  1. Although the network of national parks in Zambia offers a degree of protection for freshwater diversity, the protection status of numerous systems outside of these parks requires further action. The biodiversity associated with its freshwater systems, both lotic and lentic, is unique, covering a climatic gradient from tropical to subtropical across the Zambezi and Congo basins. Recent Zambian legislation allows for the delineation of water resource protection areas (WRPAs), with one of the criteria being that they include aquatic areas of ecological importance (AEIs).
  2. In this study, a systematic conservation planning approach was used to identify aquatic AEIs objectively. Importantly, the approach included a rigorous and iterative stakeholder engagement and review process.
  3. The conservation planning software marxan was chosen because of its ability to integrate upstream–downstream connectivity. In total, 5,671 planning units (sub-catchments with an average area of approx. 14,000 ha) were populated with 77 biodiversity features: data were drawn from a wide range of sources, and included fishes, semi-aquatic mammals, molluscs, amphibians, and ecotonal physiographic features, such as waterfalls. Sub-catchments were preferentially chosen using a combination of area- and distance-weighted boundary costs.
  4. The final solution highlights critical clusters in each of the major freshwater ecoregions in Zambia, with all conservation targets being met. Results show that although the existing protected area network also coincides with identified aquatic AEIs, approximately 80% of all aquatic AEIs fall outside of formally protected areas.
  5. The outcomes of this process serve as one of three prioritization layers (the other two being water provision and sensitivity to human impacts) that are integrated in a larger study to select and prioritize WRPAs.

A polyclonal IgY preparation, purified from yolk of immunized hens, is shown to be useful for the detection of Loma salmonae in Pacific salmon. Purity of the IgY was confirmed by SDS-PAGE and western blots of yolk protein extracts. Reactivity of the IgY preparation to L. salmonae was determined through indirect-fluorescent antibody tests on purified spores and by immunohistochemistry. Developing xenomas in histological sections of heart and gill tissue of infected chinook salmon (Oncorhynchus tshawytscha) were visualized as early as 4 weeks post-exposure (PE). Non-lethal detection of L. salmonae was obtained by indirect-fluorescent microscopy of acetone-fixed gill smears. The IgY preparation was shown to react with another Loma sp., L. branchialis. In contrast, no reactivity was observed with other microsporidian parasites of fish such as Pseudoloma neurophilia and Glugea anomala. Yolk derived IgY may be useful in the detection of other fish pathogens.  相似文献

  • 1. Growing concern associated with threats to the marine environment has resulted in an increased demand for marine reserves that conserve representative and adequate examples of biodiversity. Often, the decisions about where to locate reserves must be made in the absence of detailed information on the patterns of distribution of the biota. Alternative approaches are required that include defining habitats using surrogates for biodiversity. Surrogate measures of biodiversity enable decisions about where to locate marine reserves to be made more reliably in the absence of detailed data on the distribution of species.

Intertidal habitat types derived using physical properties of the shoreline were used as a surrogate for intertidal biodiversity to assist with the identification of sites for inclusion in a candidate system of intertidal marine reserves for 17 463 km of the mainland coast of Queensland, Australia. This represents the first systematic approach, on essentially one‐dimensional data, using fine‐scale (tens to hundreds of metres) intertidal habitats to identify a system of marine reserves for such a large length of coast. A range of solutions would provide for the protection of a representative example of intertidal habitats in Queensland.

The design and planning of marine and terrestrial protected areas systems should not be undertaken independently of each other because it is likely to lead to inadequate representation of intertidal habitats in either system. The development of reserve systems specially designed to protect intertidal habitats should be integrated into the design of terrestrial and marine protected area systems.

Various ocean‐climate models driven by increased greenhouse gases and higher temperatures predict a decline in oceanic dissolved oxygen (DO) as a result of greater stratification, reduced ventilation below the thermocline, and decreased solubility at higher temperatures. Since spreading of low oxygen waters is underway and predicted to increase, understanding impacts on higher trophic levels is essential. Within the California Current System, shoaling of the oxygen minimum zone (OMZ) is expected to produce complex changes. Onshore movement of the OMZ could lead to habitat compression for species with higher oxygen requirements while allowing expansion of species tolerant of low bottom DO. As part of annual groundfish surveys, we sampled catch across a range of conditions from the upper to the lower limit of the OMZ and shoreward across the continental shelf of the US west coast. DO ranged from 0.02 to 4.25 mL L?1 with 642 stations (of 1020 sampled) experiencing hypoxic conditions in 2008–2010. Catch and species richness exhibited significant and positive relationships with near‐bottom oxygen concentration. The probability of occurrence was estimated for four species (spotted ratfish, petrale sole, greenstriped rockfish and Dover sole) using a binomial Generalized Additive Model. The models for each species included terms for position, day of the year, salinity, near‐bottom temperature and the interaction term between depth and near‐bottom DO. Spotted ratfish and petrale sole were sensitive to changes in near‐bottom oxygen, while greenstriped rockfish and Dover sole show no changes in probability of occurrence in relation to changes in oxygen concentration.  相似文献

The most widespread and numerous inland fish in the world is likely the mosquitofish (Gambusia affinis and G. holbrooki, Poeciliidae). Much has been written about the basic biology, the current distribution and the negative impacts of non-native populations of mosquitofish. Here, we instead review the relationship of humanity with mosquitofish. First, we review the early literature on the species and aim to resolve its path towards becoming the globally dominant fish for biological control of mosquitoes. We identify the initial advocates of mosquitofish use, we examine the reasons behind their advocacy, and we document the spread of their viewpoints into and from the globally foundational mosquito control texts. Second, we identify the people and institutions that facilitated early international translocations of mosquitofish, including, among others, David Starr Jordan, the Rockefeller Foundation and the International Red Cross. Third, we discuss the reduction in mosquitofish translocation and use during and after WWII, initially stemming from the discovery and use of other methods, like DDT and later from a recognition of the negative ecological consequences of non-native mosquitofish populations. Fourth, we propose that the future utility of mosquitofish is largely in its value as a model study organism. We provide an overview of the contributions mosquitofish have made to some major fields in biology. Finally, we suggest that the value of mosquitofish as a model system should increase into the future, behind a momentum of research advances, and as human-mediated range expansion will permit access to mosquitofish by yet greater numbers of biologists worldwide.  相似文献

