The effect of the primary infection site by Fusarium graminearum and F. culmorum within wheat ears on Fusarium head blight (FHB) was investigated under controlled conditions. FHB development was assessed visually and thermographically following inoculation by: (i) spraying ears, or injecting inoculum into spikelets on (ii) tip, (iii) centre and (iv) base of the ears, separately. Fusarium infection significantly increased the temperature span within ears 6 days post inoculation (dpi), especially infections starting at the ear tip. The temperature difference between air and ear was negatively correlated to FHB severity and enabled disease detection even 29 dpi. F. culmorum caused significant higher disease severity neither reflected in the frequency of infected kernels nor in thousand kernel weight (TKW). Spray inoculations had the strongest effect on TKW, whereas tip inoculations had no effect. Centre and base inoculations had intermediate effects on TKW, although FHB levels did not differ with the same trend among inoculation scenarios. The overall low correlations among FHB severity, infected kernels and TKW are explained by the pathogen spread within ears – downwards more than upwards – and the effect on yield formation which is lower for infections of the upper parts of ears. An exponential model showed high goodness of fit for gradients of infected kernels within ears (R 2 ≥ 70) except tip infection with F. culmorum. This study confirmed that FHB is a function of the primary infection site within ears. Thermography was useful to differentiate among infection scenarios and may be applied in breeding for FHB resistance. 相似文献

ABSTRACT The need for salinity resistance in turfgrass is increasing because of the enhanced use of effluent and other low-quality water for turfgrass irrigation. Although most turfgrasses form an arbuscular mycorrhizal fungus (AMF) symbiosis, there is little information on the mycorrhization of turfgrass species. Therefore, the aim of this study was to determine the effects of three AMF species, Glomus intraradices Schenck & Smith, Glomus etunicatum Becker & Gerdemann, and Glomus deserticola Trappe & John, and a mixture thereof on the growth, productivity, and nutrient uptake of two species of cool-season turfgrasses, Challenger Kentucky bluegrass (Poa pratensis L.) and Arid tall fescue (Festuca arundinacea Schreb.), and to relate the effects to colonization of the roots by mycorrhiza to assess the dependency of the plants (mycorrhizal dependency [MD]). Following the experimental period (4 months) and measurements, the mycorrhizal inoculated plants had significantly greater biomass production compared to that of non-inoculated plants. MD and shoot mineral contents (particularly P) differed among turfgrass hosting AMF, and the highest value (13%) occurred for P. pratensis and F. arundinacea seedlings colonized with G. intraradices and G. deserticola, respectively. The P content was highest for the F. arundinacea/mixed AMF combination compared to other treatments. We confirmed that mycorrhizal inoculation (P. pratensis/G. intraradices and F. arundinacea/mixed AMF combinations) enhanced plant productivity and nutrient uptake (especially P) even under non-optimum conditions. 相似文献

First detection of Neoscytalidium dimidiatum associated with canker disease in Egyptian Ficus trees 下载免费PDF全文
In September 2013, a canker disease of Ficus nitida and F. benjamina was reported for the first time in Assiut governorate, Egypt. Infected samples were collected from various locations. Pure cultures of a fungus were isolated on potato dextrose agar at 25°C from diseased plants. Morphological investigation and DNA sequencing showed that the causal agent was Neoscytalidium dimidiatum. A pathogenicity test conducted using 2‐year‐old plants of each host gave 70%–80% infection, and the pathogen was reisolated from the inoculated plants. A pure culture of N. dimidiatum was deposited in the culture collection of the Assiut University Mycological Centre (AUMC) under the code AUMC 9293 and the ITS sequence was placed in NCBI under accession number KX985929 . 相似文献

Enhancement of the transfection efficiency of DNA into Crocus sativus L. cells via PEI nanoparticles
Over the past decade, several natural and synthetic cationic polymers have been utilized for gene delivery into cells. Among them, polyethylenimine (PEI) was used for gene therapy successfully. The present study investigated the effect of PEI and ultrasound waves on ssDNA delivery into saffron cells. Gel retardation, dynamic light scattering (DLS) and scanning electron microscopy (SEM) assays were employed to determine the physicochemical properties of PEI/f-DNA polyplex (complex of PEI and fluorescently labeled DNA). Moreover, the cytotoxicity of PEI, PEI/f-DNA polyplex and ultrasound were investigated on saffron cells at different concentrations. The gel retardation results indicated that the formation and neutralization of the PEI/f-DNA polyplex were completed at N/P=5. The particle size distribution of the polyplexes was from 50 to 122 nm. The experimental results revealed that the cytotoxicity of the PEI/f-DNA polyplex was lower than that of PEI alone, hence the cells showed both dose- and exposure duration-dependent responses. Furthermore, the viability of saffron cells declined extremely after 5 and 10 min sonication but this reduction was not significant at 2 min exposure duration. The results also indicated that the combined utilization of ultrasound and PEI nanoparticles increased the transfection efficiency of saffron cells up to two times higher than those obtained by PEI or ultrasound separately. 相似文献

