Sugi (Cryptomeria japonica D. Don) produces secondary metabolite norlignans in xylem. Several norlignans are involved in the coloration of heartwood and defense of sapwood against microbial invasion. Their biosynthetic process should be well understood so that their properties can be exploited to improve the quality and utility of C. japonica wood. Unfortunately, information on the norlignan biosynthesis is limited because norlignans are mainly synthesized in a particular season in the transition zone (TZ) along with the heartwood formation, and is difficult to study. Although the generation of two norlignans of C. japonica, agatharesinol and (E)-hinokiresinol, has been reported, systems for producing other norlignans are not yet known. To establish a novel norlignan generating system, we examined the changes occurring in norlignans in a TZ during the process of drying a C. japonica log. On the day of felling, the TZ contained agatharesinol and (E)-hinokiresinol, which increased until they reached a maximum on day 40 after felling. Sequirin-C appeared on day 40 and increased to day 70. The generation of sequirin-C in the TZ can be used to investigate the biosynthetic process of heartwood norlignans. This study describes for the first time the changes that occur in the composition of norlignan during the drying of the TZ.  相似文献

The current expansion of the oil palm (Elaeis guineensis Jacq.) in the Brazilian Amazon has mainly occurred within smallholder agricultural and degraded areas. Under the social and environmental scenarios associated with these areas, oil palm-based agroforestry systems represent a potentially sustainable method of expanding the crop. The capacity of such systems to store carbon (C) in the soil is an important ecosystem service that is currently not well understood. Here, we quantified the spatial variation of soil C stocks in young (2.5-year-old) oil palm-based agroforestry systems with contrasting species diversity (high vs. low); both systems were compared with a ~10-year-old forest regrowth site and a 9-year-old traditional agroforestry system. The oil palm-based agroforestry system consisted of series of double rows of oil palm and strips of various herbaceous, shrub, and tree species. The mean (±standard error) soil C stocks at 0–50 cm depth were significantly higher in the low (91.8 ± 3.1 Mg C ha?1) and high (87.6 ± 3.3 Mg C ha?1) species diversity oil palm-based agroforestry systems than in the forest regrowth (71.0 ± 2.4 Mg C ha?1) and traditional agroforestry (68.4 ± 4.9 Mg C ha?1) sites. In general, no clear spatial pattern of soil C stocks could be identified in the oil palm-based agroforestry systems. The significant difference in soil carbon between the oil palm area (under oil palm: 12.7 ± 2.3 Mg C ha?1 and between oil palm: 10.6 ± 0.5 Mg C ha?1) and the strip area (17.0 ± 1.4 Mg C ha?1) at 0–5 cm depth very likely reflects the high input of organic fertilizer in the strip area of the high species diversity oil palm-based agroforestry system treatment. Overall, our results indicate a high level of early net accumulation of soil C in the oil palm-based agroforestry systems (6.6–8.3 Mg C ha?1 year?1) that likely reflects the combination of fire-free land preparation, organic fertilization, and the input of plant residues from pruning and weeding.  相似文献

Summary The effects of the addition of sulfurous acid into culture solution and of cold treatment of the solution were examined to simplify the culture of detached wheat ears. In the simplified method, detached ears could be cultured at room temperature on the liquid medium containing 100 g/l sucrose and 0.075% sulfurous acid without any sterilization. The immature seeds in detached ears cultured by this method were treated with low temperature or with chemicals known to have vernalizing effect. The chemical treatment did not affect the chilling requirement of immature embryos, although photoperiodic response and narrow-sense earliness were reduced by kinetin and trypsin. The low temperature treatment drastically affected the chilling requirement, and fully vernalized mature seeds having normal germinability were obtained by treating the detached ears in culture with low temperature from 10 days after anthesis.  相似文献

