Determination of the soil microbial biomass carbon using the method of substrate-induced respiration
Specific features of determining the carbon content in the soil microbial biomass using the method of substrate-induced respiration
(MBSIR) were studied as related to the conditions of the incubation (the glucose concentration and temperature) and pre-incubation
(the duration and temperature) of the soil samples collected in the summer (tundra gley and soddy-podzolic soils and chernozems)
and in different seasons (for the gray forest soil). The glucose concentration providing the highest substrate-induced respiration
(SIR) in the soils studied was shown to be 2–15 mg/g. The MBSIR in the soil samples collected in summer and in the soils pre-incubated for 10 and 22°C (7 days) did not significantly differ.
The MBSIR in the gray forest soil pre-incubated at 3, 6, and 10°C (winter, spring/autumn, and summer, respectively) and at 22°C (recommended
by the authors of the SIR method) was similar for the cropland in all the seasons. For the meadow, it was the same in the
winter, summer, and autumn, and, in summer, it did not differ only for the forest. For the comparative assessment of the MBSIR, soil samples from different ecosystems are recommended to be collected in the autumn or in the summer. Soil samples of 100–500
g should be pre-incubated for 7 days at 22°C and moisture of 60% of the total water capacity; then, 1-2 g soil should be incubated

Inhibitory effects of the Noble muscadine grape extracts and the representative phytochemicals for anthocyanins (i.e., cyanidin and cyanidin-3,5-diglucoside) on two enzymes, that is, α-glucosidase and pancreatic lipase, were investigated regarding their antidiabetic activities. The study demonstrated that the anthocyanin extracts and the selected chemicals obeyed the competitive mode against the enzymes. The methanolic extracts of whole fruit and skin of the muscadine showed inhibitory activities against the α-glucosidase with their IC(50) values at 1.50 and 2.73 mg/mL, and those against the lipase at 16.90 and 11.15 mg/mL, respectively, which indicated that the muscadine extracts possessed strong antidiabetic activities. Particularly, the ethyl acetate (EtoAc) extract and the butanol (BuOH) extract exhibited much higher inhibitory activities against both enzymes than the CHCl(3) and water extracts, while the majority of anthocyanins existed in the BuOH fractions. Moreover, cyanidin exhibited a much stronger antidiabetic activity than cyanidin-3,5-diglucoside, suggesting that anthocyanins may have higher inhibitory activities after being digested. Further chromatographic analysis by high-performance liquid chromatography-mass spectrometry identified five individual anthocyanins, including cyanidin, delphinidin, petunidin, peonidin, and malvidin glycosides. 相似文献

Austral bracken Pteridium esculentum contains three unstable norsesquiterpene glycosides: ptaquiloside, ptesculentoside, and caudatoside, in variable proportions. The concentration of each of the glycosides was determined in this study as their respective degradation products, pterosin B, pterosin G and pterosin A, by HPLC-UV analysis. Samples of P. esculentum collected from six sites in eastern Australia contained up to 17 mg of total glycoside/g DW, with both ptaquiloside and ptesculentoside present as major components accompanied by smaller amounts of caudatoside. Ratios of ptaquiloside to ptesculentoside varied from 1:3 to 4:3, but in all Australian samples ptesculentoside was a significant component. This profile differed substantially from that of P. esculentum from New Zealand, which contained only small amounts of both ptesculentoside and caudatoside, with ptaquiloside as the dominant component. A similar profile with ptaquiloside as the dominant glycoside was obtained for Pteridium aquilinum subsp. wightianum (previously P. revolutum ) from northern Queensland and also P. aquilinum from European sources. Ptesculentoside has chemical reactivity similar to that of ptaquiloside and presumably biological activity similar to that of this potent carcinogen. The presence of this additional reactive glycoside in Australian P. esculentum implies greater toxicity for consuming animals than previously estimated from ptaquiloside content alone. 相似文献

The fall armyworm Spodoptera frugiperda is a polyphagous pest that causes important damage in different regions of America and mainly affects corn crops in both tropical and subtropical areas. Currently, control relies on both transgenic plants and/or chemical pesticides. In this work we describe the preparation of an indexed combinatorial library of amides and its toxic effect by contact against S. frugiperda . (E)-1-(1-Piperidinyl)-3-[4-(trifluoromethoxy)phenyl]-2-propen-1-one was the most active compound with an LD(50) = 0.793 μg mg(-1) of larva. The degree and rate of the aggregate swelling in loamy sandy, loamy, and clayey soddy-podzolic soils of northwestern Russia have been measured using the methods of image analysis. The results of the study have shown that the degree of swelling of the soil aggregates depended on the proportions of the fine (0.005 mm) and sandy fractions, and the rate of the swelling depended on the density of the soil solid phase. A reliable effect of the soil organic matter content on the swelling rate of the studied objects has been established. The aggregates of the arable soddy-podzolic soils with the highest content of organic matter, as well as the aggregates of the native soils, have been characterized by the lowest swelling rates. A method has been developed for the determination of the swelling rate of soil aggregates that has enabled the assessment of the soil structure. The regression dependences of the degree and rate of the aggregate swelling on the properties and composition of soddy-podzolic soils have been plotted. 相似文献

