Monoclonal antibodies (MAbs) to the pesticide propoxur were produced from haptens with carboxylic spacer arms of different lengths introduced at the carbamate group of the analyte structure. MAbs were subsequently characterized in the conjugate-coated format using these immunizing haptens and newly synthesized compounds as homologous and heterologous assay conjugates, respectively. Appropriate combinations of immunoreagents resulted in competitive enzyme immunoassays (ELISA) with I(50) values in the low nanomolar range (6.5-17.9 nM). A modification of the conjugate-coated format consisting of the simultaneous incubation of the MAb and the peroxidase-labeled secondary antibody in the presence of the analyte resulted in an assay with an I(50) value of 4.4 nM. This one-step conjugate-coated ELISA format is as simple and fast as the antibody-coated format but without the need of synthesizing enzyme-hapten conjugates. Major N-methylcarbamate pesticides were not recognized by the MAb. This immunoassay should reasonably allow the rapid, low-cost, and sensitive determination of propoxur in food, soils, and the environment at levels of regulatory and practical importance.  相似文献

The overall objective of this research was to find a new way to valorize rye bran, by producing a gellifier from the enzymatic solubilization of arabinoxylans (AX). The effects of three pure endo-xylanases from Aspergillus niger (Xyl-1), Talaromyces emersonii (Xyl-2), and Bacillus subtilis (Xyl-3) and of Grindamyl S100 (GS100), a commercial enzyme preparation containing a Xyl-1 type endo-xylanase, were tested on rye bran to study the solubilization of water-unextractable arabinoxylans (WUAX). Eight different extrusion-treated rye brans were also used as substrates to find the best physical treatment to facilitate enzymatic arabinoxylan (AX) solubilization. Arabinoxylans were better solubilized from the bran extruded at high temperature using Xyl-3. This enzyme was then tested in combination with pure (1,4)-beta-d-arabinoxylan arabinofuranohydrolase (AXH) and endo-beta-d-glucanase or ferulic acid esterase (FAE), from A. niger. Only beta-glucanase in combination with Xyl-3 improved the AX extraction, but it did not have a marked effect on the viscosity of the extracts. Xyl-3 was then tested on a high-shear-treated rye bran, and results were compared to those obtained with the high-temperature-extruded rye bran. The high-shear treatment did not improve the bran AX enzymatic solubilization. The combination of FAE with Xyl-1 or Xyl-3 did not improve the AX extraction from untreated and high-shear-treated rye bran. Finally, to study the gelation capacity of the enzymatically solubilized AX, the effect of the hydrogen peroxide/horseradish peroxidase (H(2)O(2)/POD) was tested on the Xyl-3 high-temperature-extruded bran extracts. Solubilized AX did not gel in the presence of the oxidizing system.  相似文献

Knowledge of relationships among the cultivated and wild species of Cerasus, a subgenus of Prunus L., is important for recognizing gene pools, identifying pitfalls in germplasm collections and developing effective conservation and management strategies. In this study, morphological characterization of wild Cerasus subgenus species naturally growing in Iran, including P. avium L. (mazzard), P. mahaleb L., P. brachypetala Boiss., P. incana Pall., P. yazdiana Mozaff., P. microcarpa Boiss. subsp. microcarpa, P. microcarpa Boiss. subsp. diffusa and P. pseudoprostrata Pojark. and three commercial species, sweet cherry (P. avium L.), sour cherry (P. cerasus L.) and duke cherry (P. × gondouinii Rehd.) was investigated. Twenty-nine variables were recorded in 95 accessions of these species to detect similarities among them. All studied characteristics were showing a high degree of variability, but it was highly pronounced for plant height, leaf characters especially for dimensions and pubescence as well as for petiole and peduncle length. The majority of significant correlation coefficients were found in the characteristics representing plant size and growth habit with leaf size. Principal component analysis was performed for phenotypical diversity determination and grouping of species. Characters with high discriminating values were those related to growth habit, height, compactness, trunk diameter, leaf dimensions, petiole length, leaf tip and serration, flower color, peduncle length, fruit shape and stone shape. Based on the cluster analysis, species included in two main clusters, first those with large structure (sweet, sour and duke cherries, mazzard and mahaleb), second the other species with small structure which might be potentially suitable for rootstock breeding programs. Scatter plot using two first factors also confirmed the grouping by cluster analysis. The conservation of the highly diverse native populations of Iranian wild Cerasus species is recommended.  相似文献

