Seoul National University (SNU) miniature pigs represent a closed colony with 24 founder pigs and a well preserved pedigree. Characterization using mRNA sequence analysis was conducted for 6 swine leukocyte antigen (SLA) loci in parental or founder pigs, and 17 defined alleles were detected. Based on these complete coding sequences, 17 sequence specific primers (SSPs) were designed for polymorphic sites. To validate the specificity of each allele SSP, the PCR‐SSP was conducted with defined allele clones as templates. PCR‐SSP was conducted with the hot start polymerase and touch‐down PCR. The parental or found SNU miniature pigs showed overall SLA class I and II heterozygotes. Using the established PCR‐SSP method, we conducted SLA typing for breeding stock including 2 pedigreed pigs and identified the novel SLA class II homozygote haplotye (DRA*0201, DRB1*0403, DQA*0102 and DQB1*0701) and 2 SLA homozygote pig lines: SLA class I Hp‐3.0 and class II Hp‐0.3, and SLA class I Hp‐2.0 and class II Hp‐0.2. We thought that our PCR‐SSP SLA typing method could be applicable for new SLA homozygote line establishment by assignment and scheduled breeding.  相似文献

Seoul National University (SNU) miniature pigs are originated from the Minnesota miniature pig. This study was conducted to investigate the maternal origin of SNU (Minnesota) miniature pigs and their phylogenetic relationships by analyzing the mitochondrial DNA (mtDNA) D‐loop (control region) sequence. Two mtDNA D‐loop sequences of the SNU miniature pigs were identified. On an unweighted pair‐group method with an arithmetic mean (UPGMA) phylogenetic tree analysis, the large white was the pig breed closest to the SNU miniature pig, and the pairwise distance analysis showed the same result. While mtDNA sequences of 4 pig breeds which were used to establish Minnesota miniature pig were not known, our result might be different from the history of the Minnesota miniature pig development. In conclusion, we thought that some haplotypes of the Minnesota miniature pig maternally were originated from the Large white pig, or that wild pigs had similar mtDNA sequences to the Large white pig, and all SNU miniature pigs were derived from this colony.  相似文献

Poly(ethylene 2,6-naphthalate)/multi-walled carbon nanotube (PEN/MWNT) nanocomposites are prepared by in situ condensation polymerization in the presence of various acid-treated MWNT (a-MWNT) contents and their morphology, rheological and mechanical properties are investigated as a function of the a-MWNT content. SEM image of a plasma-etched nanocomposite exhibits that a-MWNTs are dispersed well in the PEN matrix by forming an interconnected network structure. Accordingly, rheological properties such as complex viscosities and shear moduli of PEN/a-MWNT nanocomposites at the terminal region of low frequency are much higher than those of pure PEN. Glass transition temperatures of nanocomposites also increase with the increment of the a-MWNT content, which stems from the reduced chain mobility due to the specific interaction between a-MWNTs and PEN matrix. Dynamic and tensile mechanical properties of nanocomposites are also higher than those of pure PEN and they increase with the increment of the a-MWNT content. The highly improved mechanical properties of PEN/a-MWNT nanocomposites are explained to originate from the interconnected network structure of a-MWNTs in PEN matrix as well as the strong interfacial adhesion between a-MWNTs and PEN matrix.  相似文献

In this study, a three-dimensional (3D) poly(lactide-co-glycolide) (PLGA) microfibrous scaffold with high porosity (ca. 90 % porosity) was developed for evaluating its performance in tissue engineering application. A dope solution of PLGA/polyethylene oxide (PEO) blend was electrospun into a methanol coagulation bath for fabricating highly porous 3D PLGA scaffold and a salt leaching method was used for making interconnected pores of 100?C200 ??m size inside the scaffold. The morphological structure, pore size and porosity of the microfibrous scaffold were determined, and compared with twodimensional (2D) mat-type and 3D sponge-type of PLGA scaffold. Also, swelling ratio, water uptake and compressive strength were compared in order to elucidate the structure-property relationships of different types of the scaffolds, especially in a wet condition. As a result of scanning electron microscopy (SEM) observation, normal human dermal fibroblasts (nHDF) were migrated, attached, and proliferated well inside the 3D scaffold. MTT assay confirmed that the highly porous 3D PLGA microfibrous scaffold had superior cell adhesion and proliferation abilities due to fibrous structure of large specific surface area, and interconnected pore structure. Therefore, this high performance 3D PLGA scaffold can have a high potentiality for application in tissue engineering in comparison with conventional PLGA scaffolds.  相似文献

The effects of the combined treatment of drying and heat treatment using superheated steam (SHS) were studied relative to the changes of the major chemical components in larch wood. The green lumber was dried and heat-treated in SHS conditions of 250 °C and 0.5 MPa for 18 h, and the relative percentage contents of sugars, lignin, and extractives were investigated and compared with the relative percentage contents in the lumber heat-treated in hot air conditions of 250 °C and atmospheric pressure for 18 h. After both heat treatment methods, the relative percentage contents of xylan, mannan, galactan, and arabinan were greatly decreased, whereas that of the Klason lignin was increased, additionally that of glucan and extractives remained almost unchanged. Lignin may bind with furan compounds decomposed from hemicellulose following heat treatment, thus contributing to the increase in the apparent relative percentage contents of the Klason lignin. In addition, the condensate collected in the condenser after combined drying and heat treatment using SHS was investigated qualitatively and quantitatively by high-performance liquid chromatography (HPLC). A large amount of furfural and acetic acid decomposed from hemicellulose was detected and some sugar components composed of cellulose and hemicellulose were detected in the liquid condensate.  相似文献

