Turbidity has been used as an effective indicator of water quality and it is regulated by national control agencies as a water potability parameter. This work proposes a simple, low-cost, and easily reproducible method based on digitally processed snapshots of electromagnetic radiation beam through a scattering medium in order to measure turbidity. The formazin polymer was used as the standard reference in the experiments to prepare samples from 0.1 to 100 NTU for high turbidity range and from 0.02 to 10 FNU for low turbidity range. The device design is comprised of a webcam and an LED/laser as a light sensor and a radiation source, respectively. The captured scattered light snapshot can be decomposed digitally in color components values and correlated with the turbidity parameter. It is possible to obtain differently device performances changing the light sensor device configuration. The linear regression analyses have shown a distinct relationship among red, green, blue, and grayscale components and the turbidity. At high range, the green component present the best LOD and LOQ values 0.64 and 2.10, respectively, operating with an ordinary webcam and white LED. Nevertheless, the best device performances were obtained with dedicated Raspberry Pi camera modules and white LED for low range turbidity reaching LOD=0.027 and LOQ=0.087 FNU. The figures of merit show recovery between (97.50?101.95%), repeatability (2.11%), within-laboratory reproducibility (2.25%), and limits of quantification (0.087 FNU). The achieved performance device shows the effectiveness of digitally processed snapshots obtained with digital cameras for turbidity measurements.  相似文献

Previous studies of afforestation in Patagonia indicate that 30–50 % tree cover produces positive effects on the pasture. This coverage level is achieved by applying pruning and strong thinning to reduce the volume of timber production per area unit. From an economic standpoint, in order to not reduce the income level of the system, it is necessary to find tree species that could replace ponderosa pine (Pinus ponderosa) and maximize income per volume unit of wood. We evaluated the feasibility of implementation of five broadleaved tree species (native and exotic) with higher intrinsic wood quality than ponderosa pine. We tested the influence of tree cover generated by a framework of silvopastoral plantation on the increase in survival, regrowth and absolute increase in height, and compared ecophysiological variables (net photosynthetic activity, stomatal conductance, intrinsic water use efficiency) of different species to the status of a traditional plantation without tree cover. Additionally values of air temperature and relative humidity were registered under both cover conditions. Preliminary results support the conclusion that three of these five species could be considered as alternatives to ponderosa pine species for establishing silvopastoral systems with higher timber value. Our Based on the results, the use of tree cover generated under the current plantations of P. ponderosa could increase the success of the establishment of these species.  相似文献

Different theories have been brought forward to explain the commonly observed δ15N enrichment with depth in soil profiles, including the discrimination against 15N during N decomposition and the buildup of 15N-enriched microbial residues. A combination of soil organic matter (SOM) size and density fractionations, 15N determinations, and phospholipid fatty acid (PLFA) analyses was conducted on soils from a pristine N-limited Nothofagus forest in southern Chile. The purpose of this study was to investigate which SOM fractions mostly reflect the 15N-enrichment pattern and to link 15N SOM enrichment with microbial community composition. Nitrogen-15 enrichments were greater for the microaggregate (<150 μm) than for the macroaggregate (>150 μm) size fraction, with Rayleigh isotope enrichment factors averaging −8.5‰ and −3.7‰, respectively. The macro-organic matter density fractions (>150 μm) showed intermediate enrichment factors of −5.1‰ and −7.3‰ for the light (<1.37 g cm−3) and heavy (>1.37 g cm−3) fraction, respectively. The abundance of fungal and bacterial PLFAs was significantly higher in the microaggregate compared to the macroaggregate size fraction, but their relative abundance did not change between aggregate size fractions. Our data link differential 15N enrichment of SOM fractions to “total” microbial abundance and, as such, corroborates existing theories of microbial-induced 15N enrichment. Isotopic fractionation during microbial N decomposition processes alone could not explain the large 15N enrichment in the microaggregate size fraction (−8.5‰) and the heavy density fraction (−7.3‰). We therefore suggest that microbial turnover and accretion of 15N-enriched microbial (especially fungal) compounds was an additional driver for 15N enrichment of this soil profile.  相似文献

CBA/J mice were used in the present study to establish differences between the immune response to three chlamydial strains: AB7 (Chlamydia psittaci wild-type strain), 1B (C. psittaci vaccinal strain) and iB1 (C. pecorum). The evolution of chlamydial infection was evaluated in each strain by studying the clinical signs, the number of bacteria isolated from the spleen and the pathology of the liver. Three aspects of the immune response were then studied: the characterization of the infiltrate of leukocytes in the liver, the percentages of T- and B-cells, macrophages and neutrophils in the spleen, and the presence of cytokines in the serum. Infection followed a different course in the C. psittaci-infected mice; 1B-infected mice showed milder levels in all the parameters analysed than their AB7-infected counterparts. The resolution of infection was earlier in 1B-infected mice and, although the immune response to both strains was Th1-like, a more intense CD8+ T-cell response and an earlier presence of TNF-alpha in serum were observed in this group. C. pecorum infection was controlled mainly by a non-specific immune response, since these mice showed no signs of a systemic specific immune response. Neutrophil depletion experiments showed that these cells play a very limited role in the non-specific response against C. pecorum.  相似文献

