The study was conducted to clarify how early high plane of nutrition related to metabolic imprinting affected growth, carcass characteristics, and meat quality of grass-fed Wagyu (Japanese Black cattle). Wagyu steers were allocated randomly into 2 dietary groups: (1) steers fed milk replacer (crude protein 26.0%, crude fat 25.5%; maximum intake 0.6 kg/d) until 3 mo of age and then fed roughage (orchard grass hay) ad libitum from 4 to 10 mo of age (roughage group, RG; n = 11); (2) steers fed milk replacer (maximum intake of 1.8 kg/d) until 3 mo of age and then fed a high-concentrate diet from 4 to 10 mo of age (early high nutrition, EHN; n = 12). After 11 mo of age, all steers were fed roughage ad libitum until 31 mo of age and then slaughtered. Growth performance, carcass traits, longissimus muscle (LM) meat quality and intramuscular fat (IMF) content, plasma insulin-like growth factor I (IGF-I) concentration, and bone mineral density were measured. Body weight was greater in EHN steers (571 kg) than RG steers (520 kg; P < 0.01). Plasma IGF-I levels were higher in EHN steers than in RG steers at 3, 10, and 14 mo of age (P < 0.01, P < 0.005, P < 0.001, respectively); however, plasma IGF-I levels were lower in EHN steers compared with RG steers at 30 mo of age (P < 0.01). The total weight of the muscles and bones of the left half of the carcass was not different between the 2 groups (P = 0.065). Five of the 19 muscles investigated (semimembranosus, P = 0.036; infraspinatus, P = 0.024; supraspinatus, P = 0.0019; serratus ventralis cervicis, P = 0.032; serratus ventralis thoracis, P = 0.027) were heavier in EHN steers. Total fat weight in the left half of the carcass was 30% greater (P = 0.025) in HNE carcasses. Subcutaneous and perirenal fat weights were 53% and 84% greater (P = 0.008, P = 0.002, respectively) in EHN carcasses. The LM IMF content was greater in EHN loins (13.2%) compared with RG loins (9.4%) at 31 mo of age (P = 0.038); however, no differences were found for shear force, tenderness, and cook loss. ABSTRACT: To understand the mechanism of the behavioral response in the capture process of how fish recognize fishing gear and then how they can avoid the gear, the visual acuity of Pacific saury Cololabis saira was investigated by histological examination of the retina of individuals in the size range of 75–365 mm fork length (FL). The contour map of cone density distribution shows that the highest cone density is located in the temporal area of the retina, which indicated the visual axis as the forward direction. The visual acuity (VA) depends both on the focal length of the lens and the number of cones in the retina. The lens diameter increased linearly from 1.40 to 4.73 mm with fish growth, while the cone density decreased gradually from 765 to 378 cells/0.01 mm2 . Our results show that the visual acuity increases proportionately from 0.057 to 0.140 for individuals ranging in FL from 75 to 365 mm as expressed by the equation VA = 0.0065 × FL 0.5271 ( r 2 = 0.9624). 相似文献

Quercus serrata Thunb. ex Murray is a widespread deciduous oak in China, the Korean Peninsula, and Japan, and a strong isoprene emitter. Establishing accurate inventories of this species and estimating net carbon budgets, including biogenic volatile organic compounds (BVOC), necessitates detailed evaluation of BVOC emission and oxidation characteristics. Emissions of isoprene, the most abundant BVOC, presumably contribute to atmospheric chemistry through the formation of photochemical oxidants and secondary organic aerosols. We built an isoprene flux monitoring system to simultaneously reveal characteristics of the flux and fate of isoprene at multiple locations in Q. serrata forests. We used proton transfer reaction mass spectrometry (PTR-MS) and an automated closed chamber to measure isoprene emissions from soil and leaves in a warm-temperate Q. serrata forest. We used a relaxed eddy accumulation system with PTR-MS to simultaneously measure the canopy flux. In continuous foliage chamber measurements, we observed daily variations of isoprene emissions and continuous nocturnal emissions from leaves. Nocturnal emissions exceeded 25 % of total daily leaf emissions and were relatively high at sunset and low at sunrise. These results suggest that nocturnal emissions from mature trees may not be negligible. When leaf emissions were high in the daytime, the canopy isoprene flux tended to plateau at an upper limit. Observations of isoprene concentrations and gradients suggest that the plateau was caused by acceleration of isoprene oxidation, and sequential formation of secondary organic aerosols may occur near the leaf just after emission. Elucidation of these linkages may require continuous field measurements with a simultaneous multi-flux monitoring system. 相似文献

