In soils of arid and semiarid climates, dissolution of primary (lithogenic) carbonate and recrystallization with CO2 from soil air leads to precipitation of pedogenic carbonates and formation of calcic horizons. Thus, their carbon isotope composition represents the conditions prevailing during their formation. However, the widespread use of the isotopic signature (δ13C, δ18O, Δ14C) of pedogenic carbonates for reconstruction of local paleovegetation, paleoprecipitation and other environmental conditions lacks knowledge of the time frame of pedogenic carbonate formation, which depends on climatic factors. We hypothesized that temperature-dependent biotic processes like plant growth and root and rhizomicrobial respiration have stronger influence on soil CaCO3 recrystallization than abiotic temperature-dependent solubility of CO2 and CaCO3.To assess the effect of temperature on initial CaCO3 recrystallization rates, loess with primary CaCO3 was exposed to 14CO2 from root and rhizomicrobial respiration of plants labeled in 14CO2 atmosphere at 10, 20 or 30 °C. 14C recovered in recrystallized CaCO3 was quantified to calculate amounts of secondary CaCO3 and corresponding recrystallization rates, which were in the range of 10−6-10−4 day−1, meaning that 10−4-10−2% of total loess CaCO3 were recrystallized per day. Increasing rates with increasing temperature showed the major role of biological activities like enhanced water uptake by roots and respiration. The abiotic effect of lower solubility of CO2 in water by increasing temperature was completely overcompensated by biotic processes. Based on initial recrystallization rates, periods necessary for complete recrystallization were estimated for different temperatures, presuming that CaCO3 recrystallization in soil takes place mainly during the growing season. Taking into account the shortening effect of increasing temperature on the length of growing season, the contrast between low and high temperature was diminished, yielding recrystallization periods of 5740 years, 4330 years and 1060 years at 10, 20 and 30 °C, respectively. In summary, increasing CaCO3 recrystallization rates with increasing temperature demonstrated the important role of vegetation for pedogenic CaCO3 formation and the predominantly biotic effects of growing season temperature.Considering the long periods of pedogenic carbonate formation lasting to some millennia, we conclude that methodological resolution of paleoenvironmental studies based on isotope composition of pedogenic carbonates is limited not by instrumental precision but by the time frame of pedogenic carbonate formation and hence cannot be better than thousands of years.  相似文献

Adherence by Helicobacter pylori increases the risk of gastric disease. Here, we report that more than 95% of strains that bind fucosylated blood group antigen bind A, B, and O antigens (generalists), whereas 60% of adherent South American Amerindian strains bind blood group O antigens best (specialists). This specialization coincides with the unique predominance of blood group O in these Amerindians. Strains differed about 1500-fold in binding affinities, and diversifying selection was evident in babA sequences. We propose that cycles of selection for increased and decreased bacterial adherence contribute to babA diversity and that these cycles have led to gradual replacement of generalist binding by specialist binding in blood group O-dominant human populations.  相似文献

A highly regular mesh of hexagonal boron nitride with a 3-nanometer periodicity and a 2-nanometer hole size was formed by self-assembly on a Rh(111) single crystalline surface. Two layers of mesh cover the surface uniformly after high-temperature exposure of the clean rhodium surface to borazine (HBNH)3. The two layers are offset in such a way as to expose a minimum metal surface area. Hole formation is likely driven by the lattice mismatch of the film and the rhodium substrate. This regular nanostructure is thermally very stable and can serve as a template to organize molecules, as is exemplified by the decoration of the mesh by C60 molecules.  相似文献

