Two pea (Pisum sativum L.) cultivars were compared: cv Lincoln and cv Douce de Provence. Seedlings grown for 14 d on standard medium were challenged for 21 d with salt using a split-root system. This protocol allowed salt-treated plants to absorb nutrients through a part of their root system maintained in control medium (C), the other part of the root system being placed in medium added with 75 mM NaCl (S). Full salt treatment (S/S) resulted in severe but non-lethal growth inhibition, high concentration of Na+ and Cl in leaves, and decrease in leaf K+ and chlorophyll contents. The two latter effects were more pronounced in Lincoln than in D. Provence. Growth inhibition was partially (Lincoln) or totally (D. Provence) alleviated in S/C configuration, and K+ content was less diminished than in full salt treatment. S/C treatment mitigated Na+ and Cl accumulation in Lincoln, but not in D. Provence. Thus, in the latter cultivar, growth inhibition by salt in S/S condition likely did not result from excessive Na+ and Cl accumulation in leaves. Increased electrolyte leakage from leaf tissues evidenced damages to leaf cell plasma membrane of both cultivars in S/S condition. However, damages to chloroplasts, as inferred from chlorophyll loss, were much pronounced in Lincoln than in D. Provence. Antioxidant enzymic activities in leaves were measured as proxies for oxidative stress. Catalase activity was stimulated by S/S treatment in both cultivars, but superoxide dismutase (Fe and Cu/Zn isoforms) and gaiacol peroxidase activities were augmented only in Lincoln. The absence of superoxide dismutase activity stimulation by salt in D. Provence could signify either that constitutive activity was sufficient to ensure protection against oxidative stress, or that intrinsic salt tolerance of this cultivar mitigated cellular oxidative stress. Thus, intraspecific variability for salt response exists between pea cultivars presenting similar growth sensitivity to salt.  相似文献

Basement membrane of glomerular mesangium (BMG) is one of important components which play a key role to support of the capillary loops in a renal glomerulus and completeness of BMG due to interaction of ureteric bud and metanephric mesenchyme during glomerulogenesis. As laminin contribute in extra cellular matrix and especially in basement membrane, the aim of the present study was to demonstrate the distribution of this molecule so, in this investigation specific antibody against laminin have been used in light microscopy to study development of BMG of fetal and postnatal mouse glomerular mesangium. Female inbred Balb/c mice were selected and were kept under normal condition and finding vaginal plug was assumed as day zero of pregnancy. Two pregnant mice were sacrificed by cervical dislocation in one of gestational days 13-18, respectively and their fetuses were fixed, serially sectioned and by using antibody against laminin in BMG were carried out. The same process was used for kidneys preparation at 15 postnatal days. Present data revealed that laminin showed weak reaction on day 14 of gestation. The amount of laminin increased continuously until next days of fetal life and primary of 10 days postnatal in BMG. After this period, laminin reaction did not show significant change in newborns. These data indicate that laminin appears just during the glomerulogenesis and because of continuity with vasculature which is required for Extra Cellular Matrix (ECM) and glomerular endothelial cell differentiation, laminin, is the one of major structural proteins in BMG.  相似文献

By using an agar dilution assay, the antifungal activity of aqueous extracts prepared from Allium cepa (onion; AOE) and Allium sativum (garlic; AGE) were evaluated against Malassezia furfur (25 strains), Candida albicans (18 strains), other Candida sp. (12 strains) as well as 35 strains of various dermatophyte species and compared with the activity of a known antifungal drug, ketoconazole (KTZ). All the AOE, AGE and KTZ were found to be able to inhibit growth of all fungi tested in a dose-dependent manner with maximum of 100% at defined concentrations. The results indicate that onion and garlic might be promising in treatment of fungal-associated diseases from important pathogenic genera Candida, Malassezia and the dermatophytes.  相似文献

