Diet analysis of small mammal pests: A comparison of molecular and microhistological methods 下载免费PDF全文
Knowledge of what pest species are eating is important to determine their impact on stored food products and to plan management strategies accordingly. In this study, we investigated the food habits of 2 rodents, Rattus rattus (ship rat) and Mus musculus castaneus (house mouse) as well as an insectivore, Suncus murinus (shrew), present in human dwellings. Both a microhistological approach and a DNA barcoding approach were used in the present study. Following DNA extraction, amplification was performed using group‐specific primers targeting birds, plants and invertebrates. Resulting polymerase chain reaction products were sequenced and analyzed to identify the different prey species present in the gut contents. The findings from the application of both techniques were in agreement, but the detection of prey type with each technique was different. The DNA barcoding approach gave greater species‐level identification when compared to the microhistological method, especially for the invertebrate and avian prey. Overall, with both techniques, 23 prey taxa were identified in the gut contents of the 3 species, including 15 plants, 7 insects and a single bird species. We conclude that with a selection of suitable “barcode genes” and optimization of polymerase chain reaction protocols, DNA barcoding can provide more accurate and faster results. Prey detection from either technique alone can bias the dietary information. Hence, combining prey information of both microhistological analysis and DNA barcoding is recommended to study pest diet, especially if the pest is an omnivore or insectivore species. 相似文献

Interspecific comparisons of bioassay and biochemical data suggest two major patterns of target site resistance to carbamates and organophosphates. Pattern I resistance, which is generally more effective for carbamates, has been shown in two sub-species of mosquitoes to be due to a particular Gly-Ser mutation in the oxyanion hole within the active site of one of their two acetylcholinesterase enzymes. Intriguingly, different substitutions at the equivalent site confer organophosphate hydrolytic ability on other esterases responsible for metabolic resistance in some other species. In the case of the aphid, Myzus persicae, Pattern I resistance is due to a Ser-Phe mutation in the vicinity of the acyl pocket of acetylcholinesterase. Pattern II resistance is at least as effective for organophosphates as it is for carbamates and may even be specific to organophosphates in some cases. Molecular studies on this pattern of resistance in three higher Diptera show that it is due to changes that constrict the acetylcholinesterase active site gorge and limit binding of the insecticide to the catalytic residues at the base of the gorge. One case of Pattern II resistance in the mosquito, Culex tritaeniorhynchus, involves the same site near the acyl pocket of acetylcholinesterase, albeit a different substitution, as that involved in Pattern I resistance in M. persicae. 相似文献

program conducted between January 2005 and August 2008. A 51-question survey was conducted in 11 villages involved in the
Newcastle disease vaccination program in Chibuto, Mozambique. The mean flock size was significantly higher in households that
provided their chickens with feed (15.0) than chickens that only scavenged (8.7; P = 0.0001). The mean flock size was significantly higher in households with vaccinated chickens (16.9) than those with unvaccinated
chickens (10.0; P = 0.0005). The average number of chicks hatched during the most recent brooding was significantly higher in households that
fed their chickens (9.2) than chickens that only scavenge (6.9; P = 0.0335). The mean hatch rate was significantly higher in households with vaccinated chickens (0.8) than those with unvaccinated
chickens (0.7; P = 0.0324). It was determined that unvaccinated chickens are approximately five times more at risk to die of Newcastle disease
(odds ratio = 4.79). This study supported the efficacy of the I-2 Newcastle disease vaccine as shown by the increased average
flock size and decreased incidence of chicken mortality due to Newcastle disease. The level of farmer involvement and ongoing
commitment by community vaccinators suggest that the Chibuto vaccination program is likely to be sustainable in the medium-