Abstract Due to the discrepancy in metabolic sodium (Na) requirements between plants and animals, cycling of Na between humans and plants is limited and critical to the proper functioning of bio‐regenerative life support systems, being considered for long‐term human habitats in space (e.g., Martian bases). This study was conducted to determine the effects of limited potassium (K) on growth, Na uptake, photosynthesis, ionic partitioning, and water relations of red‐beet (Beta vulgaris L. ssp. vulgaris) under moderate Na‐saline conditions. Two cultivars, Klein Bol, and Ruby Queen were grown for 42 days in a growth chamber using a re‐circulating nutrient film technique where the supplied K levels were 5.0, 1.25, 0.25, and 0.10 mM in a modified half‐strength Hoagland solution salinized with 50 mM NaCl. Reducing K levels from 5.0 to 0.10 mM quadrupled the Na uptake, and lamina Na levels reached ‐20 g kg?1 dwt. Lamina K levels decreased from ‐60 g kg?1 dwt at 5.0 mM K to ‐4.0 g kg?1 dwt at 0.10 mM K. Ruby Queen and Klein Bol responded differently to these changes in Na and K status. Klein Bol showed a linear decline in dry matter production with a decrease in available K, whereas for cv. Ruby Queen, growth was stimulated at 1.25 mM K and relatively insensitive to a further decreases of K down to 0.10 mM. Leaf glycinebetaine levels showed no significant response to the changing K treatments. Leaf relative water content and osmotic potential were significantly higher for both cultivars at low‐K treatments. Leaf chlorophyll levels were significantly decreased at low‐K treatments, but leaf photosynthetic rates showed no significant difference. No substantial changes were observed in the total cation concentration of plant tissues despite major shifts in the relative Na and K uptake at various K levels. Sodium accounted for 90% of the total cation uptake at the low K levels, and thus Na was likely replacing K in osmotic functions without negatively affecting the plant water status, or growth. Our results also suggest that cv. Ruby Queen can tolerate a much higher Na tissue concentration than cv. Klein Bol before there is any growth reduction. 相似文献

Dry bean along with rice is a staple food for the population of South America. In this tropical region beans are grown on Oxisols and phosphorus (P) is one of the most yield limiting factors for dry bean production on these soils. A greenhouse experiment was conducted to evaluate P use efficiency in 20 elite dry bean genotypes grown at deficient (25 mg P kg?1 soil) and sufficient (200 mg P kg?1) levels of soil P. Grain yields and yield components were significantly increased with P fertilization and, interspecific genotype differences were observed for yield and yield components. The grain yield efficiency index (GYEI) was having highly significant quadratic association with grain yield. Based on GYEI most P use efficient genotypes were CNFP 8000, CNFP 10035, CNFP10104, CNFC 10410, CNFC 9461, CNFC 10467, CNFP 10109 and CNFP 10076 and most inefficient genotypes were CNFC 10438, CNFP 10120, CNFP 10103, and CNFC 10444. Shoot dry weight, number of pods per plant, 100-grain weights and number of seeds per pod was having significant positive association with grain yield. Hence, grain yield of dry bean can be improved with the improvement of these plant traits by adopting appropriate management practices. Soil pH, extractable P and calcium (Ca) saturation were significantly influenced by P treatments. Based on regression equation, optimum pH value in water was 6.6, optimum P in Mehlich 1 extraction solution was 36 mg kg?1 and optimum Ca saturation value was 37% for dry maximum bean yield. 相似文献

Homogenates were prepared from the leaves of hydroponically cultivated sunflowers (Helianthus annuus, L.) under deficient, normal and toxic B conditions and the polyphenol oxidase activity together with the ortho‐diphenolic and micronutrient content was measured every week from one month to flowering. B deficiency slightly depressed polyphenol oxidase activity and B toxicity decreased it significantly at all times, the response fitting a line versus the logarithm of the B concentration in solution. We found a positive correlation between polyphenol oxidase activity and the B content in leaves. B action on enzymatic activity seemed not to be caused by direct interaction with substrates as the infiltration of B into deficient leaves did not restore the normal activity According to our results we suggest that polyphenol oxidase activity may be a valid functional parameter to indicate the nutritional status of B in sunflower plants, if its specificity for B can be demonstrated. The ortho‐diphenolic content did not change with the B levels used in our experiments. Neither did we find any correlation between enzymatic activity and the o‐diphenolic content of the leaves. 相似文献
