Proteomic analysis of mare uterine flush fluid provides a minimally invasive technique for studying protein changes associated with the oestrous cycle. The aim of this study was to identify differentially abundant proteins in the uterine flush fluid of mares in oestrus and dioestrus. In this study, uterine flush fluid samples were collected from eight reproductively healthy mares in either oestrus (n = 5) or dioestrus (n = 3). Proteomic analysis was performed using liquid chromatography‐tandem mass spectrometry. Of 172 proteins identified, six proteins (immunoglobulin lambda‐like polypeptide 1, haemoglobin subunit alpha, alpha‐1B‐glycoprotein, serotransferrin, apolipoprotein A‐1, and haemoglobin subunit beta) were significantly more abundant in oestrus. These proteins may contribute to the endometrial defence system through roles in inflammation, immunity or antimicrobial activity. In other species, some of these proteins have been described as immunoglobulins, negative acute phase proteins or defence agents against micro‐organisms. During dioestrus, immunoglobulin alpha‐1 chain C region‐related, complement factor I, CD 109 antigen and uterocalin, were significantly more abundant. Research in other species suggests that these four proteins contribute to the immune response through proposed immunoregulatory characteristics, complement system involvement or roles in B cell–T cell interactions. In conclusion, ten differentially abundant proteins were identified in the uterine flush fluid of mares in oestrus and dioestrus. Targeted studies on these proteins could elucidate their role in uterine defence mechanisms during the oestrous cycle in the mare.  相似文献

The aim of this study was to estimate the current level of inbreeding in the German cow population and for bull dams born in Germany, to find out sires most related to different subsets of their breed and to demonstrate the negative effect of homozygosity in the case of complex vertebral malformation (CVM). Further on, the application of optimum genetic contribution (OGC) theory for the selection of bull dams and bull sires in different breeding scenarios was investigated. Levels of inbreeding for the cow population were in a low range from 0.97% to 1.70% evaluating birth years from 1996 to 1999 in a total dataset of 244,427 registered Holstein cows. The inbreeding coefficient of 8030 bull dams was much higher, i.e. 3.71%, for the birth year 1999. Increases in inbreeding of 0.19% per year indicated an effective population size of only 52 animals. Individual sires like R.O.R.A. Elevation and Hannoverhill Starbuck were highly related to potential bull dams with coefficients of relationship of 13.4% and 12.9%, respectively, whereas P.F. Arlinda Chief (16.3%) and Carlin-M Ivanhoe Bell (16.1%) were highest related to the best available AI sires. Coefficients of relationship were calculated by classes of estimated breeding values (EBV) for production traits showing highest values above 7% in the two highest EBV-classes. The optimum genetic contribution theory using official EBVs and approximative, for zero inbreeding corrected EBVs, was applied for elite matings in a breeding program embracing 30 young bulls per year to find the optimal allocations of bull sires and bull dams. Compared with the actual breeding program applied in practice, OGC-theory has the potential to increase genetic gain under the same constraint for the increase of average relationship by 13.1%. A more relaxed constraint on increase in inbreeding allowed even higher expected genetic gain whereas a more severe constraint resulted in more equal contributions of selected bull sires. Contributions from 21 selected bull sires and 30 selected bull dams for a scenario at 5% constrained relationship were used to develop a specific mating plan to minimise inbreeding in the short term in the following generation applying a simulated annealing algorithm. The expected coefficient of inbreeding of progeny was 66.3% less then the one resulting from random mating. Mating programs can address inbreeding concerns on the farm, at least in the short term, but long-term control of inbreeding in a dairy population requires consideration of relationships between young bulls entering AI progeny test programs. Significantly better EBVs of CVM-free bulls compared with CVM-carriers for the paternal fertility justify the application of OGC for elite matings.  相似文献

Although livestock experience many stressors throughout their life, one of the most commonly experienced, and most difficult to control, is stress caused by fluctuations in environmental temperatures that extend beyond the thermoneutral (TN) zone for an animal. In swine, cold stress has long been recognized as a main cause of neonatal morbidity and mortality. A possible explanation for this increased morbidity and mortality may be related to their inability to generate a febrile response. Previously, we reported that the acute phase immune response, including the generation of fever, after exposure to lipopolysaccharide (LPS; Escherichia coli O111: B4; Sigma-Aldrich, St Louis, MO, USA) is substantially altered in neonatal pigs maintained in a cold environment (ie, 18°C). Neonatal pigs that were maintained in a cold environment and administered LPS experienced a period of hypothermia coupled with altered endocrine and proinflammatory cytokine responses that could prove detrimental. In cattle, we previously reported differences in the acute phase immune response of two diverse breeds of Bos taurus cattle (Angus and Romosinuano) when maintained under TN conditions and exposed to LPS. More recently we have reported that differences in the stress and immune responses of Angus and Romosinuano heifers varies, depending on whether the cattle were housed at either TN or heat stress air temperatures. Our data clearly show that even intermittent periods of heat stress similar to that experienced in production environments can have significant effects on the stress and innate immune responses of cattle. Understanding the effect of thermal stress on livestock is critical to developing and implementing alternative management practices to improve their overall health and well-being.  相似文献

Genetics of resistance to soybean cyst nematode (SCN), Heterodera glycines Ichinohe is very complex. Crosses involving PI 437654, which is resistant to all races of cyst nematodes with other sources of resistance (Peking, PI 88788, and PI 90763) indicated that resistance to race 3 was controlled by four genes, two of which were dominant resistance genes and the other two were recessive resistance genes. For race 5, a four gene model with two recessive and two dominant resistance genes in epistasis has been proposed. For race 14, the results suggested a three gene model with one dominant and two recessive alleles. Several other plant introductions have been isolated which have different genes conditioning resistance. Most of the currently grown soybean varieties derived resistance from Peking and/or PI 88788. Resistance to SCN in these soybean varieties has broken down because of the emergence of several new races and populations of SCN. The use of PI 437654 or Hartwig and other plant introductions with different genes for resistance will broaden genetic diversity and stabilize yield.  相似文献