Differentiated cells were recognized in calli derived from needles of Torreya nucifera and in calli derived from immature zygotic embryos of Cryptomeria japonica. Some differentiated cells resembled tracheary elements of primary xylem with spiral or reticulate thickening of cell walls. Other cells resembled tracheary elements with thick cell walls and bordered pits, which are features of secondary xylem. These tracheary elements were formed in cell clusters. Tracheary elements in calli of T. nucifera formed more highly developed structures, such as bordered pits and spiral thickening, than those of C. japonica. Cultured cells derived from conifers might provide a good model for studies of the differentiation of secondary xylem in vitro. 相似文献

Tetraploid fish, which are considered as key resources of diploid gametes for further breeding and ploidy manipulation, can be artificially induced by inhibition of the mitotic cell division with hydrostatic pressure or temperature treatments. Although many attempts have been made to induce artificial tetraploid strains, successful establishment of viable and fertile tetraploid strains are rare. In pond loach, Misgurnus anguillicaudatus, natural tetraploid individuals are distributed in wild populations and diploid gametes from the tetraploid fish have been used for the induction of polyploid individuals, but artificially induced tetraploid strains have not been established yet. In the present study, we optimised starting timing of the heat-shock treatment (41 °C for 2 min) to inhibit a mitotic cell division in fertilised eggs of the normal diploid pond loach between 21 and 51 min after insemination at 20 °C. After the treatment, we observed external appearance of hatching larvae and flow cytometrically determined ploidy status of the resultant larvae. Although tetraploid and diploid/tetraploid mosaic larvae were obtained, the optimum timings for induction of tetraploidy varied amongst crosses. Various kinds of ploidy such as haploidy, diploidy, triploidy, pentaploidy, hexaploidy, aneuploidy and mosaic were detected in non-optimum heat-shock timings for tetraploidisation. Survivors, a tetraploid and a diploid/tetraploid mosaic male, matured at the age of 1-year-old, but they produced functional haploid spermatozoa. 相似文献

wooden houses and evaluated the accuracy thereof by carrying out numerical simulations by shaking table tests. The distinct
element method was adopted as the basic theory of our numerical analysis. This research is the first approach in which the
extended distinct element method was used for Japanese timber post-and-beam construction. The size of the analysis model is
a 5.5 m × 5.5 m, two-story real-size wooden house. The three analytical models were developed in terms of the strength of
exterior mortar walls. The simulation results were compared with the shaking table test results. One of the collapsing processes
of the numerical simulation corresponds well to the experiment results. Assessment of the possibility of collapse for real-size

We undertook a three‐dimensional reconstruction of intramuscular collagen networks of bovine muscle using an immunohistochemical/confocal laser‐scanning microscopic method. By immunohistochemical staining, type I and III collagens were observed mainly in the perimysium, while type IV collagen was observed in the endomysium. On the other hand, type V and VI collagens were observed in both the perimysium and endomysium. By confocal laser‐scanning microscopy, the collagen observed in the perimysium was three‐dimensionally reconstructed as plate‐shaped layers whereas the collagen observed in the endomysium surrounded myofibers. The three‐dimensionally reconstructed observations using immunohistochemical/confocal laser‐scanning microscopic method is useful for investigating collagen networks in muscle. 相似文献

The objective of this article is to compare feed cost, palatability and environmental impacts among feeding systems of high concentrate (HC), high hay (HH) and grass‐only‐fed (Gof) groups. Feed cost was the sum of costs paid for feed intake times the price of feed per kilogram. Palatability was measured by a panel taste test using HH and Gof beef and analyzed for differences. Environmental impacts were calculated based on 1 kg of Japanese beef yield of CO2 equivalents (eq) and animal end weights at each feeding stage. Results showed that the HH and Gof feeding systems could significantly reduce feed costs by approximately 60% and 78%, respectively, from the HC. In the panel taste test, 50% and 47.50% of panelists indicated that HH beef was ‘extremely delicious’ and ‘acceptable,’ respectively, while 15% indicated that Gof beef was ‘extremely delicious’; 62.50% indicated that Gof beef was ‘acceptable.’ Environmental impacts of each feeding system in terms of CO2 equivalents (eq) were 9.32, 6.10 and 2.04 tonnes of eq for the HC, HH and Gof, respectively. Swimming performance of jack mackerel Trachurus japonicus (18.2 ± 0.8 cm fork length (FL), n = 185) was examined in a flume tank by measuring the stride length at low and high tail beat frequencies with electromyogram monitoring and a muscle twitch experiment. Stride length was analyzed by monitoring the tail beat frequency according to the swimming speed at different temperatures of 10, 15 and 22 °C. In the electromyographic observations, the initiation of ordinary muscle activity occurred between 71.4 and 99.6 cm/s, that is 3.7 to 5.3 FL/s, when the tail beat frequency was over 6 Hz. The swimming speeds increased rectilinearly with the tail beat frequency at each water temperature both for the low and high tail beat frequency. Lower stride length was observed at the lowest temperature (10 °C) tested. The forced swimming exercise significantly affected the muscle contraction time to become longer than the control fish, which indicated a reduction of the maximum swimming speed performance. 相似文献
