Agricultural soil is a major source of nitrous oxide (N2O), nitric oxide (NO) and ammonia (NH3). Little information is available on emissions of these gases from soils amended with organic fertilizers at different soil water contents. N2O, NO and NH3 emissions were measured in large-scale incubations of a fresh sandy loam soil and amended with four organic fertilizers, [poultry litter (PL), composted plant residues (CP), sewage sludge pellets (SP) and cattle farm yard manure (CM)], urea fertilizer (UA) or a zero-N control (ZR) for 38 days. Fertilizers were added to soil at 40, 60 or 80% water-filled pore space (WFPS). The results showed that urea and organic fertilizer were important sources of N2O and NO. Total N2O and NO emissions from UA ranged from 0.04 to 0.62%, and 0.23 to 1.55% of applied N, respectively. Total N2O and NO emissions from organic fertilizer treatments ranged from 0.01 to 1.65%, and <0.01 to=" 0.55%=" of=" applied=" n,=" respectively.=" the=" lower=">2O and NO emissions from CP and CM suggested that applying N is these forms could be a useful mitigation option. Comparison of the NO-N/N2O-N ratio suggested that nitrification was more dominant in UA whereas denitrification was more dominant in the organic fertilizer treatments. Most N was lost from PL and UA as NH3, and this was not influenced significantly by WFPS. Emissions of NH3 from UA and PL ranged from 62.4 to 69.6%, and 3.17 to 6.11% of applied N, respectively.  相似文献

In the realm of plant genomics, metabolic profiling has become a valuable tool with which to assess the effect of genetic and/or environmental factors on plant development. This paper reports the first application of metabolic profiling on differentiating xylem tissue of loblolly pine. A protocol is presented for the analysis of loblolly pine xylem tissue. The effects of sample preparation, extraction, and derivatization on the corresponding metabolite profiles and yields have been investigated and are reported. Gas chromatography-mass spectroscopy has been used to quantify >60 polar and lipophilic metabolites from wood-forming tissue. It was possible to assign chemical structures to approximately half of these compounds. Comparison of six loblolly pine genotypes, three high cellulose (50-52%) and three medium (45-48%) cellulose, showed distinct metabolic profiles. Principal component analysis enabled the assignment of metabolic phenotypes using these large data sets. Metabolic phenotype clustering occurred in which the three high-cellulose genotypes were segregated from the medium-cellulose genotypes. These results demonstrate the use of metabolic profiling for the study of wood-forming tissue and as a tool in functional genomics.  相似文献

Bran extracts of Akron wheat grown at four nonirrigated and one irrigated testing locations were examined and compared for their free radical scavenging properties against the 2,2-diphenyl-1-picrylhydrazyl radical (DPPH(*)) and the radical cation ABTS(*)(+), chelating capacities, and total phenolic content (TPC) to determine the potential effects of environmental factors on the antioxidant properties of hard winter wheat. The environmental factors included total solar radiation, average daily solar radiation, and number of hours exceeding 32 degrees C. The results showed that bran samples from different growing locations may significantly differ in their radical scavenging activities against both DPPH(*) and ABTS(*)(+), chelating capacities, and TPC. A significant negative correlation was detected between the chelating activities of the bran samples from the four nonirrigated locations and total solar or daily average solar radiation (r = -0.999 and P = 0.001). These data suggest potential influences of growing conditions on the antioxidant properties of hard winter wheat and the possibility of producing wheat that is strong in a selected antioxidant property by optimizing the growing conditions of a selected wheat variety. More research is required to further investigate the relationship among antioxidant properties and environmental factors using different wheat varieties and larger sample sizes.  相似文献

