales, only a single-layer of spermatogonia was observed. However, spermatozoa were observed in seminiferous tubules in 2/13 of mature common minke and 4/4 of mature Bryde's whales with the low or undetectable T levels. These results indicate that the low serum T concentrations reflect the inactivity of spermatogenesis in both baleen whales, and that it is not possible to assess gonadal activity in either common minke or Bryde's whales using serum sex hormone concentrations during the feeding season.  相似文献

The aim of this study was to investigate the relationship between the first ovulation within 3 weeks postpartum and subsequent ovarian cycles and fertility in high producing dairy cattle in Hokkaido, Japan. In Experiment 1, 110 cows (44 primiparous and 66 multiparous) were used to determine the effects of the first ovulation within 3 weeks postpartum on subsequent ovarian cycles. Milk samples were collected twice weekly from 7 to 100 days postpartum. The first ovulation was identified by an increase in milk progesterone (P4) to more than 1 ng/ml within 3 weeks postpartum. The numbers of cows showing ovulation and anovulation within 3 weeks postpartum were 31 (70.5%) and 13 (29.5%) in the primiparous cows and 35 (53.0%) and 31 (47.0%) in the multiparous cows, respectively. The patterns of ovarian resumption after calving were classified into two types (normal ovarian cycles and abnormal ovarian cycles) on the basis of milk P4 concentrations. Initiation of normal ovarian function in cows ovulated within 3 weeks postpartum occurred earlier than in anovulated cows regardless of the number of calvings (primiparous, 27.8 days vs. 44.4 days; multiparous, 30.6 days vs. 55.7 days; P<0.01). Out of the multiparous cows that ovulated within 3 weeks postpartum, initiation of normal ovarian function followed by a normal luteal phase was earlier than when it was followed by an abnormal luteal phase (25.5 days vs. 40.4 days; P<0.05). Milk P4 concentrations after the first ovulation were lower than those after the second ovulation in both the primiparous and multiparous cows (P<0.05). In Experiment 2, 22 multiparous cows were used to determine the effects of the first ovulation within 3 weeks postpartum on subsequent fertility. Blood samples were collected once a week from 0 to 3 weeks postpartum. The interval from parturition to first service in ovulated cows was shorter than in anovulated cows (68.4 days vs. 94.8 days; P<0.05). The conception rate by 100 days after calving tended to be higher in ovulated cows than in anovulated cows (50.0% vs. 16.7%, P=0.09). In conclusion, our data strongly suggests that ovulation within 3 weeks postpartum is a crucial phenomenon for subsequent resumption of ovarian function and conception, and thus it can be used as an index of subsequent reproductive performance.  相似文献