Management of anthracnose caused by Colletotrichum spp. is the most important issue for the tropical fruit industry because of resulting financial losses. Antifungal effects of gum arabic (GA) (10%), lemongrass oil (LG) (0.05%), cinnamon oil (CM) (0.4%), and their combinations were investigated in vitro and in vivo for controlling postharvest anthracnose of banana and papaya. LG at 0.05% and 0.4% CM showed fungicidal effects against Colletotrichum musae and Colletotrichum gloeosporioides, causal organisms of banana and papaya anthracnose, respectively. GA alone did not show any fungicidal effects while the combination of 0.05% LG and 0.4% CM with Ten percent GA alone showed more fungicidal effects. However, potato dextrose agar (PDA) medium modified with 10% GA combined with 0.4% CM showed the most promising results among all treatments against C. musae and C. gloeosporioides in suppressing the mycelial growth (73.4%) and (70.0%) and spore germination inhibition (88%) and (85%), respectively. In vivo studies also revealed that 10% GA combined with 0.4% CM was the optimal concentration in controlling decay (80%) and (71%), showing a synergistic effect in the reduction of C. musae and C. gloeosporioides, respectively, in artificially inoculated bananas and papayas. The results regarding quality evaluation also confirmed the efficacy of 10% GA combined with 0.4% CM coatings since ripening was significantly delayed, in terms of percentage weight loss, fruit firmness, soluble solids concentration and titratable acidity. The results suggest the possibility of using 10% gum arabic combined with 0.4% cinnamon oil as a biofungicide for controlling postharvest anthracnose in major tropical fruit such as banana and papaya.  相似文献

Acer velutinum Boiss is a valuable tree species native to Iran, and its seeds possess physiological dormancy that hampers seedling production in the nursery for large-scale reforestation efforts. The aim of this study was to determine the optimal dormancy breaking treatments for A. velutinum seeds. We conducted a factorial experiment involving six seed lots collected along an elevation gradient from 300 to 1800 m at 300 m interval and four cold-moist stratification periods (0, 4, 8 and 16 weeks) at 4℃ and 70% relative humidity. The result shows that the germination of cold-moist stratified seeds was significantly (p < 0.0001) higher than the control for all seed lots. The highest germination capacity was recorded after 16 weeks of cold-moist stratification for all seed lots (68% 88% depending on the seed lot) except those collected from mid altitude sites (600 and 900 m) that germinated equally well (≥ 75%) after 4and 8-week of clod-moist stratification compared to the other seed lots. The mean germination time was significantly shorter (12 to 19 days, depending on the seed lot) for seeds stratified for 16 weeks than for untreated seeds. It can be concluded that: (1) cold-moist stratification for 16 weeks is the best pre-sowing treatment for breaking dormancy in A. velutinum seeds; and (2) seeds should be collected from mid altitude sites (600 and 900 m) to get more than 80% germination within 15 days, and these seed lots even required shorter cold-moist stratification period(eight weeks) than other seed lots.  相似文献

We observed robust coupling between the high- and low-frequency bands of ongoing electrical activity in the human brain. In particular, the phase of the low-frequency theta (4 to 8 hertz) rhythm modulates power in the high gamma (80 to 150 hertz) band of the electrocorticogram, with stronger modulation occurring at higher theta amplitudes. Furthermore, different behavioral tasks evoke distinct patterns of theta/high gamma coupling across the cortex. The results indicate that transient coupling between low- and high-frequency brain rhythms coordinates activity in distributed cortical areas, providing a mechanism for effective communication during cognitive processing in humans.  相似文献

