The response of bean plants (Phaseolus vulgaris L.) to different levels of arsenic (As) and salinity was investigated, including the processes of uptake, distribution, and accumulation of As and the effect of arsenite and salinity on plant growth and fruit production. The experiment was performed in soilless culture at two levels of As: 2 and 5 mg As L‐1 [added as sodium arsenite (NaAsO2)], and three saline levels [only sodium chloride (NaCl) was added]: 1,000,2,000, and 4,000 μS#lbcM‐1. Arsenic uptake and concentration in root increased upon increased NaAsO2 concentration in the nutrient solution. However, the increase in the As root content was not proportional to the As level in the nutrient solution. High levels of salinity in solution decreased As uptake and the concentration of As in root, stem, and leaf. Upon uptake, As was readily translocated to the aerial organs and approximately half of the absorbed As was transported to the upper parts of the bean plants. The As concentration in fruit always remained below the recommended limit for As content in fruit and edible vegetal products. While salinity did not significantly affect plant growth, arsenite was found to be phytotoxic to the bean plants.  相似文献

We describe the magnetic resonance (MR) imaging aspects of normal canine optic nerve, the diameter of the optic nerve as measured on MR images, and optimal MR sequences for the evaluation of the optic nerve using a 0.2 T MR unit. Three millimeter contiguous slides of the normal canine orbital region were acquired in transverse and dorsal oblique planes using a variety of tissue weighting sequences. It was apparent that detailed anatomic assessment of the optic nerve can be performed with low‐field MR imaging, but none of the sequences provided unequivocal superior image quality of the optic nerve. The mean diameter of the optic nerve sheath complex was 3.7 mm and of the optic nerve 1.7 mm. The intraorbital and intracanalicular parts of the optic nerve are consistently visible and differentiation between the optic nerve and optic nerve sheath complex is possible using low‐field MR systems.  相似文献

The different products of Eichhornia crassipes leaves including dried E. crassipes powder (DEP), hot‐water treated E. crassipe (HTE), hot‐water extract of E. crassipe (ECE) and dreg of hot‐water extract of E. crassipe extract (dECE) were produced and incorporated into the diet of the prawn, Macrobrachium rosenbergii, as an immunostimulant. Results showed that prawn fed the HTE‐, ECE‐ and dECE‐containing diets for 4 months had increased total haemocyte count, different haemocyte count, phenoloxidase activity, respiratory bursts, superoxide dismutase activity, glutathione peroxidase activity especially in HTE and ECE treatments. The phagocytic activity and clearance efficiency against Lactococcus garvieae of prawn fed the HTE‐ and ECE‐containing diets were significantly higher than those of prawn fed the control diet at 2–4 months. The relative percentage survival of prawn fed the DEP‐, HTE‐, ECE‐ and dECE‐containing diets for 4 months following 144 h challenging with L. garvieae were 19.0%, 38.1%, 38.1% and 33.3%. It was concluded that E. crassipes leaves containing an active component which was easily extracted by hot water can enhance innate immunity and resistance against pathogen of M. rosenbergii by dietary long‐term administration, and the administration of HTE in the diet was the best strategy due to the availability and convenience.  相似文献

Lactic acid bacteria (LAB) are suitable for rice straw silage fermentation, but have been studied rarely, and rice straw as raw material for ensiling is difficult because of its disadvantages, such as low nutrition for microbial activities and low abundances of natural populations of LAB. So we investigated the effect of application of LAB and chemical additives on the fermentation quality and microbial community of wilted rice straw silage. Treatment with chemical additives increased the concentrations of crude protein (CP), water soluble carbohydrate (WSC), acetic acid and lactic acid, reduced the concentrations of acid detergent fiber (ADF) and neutral detergent fiber (NDF), but did not effectively inhibit the growth of spoilage organisms. Inoculation with LABs did not improve the nutritional value of the silage because of poor growth of LABs in wilted rice straw. Inoculation with LAB and addition of chemical materials improved the quality of silage similar to the effects of addition of chemical materials alone. Growth of aerobic and facultatively anaerobic bacteria was inhibited by this mixed treatment and the LAB gradually dominated the microbial community. In summary, the fermentation quality of wilted rice straw silage had improved by addition of LAB and chemical materials.  相似文献

Forty‐eight castrated male goats were used to determine the effects of feeding green tea by‐products (GTB) on growth performance, meat quality, blood metabolites and immune cell proliferation. Experimental treatments consisted of basal diets supplemented with four levels of GTB (0%, 0.5%, 1.0% or 2.0%). Four replicate pens were assigned to each treatment with three goats per replicate. Increasing dietary GTB tended to linearly increase the overall average weight gain and feed intake (p = 0.09). Water holding capacity, pH and sensory attributes of meat were not affected by GTB supplementation, while cooking loss was reduced both linearly and quadratically (p < 0.01). The redness (linear; p = 0.02, quadratic; p < 0.01) and yellowness (quadratic; p < 0.01) values of goat meat were improved by GTB supplementation. Increasing dietary GTB quadratically increased protein and decreased crude fat (p < 0.05), while linearly decreased cholesterol (p = 0.03) content of goat meat. The proportions of monounsaturated fatty acid, polyunsaturated fatty acid (PUFA) and n‐6 PUFA increased linearly (p < 0.01) and n‐3 PUFA increased quadratically (p < 0.05) as GTB increased in diets. Increasing dietary GTB linearly increased the PUFA/SFA (saturated fatty acid) and tended to linearly and quadratically increase (p ≤ 0.10) the n‐6/n‐3 ratio. The thiobarbituric acid‐reactive substances values of meat were lower in the 2.0% GTB‐supplemented group in all storage periods (p < 0.05). Dietary GTB linearly decreased plasma glucose and cholesterol (p < 0.01) and quadratically decreased urea nitrogen concentrations (p = 0.001). The growth of spleen cells incubated in concanavalin A and lipopolysaccharides medium increased significantly (p < 0.05) in response to GTB supplementation. Our results suggest that GTB may positively affect the growth performance, meat quality, blood metabolites and immune cell proliferation when supplemented as a feed additive in goat diet.  相似文献

