Non-edible parts of crustaceans could be a rich source of valuable bioactive compounds such as the carotenoid astaxanthin and peptides, which have well-recognized beneficial effects. These compounds are widely used in nutraceuticals and pharmaceuticals, and their market is rapidly growing, suggesting the need to find alternative sources. The aim of this work was to set up a pilot-scale protocol for the reutilization of by-products of processed shrimp, in order to address the utilization of this valuable biomass for nutraceutical and pharmaceuticals application, through the extraction of astaxanthin-enriched oil and antioxidant-rich protein hydrolysates. Astaxanthin (AST) was obtained using “green extraction methods,” such as using fish oil and different fatty acid ethyl esters as solvents and through supercritical fluid extraction (SFE), whereas bioactive peptides were obtained by protease hydrolysis. Both astaxanthin and bioactive peptides exhibited bioactive properties in vitro in cellular model systems, such as antioxidant and angiotensin I converting enzyme (ACE) inhibitory activities (IA). The results show higher astaxanthin yields in ethyl esters fatty acids (TFA) extraction and significant enrichment by short-path distillation (SPD) up to 114.80 ± 1.23 µg/mL. Peptide fractions of <3 kDa and 3–5 kDa exhibited greater antioxidant activity while the fraction 5–10 kDa exhibited a better ACE-IA. Lower-molecular-weight bioactive peptides and astaxanthin extracted using supercritical fluids showed protective effects against oxidative damage in 142BR and in 3T3 cell lines. These results suggest that “green” extraction methods allow us to obtain high-quality bioactive compounds from large volumes of shrimp waste for nutraceutical and pharmaceutical applications. 相似文献

Fishery by-products are rich in biologically active substances and the use of green and efficient extraction methods to recover these high-added-value compounds is of particular importance. In this study, head, skin and viscera of rainbow trout and sole were used as the target matrices and accelerated solvent extraction (ASE) (45–55 °C, 15 min, pH 5.2–6.8, 103.4 bars) and pulsed electric fields (PEF) (1–3 kV/cm, 123–300 kJ/kg, 15–24 h) were applied as extraction technologies. The results showed that ASE and PEF significantly increased the protein extract efficiency of the fish by-products (p < 0.05) by up to 80%. SDS-PAGE results showed that ASE and PEF treatments changed the molecular size distribution of the protein in the extracts, which was specifically expressed as the change in the area or number of bands between 5 and 250 kDa. The antioxidant capacity of the extracts was evaluated by oxygen radical absorbance capacity (ORAC) and total antioxidant capacity (ABTS) assays. The results showed that both ASE and PEF treatments significantly increased the antioxidant capacity of rainbow trout and sole skin and head extracts (p < 0.05). Ultrasound-assisted water extraction was optimized to recover gelling biopolymers and antioxidant compounds from Mastocarpus stellatus. A set of experiments following a Box–Behnken design was proposed to study the influence of extraction time, solid liquid ratio, and ultrasound amplitude on the yield, sulfate content, and thermo-rheological properties (viscoelasticity and gelling temperature) of the carrageenan fraction, as well as the composition (protein and phenolic content) and antiradical capacity of the soluble extracts. Operating at 80 °C and 80 kHz, the models predicted a compromise optimum extraction conditions at ~35 min, solid liquid ratio of ~2 g/100 g, and ultrasound amplitude of ~79%. Under these conditions, 40.3% carrageenan yield was attained and this product presented 46% sulfate and good mechanical properties, a viscoelastic modulus of 741.4 Pa, with the lowest gelling temperatures of 39.4 °C. The carrageenans also exhibited promising antiproliferative properties on selected human cancer cellular lines, A-549, A-2780, HeLa 229, and HT-29 with EC50 under 51.9 μg/mL. The dried soluble extract contained 20.4 mg protein/g, 11.3 mg gallic acid eq/g, and the antiradical potency was equivalent to 59 mg Trolox/g. 相似文献

This study presents the validation of a high-performance liquid chromatography diode array detector (HPLC-DAD) method for the determination of different mycosporine-like amino acids (MAAs) in the red alga Bostrychia scorpioides. The investigated MAAs, named bostrychines, have only been found in this specific species so far. The developed HPLC-DAD method was successfully applied for the quantification of the major MAAs in Bostrychia scorpioides extracts, collected from four different countries in Europe showing only minor differences between the investigated samples. In the past, several Bostrychia spp. have been reported to include cryptic species, and in some cases such as B. calliptera, B. simpliciuscula, and B. moritziana, the polyphyly was supported by differences in their MAA composition. The uniformity in the MAA composition of the investigated B. scorpioides samples is in agreement with the reported monophyly of this Bostrychia sp. 相似文献