Recent intensive grazing in Mongolia may be significantly reducing the infiltration rate of rangeland. This study measured infiltration rates using simulated rainfall with high raindrop impact for small plots established on steppe grassland, desert grassland, and shrubland sites in Mongolia. The response of the infiltration rate to short-term livestock removal was also investigated. On the steppe grassland, a high infiltration rate was measured on an ungrazed plot with relatively dense vegetation cover; a statistically significant correlation was found between the total surface cover and final infiltration rate, indicating that surface cover by rock fragments also increased the infiltration rate to some extent. For desert grassland and shrubland, however, the surface cover condition was not a major factor controlling the final infiltration rate. After 4 years of livestock removal, the surface vegetation cover of the ungrazed plot was greater than that of the grazed plot, but no appreciable change occurred in soil penetration resistance. These results suggest that the high infiltration rate on the ungrazed plot was maintained mainly by the recovery of surface vegetation cover after the short-term livestock removal; this may indicate a potential mechanism of recovery from desertification processes for Mongolian rangeland.  相似文献

Epicotyl length (ECL) of adzuki bean (Vigna angularis) affects the efficiency of mechanized weeding and harvest. The present study investigated the genetic factors controlling ECL. An F2 population derived from a cross between the breeding line ‘Tokei1121’ (T1121, long epicotyls) and the cultivar ‘Erimo167’ (common epicotyls) was phenotyped for ECL and genotyped using simple sequence repeats (SSRs) and single-nucleotide polymorphism (SNP) markers. A molecular linkage map was generated and fifty-two segregating markers, including 27 SSRs and 25 SNPs, were located on seven linkage groups (LGs) at a LOD threshold value of 3.0. Four quantitative trait loci (QTLs) for ECL, with LOD scores of 4.0, 3.4, 4.8 and 6.4, were identified on LGs 2, 4, 7 and 10, respectively; together, these four QTLs accounted for 49.3% of the phenotypic variance. The segregation patterns observed in F5 residual heterozygous lines at qECL10 revealed that a single recessive gene derived from T1121 contributed to the longer ECL phenotype. Using five insertion and deletion markers, this gene was fine mapped to a ~255 kb region near the end of LG10. These findings will facilitate marker-assisted selection for breeding in the adzuki bean and contribute to an understanding of the mechanisms associated with epicotyl elongation.  相似文献

The heat-induced denaturation curve of ovalbumin followed by the ellipticity at 222 nm in circular dichroism spectra was consistent with that monitored by fluorescence with thioflavin T, which is an indication of amyloid fibril formation, while other proteins such as lysozyme and ovotransferrin did not fluoresce with thioflavin T during heat denaturation. The amount of soluble aggregate formed during heat denaturation was proportional to the increase in fluorescence with thioflavin T. The binding of soluble aggregates with thioflavin T was greatly suppressed in heat-denatured ovalbumin in the presence of thioflavin T. The similar inhibition effect of thioflavin T on the gel formation of heat-induced ovalbumin was observed. These results suggest that the amyloidogenic intermolecular beta-structure is involved in the formation of soluble aggregate and gel of heat-induced ovalbumin.  相似文献

ABSTRACT

The exploitation of phosphate mines generates an important quantity of phosphate sludge that remains accumulated and not valorized. In this context, composting with organic matter and rhizospheric microorganisms offers an interesting alternative and that is more sustainable for agriculture. This work aims to investigate the synergetic effect of arbuscular mycorrhizal fungi (AMF), phosphate-solubilizing bacteria (PSB) and phospho-compost (PC), produced from phosphate-laundered sludge and organic wastes, and their combination on plant growth, phosphorus solubilization and phosphatase activities (alkaline and acid). Inoculated mycorrhizae and bacteria strains used in this study were selected from plant rhizosphere grown on phosphate-laundered sludge. Significant (p < .05) increases in plant growth was observed when inoculated with both consortia and PC (PC+ PSB+ AMF) similar to those recorded in plants amended with chemical fertilizer. Tripartite inoculated tomato had a significantly (p < .05) higher shoot height; shoot and root dry weight, root colonization and available P content, than the control. Co-inoculation with PC and AMF greatly increased alkaline phosphatase activity and the rate of mycorrhizal intensity. We conclude that PC and endophytic AMF and PSB consortia contribute to a tripartite inoculation in tomato seedlings and are coordinately involved in plant growth and phosphorus solubilization. These results open up promising prospects for using formulate phospho-compost enriched with phosphorus-solubilizing microorganisms (PSM) in crop cultivation as biofertilizers to solve problems of phosphate-laundered sludge accumulation.  相似文献