BACKGROUND: Coptotermes formosanus Shiraki was accidentally introduced into the United States from Asia. The introduction of the pest has brought significant economic consequences. During the past decade, Mississippi has become a significantly infested state, partly due to the proximity to coastal port cities such as New Orleans. This study was initiated to investigate the origin and infestation route of C. formosanus in southern Mississippi. RESULTS: Twenty‐eight colonies (21 colonies from Mississippi, six from Louisiana, one from China) were collected. Sequencing and analysis of 112 sequences revealed 15 haplotypes of cytochrome oxidase subunit II (COII) gene in the world. Two haplotypes of COII were identified in Mississippi. In addition, specific primers were designed and tested differentially to amplify characteristic fragments for verifying and surveying different genotypes of C. formosanus in the future. CONCLUSION: Of the two haplotypes identified in Mississippi, the GA group was identical to those reported previously in Georgia, Louisiana, Alabama and other infested states. The second haplotype, the AT group, was identified for the first time in southeastern United States. Sequence identity of the AT‐group C. formosanus with those reported mainly in southeastern Asian countries provided evidence of at least two introductions of C. formosanus into the United States. Agroforestry systems deposit great amounts of plant residues on soil and this leads to high levels of soil organic matter content and has increased soil biodiversity and improved its conservation. This study compares the distribution of meso and macrofaunal communities in soil and litter under cacao agroforestry systems and in a natural forest in the southern Bahia state of Brazil. Soil and litter samples were obtained in September 2003, February 2004, and August 2004 in five cacao agroforestry systems. The systems evaluated included: cacao renewed under Erythrina sp. (Erythrina poeppigiana) (CRE); cacao renewed under natural forest (Cabruca, CRF); an old cacao system under Erythrina sp. (OCE); an old cacao system under a natural forest system (Cabruca, OCF) and a cacao germplasm collection area (CGC). As a reference soil and litter under a natural forest (NF) was included. Organisms were collected over a 15-day period with a Berlese–Tullgren apparatus. The density and richness of total fauna varied distinctly according to sites, sampling time and material sampled (soil and litter). 16,409 of fauna were recovered from soil and litter samples and the density of total fauna was 2,094 individuals m−2 in the litter and 641 individuals m−2 in the soil. The richness was 11.8 in the litter and 7.5 in the soil. The cacao agroforestry systems adopted for growing cacao in the southern Bahia region of Brazil have beneficial effects on the soil and litter faunal communities, and such systems of cacao cultivation could be considered as a conservation system for soil fauna. The development of a litter layer resulted in higher abundance and diversity of soil fauna.  相似文献

A soybean lecithin‐based extender supplemented with hyaluronic acid (HA) was assayed for effectiveness to improve the quality of frozen–thawed ram semen. HA has not been tested yet in an extender containing soybean lecithin for freezing ram semen. Thus, the aim of this study was to analyse the effects of soybean lecithin at 1% or 1.5% along with HA at 0, 0.5 and 1 mg ml‐1 in a Tris‐based extender on the motion characteristics, membrane integrity (HOST), viability, GSH peroxidase (GSH‐PX) activity, lipid peroxidation and acrosomal status after freezing–thawing. Semen was collected from four Mehraban rams during the breeding season and frozen in the six lecithin×HA extenders. The extender containing 1.5% lecithin supplemented with no HA yielded higher total motility (52.5%±1.6), viability (55.8%±1.6) and membrane integrity (44.5%±1.7), but the effects of the lecithin concentration did not reach signification. Linearity‐related parameters, ALH, BCF, lipid peroxidation, GSH‐PX activity, morphology and acrosomal status were not affected by the extender composition. In general, adding HA significantly decreased sperm velocity (1 mg ml‐1 HA), total motility (only with 1.5% lecithin), viability (1 mg ml‐1 HA for 1% lecithin; both concentrations for 1.5% lecithin) and membrane integrity. In conclusion, adding HA to the freezing extender supplemented with soybean lecithin failed to improve quality‐related variables in ram semen. Increasing the lecithin content could have a positive effect, but further studies are needed.  相似文献

Plant pathogens have developed different strategies during their evolution to infect and colonize their hosts. In the same way, plants have evolved different mechanisms acting against potential pathogens trying to infect and colonize their tissues. Regulation of a wide variety of proteins is required in order to perceive the pathogen and to activate the plant defence mechanisms. The apoplast is the first compartment where these recognition phenomena occur in most plant–pathogen interactions, allowing the exchange of different molecules and facilitating inter‐ and intracellular communication in plant cells. Proteomic analysis of the apoplast in recent years has found the initial biochemical responses involved in pathogen recognition and early defence responses. However, this proteomic approach requires some specific experimental conditions to obtain an extract free of cytoplasmic proteins and nonprotein contaminants that affect the subsequent stages of separation and quantification. Obtaining the highest proportion of proteins from the apoplastic space in infected tissues requires different steps such as extraction of apoplastic washing fluids and preparation of total secreted proteins (protein precipitation, solubilization, separation and digestion). Protein identification using mass spectrometry techniques and bioinformatics tools identifying peptides for the extracellular exportation is required to confirm the apoplastic location. This review compiles the most commonly used techniques for proteomic studies, focusing on the early biochemical changes occurring in the apoplast of plants infected by a wide range of pathogens. The scope of this approach to discover the molecular mechanisms involved in the plant–pathogen interaction is discussed.  相似文献
