Pertussis toxin (PTX) is a major virulence factor in whooping cough and can elicit protective antibodies. Amino acid residues 8 to 15 of PTX subunit S1 are important for the adenosine diphosphate-ribosyltransferase activity associated with the pathobiological effects of PTX. Furthermore, this region contains at least a portion of an epitope that elicits both toxin-neutralizing and protective antibody responses in mice. The gene encoding the S1 subunit was subjected to site-specific mutagenesis in this critical region. A mutant containing a single amino acid substitution (Arg9----Lys) had reduced enzymatic activity (approximately 0.02% of control) while retaining the protective epitope. This analog S1 molecule may provide the basis for a genetically detoxified PTX with potential for use as a component of an acellular vaccine against whooping cough. 相似文献

The rotational mobility of myoglobin in situ was determined by proton nuclear magnetic resonance line width measurements of a characteristic myoglobin resonance observed in bovine heart muscle spectra. The protein diffuses intracellularly at nearly half the rate observed in dilute solution. This high mobility allows the oxygenated form of myoglobin to contribute significantly to the overall diffusive flux of oxygen in respiring heart muscle. 相似文献

Both impulsivity and novelty-seeking have been suggested to be behavioral markers of the propensity to take addictive drugs. However, their relevance for the vulnerability to compulsively seek and take drugs, which is a hallmark feature of addiction, is unknown. We report here that, whereas high reactivity to novelty predicts the propensity to initiate cocaine self-administration, high impulsivity predicts the development of addiction-like behavior in rats, including persistent or compulsive drug-taking in the face of aversive outcomes. This study shows experimental evidence that a shift from impulsivity to compulsivity occurs during the development of addictive behavior, which provides insights into the genesis and neural mechanisms of drug addiction. 相似文献

There is a pressing need to understand the consequences of human activities, such as land transformations, on watershed ecosystem
services. This is a challenging task because different indicators of water quality and yield are expected to vary in their
responsiveness to large versus local-scale heterogeneity in land use and land cover (LUC). Here we rely on water quality data
collected between 1977 and 2000 from dozens of gauge stations in Puerto Rico together with precipitation data and land cover
maps to (1) quantify impacts of spatial heterogeneity in LUC on several water quality indicators; (2) determine the spatial
scale at which this heterogeneity influences water quality; and (3) examine how antecedent precipitation modulates these impacts.
Our models explained 30–58% of observed variance in water quality metrics. Temporal variation in antecedent precipitation
and changes in LUC between measurements periods rather than spatial variation in LUC accounted for the majority of variation
in water quality. Urbanization and pasture development generally degraded water quality while agriculture and secondary forest
re-growth had mixed impacts. The spatial scale over which LUC influenced water quality differed across indicators. Turbidity
and dissolved oxygen (DO) responded to LUC in large-scale watersheds, in-stream nitrogen concentrations to LUC in riparian
buffers of large watersheds, and fecal matter content and in-stream phosphorus concentration to LUC at the sub-watershed scale.
Stream discharge modulated impacts of LUC on water quality for most of the metrics. Our findings highlight the importance
of considering multiple spatial scales for understanding the impacts of human activities on watershed ecosystem services. 相似文献

Attenuated total reflection Fourier-transform infrared spectroscopy (ATR-FTIR), followed by multivariate treatment of the spectral data, was used to classify seed oils of the genus Cucurbita (pumpkins) according to their species as C. maxima, C. pepo, and C. moschata. Also, C. moschata seed oils were classified according to their genetic variety as RG, Inivit C-88, and Inivit C-2000. Up to 23 wavelength regions were selected on the spectra, each region corresponding to a peak or shoulder. The normalized absorbance peak areas within these regions were used as predictors. Using linear discriminant analysis (LDA), an excellent resolution among all categories concerning both Cucurbita species and C. moschata varieties was achieved. The proposed method was straightforward and quick and can be easily implemented. Quality control of pumpkin seed oils is important because Cucurbita species and genetic variety are both related to the pharmaceutical properties of the oils. 相似文献

In this study, antioxidant activities in free-radical-mediated oxidative systems and the genotoxic/antigenotoxic effects of two proteins with molecular mass around 17 kDa, purified from Solanum betaceum fruits (cyphomine) and Solanum tuberosum tubers (solamarine), were investigated. Both proteins inhibited uric acid formation with IC(50) values between 55 and 60 μg/mL, and both proteins were able to reduce oxidative damage by scavenging hydroxyl radicals and superoxide anion in a dose-dependent manner. Furthermore, the DPPH? reduction assay showed SC(50) values of 55-73 μg/mL. Cyphomine and solamarine were able to retain their antioxidant activity after heat treatment at 80 °C for 15 min. Allium cepa and Salmonella /microsome assays showed no genotoxic and mutagenic effects. Solamarine showed an antimutagenic effect against a direct mutagen (4-nitro-o-phenylenediamine). Consequently, the present study showed that the investigated proteins are promising ingredients for the development of functional foods with a beneficial impact on human health and an important source for the production of bioactive peptides. 相似文献

Forest declines are usually complex multifactorial phenomena that involve interactions between different factors. The possible interaction between different types of mycelial pathogens was investigated through artificial inoculation of oak seedlings, involving two root rot basidiomycetes, Collybia fusipes and Armillaria mellea, and two Phytophthora species, P. cinnamomi and P. cambivora. These pathogens were inoculated onto young Quercus robur saplings in greenhouse conditions, either alone or combining a root rot basidiomycete with a Phytophthora species. Three out of the four Phytophthora spp.*root rot basidiomycete combinations tested resulted in significantly greater damage to the oak host than the sum of the damages induced by the individual pathogens. In late 2003, a new disease appeared in protected bean crops in southeastern Spain, causing a decrease of over 50% in production.
Several samples of affected plants were collected and analyzed and the agent of this disease was identified as the bacterium
Erwinia aphidicola, which had never been described as a pathogen previously. We attempted to determine the possible bacterium transmission through
seeds, using 120 commercial bean seeds from the same batch as that used in an affected farm, and 120 seeds from the fruiting
plants of the same farm. Seed coats, cotyledons and leaves of plants originating from them, were taken and analyzed. Several
of the developed symptoms on plants from commercial and fruiting plant seeds were internervial chlorosis, necrotic pits and
rough roots and they coincided with those observed on affected crops. Bacteria present in commercial seed cotyledons were
isolated and analyzed by biochemical and molecular tests. Results confirmed the presence of Erwinia aphidicola in four analyzed seeds; moreover, Bacillus simplex/Bacillus muralis, Pseudomonas mendocina, Pseudomonas putida and Paenibacillus polymyxa were also identified. 相似文献
