Crop rotation showed a pronounced effect on incidence of tomato damping-off and onion (Fusarium) basal rot as well as on the soil population of the two disease incitants. Out of nine rotation systems, wheat/sorghum and wheat/maize preceding tomato diminished damping-off while clover/maize, lentil/sorghum, wheat/sorghum and wheat/maize preceding tomato diminished damping-off while clover/maize, lentil/sorghum, wheat/sorghum and wheat/maize preceding onions increased basal rot incidence. First crops of the rotation system as well as the second crop were of noticeable effect onRhizoctonia solani andFusarium oxysporum f. sp.cepae as onBacillus subtilis (antagonistic to both fungi) population in soil. Almost all rotations increased population ofF. oxysporum f. sp.cepae andB. subtilis in soil while response ofR. solani was variable. The results show thatBac. subtilis did not correlate with tomato damping-off or onion basal rot. Cold stress is one of the major abiotic factors that influence the productivity and geographical distribution of many agriculturally important crops like Hevea brasiliensis. Cultivation of H. brasiliensis in India is being extended to northeastern regions, where low temperature during winter adversely affects its survival, growth, and productivity. Developing cold-tolerant genotypes is a primary requisite to maximize the productivity under such challenging environmental conditions. However, lack of methods for early evaluation of cold tolerance in the newly developed clones and the extensive time required for assessing their tolerance in the field are major constraints for clonal selection. The present study was initiated with an objective to identify and characterize cold stress responsive miRNAs from H. brasiliensis that show stronger association with cold tolerance. Next generation sequencing using Illumina HiSeq method revealed the expression of 21 and 29 conserved miRNA (from clone RRIM 600) families in cold-stressed and control samples, respectively. Forty-two novel miRNAs were identified from this study. Upon differential expression analysis, eight conserved miRNAs were found commonly expressed in both the samples. When expression analyses were performed subsequently with six selected miRNAs in two Hevea clones (viz. RRII 105 and RRIM 600), miR169 showed a strong association with cold tolerance. miRNAs such as miR482 and miR159 also exhibited association with cold tolerance. This study suggests the possibility of employing these miRNAs as markers for cold tolerance after validation in more number of genotypes with varying levels of cold tolerance. 相似文献

A greenhouse experiment was conducted to investigate the immediate effect of application of mono‐ammonium phosphate (MAP), single superphosphate (SSP), and triple superphosphate (TSP) fertilizers containing varying concentrations of Cd on (1) chemical speciation of Cd and Zn in soil solution by chemical‐equilibrium calculations (MINEQL+4.6 model), (2) growth of barley plants, (3) concentrations of Cd, P, and Zn in soil solution and plant tissue, as well as total plant accumulation of Cd, P, and Zn, and (4) monitoring pH and element changes during incubation periods following phosphate application. Results show that, in general, the pH of soil solution increased during the first 40 d of incubation, then declined. Also, at the end of incubation period, pH of soil solution was affected by fertilization source and fertilization rate. The concentration of Cd in soil solution changed with time. Phosphate fertilization (p < 0.05) or fertilizer source (p < 0.05) showed consistent effects. Also, the application of phosphate fertilizers with three rates significantly increased Zn concentrations in soil solution during the first half (0–30 d) of incubation period and then decreased but still more than in the control. In general, application of different sources of phosphate at 100 g kg–1 did not change the dominant forms of Cd in soil solution during all incubation time intervals. Speciation of Zn in the control after 30 d of incubation had changed, in comparison to 10 d of incubation, and the dominant forms were Zn2+, ZnOH+, ZnHCO3, ZnCO3(aq), and Zn(OH)2(aq). Adding phosphate fertilizer significantly increased both shoot and root dry weight compared to control, indicating P was a growth‐limiting factor in the control plants. The Zn concentrations in shoot and root were lower in the TSP‐ and SSP‐fertilizers treatment than those in the MAP and fertilizer treatments at all rates of fertilization. Adding phosphate increased the Cd : Zn and P : Zn ratios in the shoot and root tissue, with the effect being greater with increasing fertilization rate. Phosphate fertilization greatly increased the total accumulation of Cd of barley compared with the control plants (p < 0.001), with the effect being greater with increasing fertilization rate. The Eph tyrosine kinase receptors and their ephrin ligands play a central role in human cancer as their deregulated expression induces tumorigenesis with aggressive phenotypes. To evaluate their potential contribution to EphA2-ephrinA1 modulation, several colonic catabolites of dietary (poly)phenolics, known to be generated in vivo, were screened using an ELISA-based binding assay. Some of the catabolites inhibited the binding in a dose-dependent manner (IC(50) values from 0.26 to 43 μM). Functional studies on prostate adenocarcinoma cells revealed that pyrogallol and protocatechuic acid specifically antagonized ephrinA1-Fc-induced EphA2 phosphorylation at concentrations that were not cytotoxic. The active concentrations of pyrogallol appear to be close to what can be reached in vivo under physiological conditions. Finally, because of the roles played by the Eph-ephrin system not only in cancer development but also in neurodegeneration and diabetes, pyrogallol and protocatechuic acid are candidates for more detailed functional studies to elucidate their role in these pathophysiological processes. 相似文献

