Water availability and salt excess are limiting factors in Mexican mixed pine-oak forest. In order to characterise the acclimatation of native species to these stresses, leaf water (Ψw) and osmotic potentials (Ψs) of Juniperus flaccida, Pinus pseudostrobus and Quercus canbyi were measured under natural drought and non-drought conditions under two different aspects in the Sierra Madre Oriental. Factorial ANOVA revealed significant differences in Ψw and Ψs between two aspects, species and sampling dates. In general, all species showed high predawn and low midday values that declined progressively with increasing drought and soil–water loss. Seasonal and diurnal fluctuation of Ψw and Ψs were higher for J. flaccida and Q. canbyi than for P. pseudostrobus. Leaf Ψw and Ψs were mainly correlated with soil water content, while Ψs of P. pseudostrobus were hardly correlated with environmental variables. Thus, species have different strategies to withstand drought. P. pseudostrobus was identified as a species with isohydric water status regulation, while J. flaccida and Q. canbyi presented water potential patterns typical for anisohydric species. The type of water status regulation may be a critical factor for plant survival and mortality in the context of climate change. Nevertheless, for precise conclusions about the advantages and disadvantages of each type, further long-term investigations are required.  相似文献

Meadowfoam ( Limnanthes alba) is an oilseed crop grown in western Oregon. The seed meal has potential value as a biopesticide due to glucosinolate degradation products and phytoecdysteroids, a group of polyhydroxylated triterpenoids with potent activities as arthropod molting hormones. Liquid chromatography in combination with tandem mass spectrometry operated in the precursor ion mode revealed the presence of four ecdysteroid glycosides in meadowfoam seed meal. The carbohydrate sequence and the identity of the ecdysteroid aglycones, ponasterone A and 20-hydroxyecdysone, were determined by product ion scanning. Ecdysteroids were detected in the negative ion mode as [M + formate] (-) ions, which yielded [M - H] (-) and alpha-cleavage fragments with retention of hydroxyl groups in MS/MS experiments (not seen in the positive ion mode), allowing the determination of the number of hydroxyl groups in the side chain and in the steroid ring system. MS/MS of glycoside ions ([MH] (+) or [M + formate] (-)) provided carbohydrate sequence information.  相似文献

Streptococcus suis is a major cause of meningitis, sepsis and arthritis in piglets and a zoonotic agent. Survival in the blood circulation system represents a major step in pathogenesis of S. suis infections. To get further insights into the mechanisms of S. suis survival in the host, we compared a highly virulent S. suis serotype 2 strain with its non-encapsulated and suilysin-deficient mutants in their abilities to resist phagocytosis and killing by polymorphonuclear neutrophils (PMNs) and mononuclear cells. PMNs displayed a higher capacity to take up encapsulated bacteria than mononuclear cells, whereas both cell types internalized efficiently non-encapsulated S. suis. Differentiation of extracellular and intracellular survival of the WT strain revealed that in PMNs the majority of the cell-associated streptococci were intracellular, whereas in mononuclear cells the majority remained attached to the cell surface. S. suis survived mainly extracellularly, since both cells killed intracellular bacteria to a similar extent. As a consequence of different resistance to phagocytosis, only the encapsulated S. suis strains survived co-cultivation with PMNs. Comparison of the WT strain with its encapsulated suilysin-deficient mutant revealed reduced survival of the mutant after co-cultivation with PMNs. Involvement of suilysin in inhibition of phagocytosis was further confirmed by the use of anti-suilysin antibodies and recombinant suilysin. Kinetic experiments with PMNs suggested that reduced survival of the mutant strain was mainly associated with an increased uptake, whilst both strains adhered similarly. Concluding, our results indicate that the capsule and the suilysin play important roles in S. suis survival in the host by interfering with phagocytic uptake.  相似文献

CD137 plays an important role as a co-stimulatory molecule in activated T cells. Agonistic CD137 specific antibodies have been investigated as therapeutic agents to promote tumor-specific immune responses by direct activation of T cells. As part of the pre-clinical pharmacological evaluation of cynomolgus monkeys, monkey CD137 was cloned and characterized. The deduced amino acid sequence encoded a full-length gene of 254 amino acids 95% identical to human CD137. Sequence variants identified in monkey CD137 include four splice variants lacking the transmembrane domain. These variants were detectable in human including two previously unreported variants. Two missense single nucleotide polymorphisms were detected present in 42 and 50% of 36 monkeys tested. In both monkey and human, mRNA expression of full-length CD137 and splice variants were significantly increased in peripheral blood mononuclear cells (PBMCs) upon stimulation by anti-CD3 antibodies. Recombinant monkey CD137 protein was bound with high affinity by an agonistic anti-human CD137 antibody but not by an anti-mouse CD137 antibody. In summary, compared to human, monkey CD137 showed distinct extracellular domain amino acid sequence and sequence polymorphisms. Thus, antibodies directed against epitopes in this extracellular domain could have differences in pharmacologic activity between cynomolgus monkeys and human or across individual cynomolgus monkeys.  相似文献

