Smallholder farmers dominate agriculture in Nepal. These farmers have poor knowledge about agriculture and lack of support for soil management and integrated plant-nutrient systems. Focusing on the importance and need for soil-fertility management, a soil-testing mobile van program has recently been introduced in Nepal by Soil Management Directorate, Hariharbhawan. With the introduction of the mobile lab, famers can get their soil tested for nutrient deficiencies and fertilizer requirements at their doorsteps. Using mobile lab, spatial distributions of chemical properties, including pH, organic matter (OM), total nitrogen (N), available phosphorus (as P2O5), and available potassium (as K2O) were examined in soil samples taken from the 0 to 15 cm depth from selected agricultural fields in eight different districts in the mid-hills and Terai regions of Nepal. Tests conducted on 1,479 soil samples in the soil-testing mobile van revealed the following: the mean soil OM ranged from 0.01 to 1.77%; total N content ranged from 0.01 to 0.08%; mean available P2O5 ranged from 16.47 to 197.82 kg ha?1; and mean available K2O ranged from 84.3 to 422.57 kg ha?1. For each crop to be grown, farmers were provided with individual soil health reports and fertilizer recommendations (rate, amount, and type). This program not only allows scientists and farmers to work closely and share information but also serves as a model for the nation to successfully transfer technology for improving soil health and sustainability.  相似文献

Proline/hydroxyproline-rich glycoprotein (P/HRGP) level in pearl millet genotypes resistant to downy mildew increase after inoculation with the oomycete pathogen Sclerospora graminicola. Using purified P/HRGPs from pearl millet cell walls, polyclonal antibodies (Pab-P/HRGP) were raised in rabbit. Based on this antiserum, an enzyme immunoassay was developed that displays a linearity detection range from 0.01 to 10 μg P/HRGP. Western blot analysis, confirming the induction of three marker P/HRGPs in the infected resistant genotype, and immunocytochemical studies on P/HRGP localization either in epidermal peelings or in suspension-cultured cells demonstrated the specificity of the antiserum. Besides its characterization, Pab-P/HRGP was employed to screen various genotypes of pearl millet for fast, sensitive and specific detection of induced P/HRGPs upon infections. The results presented are discussed with presumed importance to downy mildew disease and the use of this new antiserum in pearl millet screening for disease resistance.  相似文献

The potential of tannins from 21 medicinal and aromatic plant leaves as antimethanogenic additives in ruminant feeds was investigated. The effect of tannin from these leaves on rumen fermentation parameters, protozoa population and methanogenesis was studied by incubating the samples [200 mg dry matter (DM)] without and with polyethylene glycol (PEG)‐6000 (400 mg DM) as a tannin binder during 24‐h incubation in the in vitro Hohenheim gas method. Based on the methane percentage estimated in the total gas produced, methane production in millilitre was calculated [methane volume (ml) = methane % × total gas produced (ml) in 24 h]. In the samples, crude protein and neutral detergent fibre (g/kg DM) ranged from 113 to 172 and from 352 to 444 respectively. The total phenol (TP; g/kg DM) content was highest in Terminalia chebula (274) followed by Hemigraphis colorata (71) and Sapindus laurifolia (51) respectively. In the remaining samples, it was <43 g/kg DM. Activity of tannins, as represented by the increase in gas volume on addition of PEG, ranged from 0 to 133%, with the highest being recorded in T. chebula. The per cent increase in methane on PEG addition was 0 for Ammi majus, Aristolochia indica, Cascabela thevetia, Ipomea nil and Lantana camara, illustrating that tannins present in these samples had no effect on methane concentration. The PEG addition increased the total protozoa count by >50% in A. indica and C. thevetica. One of the important findings of our study was that of the 21 samples screened, Entodinia population increased in 12 with PEG as compared to 7 where Holotricha increased, indicating higher susceptibility of Entodinia to tannin. There was no increase in the protozoa population with PEG when incubating Cardiospermum halicacabum, Clerodendrum inerme, Dioscorea floribunda, Nerium oleander and Selastras paniculatus, which strongly suggested that methane suppression recorded in these samples was not because of a defaunating effect of their tannins per se. The fermentation pattern reflected increased total volatile fatty acid (TVFA) concentration from 0 to 28.3% with PEG addition among the leaves. Our results confirmed further observations that methanogenesis in vitro is not essentially related to density of protozoa population. Secondly, medicinal and aromatic plants such as C. inerme, Gymnema sylvestre and Sapindus laurifolia containing tannins appear to have a potential to suppress in vitro methanogenesis.  相似文献

Differential resistance of pearl millet cultivars to downy mildew disease was correlated with the levels of -1,3-glucanase in their seeds. Higher activity of the enzyme in highly resistant cultivars and lower activity in the highly susceptible ones suggested the possible use of -1,3-glucanase as a biochemical marker for screening pearl millet cultivars for downy mildew disease. Inoculation of seedlings with the downy mildew pathogen Sclerospora graminicola resulted in increased enzyme levels in resistant cultivars. Mesocotyl and shoot regions of seedlings recorded higher levels of enzyme than the root. Isoelectric focusing revealed four basic isoforms with pI 9.6, 9.0, 8.9 and 8.2 and two acidic isoforms with pI 4.9 and 6.2 of -1,3-glucanase in pearl millet. The pI 9.6 isoform was a major isoform of the enzyme in the pearl millet seedlings with a probable developmental function. Isoforms pI 6.2 and pI 8.2 appeared to be involved in resistance and pI 4.9 isoform seemed to be involved in pathogenesis of pearl millet-downy mildew.  相似文献