This investigation was done to determine the release of potassium (K) from five calcareous soils of southern Iran using 0.025 M CaCl2, HCl and citric acid during six successive extractions and to study the K fixation capacity of the soils after K release experiment. Mineralogical study indicated that Vertisols and Mollisols were dominated with smectites; while other soils had illite, chlorite, palygorskite and smectite. Results indicated that citric acid extracted more K than CaCl2 and HCl (137 vs. 111 and 113 mg kg?1, respectively). The analysis of calcium (Ca), magnesium (Mg) and K concentrations in the solutions suggests that the exchange of K with soluble Ca and Mg (originated from dissolution of carbonates by acidic solutions) is the main mechanism of K release, but citrate is able to dissolve K-bearing minerals and release K in slightly calcareous soils. Soils with more illite released more K. Potassium fixation capacity of soils increased after extractions of soils with different extractants from 324 to 471 mg kg?1, with no significant difference. It is suggested to apply more K fertilizers in K-depleted calcareous soils and use of different solutions for extracting K from soil minerals may be a temporary and short term solution.  相似文献

Abstract

To investigate the effect of foliar application of nano-chelates of iron, zinc, and manganese subjected to different irrigation conditions on physiological traits, and yield of soybean (cultivar M9), a split plot experiment was conducted in a completely randomized block design with three replications in two crop years (2016–2017). The main plot included four levels of irrigation (I): full irrigation (I 1), irrigation withhold at flowering stage (I 2), irrigation withhold at podding stage (I 3), and irrigation withhold during the grain filling period (I 4). Also, the subplot included eight levels of foliar application with Fe, Zn, Mn, Fe?+?Zn, Fe?+?Mn, Zn?+?Mn, Fe?+?Zn?+?Mn nano-chelates, and distilled water (control). The results of combined analysis of variance suggested that the effect of irrigation and foliar application of nano-chelate was significant on all traits. Water deficit stress significantly reduced the grain yield. The minimum numbers of pods per plant, number of grains per plant, 100-seed weight per plant, leaf area index, leaf chlorophyll concentration, total dry weight of plant, and the grain yield were obtained by irrigation withhold at podding stage. Foliar application of combined nano-chelates increased the soybean resistance against water shortage more considerably than the separate consumption of these elements. Under drought stress in podding stage, the application of Fe?+?Zn led to the highest yield with a mean of 2613.84?kg ha?1 where this increase was 61.1% higher than control.  相似文献

