为了对采后香蕉的成熟度进行无损检测,提高货架品质和减少过熟造成的损失,以巴西香蕉为研究对象,以1,2-二油锡甘油-3-磷-L-丝氨酸、半乳糖神经酰胺、纤维素乙酯、乙基纤维素做为敏感材料,采用滴涂法修饰了4个石英晶体微天平(quartz crystal microbalance,QCM),测试了1~7级成熟度的香蕉的挥发性有机化合物的响应特性,并用主成分分析法和线性判别分析法对响应结果做了分析。试验表明,在4种敏感膜中纤维素乙酯对香蕉的7个成熟阶段的挥发性气体的响应最明显,响应值先随香蕉成熟度增加而增加,在4级成熟阶段时最大,随后降低。用主成分分析法对7个成熟阶段香蕉的挥发物的响应值聚类分析,97%能够正确聚类;用线性判别分析法可获得100%的识别率。据此,4种材料修饰的石英晶体微天平传感器可以用于香蕉成熟度的识别。该研究结论对香蕉成熟度自动化分级研究有一定实用价值。 相似文献

The present study was undertaken to investigate the effect of dietary supplementation with nimesulide or eugenol on N-nitrosodiethylamine (DEN)-initiated early hepatocarcinogenesis in F344 male rats. Both compounds did not alter the expression of cytochrome P450 (CYP) 2E1, the enzyme that plays a major role in the activation of DEN to genotoxic products; however, nimesulide induced the expression of CYP1A1. Western blot analysis revealed that COX-1 and COX-2 protein expressions were not modulated by DEN compared with normal controls. Furthermore, post-initiation feeding with nimesulide or eugenol did not modulate COX-2 protein expression in normal or DEN-treated rats, whereas eugenol significantly increased the liver prostaglandin E(2) (PGE(2)) levels of DEN-injected animals compared with the DEN controls. Ultimately, nimesulide or eugenol did not modify DEN-induced hepatocarcinogenesis as evidenced by insignificant changes in the number and size of preneoplastic placental glutathione S-transferase (GST-P) positive liver foci compared with the DEN controls. These results suggest that COX-2, as well as prostaglandin E(2), may play no role in the post-initiation development of DEN-induced rat hepatocarcinogenesis at an early stage. 相似文献

We investigated the possible role of metal proteinase on the fruit-body formation ofHypsizygus marmoreus. The addition of a specific metal proteinase inhibitor, phosphoramidon, to the culture medium (10g/ml) completely inhibited fruit-body formation. Metal proteinase activity in both the medium and the mycelia of this fungus increased markedly during vegetative mycelial growth, and activity was maximal 25 days after inoculation. When phosphoramidon was added to the culture medium during vegetative mycelial growth, the metal proteinase activity in the mycelium decreased to 56% of the control (without inhibitor) level. Isoelectric focusing analysis showed that two kinds of metal proteinases with a pl of 7.7 and 8.4, respectively, were obtained from 29-day-old mycelia. Uptake of phosphoramidon into the mycelia was confirmed as the result of inhibition of thermolysin activity by the mycelial extracts. The degree of inhibitor uptake into mycelia was about 2.0% and was independent of the initial concentration of the inhibitor administered. The addition of peptone and amino acids to medium treated with phosphoramidon resulted in fruit-body dry weight yields that were about 50% that of the control. 相似文献

The effects of initial tree spacing on wood density at breast height were examined for 22-year-old Japanese larch (Larix kaempferi). The experiment involved the use of three plots with different initial tree spacing densities (300, 500, and 1000 trees/ha).
For five trees selected from each plot, the total tree height, diameter at breast height, height to the base of the live crown,
and crown diameter were measured. Ring width and wood density for individual growth rings were determined by X-ray densitometry.
A mixed-effects model was applied for fitting the radial variation in wood density in relation to initial spacing. Models
having various mean and covariance structures were tested in devising an appropriate wood density model. The model, consisting
of the mean structure with quadratic age effects and heterogeneous first-order autoregressive covariance, was able to describe
the radial variation in wood density. Closer spacing of trees (1000 trees/ha) resulted in a faster increase in wood density
from the pith outward than for more widely spaced trees, indicating that initial tree spacing may influence the age of transition

Hybrids generated by crossing Kuril larch (Larix gmelinii var. japonica) and Japanese larch (L. kaempferi) are expected to have high carbon accumulation ability because of fast growth and high wood density in Hokkaido, Japan. We
estimated the amount of carbon accumulation of the hybrid larch in three progeny test plantations consisting of 21 full-sib
families and compared the results to the carbon accumulation of open-pollinated progenies of Japanese larch plus-trees (improved
Japanese larch). Gene-environment interactions were not observed for tree height and diameter at breast height but were seen
for wood density (area-weighted density, AWD). The amount of carbon accumulated per unit area (Cstand) positively correlated with the stand volume. The AWD did not correlate with the C
stand; therefore, families with high wood density can be selected independent of the C
stand. The C
stand of the best full-sib family, female parent half-sib family, and male parent half-sib family at three sites were 106.1, 84.6,
and 93.2 Cton·ha−1, respectively. All these values exceed the mean for the improved Japanese larch, which has a C