Invasive plants are one of the greatest threats to endangered insect species and a major threat to Lepidoptera in eastern North America. We investigated the effects of the invasive shrub Chinese privet (Ligustrum sinense) and two methods (mulching or hand-felling) of removing it from riparian forests on butterfly communities and compared them to untreated, heavily invaded control plots and to “desired future condition” forests that never had extensive privet cover. Privet mulching resulted in nearly twice as many butterflies as privet felling and both treatments had more butterflies two years after privet removal than untreated control plots. Butterfly communities on control plots differed from those on the two treatments and the desired future condition forests. A number of forest characteristics were evaluated but only herbaceous plant cover (excluding privet) was positively correlated with butterfly abundance, diversity and evenness. The Carolina satyr, Hermeuptychia sosybius, was the best indicator of forests where privet had never invaded. Removing Chinese privet from riparian forests in the southeastern United States greatly improved forest habitats for butterflies and evidence suggests that butterfly communities in other temperate forests could benefit from removal of extensive shrub layers dominated by a single species.  相似文献

Conservation organizations in the northeastern United States (US) recommend forest clearcutting to create shrubland habitat, which is required by many wildlife species with declining populations. The planning of habitat management programs is hampered by a lack of information on the current extent of shrubland habitat and the current rate of forest clearcutting that creates shrubland habitat. We addressed these information gaps by using a combination of automated and manual approaches to determine the extent and spatial configuration of shrubland habitat and recent forest clearcuts. We focused on the state of Rhode Island because (a) it is representative of the northeastern US in terms of the prevalence of private ownership of forests, and the ongoing decline in the populations of many shrubland wildlife species; (b) federal, state and private conservation groups are actively promoting clearcuts to create shrubland habitat; (c) many state-wide GIS databases are available; and (d) the spatial extent of the state made our results both generalizable and politically relevant. Our fine-scale mapping allowed a detailed analysis of shrubland distribution in conjunction with other available GIS layers that facilitates identification of priority areas for habitat management. We found that the extent of upland shrubland in non-coastal areas is decreasing by at least 1.5% annually. Considering the lack of consensus about conservation targets for the amount of shrubland, we propose that conservation organizations attempt to stabilize rather than expand the extent of shrubland habitat. This approach would provide an opportunity to assess whether the current extent of shrubland is sufficient to maintain reduced but stable wildlife populations that require this habitat. We propose a coordinated forest management program with targets for increased forest management on conservation lands. We found that the average patch size of shrubland created by recent clearcuts is large enough for most shrubland bird species, but too small for the New England cottontail (Sylvilagus transitionalis), which has been proposed for threatened and endangered status.  相似文献

Thirty-six Fusarium strains were grown on cracked yellow corn and evaluated for optimum fusaproliferin production, with Fusarium subglutinans E-1583 producing the highest levels (1600 microg/g). Three solvent systems were tested for extracting fusaproliferin from the cultures of F. subglutinans E-1583. Methanol gave the highest fusaproliferin recovery, followed by methanol/1% aqueous NaCl (55:45, v/v) and acetonitrile/methanol/H(2)O (16:3:1, v/v/v). Hexane partitioning was effective in removing many impurities from the crude fusaproliferin extracts prior to the liquid chromatography step. Fusaproliferin samples were further purified by high-performance liquid chromatography (HPLC) with a C18 preparatory column using a mobile phase of acetonitrile/H(2)O (80:20, v/v). The purity of the fusaproliferin was verified by analytical HPLC, GC/MS, (1)H NMR spectroscopy, and electrospray ionization (ESI) MS. The isolated fusaproliferin was shown to be free of impurities and can be used as a standard for routine analysis. Fusaproliferin was shown to be temperature-sensitive when samples were stored at room temperature (20-24 degrees C) for more than several days. After 30 days at 4 degrees C, approximately 8% of the fusaproliferin had been transformed to deacetyl-fusaproliferin; however, samples stored at -20 degrees C for 1 year contained only trace amounts of the deacetylated form.  相似文献

