BACKGROUND: Fifteen novel derivatives of D‐DIBOA, including aromatic ring modifications and the addition of side chains in positions C‐2 and N‐4, had previously been synthesised and their phytotoxicity on standard target species (STS) evaluated. This strategy combined steric, electronic, solubility and lipophilicity requirements to achieve the maximum phytotoxic activity. An evaluation of the bioactivity of these compounds on the systems Oryza sativa–Echinochloa crus‐galli and Triticum aestivum–Avena fatua is reported here. RESULTS: All compounds showed inhibition profiles on the two species Echinochloa crus‐galli (L.) Beauv. and Avena fatua L. The most marked effects were caused by 6F‐4Pr‐D‐DIBOA, 6F‐4Val‐D‐DIBOA, 6Cl‐4Pr‐D‐DIBOA and 6Cl‐4Val‐D‐DIBOA. The IC50 values for the systems Echinochloa crus‐galliOryza sativa and Avena fatua–Triticum aestivum for all compounds were compared. The compound that showed the greatest selectivity for the system Echinochloa crus‐galliOryza sativa was 8Cl‐4Pr‐D‐DIBOA, which was 15 times more selective than the commercial herbicide propanil (Cotanil‐35). With regard to the system Avena fatua–Triticum aestivum, the compounds that showed the highest selectivities were 8Cl‐4Val‐D‐DIBOA and 6F‐4Pr‐D‐DIBOA. The results obtained for 6F‐4Pr‐D‐DIBOA are of great interest because of the high phytotoxicity to Avena fatua (IC50 = 6 µM , r2 = 0.9616). CONCLUSION: The in vitro phytotoxicity profiles and selectivities shown by the compounds described here make them candidates for higher‐level studies. 8Cl‐4Pr‐D‐DIBOA for the system Echinochloa crus‐galliOryza sativa and 6F‐4Pr‐D‐DIBOA for Avena fatuaTriticum aestivum were the most interesting compounds. This study examined the effects of road transportation on metabolic and immunological responses in dairy heifers. Twenty Holstein heifers in early pregnancy were divided into non‐transported (NT; n = 7) and transported (T; n = 13) groups. Blood was collected before transportation (BT), immediately after transportation for 100 km (T1) and 200 km (T2), and 24 h after transportation (AT). The T heifers had higher (P < 0.05) blood cortisol and non‐esterified fatty acid concentrations after T1 and T2 than did NT heifers. By contrast, the T heifers had lower (P < 0.05) serum triglyceride concentrations after T1 and T2 than had the NT heifers. The serum cortisol and triglyceride concentrations returned (P > 0.05) to the BT concentrations at 24 h AT in the T heifers. The granulocyte‐to‐lymphocyte ratio and the percentage of monocytes were higher (P < 0.05) after T2 in the T heifers than in the NT heifers, suggesting that transportation stress increased the numbers of innate immune cells. T heifers had higher (P < 0.01) plasma haptoglobin concentrations than NT heifers 24 h AT. In conclusion, transportation increased cortisol secretion and was correlated with increased metabolic responses and up‐regulation of peripheral innate immune cells in dairy heifers.  相似文献

We examined whether mutation of the platelet‐derived growth factor receptor protein tyrosine kinase (PDGFR)‐α and PDGFR‐β genes contributes to their overexpression in canine vascular tumours. Genomic sequences of trans‐ or juxtamembrane regions of PDGFR‐α and PDGFR‐β were analysed with immunohistochemical staining and polymerase chain reaction‐direct sequencing using DNA from paraffin‐embedded neoplastic tissues of 27 hemangiosarcomas (HSAs) and 20 hemangiomas (HAs). Immunohistochemically, 75% of the HA cases were positive for PDGFR‐α and almost most of the HA cases were negative for PDGFR‐β. Of the HSA cases, 55.6% were negative for PDGFR‐α and 63% were strongly positive for PDGFR‐β. Among the HA cases, 1 missense mutation was detected in PDGFR‐α exon 18 and 1 in PDGFR‐β exon 17. Two HSA cases had missense mutations in exon 14 and 1 in exon 17 of PDGFR‐β. Thus, genomic mutation of trans‐ or juxtamembrane regions of PDGFRs was not the main mechanism driving the activation of receptors in HSA and HA.  相似文献

To evaluate arachidonic acid-related immunoregulatory mechanisms during long-term persistent pestivirus infection, we measured plasma contents of leukotriene C4 (LTC4), prostaglandin D2 (PGD2) and their plasma fatty acid (FA) precursor, arachidonic acid (AA), in six lambs with congenital border disease (BD). Significantly elevated average plasma LTC4 during the first half year of life was associated with increased PDG2 when compared to uninfected control lambs. Significantly elevated total plasma esterified AA stores suggest an effective BDV-mediated prostenoid immunostimulation. However, at 1 yr old, prostenoid secretion had fallen to normal (LTC4) or below normal (PGD2) levels. In contrast, there remained significantly elevated plasma esterified AA, present as available substrate for formation of these anti-viral immunoregulatory agents. These results suggested that preventing mobilization of AA from lipid stores for effective immune responses may be a viral strategy of BD virus that is associated with long term border disease effects.  相似文献

