A colorimetic assay for lipoxygenase activity has been developed. The assay is based on the detection of the lipoxygenase reaction product, linoleic acid hydroperoxide, by the oxidative coupling of 3-methyl-2-benzothiazolinone (MBTH) with 3-(dimethylamino)benzoic acid (DMAB) in a hemoglobin-catalyzed reaction. This test reaction is rapid and sensitive, and it offers advantages over other methods for detecting lipoxygenase activity. The assay is capable of detecting activity in a number of crude vegetable homogenates and should be particularly useful where a rapid visual determination of lipoxygenase activity is desired.  相似文献

Mineralization of paraprostatic cysts is reported to be uncommon. This retrospective study was performed to review the imaging findings of eight histologically confirmed canine paraprostatic cysts. Radiographic patterns of mineralization seen are described. Four of the eight dogs had mineralized cysts. Mineralization seen on radiography was not consistently seen on ultrasonography, probably due to the lack of acoustic shadowing artefact from the small depth of mineralized tissue. It is thus concluded that mineralization of paraprostatic cysts is more common than implied in the literature, and that radiography is superior to ultrasonography in identifying its presence.  相似文献

Radiographic flexor cortical lysis indicates advanced degenerative change and its earlier recognition may improve case outcome. Aims of this prospective, diagnostic accuracy study were to determine effects of radiographic beam angle and observer on accuracy of lesion detection. The sample included 36 limbs from 31 horses. Palmaroproximal–palmarodistal oblique (skyline) radiographs were acquired at standard (n = 38) and more shallow (alternate) angles (n = 58). Images were independently reviewed by four experience levels of five observers each (n = 20) for the presence and severity of flexor cortical lysis. Observers also reported their confidence in these answers. Responses were compared based on seeing a standard skyline or multiple projections. The definitive presence (or absence) and severity of lysis was based upon radiologist consensus agreement. When assessed by observer, the identification of lysis and the assessment of its severity was most similar to that of radiologists when observers of all levels of experience were able to view multiple skyline projections (P = 0.399, P = 0.174). Using multiple views to detect lysis resulted in improved sensitivity (85.3% vs. 97.2%, P < 0.001), decreased specificity (82.8% vs. 74.5%, P = 0.03), and improved interobserver agreement (86.0% vs. 90.2%, P = 0.21). On average, observers of all levels of experience became more confident viewing multiple projections (P < 0.001). Skyline radiographs using a flatter angle of incidence improve radiographic detection and characterization of flexor cortical lysis severity, may allow the beam to be tangential to the more distal portion of the navicular bone, and are recommended when flexor cortical lysis is suspected.  相似文献

Toward Heisenberg-limited spectroscopy with multiparticle entangled states   总被引:1,自引:0,他引:1  
The precision in spectroscopy of any quantum system is fundamentally limited by the Heisenberg uncertainty relation for energy and time. For N systems, this limit requires that they be in a quantum-mechanically entangled state. We describe a scalable method of spectroscopy that can potentially take full advantage of entanglement to reach the Heisenberg limit and has the practical advantage that the spectroscopic information is transferred to states with optimal protection against readout noise. We demonstrate our method experimentally with three beryllium ions. The spectroscopic sensitivity attained is 1.45(2) times as high as that of a perfect experiment with three non-entangled particles.  相似文献

  • 1. Mapping of seabed habitats is increasingly being used to identify the distribution and structure of marine ecosystems and as surrogate measures of biodiversity for marine protected area (MPA) planning. In this study, the distribution of seabed habitats to the 3 nmi limit around the Kent Group of islands, south‐eastern Australia were mapped using video ground‐truthed single‐beam acoustics at the mesoscale level (10 m to 1 km) as part of an MPA planning process.
  • 2. Six distinct seabed habitat types (continuous reef, patchy reef, sand, hard sand, sparse sponge, and seagrass) were identified based primarily on visual differences in the first and second echo and a further four (low, medium and high profile reef, and sand hills) on variations in seabed profile identified in the echogram. Extensive acoustic and video transects allowed an estimate of the broad‐scale spatial distribution of seabed habitats defined at several hierarchical levels and provided information on the cover of the dominant benthic species or assemblages.

The island group supports a range of consolidated habitats, including rocky reefs of varying profile dominated by the macroalgae Phyllospora comosa and Ecklonia radiata in depths down to around 45 m, adjacent to deeper sponge‐dominated reefs containing encrusting, erect and branching forms. Unconsolidated habitats occurred broadly through the island group, with the offshore region dominated by hard sand (sand with scallop shells and/or shell grit) and sparse sponge‐habitats (sand interspersed with low cover of sponge‐dominated assemblages). The sheltered coves were dominated by sand and seagrass habitats consisting of beds of the seagrasses Halophila australis, Zostera tasmanica and Posidonia australis, with variations in species composition, patchiness and percentage cover evident within and between coves.

