The aim of this study was to determine the blood flow in the carotid artery during halothane anaesthesia by means of duplex-sonography. Seventeen warm blood horses were evaluated in dorsal and lateral recumbency and the results were compared with the values of the same horses resting and under sedation. The cross sectional area of the vessels, the time-averaged maximal blood velocity, time-averaged maximal blood flow and the resistance index were determined and the flow profile was evaluated. During halothane anesthesia the total blood flow shows a significant increase which is not dependent on the positioning of the horse. Mean blood velocity is decreased by sedation and significantly increased during anesthesia. Reduced peripheral resistance is expressed by a decline of the resistance index. After sedation and during recumbency there is an increase in diameter of the carotid artery. 相似文献

A deterministic–stochastic combined model named HydroGEN was developed, as described in a companion paper (Part I: Model development), to enable the simulation of demanded daily volumes and hourly flow rates during peak periods in pressurized irrigation delivery networks. The model was applied to a pilot large-scale irrigation system located in southern Italy for calibration and for testing its reliability in analyzing the operation of large-scale pressurized delivery systems through the simulated flow configurations. Daily input data on rainfall, temperature, solar radiation, wind speed and relative humidity were gathered from a meteorological station located within the study area, whereas information on local irrigation management practices were collected through interviews with farmers and from extension specialists. The model was tested at different management levels, from district to sector and hydrants. The model testing was supported by the use of high-resolution remote-sensing imagery acquired on a single overpass date in 2006 and then classified and recoded following a ground-truthing campaign conducted during the same year. Simulations were performed to identify the 10-day peak-demand period and to generate the hydrographs of daily volumes and of hourly flow rates. Results from the different simulations were compared with historical datasets of irrigation volumes and discharges recorded during the 2008 and 2009 seasons at the upstream end of the irrigation network under study, at a sector level during the 2007 season and at selected delivery hydrants during the 2005 season. Some discrepancies between simulated and recorded data were noted that can be related to small errors in estimating crop and soil parameters, application efficiency at field level, as well as to large variability in irrigation management practices followed by local farmers. Overall, the results from testing showed that the model is capable of forecasting with good accuracy the timing of peak-demand periods, the irrigation volumes demanded during the season, as well as the hydrographs of daily volumes and hourly flow rates withdrawn by farmers during these peak-demand periods, especially when it is applied to large multi-cropped command areas. 相似文献

The objective of the current study was to determine the sensitivity and specificity of real-time polymerase chain reaction (real-time PCR) for feline hemoplasmas when applied to DNA extracted from dried whole-blood smears in comparison to that for DNA extracted from liquid whole blood. Blood samples were collected into ethylenediamine tetra-acetic acid tubes from 305 cats with possible or suspected hemoplasmosis, and dried blood smears from each sample were prepared. DNA was extracted from blood smears and a 160-microl aliquot of each liquid blood sample by using a robotic extractor and was subjected to real-time PCR for feline glyceraldehyde-3-phosphate dehydrogenase (liquid blood), 18S ribosomal RNA (dried blood), and "Candidatus Mycoplasma haemominutum", Mycoplasma haemofelis, and "Candidatus Mycoplasma turicensis" DNA. When using the results for liquid whole blood as the gold standard, the sensitivity of each assay for "Ca. M. haemominutum", M. haemofelis, and "Ca. M. turicensis" was 49 of 66 (74%), 11 of 13 (85%), and 11 of 20 (55%), respectively. The specificity of each assay was 224 of 234 (96%), 287 of 287 (100%), and 280 of 280 (100%), respectively. When possible, liquid blood samples should be submitted for detection of feline hemoplasmas by using real-time PCR. The improved sensitivity of real-time PCR on blood smears for M. haemofelis compared with that of the other hemoplasma species may reflect the higher organism burdens associated with infection with this species. 相似文献

A 13-year old Thoroughbred cross gelding (528 kg) underwent lameness investigation. Because of his temperament, general anesthesia was required to facilitate ultrasound of the left fore fetlock and intra-articular medication of three joints. Anesthesia was induced with ketamine/diazepam after acepromazine/detomidine premedication. Anesthesia was maintained for 40 minutes with a guaifenesin/detomidine/ketamine intravenous infusion. Recovery from anesthesia was initially uneventful, although of a moderate duration (70 minutes). Once standing, the horse proceeded to box walk in an agitated state and became recumbent on two occasions. The horse was manually restrained, at which time its rectal temperature was 41.8°C. Cooling measures were employed (fans, ice-water enemas, wet rugs, intravenous fluid therapy (IVFT), and topical application of surgical spirit) until rectal temperature reached 38.7°C. IVFT was continued for a further 16 hours. Four days after recovery from anesthesia, bilateral triceps, deltoideus, trapezius, and rhomboideus muscle swelling was observed. Blood creatinine kinase was elevated (24,898 IU/L). Treatment for postanesthetic myopathy was initiated (hot packing of the muscle groups, topical dimethylsulfoxide [DMSO] cream application, and oral phenylbutazone). Myoglobinuria was not observed at any time. Muscle swelling decreased over the following 3 days. The horse was discharged on day 11 and has since returned to work. 相似文献