Physical properties of shark gelatin were examined during gel formation and postgelation in comparison with pig gelatin. Samples with various concentrations and pH values were evaluated by breaking strength, dynamic viscoelasticity, and dynamic light scattering. Sol-gel and gel-sol transition temperatures for shark gelatin were remarkably lower than those for pig gelatin. Shark gelatin gel shows a narrower pH range to form a stable gel compared with pig gelatin. Melting enthalpy of shark gelatin gel was greater than that of pig gelatin gel, and G' of shark gelatin gel changed more extensively with rising temperature in comparison with pig gelatin gel. It is concluded that shark gelatin has different characteristics from pig gelatin not only for gel characteristics but also for the solution property. 相似文献

N,N'-bis(2-chloroethyl)-N-nitrosourea (BCNU) is one of the major drugs used in chemotherapy against malignant gliomas due to its effects, such as induction of bifunctional alkylation of DNA and formation of interstrand DNA cross-linkages, and induces cortical malformations in the fetal and neonatal rat brain. In this study, pregnant rats were treated with 7.5 mg/kg of BCNU on gestational day 13 (GD 13), and their fetuses were collected from 12 to 72 hours after BCNU treatment in order to examine the timecourses of morphological and immunohistochemical changes in neural progenitor cells in the developing brain. The number of pyknotic cells in the telencephalon peaked at 24 h and then gradually decreased until 72 h. The majority of these pyknotic cells were positive for cleaved caspase-3, a key executioner of apoptosis. The pyknotic cells showed the ultrastructural characteristics of apoptosis. The number of p53-positive cells began to increase prior to the appearance of apoptotic cells and p21-positive cells. The number of phosphorylated-histone H3-positive cells (mitotic cells) decreased from 24 to 36 h. The number of Iba1-positive cells (microglial cells) in the telencephalon increased from 12 to 48 h. These results suggest that BCNU induces p53-dependent apoptosis and reduces proliferative activity, resulting in reduction of the weight of the telencephalon and the thickness of the telencephalic wall in the fetal brain. This study will help to clarify the mechanisms of BCNU-induced fetal brain toxicity. 相似文献

The renal biopsy tissue from a 9-month-old, male Pyrenean Mountain dog with
renal disorder and severe proteinuria was examined. Ultrastructural examination revealed
multilaminar splitting and fragmentation of the glomerular basement membrane (GBM) and
diffuse podocyte foot process effacement. Immunofluorescent staining for α(IV) chains
revealed presence of α5(IV) and complete absence of α3(IV) and α4(IV) chains in the GBM.
Immunohistochemistry also revealed decreased and altered expression of nephrin and podocin
in the glomeruli compared with normal canine glomeruli. These results suggested that the
glomerular disease of the present case might be consistent with canine hereditary
nephropathy resembling human Alport syndrome caused by genetic defect of type IV collagen,
and indicated possible contribution of podocyte injury to severe proteinuria in this

We previously established 3 cell lines (PLS10, PLS20 and PLS30) from a chemically-induced prostate carcinoma in F344 rats, and demonstrated high potential for metastasis in nude mice. In the present study, we investigated the feasibility of establishing an orthotopic model using the 3 rat prostate cancer cell lines in immunocompetent rats with the aim of resolving species-mismatch problems and defects of immune systems. The PLS10, PLS20 and PLS30 cell lines were injected into the ventral prostates of 6-week-old rats, which were then sacrificed at experimental weeks 4 and 8. Tumor mass formation was found in rats with PLS10, but not in those with PLS20 or PLS30. Additionally, metastatic carcinomas could be detected in lymph nodes and lungs of PLS10-inoculated rats. Genetic analysis demonstrated K-ras gene mutations in PLS10 and PLS20, but not in PLS30 cells. There were no mutations in p53 and KLF6. In conclusion, we established a syngeneic orthotopic model for prostate cancer in immunocompetent rats simulating human castration-resistant prostate cancer (CRPC), which should prove useful for development and validation of therapeutic agents, especially with immunotherapy. 相似文献

A malignant basal cell tumor was found in the skin of the abdomen of a female Djungarian hamster of unknown age and weighing 40 g. Histologically, the tumor mass was composed of cells resembling the basal cells of the epidermis, and these cells exhibited solid type proliferation. In the tumor tissue and necrotic foci, horn cysts were observed. Immunohistochemically, cytokeratin was present in the tumor cells and horncysts. By electron microscopic examination, the tumor cells had scanty cell organelles and a few desmozomes. This paper describes a rare malignant basal cell tumor in a Djungarian hamster. 相似文献
