The aim of this work was to evaluate the chemical properties of the residues from erva-mate processing and also to determine the candy-making performance with addition of residues from erva-mate on consumers' acceptance and purchase intent of this new product. The candies containing different amounts of mate powder were evaluated through overall acceptability test and purchase intent. Mate powder showed high contents of dietary fiber, total ash, and total polyphenols. The total dietary fiber content of the mate candies ranged from 5.7 to 6.29% on a dry matter basis. Supplementation with mate powder caused significant increases in polyphenol and mineral contents of mate candies. The incorporation of mate powder increased the hardness of the candies and produced desirable results in their nutritional characteristics. The sensory tests indicated that mate candies were acceptable and approved in relation to purchase intent. 相似文献

Criteria for durum wheat quality are continuously evolving in response to market pressure and consumer's preference. Specific attributes of durum wheat for different end products require more rapid and objective means to grade and classify wheat parcels based on processing potential. A total of 10 durum wheat cultivars were compared for compositional, protein, and starch characteristics. Mean values for the gross composition differed for total protein, gluten, and starch. Two-dimensional electrophoresis (2DE) analysis showed the proteome diversity among the cultivars. As shown by the principal component analysis (PCA) applied to 2DE data of gliadin and glutenin fractions, cultivars differed mainly from the number of proteins and levels of protein expression. As determined by the rapid viscoanalyzer (RVA), swelling power, starch damage, amylose content, and starch pasting properties of 10 cultivars differed significantly. 2DE fingerprinting and amylose content seemed to distinguish specific cultivars being useful tools for selecting suitable durum wheat cultivars for pasta making. 相似文献

The Mediterranean diet reduces the risk of coronary artery disease as a consequence of its high content of antioxidants, namely, hydroxytyrosol (HT) and oleuropein aglycone (OleA), typical of virgin olive oil. Because intercellular and vascular cell adhesion molecules (ICAM-1 and VCAM-1) and E-selectin are crucial for endothelial activation, the role of the phenolic extract from extra virgin olive oil (OPE), OleA, HT, and homovanillyl alcohol (HVA) on cell surface and mRNA expression in human umbilical vascular endothelial cells (HUVEC) was evaluated. OPE strongly reduced cell surface expression of ICAM-1 and VCAM-1 at concentrations physiologically relevant (IC50 < 1 microM), linked to a reduction in mRNA levels. OleA and HT were the main components responsible for these effects. HVA inhibited cell surface expression of all the adhesion molecules, whereas the effect on mRNA expression was weaker. These results supply new insights on the protective role of olive oil against vascular risk through the down-regulation of adhesion molecules involved in early atherogenesis. 相似文献

Farnesol is a sesquiterpene alcohol that modulates cell-to-cell communication in Candida albicans. In recent years, several studies have shown that this molecule presents inhibitory effects against non-albicans Candida species, Paracoccidioides brasiliensis and bacteria. The present study aimed at determining the effect of farnesol on the growth of strains of the Cryptococcus neoformans species complex, through microdilution assays. In addition, the effect of farnesol on the synthesis of phospholipase and protease - important virulence-associated enzymes - by C. neoformans and Cryptococcus gattii was also investigated. A total of 36 strains were studied, out of which 20 were from veterinary sources, 8 were from human cases and 8 were from a reference collection. The minimum inhibitory concentrations (MICs) were determined in accordance with the M27-A3 protocol as described by the CLSI and farnesol was tested at a concentration range of 0.29-150μM. Phospholipase and protease activities were evaluated through growth on egg yolk agar and spectrophotometry, respectively, after pre-incubating the strains at different farnesol concentrations (MIC/4, MIC/2 and MIC). It was observed that farnesol presents an inhibitory activity against C. neoformans and C. gattii (MIC range: 0.29-75.0μM). Although farnesol did not significantly alter phospholipase activity, a tendency to decrease this activity was observed. Concerning protease, no statistically significant differences were observed when comparing the production before and after pre-incubation at different farnesol concentrations. Based on these findings, it can be concluded that farnesol has in vitro inhibitory activity against C. neoformans and C. gattii, but has little impact on the production of the analyzed virulence factors. 相似文献

A pool of selected lactic acid bacteria was used to ferment suspensions of rye flour. Two-dimensional electrophoresis showed that 109 of the 129 ethanol-soluble rye polypeptides were hydrolysed almost totally by lactic acid bacteria. Matrix-assisted laser desorption ionization—time of flight mass spectrometry and reversed-phase high performance liquid chromatography analysis confirmed the hydrolysis of prolamins. After 48 h fermentation, no prolamin polypeptides were recognized by R5-Western analysis. HPLC analysis of glutelin polymers showed a very low bacterial proteolysis but a pH dependent hydrolysis probably due to activation of rye enzymes. Prolamins were extracted from rye flour and used to produce a peptic–tryptic (PT)-digest for in vitro tests with K 562 (S) sub-clone and Caco-2/TC7 cells of human origin. The PT-digest was also treated with lactic acid bacteria before assay. The Minimal Agglutinating Capacity increased ca. 8-times when K 562 (S) sub-clone cells were exposed to rye PT-digest treated with lactic acid bacteria. Hydrolysis of rye PT-digest by lactic acid bacteria decreased the toxicity of PT-digest itself towards Caco-2/TC7 cells as estimated by cell viability, caspase-3 activity and release of nitric oxide. Rye prolamins and glutelins were extracted from doughs and subjected to PT digestion. Compared to PT-digests from chemically acidified dough, coeliac jejunal biopsies exposed to the PT-digest from the dough fermented by lactic acid bacteria did not show an increase of the infiltration of CD3+ intraepithelial lymphocytes. The same was found for epithelial cell Fas expression. Long-time fermentation of dough by selected lactic acid bacteria could be considered as a potential tool to decrease the risk of rye contamination of gluten-free products for coeliac patients. 相似文献

