This prospective, multicentre, non-blinded, open study followed 46 cats with diabetes mellitus during treatment with porcine lente insulin (also known as porcine insulin zinc suspension, Caninsulin, Intervet) for 16+/-1 weeks (stabilization phase), with additional monitoring of some cats (n=23) for a variable period. At least three of the following were present at initial presentation: appropriate history of clinical signs consistent with diabetes mellitus, glucosuria, blood glucose greater than 15 mmol/l and fructosamine greater than 380 micromol/l. Insulin treatment was started at a dose rate of 0.25-0.5 IU/kg body weight twice daily, with a maximum starting dose of 2 IU/injection. Twenty-eight of the cats were classed as reaching clinical stability during the study, in 23 of these cats this was during the stabilization phase. Seven cats went into remission during the stabilization phase and one of the cats in week 56. Clinical signs of hypoglycaemia, significantly associated with a dose of 3 units or 0.5 IU/kg or more per cat (twice daily), were observed in nine of the 46 cats during the stabilization phase and concomitant biochemical hypoglycaemia was recorded in most cases. Biochemical hypoglycaemia, recorded in 6% of the blood glucose curves performed during the stabilization phase, was significantly associated with a dose rate of 0.75 IU/kg or more twice daily. This further highlights the need for cautious stepwise changes in insulin dose. The protocol used in the present study is suitable for and easy to use in practice. AIM: To describe features of the morphology of the carpus, quantify the thickness of hyaline and calcified cartilage, and to describe the morphology and density of subchondral bone in the third carpal bone (C3) of young Thoroughbred horses in early training. METHODS: C3 of seven 2-year-old horses in training and seven untrained horses matched for age, sex and breed were assessed by gross appearance, computed tomography, fine-structure radiography, image analysis of high-resolution photographs, and histology. RESULTS: Macroscopic lesions in cartilage were few and mild, and not significantly different between groups. High bone mineral density (BMD), in some cases typical of cortical bone, was confined to the dorsal load path, and was significantly higher in trained than in untrained horses (p<0.01). In the most dorsoproximal aspect of the radial articular facet, apparently outside the dorsal load path, the BMD in both trained and untrained horses was significantly less than in other regions of interest (ROIs). Adaptive increase in density was associated with thickening of the (junctions of) trabeculae oriented proximo-distally. Hyaline cartilage was thicker (p<0.001) in the concavity of the radial articular facet than dorsal or palmar to it, and was thicker in the trained than untrained group (p=0.007). No such differences were detected in the thickness of articular calcified cartilage (ACC). CONCLUSIONS: The rapid response of bone in C3 to relatively small amounts of high-speed exercise was confirmed. A previously unreported increase in thickness of hyaline cartilage was evident, perhaps indicating that this tissue may be more responsive than hitherto thought, at least to particular types of exercise at particular times. These changes occurred with little evidence of abnormality, and thus appeared to be adaptive to the exercise regimen. The model developed should be used for further definition of the exercise stimulus required to produce adaptive, protective changes in sites susceptible to athletic injury. CLINICAL RELEVANCE: The data will serve as reference for use in subsequent imaging studies in which sophisticated aids such as magnetic resonance imaging (MRI) may be used to predict carpal lesions. 相似文献