Antistaling properties of a bacterial maltogenic amylase, sodium carboxymethylcellulose (CMC), and vital wheat gluten on quality of corn tortillas were evaluated during 14 days of storage. Amylopectin recrystallization was the driving force behind the staling of corn tortillas. Increasing levels of recrystallized amylopectin measured by differential scanning calorimetry (DSC) correlated significantly with increased tortilla stiffness (r = 0.43) and reduction in tortilla pliability (r = ‐0.42) during storage. Maltogenic amylase (275–1,650 activity units) made tortillas less stiff but did not preserve pliability and extensibility as effectively as CMC (0.25–0.5%). The combination of 825 MANU of maltogenic amylase (to interfere with intragranular amylopectin recrystallization) and 0.25% CMC (to create a more flexible intergranular matrix than retrograded amylose and amylopectin) produced less stiff, equally flexible, and less chewy tortillas than did 0.5% CMC. Vital wheat gluten was not as effective as CMC in preserving tortilla flexibility or as good as the maltogenic amylase in reducing stiffness. Further research is required to optimize the addition of maltogenic amylases in continuous processing lines that use fresh masa instead of nixtamalized corn flour (NCF) and to determine how these amylases interfere with amylopectin recrystallization.  相似文献

The derivatization followed by reductive cleavage (DFRC) method cleaves alpha- and beta-ethers in lignins but leaves lignin gamma-esters intact. When applied to grasses, which contain p-coumarate esters on their lignins, esterified monolignol derivatives are released. Saturation of the p-coumarate double bond occurs during DFRC, so the released products are 4-acetoxycinnamyl 4-acetoxyphenylpropionates. Synthesis of the esters allowed determination of response factors for the released products. Maize and bamboo lignins released 221 and 38 micromol/g of p-coumarate-derived esters. The sinapyl ester was much more abundant than the coniferyl one. The bamboo and maize lignin S/G ratios in the conjugates were 12 and 38 times greater than those of the normal monomers released by DFRC, evidence of a strong selectivity for acylation of syringyl units. Of three possible biochemical mechanisms for incorporating p-coumarates into lignin, evidence is mounting that the process involves incorporation of preacylated monolignols into the normal lignification process.  相似文献

The objective was to estimate the potential risk of N leaching into the groundwater under various types of agriculture by using lysimeter experiments on the nitrogen(N)‐cycle of various soil types. Results were obtained with 12 weighable, monolithic lysimeters with a surface area of 1 m2, a total depth of 3 m, and free drainage. Mean annual N‐leaching losses of 5 to 44 kg ha—1 and nitrate concentrations of the seepage water (leachate) between 80 and 200 mg l—1 were measured during the period of intensive agricultural use. On fallow land with a well‐established grass vegetation, some nitrate was removed by the plants. As a result, the nitrate concentrations in the leachate were reduced significantly. Ecological farming measures generally reduced N leaching losses and kept the N‐concentration in the leachate below the German threshold value for drinking water with 50 mg l—1 nitrate. However, ploughing in of clover or leguminous vegetation and the application of farmyard manure in autumn caused the nitrate concentration in the leachate to rise significantly above the mentioned threshold value.<?show $6#>  相似文献

This study aimed to determine the genetic variability of isolates of rotting of pineapple fruitlet core in Brazil on the states of Paraiba, Pernambuco and Rio Grande do Norte, based on phylogenetic analysis of the RPB2 gene, morphocultural markers and aggressiveness of the isolates. The phylogenetic reconstruction of maximum parsimony and bayesian inference of the isolates were performed. Twenty-seven phylogenetic lineages were characterized with morphocultural markers on potato dextrose agar and synthetic nutrient-poor agar. The aggressiveness of these lineages were characterized in leaves and fruits of pineapple ‘Pérola’ cultivar. The Phylogenetic reconstruction showed close relationship between isolates of this study and phylogenetic lineages of F. guttiforme, F. ananatum and F. oxysporum by analysis of RPB2 gene. Phylogenetic lineages of this study shared significant morphocultural markers similar to those described for this species. Overall, the lineages related with Fusarium oxysporum Species Complex were more aggressive to the fruits of the Fusarium lineages related with Fusarium fujikuroi Species Complex. It is possible that F. ananatum and F. guttiforme or other lineages closely related to these species or F. oxysporum are present in the fields causing pineapple rotting fruitlet core in fruits of pineapple ‘Pérola’ in Brazil. The used markers determined high genetic variability in isolates analyzed in leaves and pineapple fruit ‘Pérola’ in the states of Paraíba, Pernambuco and Rio Grande do Norte of Brazil and the pathogenic lineages analyzed were better adapted to the fruits than to the detached leaves in pineapple ‘Pérola’ analyzed.  相似文献

An 8-year-old male castrated Domestic Short-haired cat was examined for a 1-week history of blepharospasm and mucoid ocular discharge OS. Examination revealed ulcerative keratitis with stromal loss, stromal infiltrate, corneal edema, perilimbal vascularization and miosis. Cytology of the cornea revealed multiple dichotomously branching, septate fungal hyphae and severe, predominantly neutrophilic inflammation. PCR of the cytology samples confirmed the presence of Aspergillus flavus while fungal and bacterial cultures were negative. Treatment with topical 1% voriconazole solution was successful in resolving the keratomycosis.  相似文献
