We carried out this study to determine for how long small ruminant lentivirus (SRLV)‐specific antibodies can be detected by three commercial ELISA kits in goat kids after suckling infected does in field conditions. Forty‐one kids born to SRLV‐seropositive asymptomatic does were blood sampled prior to colostrum consumption, and then weekly for 6 months in total. The sera were screened with three commercial ELISA kits: whole‐virus ELISA (wELISA), recombinant transmembrane and capsid antigen ELISA (TM/CA‐ELISA), and surface antigen ELISA (SU‐ELISA). All but one kid were seronegative in all three ELISAs right after birth. At the age of 1 week all kids turned seropositive in wELISA, 39 kids (95%) in TM/CA‐ELISA, and 35 kids (85%) in SU‐ELISA. All seropositive kids turned seronegative in wELISA by the 15th week, and in SU‐ELISA by the 19th week (median of 8 weeks in both ELISA), whereas in TM/CA‐ELISA five kids (13% of 39 initially seropositive) were still seropositive at the age of 6 months (median of 11 weeks). Antibody levels at the age of 1 week proved significantly linked to the duration of maternal antibodies in all three ELISAs and could be employed to predict for how long maternal antibodies would remain detectable. 相似文献

In the presented study we evaluated the hematological changes in samples of blood obtained from 248 dogs naturally infected with large Babesia. The evaluation included red blood cell count, hemoglobin concentration, hematocrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC), leucocyte counts, thrombocyte counts, mean platelet volume (MPV), morphology of erythrocytes and leucogram. The most common disorders in affected dogs were thrombocytopenia and anisocytosis. The count of erythrocytes below reference values was detected in 26.2% of dogs and 31.4% of affected animals presented hematocrit below the reference values. Hemoglobin concentration below the reference values was noted in 29% of dogs, an increase of MCHC above normal values was detected in 21% of examinated dogs and MCV below normal values was recognized in 2% of dogs. 60.5% of dogs presented anisocytosis, 25% poikilocytosis, 23.8% polychromasia, 19.7% hypochromia and 4.4% erythroblastosis. Thrombocytopenia was detected in 99.5% of dogs, but only 15.3% of examined animals showed increase of MPV, which suggests a response of the bone marrow. 36.3% of dogs had neutropenia, and 21.8% presented a left shift, 14.9% had the lymphocytosis and 7.2% lymphopenia. 相似文献

Female mutant restricted ovulator (RO) chickens of the White Leghorn strain carry a naturally occurring single nucleotide mutation in the very low density lipoprotein receptor (VLDLR) gene. Due to this mutation, RO hens fail to express a functional VLDLR protein on the oocyte membrane, which results in an impaired uptake of circulating yolk precursor macromolecules. Mutant RO hens subsequently develop hyperlipidemia and generally fail to lay eggs due to follicular atresia. In the dog, the blood from the antebrachial part of the cephalic vein is typically drained by three vessels: the axillobrachial, the omobrachial and the proximal segment of the cephalic vein. In this work, two anatomical variations affecting the omobrachial and the proximal part of the cephalic vein in dogs are reported. The bilateral absence of the omobrachial vein was observed in two adult mongrel dogs and the lack of the proximal segment of the cephalic vein in one dog. These differences suggest that a certain degree of variation is normal in the layout of the superficial veins of the thoracic limb in the dog and implies that the blood egress from the cephalic vein in the dog, as in other species, is sometimes accomplished by only two veins. 相似文献

- A review of the long‐term changes and variations in benthic communities and the current status of the marine invasive species (MIS) in shallow waters of the Yellow Sea (Chinese sector) and the Sea of Japan (Russian and partly Korean sectors) is presented. This paper reflects on the progress and lessons learned, recommending actions for the future about the conservation of biodiversity.
- In the Bohai Sea, the benthic ecosystem has been degenerating due to anthropogenic activities such as overfishing and pollution since the 1950s. The dominant position of K‐strategy species is gradually being lost and replaced by R‐strategy species. In the Yellow Sea, the macrobenthic community is different from other areas due to the Yellow Sea Cold Water Mass. Many economic species have been destroyed, and the biotic structure has changed significantly due to overfishing and climate change.
- In the Russian sector of the Sea of Japan, the macrobenthic communities in the shallow‐water soft bottom have generally been in a stable condition for the last decades, except for some heavily polluted or disturbed areas due to dredging operations. The abundance of select large invertebrate species has changed considerably due to commercial fishing and poaching. Variations in macro? and meiobenthic communities under aquaculture conditions have occurred on a local scale during the last five decades.
- MIS show obvious differences between China and Russia in the following aspects: introduction pathways of MIS, composition and number of non‐native species, threats and impacts of MIS to native communities and ecosystems, and economic and public health impacts.
- Long‐term monitoring programmes should be developed to reveal future biotic changes and to separate the effects of cyclic variations of benthic communities from the impacts of pollution and eutrophication. Standardization of sampling procedures is required to compare changes/alternations in benthos across various regions worldwide.

