In alginate-assimilating bacteria, alginate is depolymerized to unsaturated monosaccharide by the actions of endolytic and exolytic alginate lyases (EC 4.2.2.3 and EC 4.2.2.11). The monosaccharide is non-enzymatically converted to 4-deoxy-l-erythro-5-hexoseulose uronic acid (DEH), then reduced to 2-keto-3-deoxy-d-gluconate (KDG) by a specific reductase, and metabolized through the Entner–Doudoroff pathway. Recently, the NADPH-dependent reductase A1-R that belongs to short-chain dehydrogenases/reductases (SDR) superfamily was identified as the DEH-reductase in Sphingomonas sp. A1. We have subsequently noticed that an SDR-like enzyme gene, flred, occurred in the genome of an alginolytic bacterium Flavobacterium sp. strain UMI-01. In the present study, we report on the deduced amino-acid sequence of flred and DEH-reducing activity of recombinant FlRed. The deduced amino-acid sequence of flred comprised 254 residues and showed 34% amino-acid identities to that of A1-R from Sphingomonas sp. A1 and 80%–88% to those of SDR-like enzymes from several alginolytic bacteria. Common sequence motifs of SDR-superfamily enzymes, e.g., the catalytic tetrad Asn-Lys-Tyr-Ser and the cofactor-binding sequence Thr-Gly-x-x-x-Gly-x-Gly in Rossmann fold, were completely conserved in FlRed. On the other hand, an Arg residue that determined the NADPH-specificity of Sphingomonas A1-R was replaced by Glu in FlRed. Thus, we investigated cofactor-preference of FlRed using a recombinant enzyme. As a result, the recombinant FlRed (recFlRed) was found to show high specificity to NADH. recFlRed exhibited practically no activity toward variety of aldehyde, ketone, keto ester, keto acid and aldose substrates except for DEH. On the basis of these results, we conclude that FlRed is the NADH-dependent DEH-specific SDR of Flavobacterium sp. strain UMI-01. 相似文献

The flavor volatiles in three Japanese rice cultivars, Nihonbare, Koshihikari, and Akitakomachi, during cooking were directly extracted by using a modified headspace solid‐phase microextraction (SPME) method and analyzed by gas chromatography‐mass spectrometry (GC‐MS). A total of 46 components were identified, including aldehydes, ketones, alcohols, and heterocyclic compounds, as well as fatty acids and esters, phenolic compounds, hydrocarbons, etc. The amount of key odorant compounds increased with cooking, while the amount of low‐boiling volatiles decreased. The similarities and differences of the three rice cultivars were determined through a comparison of their volatile components. Nihonbare was characterized by a higher amount of indole but an absence of the chemical class of fatty acid esters. In contrast, both Koshihikari and Akitakomachi had a higher amount of 4‐vinylphenol and an abundance of those esters. Koshihikari and Akitakomachi were quite similar in regard to those flavor volatiles. Furthermore, the observations in the research may suggest that the volatile components at cooking stage (I) were the representatives of the flavor volatiles of uncooked rice, while the volatile constituents at cooking stage (IV) were the representatives of the flavor volatiles of cooked rice. 相似文献

The present study aimed to clarify the effect of timing and type of supplementary grain in grazing dairy cows on herbage dry matter intake (HDMI), nitrogen utilization and milk production. Eight lactating cows were allowed to graze from evening to morning during three seasonal periods (spring, summer, autumn). They were randomly allocated to four treatments (timing: pre‐ (Pre) or post‐grazing (Post), for large grain allotments consisting of 75% of daily grain offered; grain type: barley or corn) in 4 × 4 Latin square designs in each period. In the spring period, HDMI was greater for cows fed corn than those fed barley (P = 0.005), whereas cows in the Pre treatment had a similar HDMI, higher (P = 0.049) urinary purine derivative concentration and greater (P = 0.004) milk yield compared with cows in the Post treatment. In the summer and autumn periods, timing treatments did not affect HDMI, nitrogen utilization or milk production, but cows supplemented with barley had higher urinary purine derivatives concentration (P < 0.05) and milk yield (P < 0.05) compared with those supplemented with corn. The results indicate that large grain allotments immediately before evening grazing during early grazing seasons increased ruminal microbial protein synthesis and milk production without reducing HDMI regardless of grain type. 相似文献

