A wild strain of Agrobacterium rhizogenes was used to regenerate twelve in vitro plant lines from different hairy roots of H. perforatum (St. John's Wort). The production of the main bioactive constituents was observed even though their yields varied in the different plant lines. Two lines were selected for the hyperoside production (4.9-4.6 mg/gdw) while nine were characterized by significant yields of chlorogenic acid (ranged from 0.47 to 1.09 mg/gdw). Furthermore, one out of twelve lines showed a 10-fold higher hypericin content (0.25 mg/gdw) than that reported for the in vitro shoots in the literature. Morphological and phytochemical features were determined in order to select H. perforatum genotypes enriched in valuable bioactive compounds. 相似文献

Sulfur (S) fertilization has been long-known to influence the amounts of total free amino acids in plants. To determine the impact of S deficiency in wheat on the concentration of, in particular, free asparagine, the spring wheat cultivar 'Star' was grown in a laboratory scale (5 L pot) at five different levels of S fertilization. After maturity, the kernels were milled into white flours (1-5) and analyzed for their contents of total S and total nitrogen as well as for free amino acids and glucose, fructose, maltose, and sucrose. Extremely high concentrations of free asparagine (Asn; 3.9-5.7 g/kg) were determined in flours 1 and 2 (30 and 60 mg of S), whereas much lower amounts (0.03-0.4 g/kg) were present in flours grown at higher S levels. The amounts of the reducing carbohydrates were, however, scarcely affected by S fertilization. In agreement with the high amount of Asn in flours 1 and 2, heating of both flours led to the generation of very high amounts of acrylamide (1.7-3.1 mg/kg) as well as of 3-aminopropionamide (40-76 mg/kg). Similar concentrations were measured in crispbread prepared from both flours. Application of rheological measurements on doughs prepared from each flour and a determination of the loaf volume of bread baked therefrom clearly indicated that flours 1 and 2 would be excluded from commercial bread processing due to their poor technological properties. Two methods for the determination of dichlorvos in durum wheat by electrochemical assay and gas chromatography, respectively, have been developed. Dichlorvos, an organophosphorus anticholinesterase pesticide, was extracted from wheat with hexane, and the filtered extract was directly analyzed by gas chromatography with nitrogen-phosphorus flame detection (NPD). Recoveries of dichlorvos from milled wheat spiked at 0.25-1.5 microg/g ranged from 96.5 to 100.9%, and the limit of detection was 0.02 microg/g. The electrochemical assay was based on the detection of choline, the acetylcholinesterase product, via a choline oxidase biosensor. An aliquot of the filtered hexane extract was partitioned with phosphate buffer solution, and the organic layer was evaporated prior to electrochemical analysis. A limit of detection of 0.05 microg/g of dichlorvos was obtained with mean recoveries of 97-103% at spiking levels of 0.25-1.5 microg/g. A good correlation (r = 0.9919) was found between the results obtained with the electrochemical and those obtained with the gas chromatographic methods. The electrochemical method was peer-validated in two laboratories that analyzed 10 blind samples (5 duplicates), including a blank and 4 spiked samples with dichlorvos at levels of 0.25, 0.60, 1.00, and 1.50 microg/g. Marisma, one of the largest salt-marsh alluvial areas in SW Spain, has been reclaimed since 1970 by artificial drainage and
amendment with phosphogypsum (PG) so as to reduce Na+ saturation. Within the reclaimed area, two 250- × 20-m plots were treated as follows: (1) amendment with 25 Mg/ha of PG every
2 to 3 years between 1979 and 2003 (plot PY); (2) like PY but PG treatment stopped after 1997 (plot DR). A contiguous virgin
Marisma salt-marsh plot (MV), neither drained nor amended, was the control. In MV, soil microbial biomass C, most enzyme activities
and total organic C content were much greater than in PY and DR soils, despite the salinity stress. The decrease in soil organic
matter content in PY and DR soils was likely due to cotton-cropping practices, which favoured the organic C mineralisation
and nutrients removal by crops. Microbial activity of MV soil, probably stimulated by the rhizodepositions of the natural
vegetation, did not suffer from the osmotic effect due to the raising of soil solution ionic strength. Microbial quotient
could be ranked as MV > PY > DR, whereas the metabolic quotient had an opposite trend. Thus, both quotients suggested that
the interruption of PG amendment was not favouring microbial activity. Principal component analysis clearly identified the
chemical and biochemical soil properties mostly affected by the reclamation and also indicated the longer PG amendment in
PY plot. Stepwise discriminant analysis identified two physiologically different types of soil microflora, one less active

