Relative retention, volatility, and temporal release of volatile compounds taken from aldehyde, ester, and alcohol chemical classes were studied at 70 degrees C in model systems using equilibrium static headspace analysis and real time dynamic headspace analysis. These systems were medium-chain triglycerides (MCT), sunflower oil, and two structured systems, i.e., water-in-oil emulsion and L2 phase (water-in-oil microemulsion). Hydrophilic domains of the emulsion type media retained specifically the hydrophilic compounds and alcohols. Four kinetic parameters characterizing the concentration- and time-dependent releases were extracted from the aroma release curves. Most of the kinetic parameter values were higher in structured systems than in oils particularly when using MCT. The oil nature was found to better control the dynamic release profiles than the system structures. The release parameters were well-related (i) to the volatile hydrophobicity as a function of the oil used and (ii) to the retention data in the specific case of the L2 phase due to a specific release behavior of alcohols.  相似文献

Biofumigation is increasingly viewed as a potentially useful technique for controlling soil-borne crop pathogens, but its efficacy has not systematically been demonstrated at field scale. We investigated the differences in efficacy observed in the field, by analysing the mechanisms by which a Brassica cover crop can act as a biofumigant crop in the prevention of soil-borne disease development. We hypothesised that the biofumigant crop might have a negative effect on soil-borne pathogens whilst growing, and that the pulverisation of this crop and the incorporation of its residues into the soil may enhance this effect. We tested this hypothesis by carrying out three field experiments in 2006, 2007 and 2008 in which Brassica juncea (brown mustard) was managed in different ways within a sugar beet–winter wheat rotation and analysing effects on sugar beet root rot caused by Rhizoctonia solani. Three treatments were studied: mustard pulled out at flowering (MP), mustard crushed at flowering and incorporated into the soil (MC) and bare soil (BS) as a control. We assessed the effect of each treatment on root rot incidence and severity at harvest. Over the 3 years of the experiment, disease incidence was significantly higher on BS plots than on the other plots and was significantly higher on MP plots than on MC plots. MC treatment gave a significantly lower mean conditional severity (severity calculated for diseased beets only) than the BS and MP treatments. Mustard residue incorporation was consistently effective at decreasing disease incidence from year to year (43, 44 and 47% efficacy, as determined by comparison with the disease incidence on BS plots, in 2006, 2007 and 2008, respectively), but the efficacy of growing mustard was variable (36, 16 and 39% efficacy in 2006, 2007 and 2008, respectively). These findings provide insight into the mechanisms by which biofumigant crops may affect soil-borne diseases. These findings have implications for the possible use of biofumigant crops as a biological method for controlling soil-borne diseases at the field scale.  相似文献

Cacao (Theobroma cacao L.) is the main source for chocolate with an annual production of four million tons worldwide. This Neotropical tree crop was domesticated in Mesoamerica as far back as 3,000 years ago. Knowledge of genetic diversity and population structure in farmer varieties of cacao in the center of domestication is essential for sustainable production of fine-flavored cacao beans and contributes to in situ/on-farm conservation of farmer varieties. Based on 70 single nucleotide polymorphism markers, we analyzed 84 fine-flavored farmer varieties collected from traditional cacao farms in Honduras and Nicaragua. The study also included 31 clones from the international cacao collections to serve as references. The SNP based multilocus matching identified six synonymous groups, including 14 Criollo and two Amelonado varieties. A moderately high level of genetic diversity was observed in these farmer varieties, indicating the possibility to further explore intra-population variation and breed for fine-flavored cocoa. Multivariate analysis showed clustering of the 84 farmer accessions in five genetic groups: ancient Criollo, Amelonado, Trinitario (including Nicaragua Trinitario and Honduras Trinitario) and Upper Amazon Forastero (only one accession). The Honduras Trinitario differed from the Nicaragua Trinitario group. The clustering results largely supported the perceived classification of cacao by local farmers and researchers, which was mainly based on morphological traits. However, the well known traditional variety “Indio” in this region was identified as synonymous with Amelonado. Parentage analysis showed that the variety “Indio” (or Amelonado) contributed more to the Trinitario type farmer varieties, whereas ancient Criollo had less influence. The present study demonstrates the efficacy of using a small set of SNP makers for cacao germplasm characterization, and further depicts the diverse origins and parentage in farmer varieties from Mesoamerica. This information thus will be highly useful for conservation and utilization of cacao germplasm from this region.  相似文献

The ability of Pseudomonas putida BTP1 to induce resistance in bean to Botrytis cinerea was demonstrated in soil experiments on plants pre-inoculated at the root level with the bacteria before challenge with the leaf pathogen. As a first step to characterize the molecules from BTP1 responsible for induction of systemic resistance in bean, heat-killed cells and supernatant from culture in an iron-limited medium were tested for their protective effect. Most of the resistance-eliciting activity of the strain was retained in the crude cell-free culture fluid. In vivo assays with samples from successive fractionation steps of the BTP1 supernatant led, (i) to the conclusion that salicylic acid, pyochelin and pyoverdin, previously identified as Pseudomonas determinants for induced systemic resistance (ISR), were not involved in systemic resistance triggered by BTP1, and (ii) to the isolation of fractions containing one main metabolite that retained most of the resistance-inducing activity in bean. Although this molecule remains to be structurally characterized, its isolation is an addition to the range of determinants from plant growth-promoting rhizobacteria (PGPR) known to stimulate plant defences.  相似文献

