Brucellosis control and eradication requires serological tests and vaccines. Effective classical vaccines (S19 in cattle and Rev 1 in small ruminants), however, induce antibodies to the O-polysaccharide of the lipopolysaccharide which may be difficult to distinguish from those resulting from infection and may thus complicate diagnosis. Rough attenuated mutants lack the O-polysaccharide and would solve this problem if eliciting protective immunity; the empirically obtained rough mutants 45/20 and RB51 have been used as vaccines. Strain 45/20 is reportedly unstable and it is not presently used. RB51 is increasingly used instead of S19 in some countries but it is rifampicin resistant and its effectiveness is controversial. Some controlled experiments have found good or absolute protection in adult cattle vaccinated orally (full dose) or subcutaneously (reduced dose) and in one field experiment, RB51 was reported to afford absolute protection to calves and to perform better than S19. Controlled experiments in calves, however, have shown reduced doses of RB51 to be ineffective, full doses only partially effective, and RB51 less effective than S19 against severe challenges. Moreover, other observations suggest that RB51 is ineffective when prevalence is high. RB51 is not useful in sheep and evidence in goats is preliminary and contradictory. Rough mutants obtained by molecular biology methods on the knowledge of the genetics and structure of Brucella lipopolysaccharide may offer alternatives. The B. abortus manBcore (rfbK) mutant seems promising in cattle, and analyses in mice suggest that mutations affecting only the O-polysaccharide result in better vaccines than those affecting both core and O-polysaccharide. Possible uses of rough vaccines also include boosting immunity by revaccination but solid evidence on its effectiveness, safety and practicality is not available. 相似文献

BACKGROUND
Cultivated bread wheat (Triticum aestivum L.) spontaneously hybridizes with wild/weedy related Aegilops populations, but little is known about the actual rates at which this hybridization occurs under field conditions. It is very important to provide reliable empirical data on this phenomenon in order to assess the potential crop–wild introgression, especially in the context of conducting risk assessments for the commercialization of genetically modified (GM) wheat, as gene flow from wheat to Aegilops species could transfer into the wild species genes coding for traits such as resistance to herbicides, insects, diseases or environmental stresses.RESULTS
The spontaneous hybridization rates between wheat and A. geniculata and A. triuncialis, which are very abundant in the Mediterranean area, have been estimated for the first time in the northern part of the Meseta Central, the great central plateau which includes the largest area of wheat cultivation in Spain. Hybridization rates averaged 0.12% and 0.008% for A. geniculata and A. triuncialis, respectively. Hybrids were found in 26% of A. geniculata and 5% of A. triuncialis populations, at rates that can be ≤3.6% for A. geniculata and 0.24% for A. triuncialis.CONCLUSION
The detection of Aegilops spp.–wheat hybrids in Aegilops populations indicates that gene flow can occur, although wheat is considered a crop with a low-to-medium risk for transgene escape. In order to obtain a better understanding of the active defense strategy of cherimoya (Annona cherimola Mill.) fruit, hydrolytic and antifungal activity, as well as expression of proteins functionally and immunogenically related to the pathogenesis-related proteins chitinase (PR-Q) and 1,3-β-glucanase (PR-2), were estimated in fruit at different ripening stages. Increase in expression of the 27 kDa constitutive chitinase and the induction of two new proteins, a 26 kDa chitinase and a 51 kDa 1,3-β-glucanase were associated with enhanced in vitro hydrolytic and antifungal activity of the acidic protein extract in ripe fruit. Ripening modified the expression of constitutive basic isoenzymes, with a sharp decrease in both relative accumulation and hydrolytic activity. Likewise, a new basic 33 kDa chitinase was induced in the over-ripe fruit, concomitant with accumulation of a basic constitutive 76 kDa 1,3-β-glucanase. At this stage, the basic protein extract modified in vitro growth inhibition of Botrytis cinerea. Short-term high CO2 treatment delayed fruit ripening and maintained a similar distribution of activity and isoenzymatic pattern in both protein fractions to that in unripe fruit. These results indicate that the changes in the pattern of defense proteins and hydrolytic activity in cherimoyas appear to be associated with ripening. Moreover, unlike the constitutively expressed isoenzymes, only the transitorily induced chitinases and 1,3-β-glucanases were associated with an active defense-related response. 相似文献

Context
Multi-scale approaches to habitat modeling have been shown to provide more accurate understanding and predictions of species-habitat associations. It remains however unexplored how spatial and temporal variations in habitat use may affect multi-scale habitat modeling.Objectives
We aimed at assessing how seasonal and temporal differences in species habitat use and distribution impact operational scales, variable influence, habitat suitability spatial patterns, and performance of multi-scale models.Methods
We evaluated the environmental factors driving brown bear habitat relationships in the Cantabrian Range (Spain) based on species presence records (ground observations) for the period 2000–2010, LiDAR data on forest structure, and seasonal estimates of foraging resources. We separately developed multi-scale habitat models for (i) each season (spring, summer, fall and winter) (ii) two sub-periods with different population status: 2000–2004 (with brown bear distribution restricted to the main population nuclei) and 2005–2010 (with expanding bear population and range); and (iii) the entire 2000–2010 period.Results
Scales of effect remained considerably stable across seasonal and temporal variations, but not the influence of certain environmental variables. The predictive ability of multi-scale models was lower in the seasons or periods in which populations used larger areas and a broader variety of environmental conditions. Seasonal estimates of foraging resources, together with LiDAR data, appeared to improve the performance of multi-scale habitat models.Conclusions
We highlight that the understanding of multi-scale behavioral responses of species to spatial patterns that continually shift over time may be essential to unravel habitat relationships and produce reliable estimates of species distributions.10.

Vibrio spp. are the most common and harmful shrimp pathogenic bacteria; however, microalgae and cyanobacteria have the ability to produce antimicrobial substances against these species. In this study, the organic and aqueous extracts of 28 species of marine microalgae and cyanobacteria were screened against Vibrio campbellii M1. Two of these phytoplankton species with antibacterial activity in aqueous extracts (Dunaliella tertiolecta and Skeletonema costatum) and nontoxic to brine shrimp Artemia franciscana nauplii were used to evaluate their anti‐Vibrio effect when used as green‐water cultures in Vibrio‐challenged white shrimp Litopenaeus vannamei cultures. No differences in mortality of juvenile L. vannamei were observed between treatments tested, suggesting that the pathogenicity of V. campbellii could be related to the growth stage of shrimp. The proximal composition of D. tertiolecta and S. costatum was in the recommended range for penaeid shrimp nutrition, allowing shrimp supplemented with these microalgae to have significantly greater total length and weight than control shrimp. Shrimp supplemented with S. costatum presented the highest values of organic mass (11.48 mg/organism) and growth rate (0.31 mg/d) in comparison to D. tertiolecta. These results indicate that microalgae are not only capable of producing antibacterial compounds against Vibrio but can also help shrimp nutrition. 相似文献