Intensive sampling of a stream draining an alpine-subalpine basin revealed that depressions in pH and acid neutralizing capacity (ANC) of surface water at the beginning of the spring snowmelt in 1987 and 1988 were not accompanied by increases in strong acid anions, and that surface waters did not become acidic (ANC<0). Samples of meltwater collected at the base of the snowpack in 1987 were acidic and exhibited distinct ‘pulses’ of nitrate and sulfate. Solutions collected with lysimeters in forest soils adjacent to the stream revealed high levels of dissolved organic carbon (DOC) and total Al. Peaks in concentration of DOC, Al, and nutrient species in the stream samples indicate a flush of soil solution into the surface water at the beginning of the melt. Infiltration of meltwater into soils and spatial heterogeneity in the timing of melting across the basin prevented stream and lake waters from becoming acidic.  相似文献

Analysis of the spatial distribution of all species of conservation importance within a region is necessary to augment reserve selection strategies and habitat management in biodiversity conservation. In this study, we analyzed the spatial aggregation, spatial association, and vegetation types of point occurrence data collected from museum and herbaria records for rare, special concern, threatened, and endangered species of plants, reptiles, mammals, and birds in western Riverside County in southern California, USA. All taxa showed clumped distributions, with aggregation evident below 14 km for plants, 12 km for reptiles, 2 km for mammals, and 10 km for birds. In addition, all combinations of the different species groups showed high positive spatial association. The Santa Rosa Plateau exhibited the highest number of rare, special concern, threatened, and endangered species, and shrubland (coastal sage and chaparral) was the vegetation type inhabited by the most species. Local land use planning, zoning and reserve design should consider the spatial aggregation within and between species to determine the appropriate scale for conservation planning. The higher spatial association between species groups in this study may indicate interdependence between different species groups or shared habitat requirements. It is important to maintain diverse communities due to potential interdependence. The results of the study indicate that concentrating preservation efforts on areas with the highest number of species of concern and the restoration of native shrublands are the most appropriate actions for multiple species habitat conservation in this area.  相似文献

To optimize the extraction of sorghum proteins, several variables were examined: sample-to-solvent ratio, detergent type and concentration, reducing agent type and concentration, extraction time, and buffer pH and concentration. Samples were quantified and characterized by RP-HPLC, FZCE, and nitrogen analysis. These studies revealed that pH, detergent type, reducing agent type, and sample-to-solvent ratio all had significant effects on the levels of protein extracted. Increasing SDS concentration (2%) and solvent-to-flour ratio (20:1) with multiple 5 min extracts reduced extraction time by 35-80% while still extracting the same levels of total protein relative to the control methodology. Reproducibility using the multiple extractions was found to be excellent with relative standard deviations of <2% for consecutive extractions.  相似文献

Acute toxicity screening tests were conducted with water soluble fractions (WSFs) of a solvent refined coal (SRC-II) liquid from a pilot plant and three reference organisms: the cladoceran Daphnia magna, the fathead (FH) minnow Pimephales promelas, and larvae of the midge fly Chironomus tentans. Stock WSFs typically contained 900 to 1100 mg l?1 total carbon (TC) and 700 to 800 mg l?1 total dye complexable phenolics, with lower concentrations of aromatic and saturate hydrocarbons and N compounds. Under standard test conditions (temperature 20 °C, pH 7.3 to 8.2 and hardness 65 to 80 mg l?1 CaCO3), mean LC50 values in mg l? TC were 3.3 for daphnia, 11.l for FH minnow, and 13.7 for midge larvae. Acute toxicity was also examined under other water quality conditions (temperature 10 or 25 °C, pH 6.0 or 6.5, and hardness ? 180 to 220 mg 1?1 CaCO3). The coal liquid was less toxic to daphnids at 10 °C than at 20 or 25 °C, but response of other organisms at different temperatures varied. The pH of the liquid had little effect on toxicity values. All organisms were less susceptible in hard water. Chemical compositions of stock WSFs were similar, suggesting that temperature, pH, and hardness had little effect on solubility of major synfucl components. Dilution indexes for stock WSFs were higher than for petroleum oils, and reflect the greater solubility of chemicals from the liquified coal in freshwater.  相似文献