Captive leopard geckos (Eublepharis macularius) often present to the exotic clinic for gastrointestinal impactions, follicular stasis, or dystocia. To our knowledge, normal ultrasonographic anatomy of these lizards has not been described. The objectives of this prospective, anatomic, analytical study were to develop ultrasound techniques for this species and to describe the normal sonographic anatomy of the head, coelomic cavity, and tail. Eleven, healthy, female leopard geckos were included. A linear array 13–18 MHz transducer was used. Geckos were sedated and restrained in dorsal recumbency for coelomic structure examination and in ventral recumbency for head and tail examinations. Sagittal and transverse images were acquired and authors recorded qualitative and quantitative ultrasonographic characteristics of anatomic structures. The ventral surface of the lungs, liver, gallbladder, caudal vena cava, portal vein, ventral abdominal vein, aorta, ovarian follicles, fat bodies, tail, and brain were visualized in 10 of 11 individuals. In one individual, molt precluded ultrasonographic examination. The heart, kidneys, urinary bladder, spleen, and pancreas were not visualized. The digestive tract was observed in 10 individuals but was too small to be measured. Findings from the current study could be used as a reference for future studies of leopard geckos.  相似文献

Brucellosis control and eradication requires serological tests and vaccines. Effective classical vaccines (S19 in cattle and Rev 1 in small ruminants), however, induce antibodies to the O-polysaccharide of the lipopolysaccharide which may be difficult to distinguish from those resulting from infection and may thus complicate diagnosis. Rough attenuated mutants lack the O-polysaccharide and would solve this problem if eliciting protective immunity; the empirically obtained rough mutants 45/20 and RB51 have been used as vaccines. Strain 45/20 is reportedly unstable and it is not presently used. RB51 is increasingly used instead of S19 in some countries but it is rifampicin resistant and its effectiveness is controversial. Some controlled experiments have found good or absolute protection in adult cattle vaccinated orally (full dose) or subcutaneously (reduced dose) and in one field experiment, RB51 was reported to afford absolute protection to calves and to perform better than S19. Controlled experiments in calves, however, have shown reduced doses of RB51 to be ineffective, full doses only partially effective, and RB51 less effective than S19 against severe challenges. Moreover, other observations suggest that RB51 is ineffective when prevalence is high. RB51 is not useful in sheep and evidence in goats is preliminary and contradictory. Rough mutants obtained by molecular biology methods on the knowledge of the genetics and structure of Brucella lipopolysaccharide may offer alternatives. The B. abortus manBcore (rfbK) mutant seems promising in cattle, and analyses in mice suggest that mutations affecting only the O-polysaccharide result in better vaccines than those affecting both core and O-polysaccharide. Possible uses of rough vaccines also include boosting immunity by revaccination but solid evidence on its effectiveness, safety and practicality is not available.  相似文献

Using a 37‐year recruitment time series, we uncovered a field pattern revealing a strong, inverse relationship between bloater Coregonus hoyi recruitment success and slimy sculpin Cottus cognatus biomass in Lake Michigan (United States), one of the largest freshwater lakes of the world. Given that slimy sculpins (and deepwater sculpin Myoxocephalus thompsonii) are known egg predators that spatiotemporally overlap with incubating bloater eggs, we used recently published data on sculpin diets and daily ration to model annual bloater egg consumption by sculpins for the 1973–2010 year‐classes. Although several strong year‐classes were produced in the late 1980s when the proportion of eggs consumed by slimy sculpins was extremely low (i.e., <0.001) and several weak year‐classes were produced when the proportion of bloater eggs consumed was at its highest (i.e., >0.10–1.0), egg predation failed to explain why recruitment was weak for the 1995–2005 year‐classes when the proportion consumed was also low (i.e., <0.02). We concluded that egg predation by slimy and deepwater sculpins could have limited bloater recruitment in some years, but that some undetermined factor was more important in many other years. Given that slimy sculpin densities are influenced by piscivorous lake trout Salvelinus namaycush, the restoration of which in Lake Michigan has lagged behind those in lakes Superior and Huron, our study highlights the importance of an ecosystem perspective when considering population dynamics of fishes.  相似文献

This study was conducted to evaluate the effects of dietary supplementation of Barodon, an anionic alkali mineral complex, on growth, feed utilization, humoral innate immunity and disease resistance of olive flounder. A basal experimental diet was used as a control and supplemented with 0.1, 0.2, 0.3, 0.4, or 0.5% Barodon. Triplicate groups of fish (26.4 ± 0.2 g) were fed one of the diets to apparent satiation twice daily for 10 wk. The growth performance was enhanced (P < 0.05) linearly and quadratically in fish fed diets containing Barodon compared with that in fish fed the control. Feed utilization was significantly improved by Barodon supplementation. Serum lysozyme and antiprotease activities were increased quadratically in Barodon fed groups. Also, significantly higher superoxide dismutase activity was found in Barodon‐fed fish. Dietary supplementation of 0.1–0.3% Barodon resulted in significant enhancement of fish disease resistance against Streptococcus iniae. The findings in this study indicate that dietary supplementation of Barodon can enhance growth, feed utilization, innate immunity, and disease resistance of olive flounder and that the optimum level seems to be 0.1% in diets.  相似文献

The Mexican cichlid Cichlasoma beani is currently exploited regionally as food and can be commercialized in the aquarium trade. Natural populations of C. beani may already be negatively affected by anthropogenic alteration of the areas in which it is distributed. The aim of the present study was to examine the effect on growth, survival, and condition of C. beani cultured in three stocking densities: three (D3), six (D6), and nine (D9) fish per each 40 L tank. At the end of a 6‐wk trial the fish cultured in D3 were longer, heavier, and grew faster than the rest of the treatments but their survival was the lowest compared to D6 and D9. The mortalities were caused by a strong aggressive behavior in D3.  相似文献