It is widely accepted that the commensal gut microbiota contributes to the health and well-being of its host. The solitary tunicate Ciona intestinalis emerges as a model organism for studying host–microbe interactions taking place in the gut, however, the potential of its gut-associated microbiota for marine biodiscovery remains unexploited. In this study, we set out to investigate the diversity, chemical space, and pharmacological potential of the gut-associated microbiota of C. intestinalis collected from the Baltic and North Seas. In a culture-based approach, we isolated 61 bacterial and 40 fungal strains affiliated to 33 different microbial genera, indicating a rich and diverse gut microbiota dominated by Gammaproteobacteria. In vitro screening of the crude microbial extracts indicated their antibacterial (64% of extracts), anticancer (22%), and/or antifungal (11%) potential. Nine microbial crude extracts were prioritized for in-depth metabolome mining by a bioactivity- and chemical diversity-based selection procedure. UPLC-MS/MS-based metabolomics combining automated (feature-based molecular networking and in silico dereplication) and manual approaches significantly improved the annotation rates. A high chemical diversity was detected where peptides and polyketides were the predominant classes. Many compounds remained unknown, including two putatively novel lipopeptides produced by a Trichoderma sp. strain. This is the first study assessing the chemical and pharmacological profile of the cultivable gut microbiota of C. intestinalis.  相似文献

This study investigated the impact of small rodents on young trees during three growing seasons (2018–2020) in the Czech Republic. Tree damage by small rodents, the quantity and quality of herbaceous plant biomass and the species composition of small mammals were monitored at two sites in European beech forest plantations. The number of trees damaged during three growing seasons correlated positively with fibre content and negatively with nitrogen content in herbaceous plant biomass. The importance of winter precipitation is reflected in the positive correlation with nitrogen content in herbaceous plant biomass. The observed tree-gnawing damage correlated positively with the abundance of one rodent species only—the bank vole. The highest damage occurred after the concurrence of a dry winter and a higher number of voles. Given current climate variability, this situation could become more common in the future. Gnawing may be a limiting factor for the successful restoration of trees with thin bark—in our case, European beech. In addition, the proportion of this woody species should be increased to achieve a more stable and closer composition in the Czech Republic.

Oxygen plasma pre-treatment was applied to cotton fabric with the aim of improving the water repellency performance of an inorganic-organic hybrid sol-gel perfluoroalkyl-functionalized polysilsesquioxane coating. Cotton fabric was pre-treated with low-pressure oxygen plasma for different treatment times and operating powers. Afterward, 1H,1H,2H,2H-perfluorooctyltriethoxysilane (SiF) was applied to the cotton fabric samples using the pad-dry-cure method. The surfaces of the untreated and modified cotton fibers were characterised using Fourier transform infrared spectroscopy, Xray photoelectron spectroscopy, scanning electron microscopy, and atomic force microscopy. The water repellency of the SiF-coated fabric samples was evaluated using static and sliding contact angle measurements with water. The results show that the plasma treatment with the shortest treatment time (10 s) and the lowest operating current (0.3 A) increased the atomic oxygen/carbon ratio of the cotton fiber surface from 0.6 to 0.8 and induced the formation of a nano-sized grainy surface. Increasing the plasma treatment time and/or operating current did not intensify the surface changes of the cotton fibers. Such saturation effects were explained by the large influence of reactive oxygen atoms during the plasma treatment. The measured static water contact angles on the surface of the untreated and plasma pre-treated and SiF-coated cotton fabrics showed that the oxygen plasma pre-treatment enabled the increase of the water contact angle from 135° to ≈150°, regardless of the applied plasma treatment time and discharge power. This improvement in the hydrophobicity of the SiF coating was followed by a decrease in the sliding angle of water droplets by more than 10° compared to the plasma untreated and SiF-coated sample characterized by a water sliding angle of 45°. Additionally, measurements of the water sliding angle revealed that the increase of the static contact angle from 149° to 150° corresponded to a drop of the water sliding angle from 33 to 24°, which suggests that the plasma pre-treatment of 20 s at an operating current of 0.3 A produced the best water-repellent performance of the SiF-coated cotton fabric.  相似文献