The effect of animal feeding on milk volatile organic compounds (VOCs) of metabolic origin was tested on a hay-based diet (H), a highly diversified pasture under continuous grazing (CG), or a less diversified pasture under rotational grazing (RG). Individual milk of 24 Montbe?liarde cows (8 per treatment) were sampled after 2 weeks. Pasture-derived milk was richer (p < 0.05) in camphene, sabinene, β-caryophyllene, and skatole than H milk. Neither milk yield nor fat content affected the majority of VOCs measured. Skatole increased slightly with milk yield, while indole and cineole decreased slightly with milk fat content but with poor regression (R(2) < 0.54). Multivariate analysis showed that, on the basis of those VOCs of metabolic origin whose concentration differed between treatment (dimethyl-sulfone, skatole, toluene, undecanoic acid, 1-octadecene, benzeneacetaldehyde, octanoic acid, and 2-pentanone-4-hydroxy-4-methyl), it was possible to obtain good discriminations among feeding systems. This study is promising for a future use of VOCs of metabolic origin to trace animal feeding systems.  相似文献

To integrate the gene pool of a wild species (primarily diploid) into a cultivated pool (primarily tetraploid), a crossing between a dihaploid cultivated rose and a hybrid of Rosa wichurana allowed to obtain interspecific diploid hybrids that produced 2n pollen grains. A return to a tetraploid level sought by breeders can then be considered using sexual polyploidization, obtained by crossing a tetraploid cultivated rose with these hybrids. Application of a high‐temperature regime led to a small but significant increase in the percentage of 2n pollen grains in these hybrids of up to 4.6%. This result was obtained by applying high temperatures close to 32°C during the day to plants cultivated in a glasshouse during recurrent cycles of bloom. Crosses were made between an unreleased tetraploid hybrid tea rose, as a female, and the diploid hybrid that produces the most 2n pollen grains. Tetraploid (42.1%) and triploid (57.9%) offspring were obtained. The use of these 2n pollen grains of the first division restitution type should facilitate the introgression of complex traits of interest.  相似文献

Mycoplasma hyopneumoniae, the causative agent of porcine enzootic pneumonia, is present in swine herds worldwide. However, there is little information on strains infecting herds in Canada. A total of 160 swine lungs with lesions suggestive of enzootic pneumonia originating from 48 different farms were recovered from two slaughterhouses and submitted for gross pathology. The pneumonic lesion scores ranged from 2% to 84%.Eighty nine percent of the lungs (143/160) were positive for M. hyopneumoniae by real-time PCR whereas 10% (16/160) and 8.8% (14/160) were positive by PCR for M. hyorhinis and M. flocculare, respectively. By culture, only 6% of the samples were positive for M. hyopneumoniae (10/160). Among the selected M. hyopneumoniae-positive lungs (n = 25), 9 lungs were co-infected with M. hyorhinis, 9 lungs with PCV2, 2 lungs with PRRSV, 12 lungs with S. suis and 10 lungs with P. multocida. MLVA and PCR-RFLP clustering of M. hyopneumoniae revealed that analyzed strains were distributed among three and five clusters respectively, regardless of severity of lesions, indicating that no cluster is associated with virulence. However, strains missing a specific MLVA locus showed significantly less severe lesions and lower numbers of bacteria. MLVA and PCR-RFLP analyses also showed a high diversity among field isolates of M. hyopneumoniae with a greater homogeneity within the same herd. Almost half of the field isolates presented less than 55% homology with selected vaccine and reference strains.  相似文献

While LiDAR-based digital elevation models (DEM) are more accurate and precise than the USGS-sourced DEM that are widely used in soil mapping in the US, their high cost and other problems prohibit an easy decision of adopting them in service-oriented soil mapping conducted by a government agency like USDA-NRCS. This study compares the performances of LiDAR-based DEM and the USGS-sourced DEM in calculating slope gradient as an input for knowledge-based digital soil mapping (KBDSM), aiming to provide scientific evidence and more importantly, propose a scientific approach to evaluating the two types of DEM for KBDSM. We conducted the comparison by evaluating how closely the DEM-based slope gradient values match the field-measured values. For a small watershed in northern Vermont, US, we prepared three DEM, including a 10-m DEM interpolated from the 7.5-minute USGS topographic map, a 1-m DEM based on LiDAR points, and a 5-m DEM resampled from the 1-m DEM. When calculating slope gradient, we applied two neighborhood sizes (10 m and 30 m), two neighborhood shapes (square and circular), and three slope gradient algorithms (Evans–Young, Horn, and modified Zevenbergen–Thorne) to the three DEM. We then compared the calculated slope gradient values with the values measured by soil scientists at 159 sample locations in the study area. Statistics show that across all the tested settings, the LiDAR-based DEM perform significantly better than the USGS-sourced DEM. We conclude that LiDAR-based DEM may considerably improve the quality of inputs for KBDSM. We also find that the results from the 1-m LiDAR-based DEM and the resampled 5-m DEM do not show considerable and consistent differences.  相似文献

Common wheat adulteration of durum wheat pasta was quantified using real‐time duplex polymerase chain reaction (PCR). The total DNA content of pasta was determined by amplifying part of a wheat gene encoding a lipid transfer protein, and common wheat DNA was quantified by amplifying part of the puroindoline‐b gene. Under the conditions defined by this study, for pasta with a theoretical adulteration of 3%, the experimentally determined mean value was 2.6–3.4%, depending on drying temperature. Pure durum wheat pastas were distinguished from adulterated pastas without ambiguity. This study demonstrates the feasibility of using real‐time duplex PCR to quantify common wheat adulteration of pasta dried at high temperature, quantification that was impossible with the French official peroxidase‐marker method.  相似文献