In February 2004 the Kent Group MPA was announced, covering all waters out to the 3 nmi limit containing two areas defined as a Sanctuary Zone (‘no take’) and a Habitat Protection Zone (‘restricted take’). Overall, seabed habitat mapping generated a capability to define the boundary and size of potential MPA zones within the Kent Group of islands and was an essential component of the planning process to improve the likelihood that the MPA was comprehensive, adequate and representative (CAR).
  • 5. The need to define habitats at multiple scales within a hierarchical classification scheme that are meaningful in terms of biodiversity and CAR principles and identifiable using mapping techniques is discussed.

The antioxidant potential of eight clingstone peach cultivars was investigated by determining phenolic compounds and inhibition of low-density lipoprotein (LDL) oxidation. Cultivars low in polyphenol oxidase (PPO) were also selected to minimize enzymatic browning. Inhibition of LDL oxidation varied from 17.0 to 37.1% in peach flesh extract, from 15.2 to 49.8% in whole peach extract, and from 18.2 to 48.1% in peel extract. Total phenols were 432.8-768.1 mg/kg in flesh extract, 483.3-803.0 mg/kg in whole extract, and 910.9-1922.9 mg/kg in peel extract. The correlation coefficient between relative LDL antioxidant activity and concentration of total phenols was 0.76. Peel PPO activity was higher than flesh activity in most cultivars. The lowest PPO and specific activities were found in the Walgant cultivar, followed by Kakamas and 18-8-23. These three cultivars combine the desirable characteristics of strong antioxidant activity, low PPO activity, and lower susceptibility to browning reactions.  相似文献

A liquid chromatographic (LC) method has been developed for the determination of carbadox, desoxycarbadox, and nitrofurazones in the 10-40 ppb range in pork muscle, liver, and kidney tissues. Tissues were homogenized in absolute ethanol, and the homogenates were treated with metaphosphoric acid and reduced in volume by rotovaporization. Hexane was added to the concentrates, which were then centrifuged to remove fat. After addition of KH2PO4 to the aqueous phase and extraction with ethyl acetate, the extracts were passed through alumina columns before analysis by reverse phase LC. Overall average recoveries (10-40 ppb range) for carbadox and desoxycarbadox from spiked tissues were 53% +/- 13.6 and 61% +/- 7.2, respectively; overall average recoveries for nitrofurazone and furazolidone were 43% +/- 7.3 and 77% +/- 10.9, respectively. Before these optimum determinations, degradation by even minimal incandescent light was found to reduce recovery especially of desoxycarbadox. The results of this photochemical degradation are reported and briefly discussed.  相似文献

OBJECTIVE: To explore the relationship between family average income (FAI; an index of socio-economic status) and body mass index (BMI; a widely used, inexpensive indicator of weight status) above the healthy weight range in a region of Mainland China. DESIGN: Population-based cross-sectional study, conducted between October 1999 and March 2000 on a sample of regular local residents aged 35 years or older who were selected by random cluster sampling. SETTING: Forty-five administrative villages selected from three urban districts and two rural counties of Nanjing municipality, Mainland China, with a regional population of 5.6 million. SUBJECTS: In total, 29,340 subjects participated; 67.7% from urban and 32.3% from rural areas; 49.8% male and 50.2% female. The response rate among eligible participants was 90.1%. RESULTS: The proportion of participants classified as overweight was 30.5%, while 7.8% were identified as obese. After adjusting for possible confounding variables (age, gender, area of residence, educational level, occupational and leisure-time physical activity, daily vegetable consumption and frequency of red meat intake), urban participants were more likely to be overweight or obese relative to their rural counterparts, more women than men were obese, and participants in the lowest FAI tertile were the least likely to be above the healthy weight range. CONCLUSIONS: The proportion of adults with BMI above the healthy weight range was positively related to having a higher socio-economic status (indexed by FAI) in a regional Chinese population.  相似文献

Effects of soil amendment with crabshell chitin on the growth of white clover (Trifolium repens L.) and perennial ryegrass (Lolium perenne L.), and on populations of soil bacteria, fungi, and plant-parasitic and free-living nematodes were investigated in a pot trial. Five soil samples were collected from Te Puke (Paengaroa Shallow Sand, a Typic Hapludand) and five from Hamilton (Bruntwood silt loam, an Aquic Hapludand), New Zealand. Subsamples of each soil were either amended with chitin or unamended and planted with white clover and ryegrass. The ryegrass shoot weight in amended soil was greater (P<0.01), most probably due to N mineralised from chitin. A significantly lower (P<0.01) root: shoot ratio of ryegrass in the amended soil also suggested improved N availability, and therefore less root mass was needed to support a given shoot mass. A reduction in nodulation was observed in 12-day-old white clover seedlings (P<0.05) and also in 6-week-old seedlings (P<0.01). The shoot weight of white clover was significantly lower (P<0.05) in amended soil, possibly due to phytotoxic effects of chitin. Chitin increased (P<0.01) the populations of bacteria and fungi by 13-fold and 2.5-fold, respectively. The cyst nematode of white clover, Heterodera trifolii, was significantly reduced in chitin-amended soil, possibly due to increased levels of chitinase produced by rhizosphere microorganisms. Two other plant-parasitic nematodes, Pratylenchus spp. and Tylenchus spp., were also reduced in ryegrass roots and in soil as a result of the chitin amendment. However, the total number of free-living nematodes increased 5.4-fold in amended soil.  相似文献