The present study investigated the mediatory effects of salicylic acid (SA) in alleviating nickel (Ni) toxicity in Melissa officinalis L. One-month-old plants were exposed to different levels of Ni and SA concentrations in sand culture under greenhouse conditions. Excess Ni significantly inhibited the growth indices and dramatically increased accumulation of Ni in the leaves and roots. Exogenously SA applications (1.0 mM) led to a substantial improvement in the shoot and root fresh and dry weights. Foliar application of SA mitigated the deleterious effects of Ni and decreased its transport to the shoots. The results showed a significant loss in chlorophylls and carotenoids contents only at 500 µM of Ni. The impact of SA was not significant in terms of chlorophyll contents, while carotenoid contents of the Ni-stressed plants were significantly affected by SA. Exposure to Ni did not modify proline accumulation. Hydrogen peroxide accumulation was observed under Ni stress, while lipid peroxidation significantly decreased at the same conditions. Application of SA caused a significant decrease in electrolyte leakage of Ni-stressed plants. Due to the high potential for Ni accumulation in the roots and translocation factor values lower than 1, M. officinalis could be introduced as an excluder medicinal plant.  相似文献

This study aimed to evaluate the soil aggregate stability and selected soil quality indicators in various land uses in a semiarid region in central Iran. Random soil sampling was used to collect soil samples from surface (0–5 cm) and subsurface (5–25 cm) soil layers in rangelands of different condition classes, dry farmland and abandoned land. The aggregate size distribution indices including mean weight diameter (MWD), geometric mean diameter (GMD) and median diameter (D50) of water-stable aggregates in the collected soil samples were measured. Our findings showed that percent of macroaggregates (>0.25 mm) of the surface and subsurface layers in rangelands of different condition classes were significantly higher than dry farmlands and abandoned lands (P < 0.05). Results showed that the trend of changes in soil organic matter was similar to soil aggregate stability in different land uses in both soil layers as follows: rangeland with good condition > rangeland with poor condition > abandoned land > dry farmlands. The structural stability indices (i.e. MWD, GMD and D50) of rangelands with good condition were significantly greater than other land uses (P < 0.05). This highlights the importance of maintaining native rangeland to prevent organic matter loss, structure deterioration and soil erosion.  相似文献

Kansas and California wheat-growing regions differ dramatically in soils, climate, wheat cultivars, crop rotation patterns, and cultural practices, which could select for different fungal populations of Mycosphaerella graminicola. Our objective in this study was to use amplified fragment length polymorphism (AFLP) loci to assess the genetic diversity of M. graminicola populations within single fields in two widely separated, and geographically isolated sites in Kansas and California. Three primer-pair combinations were used to resolve polymorphism at 177 loci in 67 and 63 isolates from Kansas and California, respectively. Genotypic variability was high, which is consistent with a genetically diverse initial inoculum. There was no evidence of genetic disequilibrium in either population, with only 4.6% of the locus pairs in Kansas, and 5.4% of the locus pairs in California in detectable disequilibrium. The migration rate calculated between the two sites was as low as 1.8 individuals per generation, and significant differences in allele frequencies were observed. Therefore, these two populations do not represent mere subsamples of a larger, randomly mating population. This is a rare report of isolation by distance occurring between two North American populations of M. graminicola, indicating that at least some of these populations may be differentiating. Although genetic isolation by distance may occur, we cannot exclude movement of new gene combinations such as fungicide resistance or virulence between these two locations.  相似文献

The objective of the present study was to describe the arthroscopic anatomy of the bovine fetlock joint using one palmar/plantar and three dorsal joint approaches. A comparative anatomic, ultrasonographic and arthroscopic study using 20 cadaveric feet from 13 non-lame adult dairy cows was performed. Arthroscopy was accomplished using a rigid arthroscope to view the synovial cavities with their synovial villi and parts of the following structures: the distal ends of the metacarpal/metatarsal III/IV bones with their trochleae and sagittal ridges, synovial grooves, the articular surfaces of the proximal sesamoid bones, the proximal aspects of the first phalanges, the lateral and medial collateral ligaments, the suspensory ligament and the interdigital ligaments as parts of the interosseus muscle, the cruciate sesamoidean ligaments, the communication site between the lateral and medial pouch in the palmar/plantar area, and dorsally the septum between the lateral and the medial pouch. The technique allowed a good overall view of most relevant structures in the sound cadaver joint. Further investigations are warranted to evaluate the diagnostic, therapeutic and prognostic applications of these techniques in the treatment of septic arthritis.  相似文献