Abstract

Biochar application has been recognized as an effective option for promoting carbon (C) sequestration, but it may also affect the production and consumption of methane (CH4) and nitrous oxide (N2O) in soil. A 1-year field experiment was conducted to investigate the effects of rice husk charcoal application on rice (Oryza sativa L.) productivity and the balance of greenhouse gas exchanges in an Andosol paddy field. The experiment compared the treatments of rice husk charcoal applied at 10, 20 and 40 Mg ha?1 (RC10, RC20 and RC40, respectively), rice husk applied at 20 Mg ha?1 (RH20), and the control (CONT). Rice straw and grain yields did not significantly differ among the treatments. The seasonal cumulative CH4 emissions were 38–47% higher from RC10, RC20 and RC40 than from the CONT. However, the increases were not in proportion to the application rates of rice husk charcoal, and their values did not significantly differ from the CONT. On the contrary, the RH20 treatment significantly increased the cumulative CH4 emission by 227% compared to the CONT. The N2O emissions during the measurement were not affected by the treatments. As a result, the combined global warming potential (GWP) of CH4 and N2O emissions was significantly higher in RH20 than in the other treatments. There was a positive linear correlation between C storage in the top 10 cm of soil and the application rate of rice husk charcoal. The increases in soil C contents compared to the CONT corresponded to 98–149% of the C amounts added as rice husk charcoal and 41% of the C added as rice husk. Carbon dioxide (CO2) fluxes in the off season were not significantly different among RC10, RC20, RC40 and CONT, indicating that C added as rice husk charcoal remained in the soil during the fallow period. The CO2 equivalent balance between soil C sequestration and the combined GWP indicates that the rice husk charcoal treatments stored more C in soil than the CONT, whereas the RH20 emitted more C than the CONT. These results suggest that rice husk charcoal application will contribute to mitigating global warming without sacrificing rice yields.  相似文献

Three apple (Malus×domestica) cultivars and 11 Malus accessions have been investigated by the probe hybridization method on their mitochondrial DNA (mtDNA). The gene probes used were: coxI, coxII, atpA, atp6, and atp9. Our results revealed enough variation to characterize ten mtDNA haplotypes among the Malus genotypes examined. The taxonomic and phylogenetic implications of mtDNA polymorphism are also discussed.  相似文献

Seventy-two accessions covering six varieties of Cucumis melo were characterized by using 35 morphological characters with emphasis on shelf-life, and the relationships between shelf-life and related characters was investigated. Principal component analysis (PCA) revealed that development period of plant and fruit, size of seed and fruit, shelf-life, stem hair, flesh juiciness, netting, abscission of peduncle, rapid yellowing of epidermis at maturity, Brix value, and color of flesh and epidermis etc. were the principal characters to discriminate melon accessions examined in the present study. According to the scatter diagram, vars. acidulusand makuwa, both of which belong to the Oriental melon, are closely related because of their short growth duration, small seed, thin pericarp and poor shelf-life, while American cantaloupe (var.reticulatus) and European cantaloupe (var. cantalupensis) are rather closely related due to their climacteric fruit with orange flesh, slipped peduncle and rapid yellowing of epidermis at maturity, which is closely related with their shelf-life. PCA also indicated that var. saccharinus was closer to var. inodorus than to the other varieties, due to their requirement of long period for development, large size of seed and fruit, and half- or non-slipped peduncle. Shelf-life of melon fruit was significantly correlated with the following characters: quality of flesh, size of seed and fruit, abscission of peduncle, development periods of plant and fruit, rapid yellowing of epidermis at maturity, Brix value and color of flesh and epidermis. Accessions with good shelf-life were mostly found in vars. saccharinus and inodorus. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献
