Atlantic heaths are semi-natural habitats of high biodiversity interest which once covered large areas of the Atlantic Region. Nowadays these heathlands are dramatically reduced in many countries although they still cover wide areas in the north-west Iberian Peninsula, especially in the poorest and most socially marginal areas that are frequently affected by wildfires. We review the role of livestock grazing as a sustainable management strategy for heathlands in Europe. We have worked on a generalized conceptual framework for the management of a resource of nutritional and environmental value by drawing together evidence from studies of the livestock and the community ecology of grazed plants and the associated fauna. Key factors that influence grazing impact, such as type of livestock (animal species and breed) and their management are discussed. Goats thrive better than sheep, and horses than cattle, when heathland vegetation is the predominant resource available. Regardless of the type of livestock species managed, the low nutritive value of this vegetation hinders the maintenance of productive groups of suckler dams with offspring through the grazing season. The nutritional requirements of livestock can be met by adding improved pasture areas to heathlands. Under that strategy, sheep have the best productive performance and cattle the poorest. Management of mixed flocks with goats can lead to a more efficient use of vegetation, improve productivity and develop a patchier habitat which supports a richer associated fauna. Overall the results indicate that the sustainability of livestock grazing in these marginal lands will be achieved if they are managed effectively according to the available vegetation and their effects on the biodiversity.  相似文献

Objective

1) To determine the pharmacokinetics of tramadol hydrochloride and its active metabolite, O-desmethyltramadol (M1), after administration through different routes in female and male C57Bl/6 mice; 2) to evaluate the stability of tramadol solutions; and 3) to identify a suitable dose regimen for prospective clinical analgesia in B6 mice.

Study design

Prospective, randomized, blinded, parallel design.

Animals

A total of 18 male and 18 female C57Bl/6 mice (20–30 g).

Methods

Mice were administered 25 mg kg?1 tramadol as a bolus [intravenously (IV), intraperitoneally (IP), subcutaneously (SQ), orally per gavage (OSgavage)] over 25 hours [orally in drinking water (OSwater) or Syrspend SF (OSSyrsp)]. Venous blood was sampled at six predetermined time points over 4 to 31 hours, depending on administration route, to determine tramadol and M1 plasma concentrations (liquid chromatography and tandem mass spectrometry detection). Pharmacokinetic parameters were described using a noncompartmental model. The stability of tramadol in water (acidified and untreated) and Syrspend SF (0.20 mg mL?1) at ambient conditions for 1 week was evaluated.

Results

After all administration routes, Cmax was >100 ng mL?1 for tramadol and >40 ng mL?1 for M1 (reported analgesic ranges in man) followed by short half-lives (2–6 hours). The mean tramadol plasma concentration after self-administration remained >100 ng mL?1 throughout consumption time. M1 was found in the OSSyrs group only at 7 hours, whereas it was detectable in OSwater throughout administration. Tramadol had low oral bioavailability (26%). Short-lasting side effects were observed only after IV administration. Water and Syrspend SF solutions were stable for 1 week.

Conclusions and clinical relevance

1) At the dose administered, high plasma concentrations of tramadol and M1 were obtained, with half-life depending on the administration route. 2) Plasma levels were stable over self-consumption time. 3) Solutions were stable for 1 week at ambient conditions.  相似文献

Objective

To evaluate and compare the analgesic efficacy and adverse effects of dexketoprofen and methadone using a noninferiority trial, during the first 24 postoperative hours in dogs undergoing orthopaedic surgery.

Study design

Randomized, blinded clinical study.

Animals

A total of 38 healthy dogs undergoing orthopaedic surgery.

Methods

Dogs were premedicated with dexmedetomidine [1 μg kg–1 intravenously (IV)] followed by dexketoprofen (1 mg kg–1 IV; group DK) or methadone (0.2 mg kg–1 IV; group M). Anaesthesia was induced with propofol and maintained with isoflurane in 60% oxygen. Postoperatively, dexketoprofen was administered every 8 hours (group DK) and methadone every 4 hours (group M). Analgesia was assessed at baseline and at 1, 2, 4, 6, 18 and 24 hours after extubation using a dynamic and interactive visual analogue scale (DIVAS), the short form of the Glasgow Composite Measure Pain Scale (CMPS-SF), mechanical wound thresholds (MWTs) and plasma cortisol levels. If CMPS-SF score was ≥5, rescue analgesia was administered. Data were analysed using a general linear mixed model, Mann–Whitney U test and chi-squared test as appropriate; a p value <0.05 was considered significant.

Results

The CMPS-SF and DIVAS scores were significantly higher in group M compared with group DK and remained higher for a longer period in group M, although the differences were not clinically significant. No significant differences were found in MWT assessment between groups. Plasma cortisol level significantly increased 2 hours after extubation, without significant differences between treatments. Rescue analgesia was administered to three animals (one in group DK; two in group M).