Vibrio anguillarum, also known as Listonella anguillarum, is the causative agent of vibriosis, a deadly haemorrhagic septicaemic disease affecting various marine and fresh/brackish water fish, bivalves and crustaceans. In both aquaculture and larviculture, this disease is responsible for severe economic losses worldwide. Because of its high morbidity and mortality rates, substantial research has been carried out to elucidate the virulence mechanisms of this pathogen and to develop rapid detection techniques and effective disease‐prevention strategies. Objective To determine if an association existed between sickness, mortality and bullers in a western Canadian feedlot.
Design A retrospective epidemiological study.
Animals 78,445 male cattle that entered a 24,000-head feedlot in western Canada from 1991 to 1993.
Procedure Animal health records for bullers were collected and analysed to see if they were at greater risk of sickness and mortality than other steers, and to see if pens with a high prevalence of bullers also had a high prevalence of sickness and mortality.
Results The prevalence of bullers increased with increasing age of cattle on arrival at the feedlot (R = 0.36; P < 0.001). Sickness and mortality decreased with increasing age of cattle on arrival. However, sickness and mortality in bullers relative to other steers actually increased with increasing age on arrival suggesting an interaction existed between sickness and bullers. Bullers were significantly (P < 0.05) more likely to get sick and to die than other steers. In all cases, there was a strong temporal association between sickness and bullers, with sickness and bullers mostly occurring within the first 30 days of the feeding period. On average, pens of cattle with a high prevalence of bullers did not have a correspondingly high prevalence of sickness or mortality.
Conclusion This study suggests that sickness is an effect modifier of dominance behaviour and therefore bullers in feedlot steers. Bullers should always be checked for signs of sickness and treated accordingly. Further research is needed to investigate the effects of sickness on dominance behaviour in pens of feedlot cattle. 相似文献

ABSTRACT1. Guanidinoacetic acid (GAA) is the single endogenous precursor of creatine, which plays a critical role in energy homeostasis of cells. Since GAA is endogenously converted to creatine by methylation, it was hypothesised that the effects of dietary GAA supplementation might determine the methionine (Met) availability in corn-soybean based diets.2. A total of 540, one-day-old male Ross 308 broilers were allocated to nine dietary treatments with six replicates (10 birds each) in a 3 × 3 factorial arrangement with three graded levels of supplementary Met (+0.4 g/kg per level), whilst cystine was equal across groups, resulting in a low, medium and high level of total sulphur amino acids, and with three levels of GAA (0, 0.6 and 1.2 g/kg). Birds were fed for 42 days.3. Increasing levels of supplemental Met enhanced performance indices in all rearing periods, although there was no effect on feed conversion ratio in the grower or feed intake in the finisher periods. Final body weight was 8.8% and 14.6% higher in the birds fed medium and high Met diets, respectively, compared to the low Met level. Relative breast weight and protein content in muscle on d 25 linearly increased with higher levels of Met. At low and high Met levels, growth in the finisher phase was negatively affected by supplementing GAA at 1.2 g/kg. It was suggested that disturbances in methylation homeostasis and/or changes in Arg metabolism might explain these findings. At the end of the grower phase, muscle creatine content was higher when feeding GAA at 0.6 and 1.2 g/kg (4464 and 4472, respectively, vs. 4054 mg/kg fresh muscle in the control group).4. The effects of dietary GAA supplementation were influenced by the dietary Met level only in the finisher period, which indicates the need for proper sulphur amino acid formulation in diets when feeding GAA. 相似文献

This paper summarises and presents in context the main findings of an extensive series of studies of early training lasting 13 weeks in which the tissue responses of 2-year-old Thoroughbred horses were assessed using a combination of methods. Negligible clinical injury was detected and thus the study fulfilled the intention of investigating adaptive change rather than injury. Cancellous and cortical bone, some digital tendons, and articular cartilage responded to early training exercise to a greater or lesser degree. Clinical examination and ancillary diagnostic aids currently in veterinary clinical use are not sufficient to detect early abnormalities in metacarpo-phalangeal joint (MCPJ) cartilage found in both trained and untrained horses. Future work should centre on detection of such changes, on the precise registration of training workload, and on the manipulation of the responses of musculoskeletal tissues by careful investigation of the effects of introducing conditioning exercise at a young age. 相似文献