Inheritance and interrelationships of seed cotton and lint yields were evaluated in a diallel analysis involving seven early maturing parents of different origin and a commercial variety. Lint yield showed relatively little additive variance and low heritability, whereas lint percentage showed the opposite. Highest yields were shown by the least determinate and slowest-maturing genotypes; yields generally decreased as determinacy increased and rate of maturity accelerated. Except for date for first open boll, components of earliness showed no associated with yield.  相似文献

A study was undertaken in order to compare plasma disposition kinetic parameters of doramectin (DRM) and ivermectin (IVM) in horses after oral administration. Ten crossbreed adult horses, clinically healthy, weighing 380-470 kg body weight (bw) were selected for study. Faecal examinations were performed to determine faecal parasite egg counts. Horses were allocated to two groups of five animals to provide an even distribution considering the variables sex, body weight and faecal egg count. Group I, were treated with an oral paste formulation of IVM at 0.2 mg/kg b/w and Group II, were treated with an oral dose of 0.2 mg/kg bw of DRM prepared as paste from the injectable formulation for oral administration. Blood samples were collected by jugular puncture between 0 h and 75 days post-treatment. Plasma was separated and later solid phase extraction and derivatization samples were analysed by high performance liquid chromatography (HPLC); a computerised kinetic analysis was carried out. Data were compared using the Mann-Whitney U-test.The mean plasma concentrations of DRM and IVM after oral administration in horses were detected until 30 and 20 days, respectively. Both drugs showed similar patterns of absorption and no significant differences were found for peak concentration, the time to peak concentration, or for absorptive half-life. The terminal elimination half-life was significantly (P<0.05) longer in the DRM treated group than for the IVM treated group. The differences observed in the elimination half-life explain the longer mean residence time and high values of area under the concentration time curve for the group treated with DRM, which are 30% higher than those of the IVM group. Considering its pharmacokinetics, tolerance and anthelmintic efficacy, the oral administration of DRM, could be an alternative to IVM for the control of parasitic diseases of horses.  相似文献

MDR1 (ABCB1) P-glycoprotein exerts a protective function in the blood–brain barrier thereby limiting the entry of many drugs and other xenobiotics to the central nervous system. A nonsense mutation has been described for Collies and related dog breeds which abolishes this function and is associated with increased susceptibility to neurotoxic side effects of several drugs including ivermectin, moxidectin and loperamide. In order to evaluate the occurrence and frequency of this nt230 (del4) MDR1 mutation in Germany, we screened 1500 dogs. Frequency of the homozygous mutated genotype was highest for Collies (33.0%), followed by Australian Shepherd (6.9%) and Shetland Sheepdog (5.7%). Thirty-seven percent of the Wäller dogs and 12.5% of the Old English Sheepdogs were heterozygous for the mutant MDR1 (−) allele. Considering the predominant role of MDR1 P-glycoprotein in drug disposition and in particular for blood–brain barrier protection, MDR1 genotype-based breeding programs are recommended for improving the safety of drug therapy in these canine breeds.  相似文献

BACKGROUND: Models for the study of hematopoietic stem cells in dogs provide important information for bone marrow transplantation in humans. Recent studies have reported the importance of human umbilical cord blood (UCB) as an alternative to allogenic bone marrow for hematopoietic reconstitution. However, there are no studies on the UCB cells of dogs. OBJECTIVE: The aim of this experiment was to characterize and quantify the blood cells of the umbilical cord of dogs. METHODS: The blood of the umbilical cord of 20 neonatal dogs, delivered at term, with a median gestation time of 58 days, was collected with a 5-mL syringe containing EDTA. Total RBC, WBC, and platelet counts, HCT, hemoglobin (Hgb) concentration, and RBC indices were determined using an automatic cell counter. The differential leukocyte count was determined manually in blood smears stained with May-Grünwald-Giemsa. Reticulocyte percentages were determined on blood smears stained with brilliant cresyl blue and counterstained with May-Grünwald Giemsa. RESULTS: The MCHC and numbers of RBCs, WBCs, neutrophils, and eosinophils in UCB were lower as compared with reference values for the peripheral blood of healthy neonatal and adult dogs; whereas, the MCV and reticulocyte percentages were higher. CONCLUSION: Erythrocyte macrocytosis and hypochromasia in UCB were consistent with marked reticulocytosis and indicative of high erythropoietic activity. The results of this study are an important first step in the characterization of UCB from neonatal dogs.  相似文献

A Chlamydophila abortus-induced abortion model was carried out on the basis of the experimental infection of ewes at day 75 of gestation. The infection induced abortions and the birth of weak lambs during the last 3 weeks of pregnancy. To study the kinetics of the infection in the placenta and in other organs, infected ewes were killed at 105, 120, and 130 days of gestation and also several days after abortion or parturition. Infected ewes developed a systemic infection that caused a mild and transient pneumonia and focal hepatitis. Pathologic changes were observed in placentas at 120 day of gestation, although the lesions varied between animals and even between placentomes of the same placenta. The first placental area infected was the maternal stroma and epithelium next to the intercaruncular areas, where neutrophilic response seemed to control the infection. A substantial degree of multiplication of C. abortus was then observed in the trophoblast cells of the placentome, periplacentomal choriallantoic membranes, and hilius, with an inflammatory exudate composed mainly of neutrophils, some macrophages, and very scarce lymphocytes. After abortion, the lesions affected the intercotyledonary areas of the aborted placentas, whereas in the uterus significant lymphocyte infiltration was observed, together with a rapid decrease of the C. abortus antigen in the degenerated caruncular tissues.  相似文献
