Insulin-like growth factor 1 (IGF-1) induces skeletal muscle maturation and enlargement (hypertrophy). These responses require protein synthesis and myofibril formation (myofibrillogenesis). However, the signaling mechanisms of myofibrillogenesis remain obscure. We found that IGF-1-induced phosphatidylinositol 3-kinase-Akt signaling formed a complex of nebulin and N-WASP at the Z bands of myofibrils by interfering with glycogen synthase kinase-3β in mice. Although N-WASP is known to be an activator of the Arp2/3 complex to form branched actin filaments, the nebulin-N-WASP complex caused actin nucleation for unbranched actin filament formation from the Z bands without the Arp2/3 complex. Furthermore, N-WASP was required for IGF-1-induced muscle hypertrophy. These findings present the mechanisms of IGF-1-induced actin filament formation in myofibrillogenesis required for muscle maturation and hypertrophy and a mechanism of actin nucleation. 相似文献

ABSTRACT: A microscopic bubble generating system (MBGS) has been developed to control dissolved oxygen (DO) levels suitable for fish farming. The MBGS has been tested to confirm its capability in net pens. Water conditions in a fish farm were monitored every two hours from June to October 2004 by setting an online vertical profiling system (OVPS) close to the net pen. DO in the net pen water decreased to physiologically stressful levels for the fish during the night (4.84–5.51 mg/L), while the DO was kept in saturated conditions during the day, due to oxygen supply from phytoplankton. The MBGS was operated from the evening to the morning of the next day for 16 h, to successfully create DO-saturated conditions in the net pen water at night. By using microscopic bubbles during the warm seasons, DO levels in the net pen water could be improved to a level suitable for fish farming. However, the low DO levels (<5.0 mg/L) of the bottom water occasionally extended to the net pen layers, despite the supply of microscopic bubbles to the water. To maintain the DO of the net pen water at levels suitable for fish farming, DO supply to the net pen water and the bottom water needs to be increased, and the organically enriched sediment just below the net pens needs to be treated. 相似文献

ABSTRACT: To elucidate the hemolytic suppression roles of taurine and the necessity of dietary taurine supplementation in yellowtail Seriola quinqueradiata fed a diet without fishmeal, juvenile fish with an initial body weight of 250 g were fed for 40 weeks in floating net cages on soybean protein diets supplemented with 0, 3.0, 4.5 and 6.0% taurine. Taurine concentration of the experimental diets were 0.03, 33.9, 52.8 and 71.6 mg/g, respectively. On the 21st week, fish fed the taurine unsupplemented diet had inferior growth and feed performances, higher death, and there were incidences of green liver and hemolytic anemia. In this group, hepatic and plasma taurine concentrations, serum osmolality and osmotic tolerance of erythrocytes (EC50 value) were significantly lower, and plasma hydroperoxide concentration was markedly higher than in the taurine supplemented groups. These conditions markedly improved corresponding with the increase of dietary taurine concentration. These results indicate that taurine plays a role in hemolytic suppression through osmoregulation and biomembrane stabilization in fish. In addition, it is suggested that yellowtail requires dietary taurine as an essential nutrition for maintaining physiological condition normally. 相似文献

Bacteria cause deformation and blemishing in pearls, and we investigate this relationship. We examined pearls derived from Pinctada margaritifera, Pinctada maxima, and Pinctada fucata, and determined the location of bacteria using histopathological and immunohistochemical techniques and 16S ribosomal RNA (rRNA) sequence analyses. The most remarkable change was the inflammatory reaction located between the pearl nucleus and the nacreous layer, composed of hemocytic infiltration with melanization, periostracum, and fibrous aragonite-like structures. These anomalous changes were limited to abnormal sites, and such inflammatory reaction sites are a major factor in the formation of pearl abnormalities. Bacteria were detected from the inflammatory sites and are suspected as the causative agent. Most of these bacteria were anaerobic. 相似文献

The growth of aquaculture has negatively affected the environment due to the high levels of nitrogen excreted by farmed fish. Here we propose that modifying the nitrogen metabolism of the fish themselves using transgenic technology might solve the pollution problem. Growth hormone (GH) is known to increase protein retention and absorption, and is thought to reduce ammonia excretion. Thus, we produced transgenic Nile tilapia (Oreochromis niloticus) that over-expressed the GH gene throughout their bodies. Our findings showed that the food-conversion efficiency of the transgenic fish was 35% higher than that of their non-transgenic siblings. The rearing period required for the transgenic fish to reach a body weight of 20 g was about 75% of that required for non-transgenic fish that were fed the same type and quantity of food. The total amount of ammonium-nitrogen excreted by the transgenic fish was about 69% of that excreted by the wild-type fish over their lifetime. These results suggest that our transgenic approach has the potential to reduce the amount of nitrogen pollution caused by farmed fish. Farmed fish are exposed to risks from feed-borne chemical contamination, such as leucomalachite green (LMG) in fish meal. Consequently, the use of malachite green is prohibited in aquacultural practice. An improved understanding of the toxicity of dietary LMG provided to farmed fish is needed in order to manage risk. Oreochromis niloticus specimens were fed experimental diets containing 0, 100, 500, and 2500 μg/kg LMG for 28 days. On sampling days 7, 14, and 28, fish in the exposed groups had detectable levels of LMG. Accumulation levels were approximately 12.2 % (in the liver) and 6.2 % (in the muscle) of the LMG concentration in the feed (104, 510, and 2200 μg/kg). Hematocrit and mean corpuscular hemoglobin concentrations in the 500 and 2500 μg/kg groups were significantly elevated at 7 and 14 days. Hemoglobin in the group that received the highest dose was significantly higher than that in the control group. Significant increases in the activities of alanine aminotransferase and alkaline phosphatase were also detected in the group receiving the highest dose. Total cholesterol concentrations in all of the exposed groups were significantly higher than those in the control group. These observations of toxicity were dose dependent. Histological changes in gills and livers were observed in LMG-exposed fish. 相似文献
