Abstract A water culture pot experiment was conducted to analyze the effects of N application during the ripening period (RP) on photosynthesis, dry matter production, and its impact on grain ripening and yield in two semidwarf indica type varieties viz. Gui Zhao 2 (GZ) and BR3 (BR) compared with a japonica type variety Koganemasari (KO) under four N rates viz. 0 (N0), 10 (N10), 20 (N20), and 40 (N40) mg L?1. Results showed that N application enabled to maintain a higher leaf area and delay leaf senescence in both indica and japonica type varieties but the decrease in the rate, of leaf area was higher in the former than in the latter and the rate was reduced with increasing N rates. Flag leaf photosynthesis and SPAD values of N treated plants were higher throughout the RP, showing the presence of a significant correlation either for each variety or all the varieties together. Higher photosynthetic rate was supported by higher leaf chlorophyll (SPAD value basis) content, stomatai conductance, and N concentration in leaf blades. Top dry matter content increased with increasing N rates mainly due to mean leaf area rather than NAR except for BR during RP but it was higher in KO than in GZ and BR. Reduction of shoot weight due to translocation of dry matter to panicles during RP was suppressed by N rates both in GZ and BR while shoot weight increased in the N-treated plants in KO. The dependency of KO on current photosynthates for panicle weight was found to be almost hundred percent while the contribution of stored carbohydrates in shoot before heading to panicle weight in GZ and BR was in the range of 4-27 and 33-54%, respectively and the rest was contributed by current photosynthates. The percentage increased with increasing N rates. Percentage of ripened grains (PRG) increased with increasing N rates in GZ and BR due to the increase in dry matter production and in the photosynthetic rates of apex leaves, despite the larger spikelet number and larger hull size. However, KO showed almost no variation although it had the highest PRG among the varieties. Brown rice yield followed the same pattern as that of PRG in GZ and BR and the highest yield was produced by BR followed by GZ and KO. These results suggest that N application during RP was more effective in increasing yield in the semidwarf indica type varieties than in the japonica type variety. 相似文献

Spermatozoa concentration, ionic composition, osmolality, glucose and total protein contents of seminal plasma and sperm motility were determined in Barbus sharpeyi (Cyprinidae, Teleosotei). Spermatozoa concentration ranged from 9.77 to 20.20 × 109 spermatozoa mL?1. Osmolality (mOsmol kg?1) and ionic contents (mM L?1) of the seminal plasma were 274.5±9.0, 70.0±3.4 Na+, 28.8±0.9 K+, 101.7±3.1 Cl?, 0.9±0.1 Mg2+ and 2.1±0.1 Ca2+ respectively. Total protein and glucose were 5.3±0.2 g L?1 and 76.7±4.3 mM L?1 respectively. Sperm motility was initiated in a hypo‐osmotic condition, composed of either an ionic (KCl or NaCl) or a non‐ionic (sucrose) activation medium. Duration of sperm motility was very short: <2 min after activation in distilled water. Percentage of motile spermatozoa was significantly higher in an activation medium containing NaCl compared with that of distilled water. An activating medium containing NaCl or KCl higher than 150 mM or sucrose higher than 275 mM totally inhibited the activation of sperm motility. Immediately after sperm activation, wave(s) propagated along the flagellum, but waves were restricted to the proximal part of the flagellum (close to the head) at 1 min post activation. At abandoned mine sites, arsenic (As)- and antimony (Sb)-enriched soils are often disposed of through onsite burial or capping. In highly weathered mine sites, the mobility of As and Sb is typically controlled by iron (Fe)(Ⅲ)/Fe(Ⅱ) phases; thus, the suitability of such disposal methods and appropriate testing techniques are questionable. In the present study, leaching potentials of As and Sb were examined using the toxicity characteristic leaching procedure (TCLP), waste extraction test (WET), and WET-extended procedure (WET-EXT) at three abandoned mine site soils in Australia. The leached concentration of As regularly exceeded USEPA criteria (5 mg L-1). The highest leached concentrations of As and Sb were observed in the finest particle size fraction (< 0.053 mm) by WET-EXT (1 040 mg L-1 for As and 21.10 mg L-1 for Sb) followed by WET (800 mg L-1 for As and 20.90 mg L-1 for Sb). The TCLP method resulted in the lowest concentrations of leached As (0.000 9 mg L-1) and Sb (0.000 3 mg L-1). Crystalline and amorphous As-bearing Fe oxides were the main phases in the soils studied. However, the best correlations of leached As determined by TCLP (0.832), WET (0.944), and WET-EXT (0.961) were found with the non-specifically sorbed (NS1) As fraction. The mineralogical and sequential extraction data clearly indicate the dominant role of Fe geochemistry in controlling leachability of As and Sb. The TCLP method was unlikely to be suitable for assessing leachability, as it exhibited no relationship with leachable Fe and substantially lower leached As and Sb than the other two methods. Given the high to extremely high leachable As and Sb concentrations, most of the soil samples would not be recommended for placement in capping works, old shafts, or reduction systems (e.g., collection in drainage basins). 相似文献