Reports of livestock infections with extended-spectrum beta-lactamase-producing Escherichia coli (ESBL-E) are increasing. Based on interviews conducted over a 6-month period, we found that veterinarians in the Vietnamese province of Thai Binh prefer to prescribe colistin-based drugs (CBD) in chicken farms. We aimed to clarify whether CBD use selects for strains of colistin-resistant ESBL-E. With the cooperation of seven local households, we detected ESBL-E in chickens’ feces after treating chickens with CBD. Phylogenetic groupings and the presence of CTX-M/AmpC genes were determined, and the multi-antibiotic susceptibility of isolates was analyzed. Our results showed that ESBL-E presented in seven chickens’ feces from two households. Seventy-two percent of ESBL-E isolates harbored CTX-M9 and the phylogenetic group A; the colistin minimum inhibitory concentration (MIC) of all isolated ESBL-E ranged from 0.064 to 1 μg mL?1. Moreover, ESBL-E isolates were used to experimentally select for colistin resistance, and the effect of commercial CBD on ESBL-E was investigated. The results showed that an ESBL-E strain with a colistin MIC of 4 μg mL?1 was able to grow in media with CBD. Although CBD treatment was effective, in vitro experiments demonstrated that ESBL-E can easily acquire colistin resistance. Australian Oryza are an understudied and underexploited genetic resource for rice improvement. Four species are indigenous: Oryza rufipogon, Oryza meridionalis, Oryza australiensis are widespread across northern Australia, whereas Oryza officinalis is known from two localities only. Molecular analysis of these wild populations is required to better define the distinctness of the taxa and the extent of any gene flow between them and rice. Limited collections of these wild populations are held in seed and DNA banks. These species have potential for domestication in some cases but also have many traits of potential value in the improvement of domesticated rice. Stress tolerance (biotic and abiotic) and grain quality characteristics in these populations may be useful. 相似文献

Genotypic variation in stomatal density and size has been reported but little is known of the genetic mechanisms behind these
leaf traits. Using 101 recombinant inbred lines derived from a cross between a tropical japonica, IR69093-41-3-2-2 and an
indica variety, IR72, we conducted a field study to determine stomatal density and size and identify quantitative trait loci
(QTL) controlling these traits under lowland conditions. Ten QTLs for stomatal density and four QTLs for size were identified
across growth stages and leaf surfaces (adaxial and abaxial). The contribution of each QTL to total phenotypic variation ranged
from 9.3 to 15.2% for stomatal density and 9.7 to 14.3% for size. The allele from IR72 increased stomatal density and that
from IR69093-41-3-2-2 increased size. The expression of the QTLs for stomatal density and size differed by growth stage indicating
that these traits might be genetically controlled depending on growth stage or that each QTL had a different function by growth
stage. Significant negative genetic correlations between stomatal density and size at both vegetative (r = −0.308**) and heading (r = −0.484**) stages were observed but no common QTL for these traits was detected across growth stages and leaf surfaces.
These results indicate that the QTLs for density and size may neither be genetically linked nor pleiotropically controlled
and findings can be used as basis for selection at the leaf level on the balance of carbon and water uptake. Further study
is needed to fully understand the mechanism underlying the observed genetic association and to elucidate the function of the

Improving the antibacterial activity against Staphylococcus aureus of composite sheets containing wasted tea leaves by roasting 总被引:1,自引:0,他引:1
We used various kinds of wasted tea leaves to develop composite sheets with antibacterial properties. Antibacterial tests
showed that the number of viable bacterial cells for the sheet containing wasted green tea leaves was around 10:6–107 CFU/ml after 18 h culture compared to 108 CFU/ml for a tea-free sheet. This indicates that cell growth was signifi cantly
inhibited. For sheets containing other types of tea leaves (oolong, black, hojicha, and pu-erh), living cells were not found, indicating that these sheets had superior antibacterial effects. Living cells were also not
found in sheets containing wasted black tea leaves or roasted tea leaves at a concentration of 60% by weight after 6 h cultivation.
Therefore, roasting treatment of wasted green tea leaves was examined to improve the antibacterial activity of the sheet.
In particular, the focus was on structural conversion of catechins by heating. 相似文献

The relationship between 9,10-ketol-octadecadienoic acid (KODA), GAs and jasmonic acid (JA) and flower bud formation influenced by fruit load in apples (Malus domestica Borkh.) was investigated. The endogenous KODA and JA concentrations in apical buds in plants subjected to flower thinning treatment (FTT), under which all flowers were removed were higher than those in plants subjected to heavy crop treatment (HCT), under which the number of leaves per fruit was adjusted to 20 from 60 to 120 days after full bloom (DAFB). In contrast, the gibberellic acid concentrations [total of gibberellin A1 (GA1) and gibberellin A4 (GA4)] in FTT plants were low compared with those in HCT plants. The result suggests that KODA and JA in contrast to GAs may have opposite effects on flower bud formation which is significantly influenced by fruit load in apples and that KODA and JA may also be associated with flower bud formation in apples. 相似文献