Portable X-ray fluorescence (pXRF) spectrometry and magnetic susceptibility (MS) via magnetometer have been increasingly used with terrain variables for digital soil mapping. However, this methodology is still emerging in many countries with tropical soils. The objective of this study was to use proximal soil sensor data associated with terrain variables at varying spatial resolutions to predict soil classes using the Random Forest (RF) algorithm. The study was conducted on a 316-ha area featuring highly variable soil classes and complex soil-landscape relationships in Minas Gerais State, Brazil. The overall accuracy and Kappa index were evaluated using soils that were classified at 118 sites, with 90 being used for modeling and 28 for validation. Digital elevation models (DEMs) were created at 5-, 10-, 20-, and 30-m resolutions using contour lines from two sources. The resulting DEMs were processed to generate 12 terrain variables. Total Fe, Ti, and SiO2 contents were obtained using pXRF, with MS determined via a magnetometer. Soil class prediction was performed using the RF algorithm. The quality of the soil maps improved when using only the five most important covariates and combining proximal sensor data with terrain variables at different spatial resolutions. The finest spatial resolution did not always provide the most accurate maps. The high soil complexity in the area prevented highly accurate predictions. The most important variables influencing the soil mapping were MS, Fe, and Ti. Proximal sensor data associated with terrain information were successfully used to map Brazilian soils at variable spatial resolutions. 相似文献

Although seed plants and multicellular animals are predominantly diploid, the prominence of diploidy varies greatly among eukaryote life cycles, and no general evolutionary advantage of diploidy has been demonstrated. By doubling the copy number of each gene, diploidy may increase the rate at which adaptive mutations are produced. However, models suggest that this does not necessarily accelerate adaptation by diploid populations. We tested model predictions regarding rates of adaptation using asexual yeast populations. Productivity and resource-use efficiency in corn (Zea mays L.) are crucial issues in sustainable agriculture, especially in high-demand resource crops such as corn. The aims of this research were to compare irrigation scheduling and nitrogen fertilization rates in corn, evaluating yield, water (WUE), irrigation water (IRRWUE) and nitrogen use (NUE) efficiencies. A 2-year field experiment was carried out in a Mediterranean coastal area of Central Italy (175 mm of rainfall in the corn-growing period) and corn was subjected to three irrigation levels (rainfed and supply at 50 and 100% of crop evapotranspiration, ETc) in interaction with three nitrogen fertilization levels (not fertilized, 15 and 30 g (N) m−2). The results indicated a large yearly variability, mainly due to a rainfall event at the silking stage in the first year; a significant irrigation effect was observed for all the variables under study, except for plant population. Nitrogen rates affected grain yield plant−1 and ear−1, grain and biomass yield, HI, WUE, IRRWUE and NUE, with significant differences between non-fertilized and the two fertilized treatments (15 and 30 g (N) m−2). Furthermore, deficit irrigation (50% of ETc) was to a large degree equal to 100% of the ETc irrigation regime. A significant interaction “N × I” was observed for grain yield and WUE. The effect of nitrogen availability was amplified at the maximum irrigation water regime. The relationships between grain yield and evapotranspiration showed basal ET, the amount necessary to start producing grain, of about 63 mm in the first and 206 mm in the second year. Rainfed crop depleted most of the water in the 0–0.6 m soil depth range, while irrigated scenarios absorbed soil water within the profile to a depth of 1.0 m. Corn in a Mediterranean area can be cultivated with acceptable yields while saving irrigation water and reducing nitrogen supply and also exploiting the positive interaction between these two factors, so maximizing resource-use efficiency. 相似文献

A new proposal for U.S. farm commodity programs would allow more flexibility in planting decisions on program acreage. This work examines the effect of such a proposal on potato production acreage. These results are used to project changes in total revenue accruing to existing potato producers and to the potato industry as a whole. The results indicate that the short term impact on the potato industry as a whole would be relatively minor but the impact could be unevenly distributed across producing regions of the U.S., causing some potential local discomfort. 相似文献

Numerous gluten preparations were produced by the variation of pressure and temperature. Optimal conditions for the production of gluten films on a laboratory-scale were by suspending of gluten (1 g) in a mixture of ethanol (3 mL), glycerol (0.5 g) and conc. formic acid (10 mL), casting and drying at 40 °C. Small-scale laboratory methods for the production of gluten films by casting and moulding were developed. Film strips obtained were examined by micro-extension tests, which resulted in curves similar to extensigrams for dough and gluten and allowed the determination of the resistance to extension, extensibility and elasticity. The results demonstrated that pressure treatment of gluten in combination with variable cultivars, temperature, process parameters and additives, allow the production of films with a wide range of rheological properties – from soft and smooth to strong and hard rubber like. Finally, it was demonstrated that the addition of fibres to gluten enhanced the stability of films. Thus, high pressure treatment allows a selective modification of gluten as raw material for film production. In comparison with conventional plastic films, gluten films have considerable advantages, because they can be produced from renewable plants and they are readily biodegradable. 相似文献
