In wheat, the ability to store and remobilise large amounts of stem water soluble carbohydrates (WSC) to grain constitutes a desirable trait to incorporate into germplasm targeted to regions with frequent terminal drought. The main aim of this paper was to examine the relationships between WSC storage, grain number and grain weight across several environments. A small set of recombinant inbred lines (2–4) contrasting in stem WSC were grown in six field trials where water availability, sowing date and/or N level were manipulated, with line yields ranging from 400 to 850 g m−2 across experiments. Biomass, N and WSC concentration (WSCc, mg g−1 dry weight) and amount (WSCa, g m−2) were monitored. A resource-oriented area-based model [Fischer, R.A., 1984. Growth and yield of wheat. The objectives of this study were to establish a protocol for the isolation of metabolically viable ventricular cardiomyocytes from the rainbow trout and to determine which measures may best reflect viability 24 h after isolation. Cardiomyocytes were isolated by enzymatic digestion and maintained in cell suspension. Viability was assessed using Trypan blue dye exclusion, ATP content, oxygen consumption and lactate dehydrogenase (LDH) leakage into the medium. Viability, assessed by these measures did not significantly change over the time period of this study. ATP content did correlate significantly with oxygen consumption but not with Trypan blue exclusion. We conclude that primary cultured cardiomyocytes remain metabolically viable for at least 24 h after isolation. Also, it appears that ATP content and oxygen consumption most adequately reflect metabolic cell viability. To be confident with a culture, however, a combination of viability measures is necessary when isolating cardiomyocytes from fish.  相似文献

First decade findings on the impacts of organic matter removal and soil compaction are reported for the 26 oldest installations in the nation-wide network of long-term soil productivity sites. Complete removal of surface organic matter led to declines in soil C concentration to 20 cm depth and to reduced nutrient availability. The effect is attributed mainly to the loss of the forest floor. Soil C storage seemed undiminished, but could be explained by bulk density changes following disturbance and to decomposition inputs of organic C from roots remaining from the harvested forest. Biomass removal during harvesting had no influence on forest growth through 10 years. Soil compaction effects depended upon initial bulk density. Soils with densities greater than 1.4 Mg m−3 resisted compaction. Density recovery was slow, particularly on soils with frigid temperature regimes. Forest productivity response to soil compaction depended both on soil texture and the degree of understory competition. Production declined on compacted clay soils, increased on sands, and generally was unaffected if an understory was absent.  相似文献

Embryogenic cultures of loblolly pine (Pinus taeda L.), slash pine (Pinus elliottii Engelm.), longleaf pine (Pinus palustris Mill.) and slash pine x longleaf pine hybrids were initiated from immature seeds on an initiation medium containing 13.57 microM 2,4-dichlorophenoxyacetic acid and 2.22 microM benzylaminopurine. Embryogenic cultures proliferated and somatic embryos developed, matured and germinated following a modified protocol and media originally developed for radiata pine (Pinus radiata D. Don.) somatic seedling production. A discrete, light-sensitive pre-germination stage and a later germination (radicle emergence) stage were identified by the differential response of somatic embryos to light of different wavelengths. Different light quality treatments were applied during the pre-germination and germination steps, using cool white fluorescent bulbs or light-emitting diodes (LEDs), or both. In general, red wavelengths provided by LEDs during these steps resulted in higher frequencies of somatic embryo germination (up to 64%) and conversion (up to 50%), longer tap roots and more first-order lateral roots than the standard cool white fluorescent treatments or treatment with blue wavelengths from LEDs. In addition, exposure to red light allowed germination of somatic embryos of some clones that failed to produce germinants under fluorescent light. Germination and conversion were further enhanced by sequential application of cool white fluorescent light and red light, resulting in up to 100% germination and conversion in one experiment. Longleaf pine somatic embryos were especially responsive to the light quality treatments, resulting in the first report of somatic seedling production for this species.  相似文献

Invasive plants are one of the greatest threats to endangered insect species and a major threat to Lepidoptera in eastern North America. We investigated the effects of the invasive shrub Chinese privet (Ligustrum sinense) and two methods (mulching or hand-felling) of removing it from riparian forests on butterfly communities and compared them to untreated, heavily invaded control plots and to “desired future condition” forests that never had extensive privet cover. Privet mulching resulted in nearly twice as many butterflies as privet felling and both treatments had more butterflies two years after privet removal than untreated control plots. Butterfly communities on control plots differed from those on the two treatments and the desired future condition forests. A number of forest characteristics were evaluated but only herbaceous plant cover (excluding privet) was positively correlated with butterfly abundance, diversity and evenness. The Carolina satyr, Hermeuptychia sosybius, was the best indicator of forests where privet had never invaded. Removing Chinese privet from riparian forests in the southeastern United States greatly improved forest habitats for butterflies and evidence suggests that butterfly communities in other temperate forests could benefit from removal of extensive shrub layers dominated by a single species.  相似文献

