After more than 20 years of hatchery production of Asian seabass in Thailand, genetic information is still lacking for effective genetic management and a selective breeding programme. This study aimed to evaluate genetic status of existing hatchery populations and genetic consequences of a selective breeding attempt. We examined genetic relatedness in seven hatchery samples, including a selectively bred population (RACF‐F1), compared with three wild samples using 11 microsatellite loci. Genetic diversity and relatedness values within most hatchery samples, except for RACF‐F1, did not differ from those of wild populations (> 0.05). RACF‐F1 had the lowest allelic diversity and effective population size (Ar = 6.99; Ne = 7.8) and highest relatedness values (mean rxy = 0.075–0.204). Pairwise ΦST values, principal component analysis and model‐based cluster analyses revealed three genetically distinct hatchery groups: Eastern Thailand (CHN, RACF, NSCF and SKCF), Southern Thailand (NICA) and the Andaman Sea (STCF). Results suggest that exiting domestic populations capture reasonable amount of genetic variation and can be useful for a base population for genetic improvement programmes. In addition, given the rapid increase in relatedness that we observed in one selectively bred population, we recommend using selection methods and hatchery practices that reduce variability in family contribution in the subsequent generations.  相似文献

Thermogravimetric analysis was used to determine the oxidative stability of various edible oils (olive oil, milkfat) and triacylglycerides (triolein, trilinolein), while the effect of natural (alpha-tocopherol, ascorbic acid) and synthetic antioxidants (butylated hydroxytoluene (BHT), butylated hydroxyanisole (BHA), and tertiary butyl hydroquinone were evaluated by addition to trilinolein. Oil resistance to oxidation was obtained by measuring the increase in sample weight due to the uptake of molecular oxygen, the temperature at maximum sample weight, and the temperature at the onset of oxidation. When comparing sample weight increase, trilinolein proved to be oxidatively less stable than triolein, olive oil, and milk fat, while triolein was less stable than olive oil and milk fat. Olive oil showed significantly higher stability than milkfat when comparing the temperature at the onset of oxidation. When comparing effectiveness of antioxidants, a combination of 0.01% BHA and 0.01% BHT increased trilinolein stability the most.  相似文献

Background

The proportion of granule types in wheat starch is an important characteristic that can affect its functionality. It is widely accepted that granule types are either large, disc-shaped A-type granules or small, spherical B-type granules. Additionally, there are some reports of the tiny C-type granules. The differences between these granule types are due to its carbohydrate composition and crystallinity which is highly, but not perfectly, correlated with the granule size. A majority of the studies that have considered granule types analyse them based on a size threshold rather than chemical composition. This is understandable due to the expense of separating starch into different types. While the use of a size threshold to classify granule type is a low-cost measure, this results in misclassification. We present an alternative, statistical method to quantify the proportion of granule types by a fit of the mixture distribution, along with an R package, a web based app and a video tutorial for how to use the web app to enable its straightforward application.

Results

Our results show that the reliability of the genotypic effects increase approximately 60% using the proportions of the A-type and B-type granule estimated by the mixture distribution over the standard size-threshold measure. Although there was a marginal drop in reliability for C-type granules. The latter is likely due to the low observed genetic variance for C-type granules.

Conclusions

The determination of the proportion of granule types from size-distribution is better achieved by using the mixing probabilities from the fit of the mixture distribution rather than using a size-threshold.

The initial electron transfer dynamics during photosynthesis have been studied in Rhodobacter sphaeroides reaction centers from wild type and 14 mutants in which the driving force and the kinetics of charge separation vary over a broad range. Surprisingly, the protein relaxation kinetics, as measured by tryptophan absorbance changes, are invariant in these mutants. By applying a reaction-diffusion model, we can fit the complex electron transfer kinetics of each mutant quantitatively, varying only the driving force. These results indicate that initial photosynthetic charge separation is limited by protein dynamics rather than by a static electron transfer barrier.  相似文献

In the moist mid‐latitudes of eastern Australia, soil water dynamics, herbage production and water use efficiency (WUE) were monitored during 2006–2008, for five perennial pastures: digit grass (Digitaria eriantha), Rhodes grass (Chloris gayana), forest bluegrass (Bothriochloa bladhii), native grass (Bothriochloa macra and Rytidosperma bipartita dominant), lucerne (Medicago sativa); and two forage crops: oat (Avena fatua) and sorghum (Sorghum bicolor). Ground cover formed more quickly in Rhodes grass and lucerne (>70% ground cover in 120 and 175 days after sowing [DAS] respectively) than in forest bluegrass and digit grass (245 and 365 DAS respectively). Values of maximum extractable water (MEW) for Rhodes grass and lucerne were similar (180–242 mm), while values for digit grass and forest bluegrass (129–175 mm) were equal to or greater than those for native grass, and two annual forage crops (77–144 mm). Lucerne expressed the maximum root depth (1.46 m), while values for the tropical grasses (0.96–1.39 m) were greater than native grasses and forage crops (0.87–0.96 m). Native grasses (6.5–12 t DM/ha) had the lowest herbage production, which resulted in values of WUE that were significantly less than most other treatments (16–21 vs. 23–43 kg DM ha?1 mm?1). Digit grass (33–34 kg DM ha?1 mm?1) had higher WUE compared with the other tropical grasses (20–27 kg DM ha?1 mm?1). The data collected here suggest that a forage system comprising digit grass, lucerne and forage oat would provide high production and WUE in this environment.  相似文献