Titanium oxide (TiO2) and zinc oxide (ZnO) composite structured nanoparticles were prepared by combining a sol-gel process and a solvothermal method. Titanium (IV) isoproxide (TTIP), used as a TiO2 precursor, was dissolved in a colloidal ZnO nanoparticle solution synthesized by the sol-gel method, and TiO2 was synthesized via solvothermal synthesis onto the ZnO nanoparticles. The effects of reaction conditions such as pH, reaction temperature, and reaction time on the morphology of the composite nanoparticles and the ultraviolet (UV) absorbance of their polymer composite films were investigated. The UV absorption of the poly(vinyl alcohol) (PVA) composite film with TiO2-coated ZnO nanoparticles was higher than that of the TiO2, ZnO, and ZnO-coated TiO2 composite films. The reaction pH was found to have the strongest influence on the UV absorbance of the PVA/(TiO2/ZnO) composite film. A pH of 7.0, reaction temperature of 250 °C, and reaction time of 24 h were the optimum conditions for UV absorption.  相似文献

Pup‐loaf bread was made with 10, 30, and 50% substitution of flour with wheat starch phosphate, a cross‐linked resistant starch (XL‐RS4), while maintaining flour protein level at 11.0% (14% mb) by adding vital wheat gluten. Bread with 30% replacement of flour with laboratory‐prepared XL‐RS4 gave a specific volume of 5.9 cm3/g compared with 6.3 g/cm3 for negative control bread (no added wheat starch), and its crumb was 53% more firm than the control bread after 1 day at 25°C, but 13% more firm after 7 days. Total dietary fiber (TDF) in one‐day‐old bread made with commercial XL‐RS4 at 30% flour substitution increased 3–4% (db) in the control to 19.2% (db) in the test bread, while the sum of slowly digestible starch (SDS) plus resistant starch (RS), determined by a modified Englyst method, increased from 24.3 to 41.8% (db). The reference amount (50 g, as‐is) of that test bread would provide 5.5 g of dietary fiber with 10% fewer calories than control bread. Sugar‐snap cookies were made at 30 and 50% flour replacement with laboratory‐prepared XL‐RS4, potato starch, high‐amylose (70%) corn starch, and commercial heat‐moisture‐treated high‐amylose (70%) corn starch. The shape of cookies was affected by the added starches except for XL‐RS4. The reference amount (30 g, as‐is) of cookies made with commercial XL‐RS4 at 30% flour replacement contained 4.3 g (db) TDF and 3.4 g (db) RS, whereas the negative control contained 0.4 g TDF and 0.6 g RS. The retention of TDF in the baked foods containing added XL‐RS4 was calculated to be >80% for bread and 100% for cookies, while the retention of RS was 35–54% for bread and 106–113% for cookies.  相似文献

In the current study, we investigated whether electroacupuncture (EA) can inhibit pathological reductions in neurogenesis. Zucker diabetic fatty (ZDF) rats at 7 weeks of age were anesthetized with zoletil, and sham-acupuncture or EA at the Zusanli (ST36) and Baihui (GV20) acupoints was administered once a day for 5 weeks. In the ZDF group that received sham-EA (ZDF-Sham group), the blood glucose level was significantly increased together with age as compared to the control littermates [Zucker lean control (ZLC) rat]. In contrast, proliferating cells and differentiated neuroblasts were significantly decreased in the ZDF-Sham group compared to the ZLC group. Although EA treatment decreased blood glucose levels, this was not statistically significant when compared to blood glucose levels changes in the ZDF-Sham group. However, proliferating cells and differentiated neuroblasts were significantly increased with EA in ZDF rats as compared to those in the ZDF-Sham group. Brain-derived neurotrophic factor (BDNF) levels were significantly decreased in hippocampal homogenates of ZDF-Sham group compared to those in the ZLC group. The EA treatment significantly increased the BDNF levels compared to those in the ZDF-Sham group, and BDNF levels in this group were similar to those in the ZLC group. These results suggest that EA at ST36 and GV20 can ameliorate the reductions in proliferating cells and differentiated neuroblasts in the dentate gyrus induced by type-2 diabetes without significantly reducing blood glucose levels with increasing BDNF levels.  相似文献

The objective of the present study was to investigate the effects of three different culture media on the development of canine somatic cell nuclear transfer (SCNT) embryos. Canine cloned embryos were cultured in modified synthetic oviductal fluid (mSOF), porcine zygote medium-3 (PZM-3), or G1/G2 sequential media. Our results showed that the G1/G2 media yielded significantly higher morula and blastocyst development in canine SCNT embryos (26.1% and 7.8%, respectively) compared to PZM-3 (8.5% and 0%) or mSOF (2.3% and 0%) media. In conclusion, this study suggests that blastocysts can be produced more efficiently using G1/G2 media to culture canine SCNT embryos.  相似文献
