The inheritance of mefenoxam resistance in the oomycetePhytophthora erythroseptica, causal organism of pink rot of potato, was investigated in two successive selfed generations of single oospore isolates. Fourteen parental isolates from three mefenoxam-sensitivity phenotypes (sensitive, intermediately resistant, and resistant) were used to produce S1 and S2 progeny isolates by selfing each isolate and germinating single oospore cultures from each isolate. The two generations of isolates were tested for mefenoxam sensitivity using the radial growth inhibition assay and calculating EC50 values. Twenty-four-hundred isolates from S1 and S2 generations were tested for mefenoxam sensitivity. Results demonstrated the lack of segregation for mefenoxam sensitivity among S1 and S2 progeny isolates from the resistant and sensitive parents. The majority of the S1 progeny isolates from the parents with intermediate resistance also possessed intermediate resistance, but demonstrated substantial quantitative shifts in response to mefenoxam more often toward increased insensitivity. A few isolates from the S1 progeny were sensitive to mefenoxam, and one isolate was mefenoxam-resistant. Similarly, the majority of the S2 progeny isolates from intermediately resistant parents also possessed intermediate resistance with some quantitative shifts in sensitivity to mefenoxam similar to those observed in S1 progeny, as well as a few isolates that were sensitive to mefenoxam. These results do not support the hypothesis that resistance to mefenoxam inP. erythroseptica is controlled by a single gene exhibiting incomplete dominance. Alternative hypotheses are proposed such as the genetics of mefenoxam resistance inP. erythroseptica is probably under the control of more than one major gene and perhaps some minor genes of additive effect.  相似文献

Potato tuber tissue inoculated at cutting withFusarium spp. orErwinia carotovora and exposed to conditions favorable for infection did not heal rapidly enough to prevent decay. Seven days of wound healing at 20 C and high relative humidity prior to inoculation were necessary to provide protection againstFusarium and two days were required to protect againstErwinia. Histological examination of cut and chemically treated tuber tissue revealed that the wound healing process was only minimally affected by mancozeb, zineb-streptomycin, zineb-fir bark, captan, captan-mertect, captan-fir bark, and sodium hypochlorite. Ethylmercury p-toluene-sulfonanalide and pyrrolidine induced formation of discontinuous wound periderm after 21 days of healing. Streptomycin and captan-rhodamine had very detrimental effects on wound healing and suberin formation. In general, the seed-piece treatment chemicals tested had only minor effects on normal wound healing. It is concluded that wound healing alone may not be sufficient to controlFusarium caused seed piece decay and chemical treatment is recommended to provide protection until wound healing can occur. At least two days of wound healing are required for control of decay caused byErwinia carotovora.  相似文献

The speed and sensitivity of PCR-based assays allow shorter turnaround times for the detection of pathogens for which culture and serological methods are difficult or unavailable. PCR was performed with primer sets Cms50 and Cms72, designed previously by Millset al. (1997) through subtractive hybridization to detectClavibacter michiganensis subspeciessepedonicus (Cms). In bacterial suspensions, fewer than three cells/10 ul reaction were detected after PCR amplicons were hybridized with specific DIG-labeled DNA probes in an enzyme-linked oligonucleosorbent assay (ELOSA). In naturally infected tuber samples representing three cultivars of potato, the diagnostic sensitivity of PCR/ ELOSA was 96%, while the specificity exceeded 99%. PCR/ELOSA detectedCms in infected tuber samples with equal sensitivity regardless of colony morphology, potato cultivar, or primer sets.  相似文献

To evaluate the estrogenic potential of secoisolariciresinol diglycoside (SDG) found in linseed meal (LSM) on visceral organ mass, IGF-I, and thyroid hormone (T3 and T4) concentrations, 48 multiparous, ovariectomized ewes (54.6 ± 1.1 kg) were used in a 3 × 4 factorial arrangement. Main effects were length of LSM feeding (0, 1, 7, or 14 d) and length of exposure to estradiol-17β (E2) implant (0, 6, or 24 h prior to tissue collection). Implanting ewes with E2 for 24 h increased liver mass relative to empty body weight (EBW; g/kg EBW) compared with ewes implanted for 0 or 6 h (P ≤ 0.03), whereas feeding LSM for 14 d decreased liver mass compared with ewes fed LSM for 1 or 7 d (P ≤ 0.02). There was an LSM × E2 interaction (P = 0.01) for duodenal mass (g/kg EBW), LSM, and E2 tended (P = 0.07) to influence the stomach complex mass; however, ileal mass was not affected. Neither LSM nor E2 affected (P ≥ 0.12) CYP2C or CYP3A mRNA expression or cellularity of the liver. Exogenous E2 influenced circulating concentrations of IGF-I, T3, and T4. The estrogenic or anti-estrogenic potential of LSM is dependent upon the tissue, exposure to E2, and the duration of LSM feeding. Feeding LSM during gestation, lactation, or during the grow-finish phase warrants further investigation.  相似文献