A life cycle assessment (LCA) was conducted to estimate whole-farm greenhouse gas (GHG) emissions from beef production in western Canada. The aim was to determine the relative contributions of the cow-calf and feedlot components to these emissions, and to examine the proportion of whole-farm emissions attributable to enteric methane (CH4). The simulated farm consisted of a beef production operation comprised of 120 cows, four bulls, and their progeny, with the progeny fattened in a feedlot. The farm also included cropland and native prairie pasture for grazing to supply the feed for the animals. The LCA was conducted over 8 years to fully account for the lifetime GHG emissions from the cows, bulls and progeny, as well as the beef marketed from cull cows, cull bulls, and progeny raised for market. The emissions were estimated using Holos, a whole-farm model developed by Agriculture and Agri-Food Canada. Holos is an empirical model, with a yearly time-step, based on the Intergovernmental Panel on Climate Change methodology, modified for Canadian conditions and farm scale. The model considers all significant CH4, N2O, and CO2 emissions and removals on the farm, as well as emissions from manufacture of inputs (fertilizer, herbicides) and off-farm emissions of N2O derived from nitrogen applied on the farm. The LCA estimated the GHG intensity of beef production in this system at 22 kg CO2 equivalent (kg carcass)−1. Enteric CH4 was the largest contributing source of GHG accounting for 63% of total emissions. Nitrous oxide from soil and manure accounted for a further 27% of the total emissions, while CH4 emissions from manure and CO2 energy emissions were minor contributors. Within the beef production cycle, the cow-calf system accounted for about 80% of total GHG emissions and the feedlot system for only 20%. About 84% of enteric CH4 was from the cow-calf herd, mostly from mature cows. It follows that mitigation practices to reduce GHG emissions from beef production should focus on reducing enteric CH4 production from mature beef cows. However, mitigation approaches must also recognize that the cow-calf production system also has many ancillary environmental benefits, allowing use of grazing and forage lands that can preserve soil carbon reserves and provide other ecosystems services.  相似文献

Multiple‐batch production is the most widely practiced method of raising channel catfish. Producers are increasingly adopting intensified production practices in multiple‐batch systems by increasing stocking density and aeration rates as a means to improve cost efficiencies. Proven stocking recommendations are required for the efficient implementation of recent developments in multiple‐batch production. Twelve 0.4‐ha ponds were understocked with 17,484, 20,612, and 26,124 fingerlings/ha (mean weight = 40 g/fish) over equal weights of carryover fish (0.46 kg/fish @ 4,589 kg/ha). Fish were fed once daily to apparent satiation with a 28% protein floating feed and aerated with a single 7.4‐kW electric paddlewheel aerator. Density‐dependent significant differences were absent for gross, net, daily net yields, marketable yields (≥0.54 kg), growth (g/day), and survival. Sub‐marketable yield (<0.54 kg) and feeding rate increased significantly with increased understocking density. Economic analysis revealed increased breakeven prices and diminished net returns with increased stocking density when sub‐marketable fish were not considered as revenue. These differences in production costs and profits among the three treatments became minimal when sub‐marketable fish were included as revenue. All three density treatments attained positive annual net cash flows. This study validates channel catfish understocking densities of 17,000–26,000 fish/ha to improve cost efficiency in intensively aerated, multiple‐batch production systems.  相似文献

Spatial and seasonal variation in the abundance and composition of phytoplankton and other basal resources (bacteria, terrestrial detritus) influence the availability of essential polyunsaturated fatty acids (PUFA) for upper trophic levels in lake food webs. We studied accumulation, composition and content of fatty acids in planktivorous vendace (Coregonus albula) muscle tissue during the summer feeding period (May to late September) in six boreal lakes. Vendace muscle fatty acid content increased from ~17.7 mg/g DW to ~28.1 mg/g DW from spring to summer, corresponding to a ~59% increase in total fatty acids. PUFA accounted for 45%–65% of all fatty acids, and the most abundant fatty acid was docosahexaenoic acid (DHA, 22:6n‐3). The DHA content remained relatively constant throughout the sampling period (~7–8 mg/g DW), but the proportion of DHA decreased from ~40% in spring to ~30% in summer and autumn. The decrease in DHA proportion was due to accumulation of other fatty acids, primarily C18 PUFA (predominantly synthetised by algae) and monounsaturated fatty acids. A similar fatty acid accumulation pattern was observed in lipid storing copepods, an important prey for planktivorous vendace. The abundance of bacterial and terrestrial biomarker fatty acids in vendace muscle and prey items was low (<5% and <0.1% respectively). The increase in weight and the deposition of internal energy stores in vendace were achieved predominantly by algae‐based food chain. This has ecological implications, as vendace requires sufficient lipid stores for reproduction in autumn and survival over winter.  相似文献

Interhemispheric inhibition is thought to mediate cortical rivalry between the two hemispheres through callosal input. The long-lasting form of this inhibition is believed to operate via γ-aminobutyric acid type B (GABA(B)) receptors, but the process is poorly understood at the cellular level. We found that the firing of layer 5 pyramidal neurons in rat somatosensory cortex due to contralateral sensory stimulation was inhibited for hundreds of milliseconds when paired with ipsilateral stimulation. The inhibition acted directly on apical dendrites via layer 1 interneurons but was silent in the absence of pyramidal cell firing, relying on metabotropic inhibition of active dendritic currents recruited during neuronal activity. The results not only reveal the microcircuitry underlying interhemispheric inhibition but also demonstrate the importance of active dendritic properties for cortical output.  相似文献