Desertification is one of the main environmental and also social and economic problems facing Iran. Seventeen out of 31 Iranian provinces, which are home to approximately 70% of the total population, are affected by desertification. This study aimed to use geographic information system (GIS) and fuzzy logic for mapping environmentally sensitive areas to desertification based on Mediterranean Desertification and Land Use approach in Isfahan province, central Iran. Six desertification indicators including climate, soil, vegetation, soil erosion, groundwater, and management and policy quality were used to determine various types of environmentally sensitive areas to desertification. Seventeen desertification indices affecting the quality of each indicator were spatially mapped and assigned a value between 0 and 1 using a fuzzy logic option of ArcSDM3 software in GIS environment. Results showed that a 21·7% of the study area was classified as critical, 70% as fragile and 5·5% as potential, and 2·9% of the area was not affected by desertification. In the town of Borkhar, 64·2% of the area was classified as critical, followed by the towns of Isfahan and Nayin with 40·2% and 31·8%, respectively. Results at provincial scale indicated that the climate indicator and humidity index with a weighting mean of 0·71 and 0·77 were the most affective factors in the desertification of the study area. The developed model in this study can be used for mapping desertification status in other 16 provinces that contain desert areas. These assessments provide a GIS‐based desertification database that Iran as a member of the United Nation Convention to Combat Desertification can use to report the condition of desertification at national scale. Streptococci are frequently isolated from bovine mastitis in dairy cows with only limited information available on the antimicrobial susceptibility of these organisms. A total of 42 Streptococci isolated from 148 milk samples of normal, sub acute and acute bovine mastitis cases. Overall, 35% of the strains tested were Streptococcus dysgalactiae, Streptococcus agalactiae 26%, Streptococcus uberis 18 and 4% were Enterococcus sp. Differences between the number of isolations in acute and sub acute groups were statistically significant, (p<0.5). The antimicrobial susceptibility for these organisms was determined for the following antimicrobial agents: cephalexine, penicillin, clindamycin, cloxaciline, gentamicin, streptomycin, amoxicillin, tetracycline, kanamycin, oxytetracycline, ampicillin, chloramphenicol and erythromycin. S. agalactiae, S. dysgalactiae, S. uberis and Enterococci demonstrated high level of resistance against streptomycin, penicillin and cloxaciline. Low level of sensitivity to other tested antimicrobials was demonstrated.  相似文献

The application of post treatments in preparation of high flux membranes is expanding rapidly. In this work, several hybrid post treatments have been introduced and used for change in the water flux of polysulfone (PSf) hollow fiber membranes. Dry wet spinning method was employed for fabrication of PSf hollow fiber membrane from spinning dope in mass ratio of 15:5:80 of PSf/PVP-K90/NMP. The simultaneous effects of single and hybrid post treatments containing traditional hypochlorite; high pressure injection technique (HPI) of hypochlorite, hot air and hot water treatments on the morphology and water flux of fabricated hollow fibers has been investigated. AFM analysis and image processing of SEM microphotographs of hollow fibers were used for structural studies. The mechanical properties of hollow fibers as well as strain at break and strength also were studied. It was found that the pores size and surface roughness parameter of hollow fiber membranes have been increased after traditional hypochlorite, HPI technique and hot water treatments while decreased when heat treated in air. In general all the employed hybrid post treatments caused to increase in the pores size of hollow fibers although the pores size increase rate in the membranes treated by the hybrid post treatments involving hot air was much lower than the others. The mechanical properties of hollow fibers have been decreased after hybrid and single post treatments containing traditional hypochlorite, HPI technique and hot water treatment while slightly increased after post treatments containing hot air. It was stated that the fabricated PSf hollow fibers were considerably affected by the employed hybrid post treatments. This can be attributed to the combine effects of used post treatments.  相似文献

Laboratory findings can definitely help the patients not to enter into status, where the damage might be happen due to a miss-diagnosis based on clinical assessment alone. The secondary disease accompanied with thyroid patients should also carefully check out due to the interference which some diseases can cause in the amount of serum thyroid hormone, particularly the free thyroxin. The dilemma over thyroid clinical diagnosis occur due to variation on serum thyroid hormone which initiated by other non-thyroidal disorders which can play an important roles in metabolic disorders of thyroid hormone due to the alteration which occur on the serum level of thyroid hormone transporter proteins. The majority of serum thyroid hormones of up to 95-99% are bound to the carrier proteins mainly to Thyroxin-Binding Globulins (TBG), some transthyretin already known as pre-albumin and albumin which are all synthesis in the liver and any modification which alter their production may alter the status of thyroid hormones. It seems TBG, transthyretin and albumin carries 75, 20, 5% of thyroid hormones within blood circulation, respectively. The dilemma facing the thyroid hormones following disruption of thyroid hormone transporter protein synthesis originate from this fact that any alteration of these protein contribute to the alteration of total thyroid and free serum thyroid hormones which are in fact the biologically active form of thyroid hormones. The subsequent of latter implication result in miss-understanding and miss-diagnosis of thyroid function tests, with possible wrongly thyroid clinical care, followed by undesired therapy of otherwise healthy thyroid.  相似文献