AIM: To identify if there was a significant difference in the average daily liveweight gain (ADG) between Thoroughbred foals weaned using two weaning protocols commonly used in New Zealand. METHODS: Sixteen Thoroughbred foals were blocked for sex and age, randomly allocated into progressive (187, SD 33 days; three colts, five fillies) or abrupt (182, SD 28 days; four colts, four fillies) weaning groups, and weighed every second day for 2 weeks either side of weaning, then fortnightly from birth to 480 (SD 31) days old. ADG was calculated to examine the short-term (10 days before weaning, 5 and 10 days post weaning) and long-term (0–6 and 6–16 months of age) effect of the two weaning treatments. RESULTS: ADG was 1.10 (SD 0.091) kg/day before weaning (0–6 months of age) and 0.59 (SD 0.06) kg/day from weaning to 480 (SD 31) days old. At the start of weaning, liveweights of the progressive and abrupt weaning groups were 276.5 (SD 40.3) kg and 257 (SD 15) kg, respectively (p=0.23). For the 5-day period during weaning, irrespective of treatment, there was a significant decrease in ADG of –0.29 (SD 0.49) kg/day and –0.15 (SD 0.30) kg/day for progressive and abrupt weaning, respectively. There was no significant difference in ADG between weaning methods at any measurement period (short or long term) during and after weaning. However, there was large variation between foals in ADG in the 10 days after the weaning process, which may indicate variation in individual foals' responses to being weaned, rather than the weaning treatment. CONCLUSIONS: Weaning, irrespective of method, was associated with a decrease in ADG in the first week after weaning. The method of weaning had no effect on post-weaning ADG either short term, 10 days after weaning, or long term up to 480 days of age. Practically, it may be more important to consider maturity and liveweight as criteria for weaning foals rather than age alone. 相似文献

In parasite-host dynamics, parasites exert frequency-dependent selection on their hosts by favouring rare alleles that may confer resistance against infection. Therefore host populations that suffer strong parasite stress should maintain higher levels of genetic variability. We studied the Lumbricus terrestris-Monocystis sp. host-parasite system at a microgeographical scale. Using three polymorphic microsatellite loci on one large earthworm population sampled at 26 different sites (281 genotypes), we tested the relationship between parasite load and genetic variation in natural samples of the common earthworm L. terrestris. Our analysis yielded the following: (1) parasite load varied significantly across sites in this population; (2) there was no consistent evidence for heterozygote deficiency (observed heterozygosities ranged between 0.74 and 0.87), indicating a low level of inbreeding; (3) there was no significant genetic structuring among sample sites; (4) we could not identify a significant association between parasite load and population genetic diversity; (5) there was considerable population differentiation (15.17%) between our German samples and a Canadian L. terrestris reference population. Our study provides insight into the population genetics of one of the most economically important soil organisms on a microgeographic scale. 相似文献

The objective of this study was to evaluate the interaction of zinc source (ZnSO4 vs. zinc amino acid complex) and vitamin E level (50 IU vs. 100 IU) on performance and intestinal health of broilers exposed to a temperature challenge in the finisher period. A total of 1224 day old male Ross 308 broilers were randomly distributed among 4 dietary treatments (9 replicates per treatment). Dietary treatments were organized in a 2 × 2 factorial arrangement: two sources of zinc, 60 mg/kg of Zn as ZnSO4.7H2O or 60 mg/kg of Zn as zinc amino acid complexes (ZnAA) combined with two levels of vitamin E (50 or 100 IU/kg). Zinc and vitamin E were added to a wheat/rye-based diet that was designed to create a mild nutritional challenge. From day 28 until day 36 (finisher period), all birds were subjected to chronic cyclic high temperatures (32°C ± 2°C and RH 55–65% for 6 h daily). The combination of ZnAA and 50 IU/kg of vitamin E improved weight gain in the starter (day 0–10), finisher (day 28–36) and overall period (day 0–36) and feed conversion ratio in the starter (day 0–10) and finisher phase (day 28–36). Providing Zn as ZnAA significantly improved villus length and villus/crypt ratio in the starter, grower and finisher period and decreased infiltration of T-lymphocytes and ovotransferrin leakage in the finisher period. In conclusion, providing broilers with a diet supplemented with ZnAA and a vitamin E level of 50 IU/kg, resulted in better growth performance as compared to all other dietary treatments. Interestingly, under the conditions of this study, positive effects of ZnAA on performance did not occur when vitamin E was supplemented at 100 IU/kg in feed.