Two cyanobacterial species (Nostoc entophytum and Oscillatoria angustissima) were tested as biofertilizers, substituting the normally used chemical fertilizer, for pea plant. Inoculation of soil with a suspension of each species or a combination of the two species significantly increased the germination percentage and stimulated the other measured growth parameters and photosynthetic pigment fractions of pea. However, the soil inoculation with one cyanobacterial species and the addition of the recommended dose or half the recommended dose of chemical fertilizer were usually more effective and also increased carbohydrate and protein contents of produced pea seeds. However, biofertilization combined with half the recommended dose of the chemical fertilizer was usually more effective than the addition of the full rate of the chemical fertilizer, and this may allow saving 50% of the used chemical fertilizer. The protein profile of the produced seeds showed appearance and disappearance of some protein bands in response to fertilization treatments compared to the control. Blue green algae analyses show that N. entophytum fixed more N, produced more exopolysaccharide, and contained more auxin and cytokinin than O. angustissima, the latter contained more gibberellins. These data may explain their different influences on growth and yield of pea. 相似文献

Rabbit hemorrhagic disease virus (RHDV) is the etiological agent of a lethal and contagious disease of rabbits that remains as a serious problem worldwide. As this virus does not replicate in cell culture systems, the capsid protein gene has been expressed in heterologous hosts or inserted in replication-competent viruses in order to obtain non-conventional RHDV vaccines. However, due to technological or safety issues, current RHDV vaccines are still prepared from organs of infected rabbits. In this work, two human type 5 derived replication-defective adenoviruses encoding the rabbit hemorrhagic disease virus VP60 capsid protein were constructed. The recombinant protein was expressed as a multimer in mouse and rabbit cell lines at levels that ranged from approximately 120 to 160 mg/L of culture. Mice intravenously or subcutaneously inoculated with a single 10(8) gene transfer units (GTU) dose of the AdVP60 vector (designed for VP60 intracellular expression) seroconverted at days 7 and 14 post-immunization, respectively. This vector generated a stronger response than that obtained with a second vector (AdVP60sec) designed for VP60 secretion. Rabbits were then immunized by parenteral or mucosal routes with a single 10(9)GTU dose of the AdVP60 and the antibody response was evaluated using a competition ELISA specific for RHDV or RHDVa. Protective hemagglutination inhibition (HI) titers were also promptly detected and IgG antibodies corresponding with inhibition percentages over 85% persisted up to one year in all rabbits, independently of the immunization route employed. These levels were similar to those elicited with inactivated RHDV or with VP60 obtained from yeast or insect cells. IgA specific antibodies were only found in saliva of rabbits immunized by intranasal instillation. The feasibility of VP60 production and vaccination of rabbits with replication-defective adenoviral vectors was demonstrated. 相似文献

The purpose of this study was to assess the toxicoses and antitumor activity of metronomic chlorambucil at a dosage of 4 mg m(-2) daily in dogs with naturally occurring cancer. Thirty-six dogs were enrolled in the study. The protocol was well tolerated with no grade 3 or 4 toxicoses noted. Complete remission was achieved, and lasted over 35 weeks in three dogs (mast cell tumour, soft tissue sarcoma and thyroid carcinoma). Partial remission was noted in 1 dog with histiocytic sarcoma (39 weeks duration) for an overall remission rate of 11% (4 of 36). Stable disease was noted in 17 dogs (47%) with various other cancers. The median progression-free interval was 61 days, and the median survival time was 153 days. Chlorambucil given in a metronomic protocol showed antitumor activity in dogs with a variety of naturally occurring cancers. 相似文献

Eimeria coecicola causes intestinal coccidiosis in rabbits and, thereby, enormous economic losses in rabbit farms. Here, we investigate the final target site of E. coecicola, the appendix of rabbits, at the level of gene expression. Rabbits, orally infected with E. coecicola, begin to shed parasitic oocysts with their feces on day 5 p.i., and approximately 1.1 million oocysts are maximally shedded on day 7 p.i. At maximal shedding, the appendix has increased in size by about 2-3-folds and reveals increased hemorrhage which is associated with increases in nitrite/nitrate, malondialdehyde and catalase activity and a decrease in glutathione. Agilent 2-color oligo whole rabbit genome microarray, in combination with quantitative real-time PCR, detects 45 and 36 genes whose expression is more than 2-fold up- and down-regulated, respectively, by E. coecicola infection on day 7 p.i. The most dramatic increase by approximately 50-fold reveals the mRNA of the pro- and anti-inflammatory pleiotropic cytokine interleukin 6 (IL-6), whereas the largest decrease by approximately 13-fold is detected for mRNAs encoding for DBP, SULT3A1, CRP and glutathione-S transferase. Also, there are up- and down-regulations in the expression of genes encoding diverse regions of antibodies. Our data suggest that IL-6 plays a central role in the infection of the appendix of rabbits by E. coecicola, presumably involved in both pathological injuries, host defences and healing processes. 相似文献