OBJECTIVE: To sequence the exonic and splice site regions of the 4 desmosomal genes associated with the human form of familial arrhythmogenic right ventricular cardiomyopathy (ARVC) in Boxers with ARVC and identify a causative mutation. ANIMALS: 10 unrelated Boxers with ARVC and 2 unaffected Labrador Retrievers (control dogs). PROCEDURES: Exonic and splice site regions of the 4 genes encoding the desmosomal proteins plakophilin-2, plakoglobin, desmoplakin, and desmoglein-2 were sequenced. Sequences were compared for nucleotide sequence changes between affected dogs and the published sequences for clinically normal dogs and between affected dogs and the control dogs. Base-pair changes were considered to be causative for ARVC if they were detected in an affected dog but not in unaffected dogs, and if they involved a conserved amino acid and changed that amino acid to one of a different polarity, acid-base status, or structure. RESULTS: A causative mutation for ARVC in Boxers was not identified, although single nucleotide polymorphisms were detected in some affected dogs within exon 3 of the plakophilin-2 gene; exon 3 of the plakoglobin gene; exons 3 and 7 of the desmoglein-2 gene; and exons 6, 14, 15, and 24 of the desmoplakin gene. None of these changed the amino acid of the respective protein. CONCLUSIONS AND CLINICAL RELEVANCE: Mutations within the desmosomal genes associated with the development of ARVC in humans do not appear to be causative for ARVC in Boxers. Genomewide scanning for genetic loci of interest in dogs should be pursued.  相似文献

Metal mining is carried out in the drainage basin of Lake Kariba, Zimbabwe. In an attempt to assess the distribution of heavy metals in the lake ecosystem, the concentrations of chromium (Cr), cadmium (Cd), copper (Cu), lead (Pb), manganese (Mn), nickel (Ni), zinc (Zn), and selenium (Se) were analysed in the following ecosystem components at different locations in the lake; water, sediment, aquatic macrophytes, mussels, and carnivorous and herbivorous fish. Concentrations were generally in the same range as in other temperate and tropical aquatic ecosystems, except for Pb and Cd, which were higher in marcrophytes and fish from the lake. Apart from Cd, there were no signs of biomagnification and the highest levels of metals were found in the macrophytes and sediments. While metal contamination in fish is probably a combination of uptake from the food and an equilibrium with the surrounding water, the levels in macrophytes appear to be determined mainly by uptake from the sediment through the roots.  相似文献

OBJECTIVE: To establish a sensitive test for the detection of autoantibodies against thyroid peroxidase (TPO) in canine serum samples. SAMPLE POPULATION: 365 serum samples from dogs with hypothyroidism as determined on the basis of serum concentrations of total and free triiodothyronine (T3), total and free thyroxine (T4), and thyroid-stimulating hormone, of which 195 (53%) had positive results for at least 1 of 3 thyroid autoantibodies (against thyroglobulin [Tg], T4, or T3) and serum samples from 28 healthy dogs (control samples). PROCEDURE: TPO was purified from canine thyroid glands by extraction with detergents, ultracentrifugation, and precipitation with ammonium sulfate. Screening for anti-TPO autoantibodies in canine sera was performed by use of an immunoblot assay. Thyroid extract containing TPO was separated electrophoretically, blotted, and probed with canine sera. Alkaline phosphatase-conjugated rabbit anti-dog IgG was used for detection of bound antibodies. RESULTS: TPO bands were observed at 110, 100, and 40 kd. Anti-TPO autoantibodies against the 40-kd fragment were detected in 33 (17%) sera of dogs with positive results for anti-Tg, anti-T4, or anti-T3 autoantibodies but not in sera of hypothyroid dogs without these autoantibodies or in sera of healthy dogs. CONCLUSIONS AND CLINICAL RELEVANCE: The immunoblot assay was a sensitive and specific method for the detection of autoantibodies because it also provided information about the antigen. Anti-TPO autoantibodies were clearly detected in a fraction of hypothyroid dogs. The value of anti-TPO autoantibodies for use in early diagnosis of animals with thyroid gland diseases should be evaluated in additional studies.  相似文献