A neoplasm involving the ileo-cecal-colic junction, thymus, and tracheobronchial lymph nodes of a 7-year-old domestic cat was composed of dense sheets of round to oval mononuclear cells with oval to indented nuclei, moderate amounts of cytoplasm, and variable numbers of round eosinophilic granules. These granules are brown to black in phosphotungstic acid hematoxylin-stained sections and stain variably with the periodic acid-Schiff stain. They are 0.8 to 1.5 micron in diameter, limited by a single unit membrane, and have variable electron density. Light microscopic cellular morphology and staining characteristics as well as ultrastructural features of these cells are consistent with feline globule leukocytes. Morphologic features of the neoplastic cells in the present case are similar to those of the only other reported neoplasm of globule leukocytes which also involved the intestine of a cat. 相似文献

Starch structure and functionality have a significant impact on the utilization of cereal grains as food and feed. Starch
viscosity characteristics are used to characterize rice cooking, processing and eating quality. In order to examine the genetics
of viscosity characteristics, we developed molecular markers for five of the major enzymes involved in starch synthesis in
the endosperm: granule bound starch synthase, soluble starch synthase, rice branching enzymes 1 and 3 and starch debranching
enzyme. These markers were polymorphic in a cross between specialty rice varieties of diverging amylose content and viscosity
characteristics. Our results indicate that the Waxy locus, encoding the gene for granule bound starch synthase, has a significant effect on peak viscosity, hot paste viscosity,
cool paste viscosity, breakdown and setback viscosity. We estimate that the tightly linked (5–10 cm)locus for starch synthase
may have a lesser, additive effect.

Multienoic fatty acids, such as linolenic acid, show their ability to interact with and to penetrate into model biomembranes by biomimetic experiments performed to support the absorption route followed by n-3 fatty acid in cells. The thermotropic behavior of model biomembranes, that is, dimyristoylphosphatidylcholine multilamellar or unilamellar vesicles, interacting with linolenic acid was investigated by differential scanning calorimetry. When dispersed in liposomes during their preparation, the examined biomolecule was found to interact with the phospholipid bilayers by modifying the gel to liquid-crystal phase transition of lipid vesicles; this modification is a function of the fatty acid concentration. Calorimetric analysis was also performed on samples obtained by leaving the pure n-3 acid in contact with lipid aqueous dispersions (multilamellar or unilamellar vesicles) and then examining the thermotropic behavior of these systems for increasing incubation times at temperatures higher than the transitional lipid temperature. Linolenic acid (LNA) was able to migrate through the aqueous medium and successively to interact with the vesicle surface and to penetrate into the model membranes, following a flip-flop mechanism, with a faster and higher effect for unilamellar vesicles, caused by the larger lipid surface exposed, compared to the multilamellar ones, although due to the lipophilic nature of LNA, such a transfer is hindered by the aqueous medium. The relevance of the medium in LNA absorption has been well clarified by other biomimetic transfer experiments, which showed the LNA transfer from loaded multilamellar vesicles to empty vesicles. Taken together, the present findings support the hypothesis of a passive n-3 acid transport as the main route of absorption into cell membranes. 相似文献

BACKGROUND: Predatory mites (Amblyseius swirskii Athias‐Henriot, Typhlodromips montdorensis Schicha, Neoseiulus cucumeris (Oudemans) and Iphiseius degenerans Berlese) were investigated for their potential to act as control agents for Thrips palmi Karny. Prey consumption rates and compatibility with pesticides were assessed. RESULTS: Second‐instar larvae were the preferred life stage. Typhlodromips montdorensis consumed the most larvae (2.8) and also an average of 1.2 adult T. palmi per 5 day period. Both 24 and 48 h assessments following application of abamectin, spinosad and imazalil demonstrated mortality of predatory mites (across all species), which was significantly higher than with the other treatments (P < 0.001). Spraying with pymetrozine did not provide any increased mortality when compared with the water control. Application of thiacloprid proved detrimental only to I. degenerans. Following indirect exposure of predatory mites to pymetrozine and imazalil, no significant differences in mite mortality were obtained. Indirect exposure to spinosad was identified as the most detrimental treatment (P < 0.001) to all mites. Abamectin also proved detrimental, with only T. montdorensis showing any potential tolerance. CONCLUSION: All predatory mites investigated offer potential for controlling T. palmi. Compatibility with chemicals varied between the mites.
