Adult mouflon ewes (Ovis musimon) were treated repeatedly with therapeutic doses of albendazole (ABZ, p.o. 7.5 mg/kg of body weight/day, for five consecutive days). Animals (treated or control) were sacrificed 24 h after the fifth dose of ABZ and liver and small intestine were collected to prepare microsomes. The activities of several biotransformation enzymes were measured in both hepatic and intestinal microsomes. A significant increase in the activity and amount of cytochromes P4501A (CYP1A) was observed in both tissues of ABZ treated mouflons compared to control animals. No other biotransformation enzymes tested were affected by five ABZ doses. The in vitro biotransformation of ABZ was studied in hepatic and intestinal microsomes from ABZ treated and control mouflons. Concentrations of two main ABZ metabolites - pharmacologically active ABZ sulfoxide and pharmacologically inactive ABZ sulfone were analysed using HPLC. A significant increase in rate of formation of ABZ sulfone (which is catalysed by CYP1A) was observed in hepatic as well as in intestinal microsomes from ABZ treated animals. The enhancement of ABZ deactivation by its repeated administration may affect the anthelmintic efficacy of this drug and may contribute to the development of parasite resistance.  相似文献

Fenbendazole (FBZ), flubendazole (FLBZ) and mebendazole (MBZ) are benzimidazole anthelmintics widely used in veterinary medicine. The effects of these drugs on cytochromes P450 (CYP) were investigated in primary cultures of swine (Sus scrofa f. domestica) hepatocytes. After 48-h incubation of hepatocytes with benzimidazoles (0.1-2.5 microm), ethoxyresorufin O-deethylation (EROD), benzoxyresorufin O-dearylation (BROD), testosterone hydroxylase (6beta-TOH) and testosterone oxidase (17-TO) activities were measured and the CYP1A and 3A protein levels were determined by Western blotting. FBZ produced a significant, concentration-dependent increase of CYP1A activity (EROD) and protein level. No enhancement of CYP1A was observed after exposure to FLBZ and MBZ. All benzimidazoles tested did not cause any induction of CYP3A (BROD, 6beta-TOH, 17-TO activities and protein content). On the other hand, MBZ produced a significant, concentration-dependent decrease of CYP3A (BROD, 6beta-TOH and 17-TO) activities. Pharmacological and toxicological consequences of CYP1A induction and CYP3A inhibition should be taken into account in treatment of pigs with FBZ and MBZ.  相似文献

The total lipid content and fatty acid composition of the muscle tissue of tilapia (Oreochromis niloticus) and of hybrid red tilapia (Oreochromis sp.) from different culture systems and from the natural and artificial environment of Thailand were compared. Wild fish and fish reared under the most extensive conditions had a more favorable fatty acid profile for human consumption as they contained higher proportions of 18:3n-3, 20:5n-3, and 22:6n-3, higher n-3/n-6 PUFA ratios, and lower proportions of 18:2n-6. The muscle tissue of intensively cultured fish was characterized by increased fat deposition that was mainly saturated and monounsaturated fatty acids and 18:2n-6. It is undesirable for the consumer to reduce 20:5n-3 and 22:6n-3 in farmed tilapia and replace them with elevated 18:2n-6. It is recommended that the amount of 18:2n-6 in the feed of the intensively reared tilapia should be reduced by substituting vegetable oils rich in 18:2n-6 with oils rich in 18:1n-9 and/or 18:3n-3.  相似文献

