Primary cilia (PC) were demonstrated for the first time in fish endothelial, epithelial and fibroblast cell lines through immunofluorescence staining with the monoclonal antibody, 6-11B-1, against acetylated α-tubulin. The study was carried out with eight recently developed cell lines from the walleye, Sander vitreus (Mitchill). These were three fibroblast-like cell lines, WE-cfin11f, WE-skin11f and WE-liver3 from, respectively, the caudal fin, skin and liver, and three epithelial-like cell lines, WE-cfin11e, WE-spleen6 and WErpe from, respectively, the caudal fin, spleen and retina. Also, endothelial-like WEBA from the bulbus arteriosus and glial-like WE-brain5 from the brain were used. Immunocytochemistry revealed strong staining for acetylated α-tubulin in mitotic spindles and midbodies for all cell lines, and in PC for all cell lines except WE-skin11f. Staining of cytoplasmic microtubules (fibrils) was absent in three cell lines, including WEBA, but present in the others, especially WE-skin11f, which might have obscured PC detection in these cells. Tubacin, an inhibitor of histone deacetylase 6, induced cytoplasmic fibrils in WEBA and the intensity of their staining in WE-cfin11f. These results suggest that the cell lines might differ in their deacetylase activities, making them useful for studying this tubulin modification in teleosts, as well as for studying PC. 相似文献

Artificial insemination (AI) of sows results in a significant elevation of prostaglandin F(2α) metabolite (PGFM) levels in peripheral plasma, whereas in mated sows such elevation is not seen. The aim of this study was to investigate whether boar seminal plasma (SP) has any effect on the release of PGFM, prostaglandin F(2α) (PGF(2α) ), prostaglandin E(2) (PGE(2) ) or interleukin-6 (IL-6) by in vitro cultured porcine endometrial (epithelial - pUE and stromal - pUS), cervical (pCE and pCS) and bovine endometrial epithelial cells (bUE). This study shows that boar SP inhibits the release of PGFM, PGF(2α) and PGE(2) by porcine endometrial and cervical cells and bovine endometrial cells after 3 and 24 h incubation. Boar SP stimulated IL-6 release by pUE, pUS and even bUE after 3 h incubation. Tumour necrosis factor α (TNFα) stimulated the release of IL-6 by pUS only after 24 h incubation, but in the presence of boar SP, this stimulation was attenuated. The overall results from these in vitro studies give us possibility to understand the difference in prostaglandin response between mated and inseminated sows. Furthermore, we demonstrated that frozen-stored epithelial and stromal cells from pig endometrium, as well as from the cervix are suitable for studying the effect of SP on the release of prostaglandins. The only prerequisite is to incubate these thawed cells with arachidonic acid as a source for the synthesis of prostaglandins. ABSTRACTThis study examined if dwarf French bean (Phaseolus vulgaris L.) grown at northern latitudes develop seeds with superior yielding potential compared with plants grown at more southern latitudes. Two lines (BND3085 and BND3106) were grown at four sites across latitudes ranging from 52°N to 3°S. BND3085 and BND3106 seeds from Saskatoon grown plants (52°N) had the least damaged seed coats but the highest degree of electrolyte leakage from the seed and the lowest maximum germination rates. The Saskatoon (BND3085 and BND3106) and Nunhem (BND3106) seed lots had the lowest maximum germination rates when germinated at 28°C, 12°C, or when exposed to saline (100?mM NaCl) conditions. Saskatoon BND3085 seed produced plants with the lowest aboveground vegetative and total seed dry matter as well as the lowest pod number per plant. In contrast, plants grown from Saskatoon BND3106 seed produced 1.5-fold more total pods, 1.3-fold more total pods and 1.4-fold higher seed dry matter per plant as compared to plants grown from the Nunhem (51°N) or Arusha seed sources. This resulted in a 2.4–4.5% higher harvest index. These results suggest cultivar-specific dwarf French bean seed propagation at more northern sites may increase the productivity of the resulting crop. 相似文献

The self-fertility of chromosome-doubled haploid lines of perennial ryegrass and their first (I1) and second (I2) generation selfed offspring was studied. The overall seed set was low, 0—0.4 seeds per spike, as would be expected for chromosome-doubled haploids of a self-incompatible species. Nevertheless, significant genetic variation was observed for self-fertility within three of the six lines studied. Within one line, three I2-clones showed increased self-fertility, while in two other lines one and three I1-clones, respectively, showed significantly increased self-fertility. Selection, during selfing, of rare pollen grains with mutations in the self-incompatibility system could explain the observed changes and would lead to a general breakdown of self-incompatibility in Lolium perenne during repeated selfing of chromosome-doubled haploids. 相似文献