A study was conducted to assess comparatively the growth performance of three different indigenous goat breeds during exposure to summer heat stress. The primary objective of the study was to observe the heat stress impact on the growth performance based on the body weight changes, allometric measurements, growth hormone (GH) concentration and peripheral blood mononuclear cell (PBMC) Insulin‐like growth factor‐1 (IGF‐1) mRNA expression pattern during the summer season in comparison with the local breed (Osmanabadi). Thirty‐six ten‐month‐ to one‐year‐old female goats of Osmanabadi, Malabari and Salem Black breeds were randomly divided into six groups, OC (n = 6; Osmanabadi control), OHS (n = 6; Osmanabadi heat stress), MC (n = 6; Malabari control), MHS (n = 6; Malabari heat stress), SBC (n = 6; Salem Black control) and SBHS (n = 6; Salem Black heat stress). Body weight was recorded at weekly intervals, whereas other growth and allometric measurements and blood collection were carried out at fortnightly intervals. Breed factor significantly (p < .05) influenced only few growth variables such as body weight, body mass index (BMI) and body condition score (BCS). However, heat stress treatment significantly (p < .05) reduced all growth parameters expect BMI. Further, the heat stress significantly (p < .01) increased plasma GH concentration in goats with significantly higher (p < .05) concentration recorded in OHS. Among the stress groups, the lower (p < .05) PBMC IGF‐1 mRNA expression was recorded in OHS, while the higher (p < .05) expression was observed in SBHS indicating the extreme adaptive capability of Salem Black breed. Thus, the results indicated that the Salem Black breed performed much better compared to both Osmanabadi and Malabari breeds indicating the superior ability of this breed to adapt to heat stress challenges. The results also indicated that plasma GH and IGF‐1 gene may act as ideal biomarkers for assessing the heat stress impact on growth performance in indigenous goats.  相似文献

Allometric relationship (W1=αW2β, where α and β are the parameters) was fitted among growth components in two maize cultivars viz., Decani hybrid and Deccan 101 in order to obtain estimates of other components of the plant system which are time consuming measurements. The results of the agronomic field trial conducted at the University of Agricultural Sciences, Bangalore were used. This model's predictability was compared with linear regression model. In both the cultivars, allometric model using leaf area (LA - W2.) and leaf dry matter (LDM - W2) simulated total dry matter production (DMP - W2) by 79 to 98 % of actual values. Further allometric model fitted well to predict stem dry matter by 91 to 93 % using LDM and LA 89 to 92 % using LDW. Whereas linear regression model estimated total DMP by 95 to 96 % using cob dry matter. In case of LDM - LA association, linear regression model was found to be the best than other model. The leaf area decreased after silking in both the cultivars and the ratio of growth rates of DMP – LA ( β 2) was negative. Between cultivars, cv. Deccan 101 had higher R2 values in most of the relationships than cv. Deccan hybrid indicating the varietal difference.  相似文献

Elicitors derived from the cell wall of fungi are shown to be active in eliciting resistance in plants against a wide range of pathogens. In the present study carbohydrate components from the autoclaved spore cell wall ofAspergillus niger were prepared as aqueous suspensions and tested for defense response in pearl millet (Pennisetum glaucum (L.) R.Br.) against the oomycetous downy mildew pathogenSclerospora graminicola (Sacc.) Schroet. The aqueous suspension derived from the spore cell wall ofA. niger was used as a seed soak treatment at concentrations of 0.25, 0.5, 1.0, 1.5 and 2.0 mg ml−1 for time intervals of 3, 6, 9 and 12 h. The concentration of 0.5 mg ml−1 for a 6 h soaking period offered 94% seed germination and seedling vigor index increased to 1526. The seed germination and the seedling vigor were significantly higher than the untreated check. Spore cell wall suspension as seed treatment at a concentration of 0.5 mg ml−1 required a 3-day time interval to provide 67% protection against downy mildew. Histological and biochemical studies were conducted to elucidate the mechanism of defense response in treated seedlings uponS. graminicola infection. Resistance host response was detected in the form of lignin and callose deposition in the epidermal cell wall of pearl millet seedlings, which is the site ofS. graminicola infection. A time course study showed rapid and localized deposition of lignin and callose in epidermal cell wall of carbohydrate components-treated pearl millet seedling coleoptiles. Increased levels of the defense-related enzyme peroxidase were detected in the treated seedlings. Peroxidase activity in elicitor-treated samples reached a peak at 8 h post-infection, which was 45% more than in their respective uninoculated control. There are many socioeconomic and technological constraints that affect the production of wheat and other staple cereals in South Asia. Wheat production is one of the economic mainstays in South Asia, but the yield gap between farmers’ fields and experimental yields is wide across the region. For the last 3 years, CIMMYT and the CAZS-NR have been collaborating with farmers, NARS, and other South Asian partners to promote improved wheat varieties and new resource conservation technologies (RCTs) in farmers’ fields. Participation fostered among farmers, scientists, extension specialists, NGOs and the private sector included variety selection (PVS), and evaluation of agronomic practices. Through PVS, several farmer-preferred technologies have been identified including wheat varieties for adverse conditions in eastern Uttar Pradesh (India) and for boron deficiency in parts of Nepal. There has been considerable improvement in the access of farmers to new varieties and technologies in the rural areas. Yield increases (15–70%) have been achieved by resource-poor farmers over the existing varieties through the adoption of new varieties and RCTs. The farmers have also made substantial cost savings and achieved higher yields through resource-conserving agronomic techniques such as zero till. Seed of the new farmer-selected cultivars has been multiplied by groups of collaborating farmers and widely distributed.  相似文献