Cnidarian toxins represent a rich source of biologically active compounds. Since they may act via oxidative stress events, the aim of the present study was to verify whether crude venom, extracted from the jellyfish Pelagia noctiluca, elicits inflammation and oxidative stress processes, known to be mediated by Reactive Oxygen Species (ROS) production, in rats. In a first set of experiments, the animals were injected with crude venom (at three different doses 6, 30 and 60 µg/kg, suspended in saline solution, i.v.) to test the mortality and possible blood pressure changes. In a second set of experiments, to confirm that Pelagia noctiluca crude venom enhances ROS formation and may contribute to the pathophysiology of inflammation, crude venom-injected animals (30 µg/kg) were also treated with tempol, a powerful antioxidant (100 mg/kg i.p., 30 and 60 min after crude venom). Administration of tempol after crude venom challenge, caused a significant reduction of each parameter related to inflammation. The potential effect of Pelagia noctiluca crude venom in the systemic inflammation process has been here demonstrated, adding novel information about its biological activity. 相似文献

The aim of this study was to investigate the presence of dermatophytes and yeasts in healthy and diseased dogs. A total of 633 samples were collected from 26 healthy animals (104 samples), 131 with dermatitis (343 samples), 74 with otitis (148 samples), and 19 with ocular diseases (38 samples). Cultures from healthy animals were positive for Malassezia pachydermatis in 13.5% (7/52) of samples from skin, 42.3% (11/26) from ear, and 3.8% (1/26) from eye. Fungal growth was observed in 20.4% (70/343) samples from animals with dermatitis. Microsporum canis was the most isolated fungus (n = 39), followed by M. pachydermatis (n = 30) and Malassezia sp. (n = 3). Of the 148 samples from dogs with otitis, 90 (60.8%) were positive for M. pachydermatis, and of the clinical specimens from the conjunctiva of animals with ophthalmic disease, 2.6% (1/38) presented positive cultures for M. pachydermatis. Only 14.3% (2/14) of the positive cultures for M. pachydermatis and 40.9% (9/22) of those for M. canis were positive in the direct exam. Direct exams were positive in 84.3% (70/83) of the culture positive samples from affected ears of dogs with otitis. Malassezia pachydermatis may act as an aggravating factor in the occurrence of cutaneous diseases, or the isolation of M. canis may be associated with the onset of dermatophytosis. Fungal culture, rather than microscopic examination, should be used as the definitive diagnostic test for dermatomycoses and otitis. 相似文献

- 1. Soft‐water, oligotrophic isoetid lakes are vulnerable to eutrophication, acidification and alkalinization. As a result of these pressures a large proportion have undergone substantial deterioration in several European countries. The understanding of these systems has been limited by either a lack of lakes close to natural conditions or through receiving less focus in broader scale macrophyte surveys. This has resulted in a dearth of information on specific lake types in their natural condition. Sixty‐eight soft‐water lakes in Ireland were studied in order to achieve a better understanding of the biological and environmental conditions defining such lakes.
- 2. Eight groups of lakes were identified using cluster analysis and indicator species analysis. Three groups were representative of isoetid‐rich lakes displaying a high frequency of occurrence of Isoetes lacustris, Lobelia dortmanna or Eriocaulon aquaticum together with the almost ubiquitous Littorella uniflora. Canonical variates analysis indicated that alkalinity, total phosphorus (TP), catchment area, altitude, moors and heathlands, mean transect depth, colour and lake area were significant variables discriminating among the eight groups. Soft‐water lakes with high amounts of isoetids tended to be less exposed, have broad shallow littoral zones with a high transparency and be situated in smaller catchments. Total phosphorus and alkalinity were typically low, although one group of isoetid‐rich lakes had higher mean TP and alkalinity values. This group may contain lakes under threat from nutrient enrichment and alkalinization and also lakes that have higher TP and alkalinity naturally.

Lakes with a prevalence of isoetids supported a greater diversity of macrophytes and chironomids indicating that such lakes may represent suitable conservation targets as they act as surrogates for soft‐water lakes of high biodiversity. Other factors favouring a focus on isoetids in conservation strategies are their importance as a functional component in soft‐water lakes and their sensitivity to lake and catchment environmental change.

Spirulina is a unicellular algae with good nutritional value. Its efficiency has been shown in Artemia salina and penaeid shrimp diets. To investigate the origin of this good nutritional value in shrimp, experiments were carried out on Penaeus japonicus juveniles. Pelleted diets containing various proportions of Spirulina from 0 to 8% were fed to shrimp. Good growth, survival and pigmentation were obtained with the diet containing 8% Spirulina. When replacing Spirulina completely by another single-cell ingredient source, lactic yeast, shrimp pigmentation was markedly reduced. Otherwise, growth performances were nearly similar. To identify the active fraction of Spirulina powder, an extraction was performed to separate two fractions, the lipidic and the lipid-free. Preliminary growth results of shrimp fed diets containing these extracted fractions showed that the active fraction of Spirulina was more likely found within the lipid-free fraction, rather than the lipidic fraction. Even though Spirulina can contribute positively to formulation of shrimp diets, it is too expensive (within the French economic context) to be incorporated at high percentage. But a substitution of lactic yeast for Spirulina is feasible as far as shrimp growth and survival are concerned. 相似文献