Osteoarthritis (OA) is one of the most common and disabling chronic joint disorders affecting horses, dogs and humans. Synovial inflammation or synovitis is a frequently observed phenomenon in osteoarthritic joints and contributes to the pathogenesis of OA through formation of various catabolic and pro-inflammatory mediators altering the balance of cartilage matrix degradation and repair. Catabolic mediators produced by the inflamed synovium include pro-inflammatory cytokines, nitric oxide, prostaglandin E(2) and several neuropeptides, which further contribute to the pathogenesis of OA by increasing cartilage degradation. Recent studies suggest that substance P, corticotropin-releasing factor, urocortin and vasoactive intestinal peptide may also be involved in OA development, but the precise role of these neuropeptides in the pathogenesis of OA is not known. Since increased production of matrix metalloproteinases by the synovium is stimulated by pro-inflammatory cytokines, future anti-inflammatory therapies should focus on the synovium as a means of controlling subsequent inflammatory damage.  相似文献

To study the influence of sow dietary fat on piglet body characteristics, multiparous sows were allocated to one of four different dietary treatments: a conventional low fat (3%) diet (LF) and three high fat (6%) diets; high fat saturated (HFS), high fat oats (HFO), and high fat linseed (HFL). All sows were fed the allocated diet from weaning of the preceding litter until the day after farrowing. At farrowing, one liveborn piglet per litter (NB), was sacrificed and dissected immediately after birth. The heaviest (H) and the lightest (L) piglets in the litter were killed and dissected in the same manner at one day of age. Measurement of body length and circumference, organ weight, body chemical composition and muscle glycogen content were determined. Body measurements were adjusted to the mean body weight (1.67 kg). Dietary treatment did not have any significant influence on body components or carcass traits except for lung weight, being lower in HFO and HFL than in LF piglets. Piglet category affected almost all parameters considered, showing the lowest values for NB piglets, except for lung and circumference that were higher in NB than in L and H piglets; and length which was lower in NB than in L piglets. NB piglets had the highest amount of muscle glycogen content, no difference was found between H and L piglets. Dietary treatments influenced piglet chemical composition, showing the highest overall values of dry matter (DM), protein, and fat for the HFL piglets' carcasses. The present data provide additional information on the depletion of energy reserves; it would appear that sow dietary fat has relatively little effect on progeny since only body chemical composition was significantly influenced by HFL diet.  相似文献

This study examines the palatability and discrete nutritional evaluation of some prototype lupin protein concentrates (PC) when fed to rainbow trout. Products were developed from both Lupinus angustifolius and Lupinus luteus kernel meals with an increase in protein of 415 g/kg DM to 690 g/kg DM for L. angustifolius and 545 g/kg DM to 750 g/kg DM for L. luteus, respectively. This study completes a three-phase approach to evaluating the nutritional value of these products. The digestibility of energy, nitrogen, phosphorus and organic matter were determined in earlier studies using the diet substitution approach. The apparent digestibility of the energy from the L. angustifolius PC and the L. luteus PC along with the apparent protein digestibility were used to formulate two series of experimental diets to examine both the palatability and discrete nutritional value of the products. Serial inclusion of either PC at 0%, 10%, 20%, 30% and 40% into a typical salmonid diet specification allowed an examination of the palatability of each product. Additional negative controls, based on the 0% diets with inclusion of sulfamerazine sodium, were included in the experiment to demonstrate the capacity of the experiment to detect significant palatability issues. No significant effects of inclusion of either PC on any fish performance criteria, such as feed intake or growth, were identified. In contrast, significant reductions in feed intake and consequently growth were observed from fish fed either of the negative controls. This experiment demonstrated that each PC was highly palatable at inclusion levels up to and including 40% of the diet. Using a protein-limited-restrictively-fed experimental approach the discrete nutritional utilisation of each PC was defined. Growth of fish fed the PC treatments was not significantly different to that of the 0% reference diet. Two control diets with substitutions of cellulose to an equivalent inclusion level to that of the PC have provided an indication of the net benefit of the test ingredients. This experiment demonstrated that each PC provided equivalent nutritional value to the fish at either of the two inclusion levels (20% and 40%) evaluated. These PCs differed in their viscosity and gelling properties which may allow feed manufacturers the opportunity to manipulate the physical attributes of their feeds. Together, these studies clearly show that the prototype PCs have substantial potential as a prospective feed ingredient for the aquaculture sector.  相似文献