The present paper concerns the distribution and mobility of heavy metals (Cu, Pb, Zn and Fe) in the soils of some abandoned mine sites in Italy and their transfer to wild flora.Materials and methods
Soils and plants were sampled from mixed sulphide mine dumps in different parts of Italy, and the concentrations of heavy metals were determined.Results and discussion
The phytoremediation ability of Salix species (Salix eleagnos, Salix purpurea and Salix caprea), Taraxacum officinale and P?lantago major for heavy metals and, in particular, zinc was estimated. The results showed that soils affected by mining activities presented total Zn, Cu, Pb and Fe concentrations above the internationally recommended permissible limits. A highly significant correlation occurred between metal concentrations in soils.Conclusions
The obtained results confirmed the environmental effects of mine waste; exploring wild flora ability to absorb metals, besides metal exploitation, proved a useful tool for planning possible remediation projects.165.

Currently, specific nutrient concentration, metabolizable energy (ME) and digestible amino acids are used as feed formulation criteria. A balanced nutrient density (BND) concept based on 2 criteria of nutrient density and balanced amino acids-to-ME ratio may offer more flexibility in optimisation of profit in formulation of diets compared with current formulation based on set values per unit of feed mass. A total of 672 one-d-old off-sex male Ross 308 broiler chickens were used across two 42-d performance trials in a 3 × 2 factorial arrangement of treatments with each diet replicated 8 times (14 birds per replicate). The experimental factors were 2 nutrient density levels (low [LD] and high [HD]) and 3 digestible lysine-to-ME ratios (DLYS:ME; low, medium, and high). Low density diets had ME of 2,876 and 3,023 kcal/kg for starter and finisher, respectively, while values for HD diets were 3,169 and 3,315 kcal/kg with proportionally higher non-nitrogenated nutrients. Separate digestibility and apparent metabolizable energy (AME) assays were conducted at d 21 and 42. Digestibility assays at d 7 were conducted on birds used for performance trials. Regardless of the diet density, birds fed low DLYS:ME had a lower (P < 0.01) feed intake (d 0 to 42) than medium and high DLYS:ME. Without interaction, birds fed low and medium DLYS:ME had a similar body weight gain being the heaviest while birds low DLYS:ME were the lightest. By an interaction (P < 0.05), the highest overall FCR value was observed for birds fed LD × low DLYS:ME and improved linearly when DLYS:ME increased to the highest level reaching a limit for birds fed HD × medium DLYS:ME. Calorie conversion linearly decreased (P < 0.001) with increments in DLYS:ME. Jejunal and ileal starch and protein digestibility were affected on d 21 and 42 but not on d 7 of age. Given the independence of response on BW and feed consumption, the use of BND as a flexible system in diet formulations has the potential to enable more accurate formulation for optimisation of growth performance of broiler chickens. 相似文献

Background: Cryptosporidium parvum is an important coccidian parasite infecting many mammals, including human. This parasite can manifest as chronic severe diarrhea in immunocompromised individuals, especially those with AIDS. The present study reports the recombinant production of rP2 and rP23 antigens of C. parvum as antigens for detecting human cryptosporidiosis using indirect ELISA tests. Methods:The coding sequences of rP2 and rP23 proteins were codon-optimized, commercially synthesized and sub-cloned in the pET28a expression vector. The expressed proteins were purified by Ni-NTA column chromatography and confirmed by Western blotting. The efficacy of rP2/rP23 proteins for serodiagnosis was evaluated by positive (n = 20) and negative (n = 20) human sera, confirmed by the Ziehl-Neelsen staining as the gold standard test. Results:In ELISA test, the sera from C. parvum-infected patients reacted strongly to rP2/rP23. The sensitivity and specificity related to the diagnostic potential of rP2/rP23 in the ELISA assay were 100%. Conclusion:Our results showed that combination of rP23 and rP2 antigens in ELISA significantly increases the performance of C. parvum serodiagnosis in human cryptosporidiosis.