This study identified the role of milling and parboiling on arsenic (As) content and its species in large numbers of rice samples. Total As contents were 108 ± 33 μg/kg in polished rice grains (PR), 159 ± 46 μg/kg in unpolished rice grains (UR), 145 ± 42 μg/kg in parboiled polished rice grains (PPR) and 145 ± 44 μg/kg in parboiled unpolished rice grains (PUR). The percentages of inorganic As (iAs) were 66% ± 8% in PR and from 72% to 77% in other grain categories. The polishing process reduced the As content in the rice grains, removing outer part of the UR with high amount of As, whereas the parboiling technique transferred the semimetal content within the grain. Total As and iAs contents were not significantly different in UR, PPR and PUR, homogenizing its distribution inside the grains. The effects of two olive mill wastewater (OMW) dilutions upon physiological function and metabolic components of three mint species grown in hydroponic culture were studied. Visible symptoms, chlorosis and stunted growth, were pronounced in peppermint and spearmint; toxicity symptoms in menthe douce were few. Peppermint and spearmint displayed loss of calcium (Ca), magnesium (Mg), and potassium (K); menthe douce revealed accumulation of Ca, sodium (Na), and iron (Fe), under OMW. The loss of Ca negatively affected the development and stability of cell wall and membrane; the Mg deficiency negatively affected the photosynthetic apparatus and carbon fixation. Fv′/ Fm′ was slightly affected under OMW, whereas, ΦPSII, q P and Rfd displayed significant reductions. No clear effect of OMW on phenols and carbohydrates among the species were found. Ascorbic acid content in peppermint dropped dramatically; the reduction was lower for spearmint and menthe douce. This study aims to identify an alternative plant tissue to be used in the early diagnosis of boron (B) deficiency in chestnut (Castanea sativa Mill.). A B-deficient orchard was selected, and 16 trees were submitted to two levels of B fertilization. When flowers were in bloom, the following tissues were sampled: leaves, androgynous catkins and flowers. There was a significant increase in B content in plant tissues due to B application. In July, the highest B content was observed in flowers in B0, but leaves had the greatest content in B1. Boron content in the tissues collected in July was positively correlated with B contents in leaves sampled in September. Foliar B concentrations, irrespective of the sampling period, were correlated with chestnut productivity, while the other tissues did not. These results suggest that the leaves, sampled in bloom, were the most efficient tissue for the early diagnosis of B deficiency. 相似文献

The response of carob (Ceratonia siliqua L.) seedlings grown at different root zone temperatures affected by nitrate and ammonium nutrition was studied. When root temperatures ranged from 10 to 35°C, ammonium‐fed plants were significantly larger than nitrate‐fed plants. Ammonium‐fed plants displayed toxicity symptoms and were much smaller at 40°C root temperature in comparison with the nitrate‐fed plants grown at the same root temperature. Root/shoot ratio slightly increase with root temperature in ammonium‐ and nitrate‐fed plants in a similar way, and shoot demand per root unit decreased with root temperature between 15 and 25°C. There was a general increase in net photosynthesis with root temperature, though nitrate‐fed plants were more sensitive to low and ammonium‐fed plants to high temperatures. Increasing the root temperature of ammonium fed plants from 10 to 40°C leads to a 30% increase in the amount of photosynthates sent to the roots. The presence of ammonium resulted in the distribution of newly fixed carbon away from carbohydrates and into nitrogen compounds. Under conditions of salt stress, plants show qualitative and quantitative alterations in various organic compounds, such as nitrogen (N) compounds and organic acids. In this work, the effect of different saline levels as well as various N levels, supplied as nitrate (NO3) or as ammonium (NH4)+NO3 on the concentration of amino acids and organic acids in the leaves of tomato and cucumber plants has been studied. The effect of the source of N on individual amino acid contents varied with plant species. Most of the amino acids increased when the concentration of N in the nutrient solution was increased, except when N was added as NH4+NO3 for tomato. The effect of salt stress depended on which amino acid was considered. The data also indicate that the effect of salinity on each particular amino acid was greatly dependent on the plant species and N source. Organic acids were differently affected by salinity and by the N source, depending on the plant species. In tomato, the concentrations of short‐chain organic acids were 2–3 times higher in NO3‐supplied plants than in those grown with NH4+NO3. Finally, in cucumber, malic acid concentration increased as a function of the saline level in the medium. 相似文献