The feasibility of large-scale bloom induction of nutritionally suited natural phytoplankton species, to feed a semi-industrial nursery of edible shellfish, built on the Belgian coast, was tested.The outdoor microalgal production unit consisted of four tanks of 100 m2 surface each (two of 1 m depth and two of 0·5 m depth), equipped with different mixing devices.The cultures were run as chemostats in which seawater was enriched with commercial inorganic N, P, and Si fertilizers. Depending on the season, between 5–10 and 80% of the culture volume could be harvested daily, with algal densities ranging from 50 000 to 500 000 cells per ml.By manipulation of operational parameters such as detention time, nutrient levels and nutrient ratios (N:Si:P), unsuited or less suited species of algae (e.g. Chlorella and Phaeodactylum) could be replaced by more desirable species (e.g. Skeletonema, Nitzschia, Chaetoceros).Various biological and technological problems encountered during year round operation, including collapsing of the culture, seawater enrichment, water quality, fouling and water treatment, are commented. 相似文献

Little information is available regarding the landscape ecology of woodland invertebrate species with limited dispersal ability.
An investigation was therefore conducted within woodland fragments in an agricultural landscape for the flightless wood cricket
(Nemobius sylvestris) on the Isle of Wight, UK. The current pattern of distribution of the species, established during a field survey, was related
to measures of habitat availability and habitat isolation/fragmentation. Results revealed that wood cricket populations were
patchily distributed and mainly found in relatively large mature woodland fragments situated closely (<50 m) to another occupied
site. Although the occurrence of wood cricket was related to fragment area, isolation, habitat availability and woodland age,
a logistic regression model revealed that presence of the species was most accurately predicted by fragment isolation and
area alone. These results highlight the vulnerability of relatively immobile woodland invertebrate species, such as wood cricket,

Tugai forests are the riparian forests along the rivers in thecontinental desert regions of Central Asia, i.e. the Tarim River,Amu Darya and Syr Darya. They mainly consist of Populus euphraticaOliv., Populus pruinosa Schrenk. and Elaeagnus oxycarpa Schltdl.As a consequence of land opening campaigns, large areas of Tugaiforests were destroyed after the 1950s. Due to excessive useof water for irrigation, the remaining Tugai forests are undersevere threat. Near natural Tugai forests still exist alongthe Tarim middle reaches in the Tarim Huyanglin Nature Reserve,Xinjiang, NW China. There is a gap in understanding, how theseedlings of P. euphratica establish as trees which continuouslyconnect to the groundwater. Therefore, the set of conditionswhich must be met for germination and successful establishment,i.e. formation of Tugai forests, was investigated along a representativetransect still under natural conditions. P. euphratica seedlingsgerminate in belts during the retreat of the summer flood onfreshly deposited sites bare of other vegetation. Such germinationsites are formed by river dynamics. While germination takesplace regularly in the study area, successful establishmentis restricted to few germination events. Seedlings face droppinggroundwater levels during spring and early summer of the secondyear after germination. Therefore, for successful establishment,it is essential that the flood of the second year starts intime and is high enough, in order to replenish the groundwater.Furthermore, clayey soil layers in the subsoil may play a rolefor successful establishment, too, as they store water betterthan sandy soil layers. 相似文献

The need to integrate non-market ecosystem services into decision-making is widely acknowledged. Despite the exponentially growing body of literature, trade-offs between services are still poorly understood. We conducted a systematic review of published literature in the Nordic countries (Denmark, Norway, Sweden and Finland) on the integration of non-market forest ecosystem services into decision-making. The aim of the review was two-fold: (1) to provide an overview of coverage of biophysical and socio-economic assessments of non-market ecosystem services in relation to forest management; (2) to determine the extent of the integration of biophysical and socio-economic models of these services into decision support models. Our findings reveal the need for wider coverage of non-market ecosystem services and evidence-based modelling of how forest management regimes affect ecosystem services. Furthermore, temporal and spatial modelling of ecosystem impacts remains a challenge. We observed a few examples of multiple non-market services assessments. Integration of non-market services into decision support was performed with either biophysical or socio-economic models, often using proxies and composite indicators. The review reveals that there is scope for more comprehensive and integrated model development, including multiple ecosystem services and appropriate handling of forest management impacts. 相似文献

BACKGROUND: Surfactants are very often used for more efficient pesticide spraying, but knowledge about their influence on the leaching potential for pesticides is very limited. In the present study, the leaching of the herbicide bentazone [3‐isopropyl‐1H‐2, 1,3‐benzothiadiazin‐4(3H)‐one 2,2‐dioxide] was measured in columns with sandy loam soil with or without the addition of a non‐ionic surfactant, octylphenol ethylene oxide condensate (Triton X‐100, Triton), and an anionic surfactant, sodium dodecylbenzenesulfonate (SDBS), and in the presence of both surfactants (SDBS + Triton). RESULTS: The mobility of bentazone (B) increased in the following order: B + Triton (slowest) < B + SDBS + Triton < B < B + SDBS (fastest). When Triton X‐100 was applied to the soil together with bentazone, the leaching of bentazone in the soil decreased significantly compared with leaching of bentazone without the addition of surfactant. SDBS and Triton X‐100 neutralised their influence on the leaching speed of bentazone in the soil columns when both surfactants were applied with bentazone. CONCLUSION: From the study it can be concluded that, depending on their properties, surfactants can enhance or reduce the mobility of bentazone. By choosing a non‐ionic surfactant, bentazone mobility can be reduced, giving time for degradation and thereby reducing the risk of groundwater pollution.
