Selenium (Se) is essential for humans and animals but has no known function in plants. Excess accumulation is toxic to both plants and animals. Dietary intake of Se is low in a large number of people worldwide. This is due to low bioavailability of Se in some soils and consequently low concentrations of Se in plant tissues.Both selenate and selenite are taken up by plants and subsequently translocated around the plant. Selenate, an analogue of sulphate, is transported by the sulphate transporter family. Some plants are able to accumulate high internal concentrations of Se (hyperaccumulators); however, genetic variation in accumulation ability amongst non-accumulators such as cereals, is relatively small.Within plant tissues, Se enters the pathways for sulphate assimilation and metabolism and will replace cysteine and methionine in proteins, often with detrimental effect. Alternatively, Se may be accumulated as methylated derivatives or lost from the plant following volatilisation.Agronomic biofortification of crops with Se-containing fertilisers, which is practised in some countries, provides the best short-term solution for improving Se content of wheat. Longer-term genetic improvement, particularly by targeting substrate discrimination of transporters between selenate and sulphate, for example, may provide a means to enhance uptake and promote accumulation.  相似文献

Wetlands are important habitats not only for their unique ecological value but also because they contain organic material that is fundamental to our understanding of precedent landscape and human past. This study compares the effects of two different land-management regimes on metabolic diversity and bacterial community structure with depth in order to relate them to the process of organic matter degradation and the potential for preservation in situ of organic archaeological artefacts in wetland soils. Soil cores were collected at five depths down to 100 cm from two wetlands sites in England. Environmental variables were monitored and the metabolic capabilities of the microbial community were studied using Biolog Ecoplates®. DNA was extracted from soil, and the bacterial community structure was examined by polymerase chain reaction followed by denaturing gradient gel electrophoresis (PCR-DGGE). To determine compositional changes in the bacterial community with depth, information about specific groups of bacteria at the site with higher water table (Hatfield Moor) was obtained by cloning and sequencing of 16S rRNA genes. Biolog and DGGE analyses showed depth variation and between-site variation. Carbon substrate utilization and bacterial diversity decreased with increasing depth. The wetland soil under an arable regime in which the water levels were kept elevated, showed higher metabolic capability and bacterial richness when compared with the soil under pasture and subjected to long-standing drainage. Cloning and sequencing showed that Proteobacteria and Acidobacteria were the predominant taxa within the soil profile, but there was a clear shift in bacterial community composition with increasing depth as several taxonomic groups (δ-Proteobacteria and Spirochaetes) were only detectable at 50 cm depth. Because the site with a high and stable water table presented higher metabolic activity and bacterial diversity, it may be that saturated conditions and a high water table are not sufficient to guarantee the preservation in situ of organic material such as archaeological artefacts.  相似文献

Acetolactate synthase (ALS) from one susceptible and two chlorsulfuronresistant biotypes of Stellaria media(L.) Vill. was assayed in the presence of eight known ALS inhibitors. As expected, ALS from the chlorsulfuronresistant biotypes (R1 and R2) showed reduced sensitivity to chlorsulfuron and other sulfonylurea herbicides. The patterns of cross-resistance varied, however, indicating that the alteration in ALS that confers chlorsulfuron resistance does not confer the same level of resistance to other sulfonylurea herbicides. The resistant biotypes were highly cross-resistant to sulfometuron-methyl and DPX-A7H81, but less cross-resistant to triasulfuron. Both R1 and R2 were highly cross-resistant to DTPS (N-[2,6-dichlorophenyl]-5,7-dimethyl-1,2,4-iriazolo[1,5a]pyrimidine-2-siilfoiiamide), but only slightly cross-resistant to imazamethahenz, an imidazolinone herbicide. The differences in the patterns of cross-resistance observed presumably reflect differences in the binding affinity of the herbicides for the altered ALS. The data presented suggest, but do not confirm, that R1 and R2 contain the same ALS mutation.  相似文献

Many insecticides (e.g. DDT, lindane, pyrethroids) are hydrophobic molecules which bind extensively to biological membranes. Binding of insecticides to phospholipid bilayers frequently shows saturation, with binding increasing with increasing insecticide concentration up to some limit, beyond which no further insecticide is incorporated into the bilayer. For lindane, this limit has been shown to correspond to the concentration at which the aqueous phase becomes saturated. Simple alkanes show similar saturation phenomena, and binding of halogenated alkanes can be followed by fluorescence quenching methods. It is shown that effects of hydrophobic molecules on bilayer fluidity are small. Effects of alkanes and insecticides on the activity of the (Ca2+-Mg2+)-ATPase purified from sarcoplasmic reticulum follow from direct binding to the ATPase, at sites which are not at the lipid-protein interface of the ATPase.  相似文献

An adequate supply of nitrogen (N) is essential for the successful establishment and sustainable productivity of forest stands. N deficits may necessitate the use of artificial fertilisers. Availability of N in the inorganic forms, and the relative abundance of the NO3-N and NH4-N components, influences the species composition of natural forest vegetation. Hence it is essential to use reliable measures of soil N supply that fully reflect its ecological significance. The new Ecological Site Classification (ESC) used in British forestry employs a multi-factorial definition of soil nutrient regime (SNR), including soil N. To develop this, a soil and vegetation study was made at 89 forest sites throughout Great Britain covering the major soil types used for forestry. “Total N” levels were compared with separate pre- and post-incubation measures of the two inorganic N components as potential indices of soil N supply. Multivariate statistical analysis showed that the major discriminant chemical variables for the sampled soils were pH, calcium and NO3-N and that these were also the main variables influencing the species composition of the ground vegetation. Total N and NH4-N were less effective discriminant variables for these sites. In some infertile soils the levels of NH4-N or total N may be of greater importance, as NO3-N is usually in very limited supply. A multivariate gradient of SNR, which incorporates the NO3-N measures, has been adopted for use within the ESC system. The position of a site on this gradient can be estimated quantitatively from soil type, ground vegetation species composition and humus type. This enables soil N supply and overall SNR to be assessed in a simple but effective way that guides the operational management of British forest soils for sustainable productivity. It will also be possible to use these techniques to monitor the nutritional status of forest sites over time.  相似文献