The present study sought to quantitatively examine mucosal inflammatory and immune responses in dogs with gastritis and the relationship of these responses to infection with Helicobacter. Gastric biopsies from 30 dogs were evaluated for B- and T-lymphocytes, neutrophils, eosinophils, macrophages, and mast cells. Mucosal atrophy, fibrosis, cellularity, and severity of gastritis were graded qualitatively. Messenger-RNA (mRNA) for actin, interleukin-1beta (IL-1beta), IL-4, IL-8, and IL-10, transforming growth factor beta (TGF-beta), and interferon gamma (IFN-gamma) was quantified by polymerase chain reaction (PCR). The presence of Helicobacter spp. was determined by urease activity, histology, PCR, and enzyme-linked immunosorbent assay. mRNA for IL-1beta, IL-8, IL-10, TGF-beta, and IFN-gamma was detected in most dogs. IL-4 mRNA was detected in only 1 dog. Correlations were observed for IL-1beta versus IL-8 and IL-10; IL-8 versus IL-10, IFN-gamma, and TGF-beta; and IL-10 versus IFN-y. Mucosal pathology was related to cytokine mRNA expression (neutrophils to IL-8 and IFN-gamma, macrophages and lymphocytes to IFN-gamma, and fibrosis to IL-1beta). Gastritis was categorized as lymphoplasmacytic in all dogs, and its histologic severity correlated with atrophy, infiltration with lymphocytes and macrophages, and expression of IL-10 and IFN-gamma. Of the dogs examined, 76.7% were infected with Helicobacter spp. Infection was associated with increased expression of TGF-beta and fibrosis. Circulating anti-Helicobacter immunoglobulin G titers were higher in uninfected than infected dogs. We conclude that lymphoplasmacytic gastritis in dogs is characterized by concurrent activation of proinflammatory and immunomodulatory cytokines, with increased mRNA expression related to mucosal pathology. No significant associations between Helicobacter infection and proinflammatory cytokine expression, severity of gastritis, or differences in the pathogenicity of different Helicobacter spp. were found.  相似文献

  1. Monitoring the abundance of mobile and wide‐ranging cetacean populations for conservation management is challenging, especially when the management is focused on static protected areas. Where abundance estimates are derived from mark–recapture data, such as photo‐identification of naturally marked individuals, unpredictable movements of animals in and out of the survey area can reduce ‘capture’ probabilities and affect the precision and accuracy of resulting estimates.
  2. A Bayesian hierarchical log–linear model was applied to photo‐identification data collected in summer 2014 to derive a multi‐site abundance estimate for a population of bottlenose dolphins, Tursiops truncatus, ranging widely throughout the coastal waters of western Ireland. In addition, the effects of varying levels of sampling effort on the minimum detectable decrease in population size were examined.
  3. The abundance (median) of dolphins was estimated as 189 (coefficient of variation (standard deviation/mean), 0.11; 95% highest‐posterior density interval, 162–232). Over 50% of the well‐marked dolphins encountered throughout the study were sighted in more than one distinct coastal area, thus displaying high mobility. In addition, it was found that it would require biennial surveys to detect a 25% decline in abundance within the six‐year reporting period of the EU’s Habitats Directive.
  4. Given that the Special Area of Conservation designated for these dolphins consists of two separate areas covering a substantial portion of the west coast of Ireland, the multisite approach is appropriate for monitoring this population. It produces a more precise estimate and is well suited for sparse recapture data collected opportunistically at multiple sites, when the lack of resources prevents large‐scale surveys or when concentrating surveys on smaller localized areas fails to capture the broad range and unpredictable occurrence of the animals. The Bayesian multi‐site approach could be applied to the management of other wide‐ranging marine or terrestrial taxa.

The western and central Pacific Ocean supports the world's largest tuna fisheries. Since the 1990s, the purse‐seine fishery has increasingly fished in association with fish aggregating devices (FADs), which has increased catches of juvenile bigeye and yellowfin tunas and vulnerable bycatch species (e.g., sharks). This has raised concerns regarding the sustainability of these species’ populations and the supporting ecosystem, but may provide improved food security of Pacific Island nations through utilisation of FAD‐associated byproduct species (e.g., wahoo). An ecosystem model of the western Pacific Warm Pool Province was used to explore the potential ecological impacts of varying FAD fishing effort (±50% or 100%) over 30 years. The ecosystem has undergone a significant change in structure since 1980 from heavy exploitation of top predators (e.g., tunas) and “fishing up the food web” of high‐trophic‐level non‐target species. The ecosystem appeared resistant to simulated fishing perturbations, with only modest changes (<10%) in the biomass of most groups, although some less productive shark bycatch species decreased by up to 43%, which had a subsequent positive effect on several byproduct species, the prey of sharks. Reduction of FAD effort by at least 50% was predicted to increase the biomass of tuna species and sharks and return the ecosystem structure to a pre‐industrial‐fishing state within 10 years. Spatial disaggregation of the model and integration of economic information are recommended to better capture ecological and economic changes that may result from fishing and/or climate impacts and to develop appropriate management measures in response.  相似文献