In 1992 a cooperative project, Clover Sweden, was initiated. The aim was to study if subterranean clover could be used as a bioindicator in the different climate zones in Sweden by studying the impact of ambient ozone concentrations on this species in different parts of the country during three consecutive summer seasons. Plants of subterranean clover, Trifolium subterraneum, L., were exposed to ambient air at 24 sites from north to south Sweden. The project was designed to be compatible with the international programme, ICP Crops within the UNECE and the Convention on Long Range Transboundary Air Pollution. The results showed that subterranean clover is a useful bioindicator of ozone in all agricultural areas of Sweden, with the exception for very cool and rainy summers resulting in poor growth of the plants. In 1992, and especially in 1994, ozone injury was detected at almost all sites in Sweden, reflecting the higher ozone levels of those summers as compared to 1993, when ozone concentrations were generally low and not much injury was detected. Typical injury was chlorotic and bifacial necrotic lesions on parts of the leaf surface. It is concluded that at mean ozone concentrations of 25 ppb (24 h mean) and 30 ppb (7 h mean) there is a potential risk for injury on 10% of the leaves. When % injured leaves was plotted against AOT (Accumulated exposure Over a Threshold) using different thresholds, it became obvious that a threshold of 20 ppb ozone should be used in order to fully protect from leaf injury under Swedish conditions.  相似文献

An in vitro method has been used to examine whether secretion of renin from the juxtaglomerular apparatus is affected by changes in the sodium chloride concentration of the tubular fluid at the macula densa. Single juxtaglomerular apparatuses were microdissected from rabbits and the tubule segment containing the macula densa was perfused, while simultaneously the entire juxtaglomerular apparatus was superfused, and the fluid was collected for renin measurement. In this preparation, in which influences from renal nerves and local hemodynamic effects are eliminated, a decrease in the tubular sodium chloride concentration at the macula densa results in a prompt stimulation of the renin release rate.  相似文献

A total of 17 simple sequence repeat (SSR) markers linked to QTLs (qDTYs) governing grain yield under reproductive stage (RS) drought stress were used to assess the genetic relationship and prospecting new donors for qDTYs among 32 popular upland rice genotypes. These SSR markers generated a total of 36 alleles with an average allele count of 2.1 per locus. Polymorphic information content value of the markers ranged from 0.376 to 0.662 with an average value of 0.484. The expected heterozyogosity ranged from 0.381 to 0.632. STRUCTURE analysis divided the 32 genotypes into three sub-populations. Subsequent phenotyping revealed that all the tolerant genotypes were grouped into one sub-population, whereas the moderately tolerant and susceptible genotypes were grouped into separate sub-populations. Phylogenetic tree constructed by the unweighted neighbour-joining method also divided the genotypes into three clusters. The grouping pattern of genotypes into the clusters was similar to that into the STRUCTURE analysis, on the basis of drought tolerance level. The average value of genetic dissimilarity coefficient among the genotypes was observed to be 0.486. Furthermore, by combining genotyping data with phenotyping data, 16 new donors for 6 qDTYs were identified.  相似文献

A thermotolerant strain AKM-P6 of Pseudomonas sp. possessing plant growth-promoting properties was isolated from rhizosphere of pigeon pea grown under semiarid conditions in India. The effect of inoculation with AKM-P6 on survival and growth of sorghum seedlings at elevated temperatures (ET) was investigated under sterile and nonsterile soil conditions. Inoculation with strain AKM-P6 helped sorghum (var CSV-15) seedlings to survive and to grow at elevated temperatures (47–50°C day/30–33°C night) up to 15 days while uninoculated plants died by the fifth day of exposure to elevated temperature. Under sterile and nonsterile conditions, significantly higher root and shoot biomass were recorded in inoculated seedlings as compared to uninoculated control at ET, but this difference was nonsignificant at ambient temperature. Inoculation induced the biosynthesis of high-molecular weight proteins in leaves under elevated temperature, reduced membrane injury, and improved the levels of cellular metabolites like proline, chlorophyll, sugars, amino acids, and proteins. Scanning electron microscopy studies confirmed the colonization and establishment of the organism on the root surface. The 16SrDNA sequence of the strain AMK-P6 showed 97% homology with that of Pseudomonas aeruginosa in the existing database. The results indicate that Pseudomonas sp. strain AKM-P6 can enhance tolerance of sorghum seedlings to elevated temperatures by inducing physiological and biochemical changes in the plant.  相似文献