Many human cancers involve up-regulation of the phosphoinositide 3-kinase PI3Kalpha, with oncogenic mutations identified in both the p110alpha catalytic and the p85alpha regulatory subunits. We used crystallographic and biochemical approaches to gain insight into activating mutations in two noncatalytic p110alpha domains-the adaptor-binding and the helical domains. A structure of the adaptor-binding domain of p110alpha in a complex with the p85alpha inter-Src homology 2 (inter-SH2) domain shows that oncogenic mutations in the adaptor-binding domain are not at the inter-SH2 interface but in a polar surface patch that is a plausible docking site for other domains in the holo p110/p85 complex. We also examined helical domain mutations and found that the Glu545 to Lys545 (E545K) oncogenic mutant disrupts an inhibitory charge-charge interaction with the p85 N-terminal SH2 domain. These studies extend our understanding of the architecture of PI3Ks and provide insight into how two classes of mutations that cause a gain in function can lead to cancer. 相似文献

The contribution of local knowledge to ecological sciences has not been fully exploited: there is still a gap between the recognition of farmers' knowledge as valid and an effective use of such knowledge in activities aimed at sustainable development. This study explores the use of farmers' indicators of erosion for developing a rapid tool for water erosion assessment at field level in the West Usambara Mountains (Tanzania). Two extensive field surveys were conducted in the research area concurrently. One survey consisted of applying an established erosion assessment method, the Assessment of Current Erosion Damage (ACED). According to the erosion features observed, fields were classified into five erosion classes, from very slightly eroded to very severely eroded. The second survey consisted of recording the type and number of indicators of erosion listed by farmers and present in the fields. The number of farmers' indicators per field increased with erosion intensity, from less than four in slightly eroded fields to more than eight in severely eroded fields. All farmers' indicators were positively correlated to the ACED erosion assessment classes. However, two groups of farmers' indicators could be distinguished in terms of erosion assessment: strong indicators, which were observed in more than 70% of cases in severely eroded fields, and weak indicators, which were observed more frequently in slightly and moderately eroded fields. Weak indicators appeared to be indicative of other land degradation phenomena, such as chemical fertility decline. Strong indicators and number of indicators were used to create a field erosion assessment tool in the form of a classification tree. The tree was built using one half of the field survey data and validated using the other half. The tree consisted of a hierarchical sequence of questions. Presence of rills and number of farmers' indicators were the most important factors of the tree. The validation yielded a highly significant Spearman's rho correlation coefficient (0.81). The contingency table showed that more than 80% of very severely eroded fields were correctly classified, whereas most misclassification occurred among slightly and moderately eroded fields. Farmers include land degradation phenomena and soil fertility decline in their definition of soil erosion. Soil and water conservation planning should address this broader farmers' perception by including, e.g., soil fertility improvements beside soil conservation. The distinction between strong and weak indicators of erosion is important in recommending the right intervention in the right spot, e.g., by counteracting soil erosion where strong indicators are present and by improving chemical fertility where weak indicators are present. The classification tree is of empirical nature and may need adaptation before being applied to other areas. The proposed methodology can be easily replicated and showed a high potential to provide extensionists with a field tool for erosion assessment. The classification tree was a successful example of integrating different types of knowledge for enhancing the co-operation between stakeholders involved in the erosion-control activities. 相似文献

We report results of new research on (1) community composition of novel subtropical dry forests developing on abandoned pastures and agricultural fields in both private and protected public lands and (2) seed germination and growth rates of plantings of native tree species on degraded soils. We found that novel dry forests were dominated by introduced species, which accounted for 59 percent of the Importance Value (IV) of stands. These forests had high species dominance, with the most dominant species averaging 51 percent of the IV but reaching values as high as 92 percent. The floristic similarity between novel and mature native forests was low (5.6 percent) compared with the similarity among novel forest stands (26 percent). Collectively, the emerging novel forests had relatively high species richness (39 species/1.2 ha). After 45-60 years of growth and development, novel forests lagged mature native forests in basal area, tree density, and species richness, and lagged stands of similar age and past land use inside a protected area. Novel forest stands inside the protected Guánica Forest had higher species richness than those located outside in private lands. Most regeneration was from seed (67 percent of the new stems were single stems). The results from the germination and planting experiment show that seeds of 17 of 21 native tree species germinated in the laboratory and grew successfully in abandoned pastures when planted and watered for a period of 13 months. Our research shows that after the initial invasion and dominance of introduced species on degraded sites, the stands diversify with native species thus evolving towards new forest types with novel species combinations. 相似文献
