The prevalence of food allergies in the world population requires integrated approaches to identify new potential allergens, especially those of plant origin. The aim of this work was the allergen in vitro analysis of Lupinus albus seed proteome, a promising food protein source, and the assessment of IgE cross-reactivities with other more diffused legume species. A combination of one- and two-dimensional gel electrophoresis and immunoblotting analyses with specific IgGs for band identification and lupin-sensitized patients' circulating IgEs for allergenicity studies has been used. Two lupin proteins, namely, conglutin gamma and 11S globulin basic subunits, strongly reacted with all patients' sera. Also, cross-reactivities with the homologous polypeptides of other legume species were observed. Otherwise, no reaction at all was detected with a 2S-type lupin protein. This global electrophoretic approach has allowed the identification of a new potential lupin allergen and confirmed the cross-reactivity among the legume 11S globulin basic subunits.  相似文献

Aquatic macrophytes are well known accumulators for heavy metals, the reason why they are used as bioindicators for water quality and in phytoremediaton strategies. This study reports on the elemental concentrations in four free-floating aquatic macrophytes (S. auriculata; P. stratiotes; E. crassipes and E. azurea) growing in two water reservoirs (Santana e Vigário, Rio de Janeiro State, Brazil) of an electric power plant that receive input from the polluted Paraíba do Sul River. Filtered water samples and water suspended solids from these environments were also analysed. Inductively coupled plasma mass spectrometry was used as the principal method, allowing the determination of up to 41 elements, including the rare-earth elements (REEs) and other trace metals not assayed before in these macrophytes. The results show that all elements studied are accumulated by the macrophytes with concentration ratios (CR = [plant]: [water]) varying from about 1,000 to 200,000, based on the dry weight of the plant species. With a few exceptions, highest accumulations were observed in E. crassipes in which CRs increase in the sequence: Cu < Mo < Cr < Pb < Tl < Fe < La < Zn < Ce< Mn. Surprisingly high CRs (e.g. Ce: 74,000) and corresponding mass concentrations were observed for the rare-earth elements (e.g. ∑REE: 112 mg kg?1), also measured in the water suspended particle fraction. The results show that this fraction acts as an effective sink for trace metals in the aquatic system studied and seems to play also an important role in the transfer of metals from water to the plant species.  相似文献

Zebrafish has become a popular research model in the last years, and several diseases affecting zebrafish research facilities have been reported. However, only one case of naturally occurring viral infections was described for this species. In 2015, infectious spleen and kidney necrosis virus (ISKNV) was detected in zebrafish from a research facility in Spain. Affected fish showed lethargy, loss of appetite, abnormal swimming, distention of the coelomic cavity and, in the most severe cases, respiratory distress, pale gills and petechial haemorrhages at the base of fins. Cytomegaly was the most relevant histopathological finding in organs and tissues, sometimes associated to degenerative and necrotic changes. ISKNV belongs to the relatively newly defined genus Megalocytivirus, family Iridoviridae, comprising large, icosahedral cytoplasmic DNA viruses. This is the first case of naturally occurring Megalocytivirus infection in zebrafish research facilities, associated with morbidity. The virus has been identified based on both pathologic and genetic evidence, to better understand the pathogenesis of the infection in zebrafish and the phylogenetic relationship with other iridoviruses. Given the ability of megalocytiviruses to cross‐species boundaries, it seems necessary to implement stringent biosecurity practices as these infections may invalidate experimental data and have major impact on laboratory and cultured fish.  相似文献

Background

The major indication for antibiotic use in Danish pigs is treatment of intestinal diseases post weaning. Clinical decisions on antibiotic batch medication are often based on inspection of diarrhoeic pools on the pen floor. In some of these treated diarrhoea outbreaks, intestinal pathogens can only be demonstrated in a small number of pigs within the treated group (low pathogen diarrhoea). Termination of antibiotic batch medication in herds suffering from such diarrhoea could potentially reduce the consumption of antibiotics in the pig industry. The objective of the present pilot study was to suggest criteria for herd diagnosis of low pathogen diarrhoea in growing pigs.Data previously collected from 20 Danish herds were used to create a case series of clinical diarrhoea outbreaks normally subjected to antibiotic treatment. In the present study, these diarrhoea outbreaks were classified as low pathogen (<15% of the pigs having bacterial intestinal disease) (n =5 outbreaks) or high pathogen (≥15% of the pigs having bacterial intestinal disease) (n =15 outbreaks). Based on the case series, different diagnostic procedures were explored, and criteria for herd diagnosis of low pathogen diarrhoea were suggested. The effect of sampling variation was explored by simulation.

Results

The diagnostic procedure with the highest combined herd-level sensitivity and specificity was qPCR testing of a pooled sample containing 20 randomly selected faecal samples. The criteria for a positive test result (high pathogen diarrhoea outbreak) were an average of 1.5 diarrhoeic faecal pools on the floor of each pen in the room under investigation and a pathogenic bacterial load ≥35,000 per gram in the faecal pool tested by qPCR. The bacterial load was the sum of Lawsonia intracellularis, Brachyspira pilosicoli and Escherichia coli F4 and F18 bacteria per gram faeces. The herd-diagnostic performance was (herd-level) diagnostic sensitivity =0.99, diagnostic specificity =0.80, positive predictive value =0.94 and negative predictive value =0.96.

Conclusions

The pilot study suggests criteria for herd diagnosis of low pathogen diarrhoea in growing pigs. The suggested criteria should now be evaluated, and the effect of terminating antibiotic batch medication in herds identified as suffering from low pathogen diarrhoea should be explored.

Barley (Hordeum vulgare L.) grain germination is required to perform the malting process. Maintenance of barley seed viability during storage is crucial for the malt industry; and modern cultivars are bred for rapid grain dormancy release after physiological maturity. Low dormancy level combined with rain close to harvest induces pre-germination/pre-harvest sprouting damage. Pre-germination might not affect viability in the short term after harvest, but it could reduce potential longevity (Ki) of a barley seed lot. Ki value is inherent for each barley lot; however, its determination is time-consuming which precludes its assessment at an industrial scale. In this study we sought quantitative relationships between Ki and the pre-germination degree of barley grain lots, assessed through quality tests routinely performed by malthouses [Falling Number (FN), α-Amylase Activity and Carlsberg]. Field pre-germinated lots from one old barley cultivar (Quilmes Palomar) and artificially pre-germinated lots from major varieties currently grown in Argentina were used. Associations between Ki and values obtained from all quality tests analysed were found for Q. Palomar. However, FN was the parameter that yielded the best and simplest explanation of Ki variability. A significant positive linear Ki -FN relationship was also obtained for each modern barley cultivar.  相似文献