This aim of the study is to investigate a halophilic bacterium Hortaea sp. B15, isolated from petroleum-contaminated soil for biodegradation of phenanthrene. Hortaea sp. B15 has the ability to completely degrade phenanthrene (100 mg/L) under salinity 10% within 1-week incubation. The metabolitic product of phenanthrene was identified and assayed by using ultraviolet-visible spectrophotometer and mass spectral analysis. Result revealed that Hortaea sp. B15 metabolized phenanthrene to form 9,10-phenanthrene quinone, salicylic acid, and gentisic acid. Hortaea sp. B15 has an efficient utilization of phenanthrene in high-saline liquid medium. All the results indicated that the fungus has a promising application for the study of high-molecular-weight PAH biodegradation and contaminated saline-alkali soil bioremediation. 相似文献

Non-woody biomass species have high-energy potentials, which could be used for bioenergy production. Invasive species are species spreading into areas, where they are not native, consequently causing environmental and economic problems. Therefore, the present study evaluated the proximate, ultimate, chemical, and fuel characteristics of wood and charcoal of three invasive non-forest tree species in Saudi Arabia: Calotropis procera, Rhazya stricta, and Phragmites australis, which were compared with the wood of Acacia tortilis, a preferable local fuelwood. All these data were discussed to investigate the possibility of using the invasive plants for energy production. The thermal behavior of wood was analyzed using thermo-gravimetric and derivative thermo-gravimetric methods. Overall, compared with the wood of A. tortilis, the woods of R. stricta and P. australis are suitable for energy production. Culex pipiens mosquitoes are vectors to many viruses and can transmit diseases such as filariasis and avian malaria. The present study evaluated the larvicidal activity of marine-derived endophytic fungi Aspergillus nomius and Aspergillus flavus from the soft coral Sarcophyton ehrenbergi along with two known cyclodepsipeptide compounds, scopularide A (1) and B (2), isolated from A. flavus extract, against third-instar larvae of C. pipiens, using distilled water as a negative control and toosenedanin as a positive control. The structures of the isolated compounds were confirmed by various spectroscopic analyses. The lethal concentrations (LC50 and LC90) were calculated by probit analysis. Scopularide A was the most potent after 96 h treatment, with LC50 and LC90 values of 58.96 and 994.31 ppm, respectively, and with 82.66% mortality at a concentration of 300 ppm. To unravel the biochemical mechanism of the tested extracts and compounds, their effects against protease, chitinase, phenoloxidases and lipase enzymes from the whole-body tissue of C. pipiens were evaluated after 72 h treatment at LC50 dose. Superior activity was observed for A. flavus extract against all tested enzymes. A molecular docking study was conducted for scopularide A and B on the four tested enzymes, to further verify the observed activity. Results revealed good binding affinities for both compounds as compared to the docked ligands, mainly via a number of hydrogen bonds. This was the first study to report the isolation of endophytic fungi A. flavus and A. nomius from the marine soft coral S. ehrenbergi. The endophytic fungal extract of A. flavus was found to be a promising source for a natural larvicidal agent against C. pipiens populations. 相似文献

为实现在自然环境条件下对苦瓜叶部病害的目标检测,该研究提出了一种基于改进的更快速区域卷积神经网络(Faster Region with Convolutional Neural Network Features,Faster R-CNN)的苦瓜叶部病害目标检测方法。Faster R-CNN以残差结构卷积神经网络ResNet-50作为该次试验的特征提取网络,将其所得特征图输入到区域建议网络提取区域建议框,并且结合苦瓜叶部病害尺寸小的特点,对原始的Faster R-CNN进行修改,增加区域建议框的尺寸个数,并在ResNet-50的基础下融入了特征金字塔网络(Feature Pyramid Networks,FPN)。结果表明,该方法训练所得的深度学习网络模型具有良好的鲁棒性,平均精度均值(Mean Average Precision,MAP)值为78.85%;融入特征金字塔网络后,所得模型的平均精度均值为86.39%,提高了7.54%,苦瓜健康叶片、白粉病、灰斑病、蔓枯病、斑点病的平均精确率(Average Precision,AP)分别为89.24%、81.48%、83.31%、88.62%和89.28%,在灰斑病检测精度上比之前可提高了16.56%,每幅图像的检测时间达0.322 s,保证检测的实时性。该方法对复杂的自然环境下的苦瓜叶部病害检测具有较好的鲁棒性和较高的精度,对瓜果类疾病预防有重要的研究意义。 相似文献