Conclusion and clinical relevance

We conclude that 1 mg kg–1 IV dexketoprofen administered every 8 hours during the first 24 hours postoperatively is noninferior to methadone in controlling pain after orthopaedic surgery in dog, although frequent pain assessments are recommended to adjust the analgesia plan.  相似文献

A 180‐day experiment was carried out under controlled conditions to compare the effects of three commercial dry diets on survival, growth and body composition of juvenile Pacifastacus leniusculus from the onset of exogenous feeding (stage 2). Protein levels in the diets were 55.5% [high‐protein content (HP) diet], 40.3% [medium‐protein content (MP)] and 33.2% [low‐protein content (LP)]. Diets were supplemented with restricted amounts of live fresh Artemia nauplii up to day 120. No significant differences in survival among diets were found along the trial, with final values around 68.5%. During the live feed supplementation, growth was similar with the three diets. At the end of the trial, weight with HP or with MP was similar (around 2.32 g) and significantly higher than with LP (1.97 g). Thus, a diet with 40% of protein can be suitable, as it allowed similar results as the diet containing 55%. Levels of protein in diets and in crayfish muscle were positively correlated (r=0.8). This study also shows that an intensive rearing up to 180 days with good survival and growth rates is possible using dry diets for finfish species supplemented with live feed.  相似文献

A group of virulent Aeromonas hydrophila, A. sobria, and A. veronii biovar sobria strains isolated from humans and fish have been described; these strains classified to serotype O11 are serologically related by their lipopolysaccharide (LPS) O-antigen (O-polysaccharide), and the presence of an S-layer consisting of multiple copies of a crystalline surface array protein with a molecular weight of 52 kDa in the form of a crystalline surface array which lies peripheral to the cell wall. A. hydrophila strain AH-1 is one of them. We isolated the LPS from this strain and determined the structure of the O-polysaccharide, which was similar to that previously described for another strain of serotype O11. The genetics of the O11-antigen showed the genes (wbO11 cluster) in two sections separated by genes involved in biosynthesis and assembly of the S-layer. The O11-antigen LPS is an example of an ABC-2-transporter-dependent pathway for O-antigen heteropolysaccharide (disaccharide) assembly. The genes involved in the biosynthesis of the LPS core (waaO11 cluster) were also identified in three different chromosome regions being nearly identical to the ones described for A. hydrophila AH-3 (serotype O34). The genetic data and preliminary chemical analysis indicated that the LPS core for strain AH-1 is identical to the one for strain AH-3.  相似文献

ObjectiveTo compare the cardiorespiratory effects and quality of induction of and recovery from anaesthesia following etomidate or alphaxalone-HPCD IV.Study designRandomized ‘blinded’ cross-over study. Twenty-four hours was allowed between phases.AnimalsEight healthy adult Beagles (four male, four female).MethodsDogs were anaesthetized with sevoflurane for instrumentation, then allowed to awake. They then received etomidate (treatment E) or alphaxalone-HPCD (treatment A) intravenously to effect. Heart rate (HR), body temperature, invasive arterial pressures (AP), systemic vascular resistance index (SVRI), stroke volume index, cardiac index (CI), contractility, respiratory rate, central venous pressure, and capnometry were obtained before anaesthetic induction (baseline), 30 seconds and 1 minute after induction, after intubation, one minute after intubation, and for every 5 minutes afterwards until the dog began to swallow and the trachea was extubated. Arterial bloods were taken for analyses before induction, after intubation and every 10 minutes thereafter. The dogs breathed room air. The quality of induction of and recovery from anaesthesia were scored categorically. Statistical analyses used anova for repeated measures, paired t-tests or Wilcoxon signed rank-test as relevant. Significance was set at p < 0.05.ResultsThe induction doses required were (mean ± SD) 2.91 ± 0.41 mg kg?1 and 4.15 ± 0.7 mg kg?1 for treatment E and A respectively. No significant changes in cardiovascular parameters were observed with treatment E. Treatment A resulted in statistically significant increases in HR and CI and reductions of APs and SVRI. Time to extubation was longer with treatment A (25 ± 7 minutes) than with treatment E (17 ± 4 minutes). Dogs became hypoxic with both treatments. The quality of induction and recovery were excellent with treatment A, but significantly less satisfactory with treatment E (recovery score, treatment E median 1, range 0–2; treatment A median 0, range 0–1).Conclusions and clinical relevanceAlphaxalone-HPCD caused significant tachycardia and increase in CI, and statistically (but not clinically) significant decreases in APs and SVRI. Etomidate caused no statistically significant cardiovascular changes. Quality of recovery was better with alfaxalone-HPCD. Both agents caused short-lived hypoxia, and oxygen supplementation is advisable.  相似文献

This study investigates the applicability of on-line coupling of capillary electrophoresis with electrospray ionization tandem mass spectrometry (CZE-ESI-MS) for the separation and characterization of alpha- and beta-acids and oxidized hop acids from crude extracts of different hop varieties. CZE-ESI-MS with negative-ion electrospray ionization proved to be a suitable technique for the determination of these types of natural compounds and their oxidized derivatives. The CZE parameters (pH, concentration, and buffer type) and ESI-MS parameters (nature and flow rate of the sheath liquid, nebulizer pressure, drying gas flow rate, temperature, and compound stability) were optimized. The optimized method provides the potential for a fast qualitative determination of hop acids and their oxidation compounds. The method was also applied to the determination of iso-alpha-acids in beer.  相似文献