White-nose syndrome (WNS) is a condition associated with an unprecedented bat mortality event in the northeastern United States. Since the winter of 2006*2007, bat declines exceeding 75% have been observed at surveyed hibernacula. Affected bats often present with visually striking white fungal growth on their muzzles, ears, and/or wing membranes. Direct microscopy and culture analyses demonstrated that the skin of WNS-affected bats is colonized by a psychrophilic fungus that is phylogenetically related to Geomyces spp. but with a conidial morphology distinct from characterized members of this genus. A fresh approach is presented to address the increasingly urgent need for alternative land management strategies in savannas. We illustrate how fine‐scale information of soil characteristics can be used to enable a more precise delimitation of sites suitable for different forms of land stewardship, including agro‐pastoral activities, forestry, biodiversity and tourism. By collating data from previous soil surveys, and augmenting this with targeted new surveys, we produce the first national data set of soil properties for the lowland savannas of Belize. Most of these soils are typical of savanna soils worldwide, that is acidic (mean surface pH = 5.7), nutrient‐poor (mean surface total exchangeable bases [TEB] = 3.4 cmol kg?1) and coarse‐textured (mean surface clay = 13.0%). Nevertheless, there is a marked spatial variability across the country in these soil properties. Some soils exhibit unusually high subsurface clay fractions (max = 73%), whilst other sites have exceptionally high pH in lower horizons (max = 8.4). Cluster analysis is used to group sites with similar soil properties. Across 79 sites, there is a clear division between soils with high clay percentages and those with coarser textures. These are sub‐divided into five groups based on further differences in parent material, chemical properties and site characteristics. Mapping the locations of these groups enables more specific land use recommendations to be made. This ability to make targeted land use recommendations from fine‐scale soil information represents an advance over the previous national land use policy, where all savanna lands were considered unsuitable for any form of agriculture. Well-being is increasingly being promoted and used to describe social progress. However, tension exists between framings that focus on enhancing individual well-being (living well) and societal or collective framings of well-being (living well together). Well-being is central to Aotearoa New Zealand's COVID-19 response and recovery. The COVID-19 pandemic reopened debates about what kind of society people want to live in. Our research explored the ‘shared typical’ or commonality of experiences of the first wave of COVID-19 response in Aotearoa New Zealand. Semi-structured interviews provided insights into a wide range of concerns participants faced and what that meant for their well-being and the well-being of Aotearoa New Zealand. We found that well-being is both multidimensional and hierarchical, and while people talked about their own well-being, it was often in the context of broader social well-being. These findings support research showing that well-being is relational. We suggest that Indigenous models of well-being are well placed to inform policy strategies enabling holistic well-being, but this needs to be done in ways that pair Indigenous and Western knowledge, rather than integrating or assimilating this knowledge into Western science approaches. 相似文献

This study describes the influence of 25% human serum albumin (HSA) supplementation on serum albumin level, total protein (TP), colloid osmotic pressure (COP), hospital stay, and survival in dogs with septic peritonitis. Records of 39 dogs with septic peritonitis were evaluated. In the HSA group, initial and post-transfusion TP, albumin, COP, and HSA dose were recorded. In the non-supplemented group, repeated values of TP, albumin, and COP were recorded over their hospitalization. Eighteen dogs survived (53.8% mortality). Repeat albumin values were higher in survivors (mean 23.9 g/L) and elevated repeat albumin values were associated with HSA supplementation. Repeat albumin and TP were higher in the HSA supplemented group (mean 24 g/L and 51.9 g/L, respectively) and their COP increased by 5.8 mmHg. Length of hospitalization was not affected. Twenty-five percent HSA increases albumin, TP, and COP in canine patients with septic peritonitis. Higher postoperative albumin levels are associated with survival. 相似文献

Summary A 0–4 scoring system to quantify scald (Rhynchosporium secalis) infection is suggested. Scores 1, 2, 3 and 4 allocated to represent 1/4, 1/2, 3/4 and 4/4 of the crop canopy scalded are easy to comprehend and intermediate scores e.g. 0.5, 1.5, 2.5 and 3.5 give it the breadth of a quantitative scale. Scores on a large number of lines showed a high degree of repeatability and were found to be highly correlated with the log transformed values of the actual leaf area damage. Although it was suggested that predictions of leaf area damage at scores 3–4 should be applied with caution, broad generalization of the scores in discriminating the amount of disease were shown to be soundly based and offered plant breeders a tool to standardize the evaluation of scald resistance in field plots on a large scale with this quick and reliable scoring system. 相似文献