ABSTRACT

Our objective was to produce a unique feed ingredient from underutilized walleye pollock (Theragra chalcogramma) and pink salmon (Oncorhynchus gorbuscha) testes. Protein content in meals from both species (72% and 80%, respectively) were above the values found in high quality herring meals (~70%), but both were poor in some essential amino acids, e.g., methionine. Additionally, both were good sources of the amino acid taurine (1.7 and 2.2% of meal, respectively). Pollock meal was very rich in phospholipids (82% of total lipids) and in DHA (28 mg/g meal) and EPA (18 mg/g meal), indicating potential as an ingredient in larval starter diets. The purine contents in both pollock and salmon testes meals were more than 10 times the concentrations found in other fish byproducts or commercial fishmeals. The high concentrations of purines found in these testes, especially in the salmon meal, make it an ideal candidate for an immune system stimulant when added to dietary formulations.  相似文献

Balanced plant nutrition is essential to achieve high yields of canola (Brassica napus L.) and get the best economic return from applied fertilizers. A field study was conducted at nine site‐years across eastern Canada to investigate the effects of nitrogen (N), sulfur (S) and boron (B) fertilization on canola nutrient uptake, nutrient balance, and their relationship to canola yields. The factorial experiment consisted of four N rates of 0 (N0), 50 (N50), 100 (N100), and 150 (N150) kg ha?1, two S rates of 0 (S0) and 20 (S20) kg ha?1, and three B treatments of 0 (B0), 2 kg ha?1 at preplant (B2.0P), and 0.5 kg B ha?1 foliar‐applied at early flowering stage (B0.5F). Each site‐year used the same experimental design and assigned treatments in a randomized complete block design with four replications. Fertilizer S application greatly improved seed yields at six out of nine site‐years, and the highest N use efficiency was in the N150+S20 treatment. Sulfur application generally increased seed S concentration, seed S removal, and plant total S uptake, while B fertilization mainly elevated straw B concentration and content, with minimal effect on seed yields. At the early flowering stage, plant tissue S ranged from 2.2 to 6.6 mg S g?1, but the N : S ratio was over or close to the critical value of 12 in the N150+S0 combination at five site‐years. On average across nine site‐years, canola reached a plateau yield of 3580 kg ha?1 when plants contained 197 kg N ha?1, 33 kg S ha?1 and 200 g B ha?1, with a seed B content of 60 g B ha?1. The critical N, S, and B values identified in this work and their potential for a posteriori nutrient diagnosis of canola should be useful to validate fertilizer requirements for canola production in eastern Canada.  相似文献

The influence of two experimental soil treatments, Z93 and W91, on nitrogen transformations, microbial activity and plant growth was investigated in soil microcosms. These compounds are commercially marketed fermentation products (Agspectrum) that are sold to be added to field soils in small amounts to promote nitrogen and other nutrient uptake by crops in USA. In laboratory microcosm experiments, soils were amended with finely ground alfalfa-leaves or wheat straw, or left unamended, in an attempt to alter patterns of soil nitrogen mineralization and immobilization. Soils were treated in the microcosms with Z93 and W91 at rates equivalent to the recommended field application rates, that range from 0.2 to 1.1 l ha−1, (0.005-0.03 μl g−1 soil). We measured their effects on soil microbial activity (substrate-induced respiration (SIR), dehydrogenase activity (DHA) and acid phosphatase activity (PHOS)), soil nitrogen pools (microbial biomass N, mineral N, dissolved organic N), and transformations (net N mineralization and nitrification, 15N dilution of the mineral N pool, and accumulation of mineral N on ion-exchange resins), and on wheat plant germination and growth (shoot and root biomass, shoot length, N uptake and 15N enrichment of shoot tissues), for up to 56 days after treatment. To follow the movement of nitrogen from inorganic fertilizer into plant biomass we used a 15N isotopic tracer. Most of the soil and plant responses to treatment with Z93 or W91 differed according to the type of organic amendment that was used. Soil treatment with either Z93 or W91 influenced phosphatase activity strongly but did not have much effect on SIR or DHA. Both chemicals altered the rates of decomposition and mineralization of organic materials in the soil, which was evidenced by significant increases in the rates of the decomposition of buried wheat straw, and by the acceleration of net, rates of N mineralization, relative to those of the controls. Soil nitrate availability increased at the end of the experiment in response to both chemical treatments. In alfalfa-amended soils, the final plant biomass was decreased significantly by treatment with W91. Increased plant growth and N-use efficiency in straw-amended soil, resulting from treatments with Z93 or W91, was linked to increased rates of N mineralization from indigenous soil organic materials. This supports the marketing of these compounds as promoters of N uptake at these low dosage inputs.  相似文献
