Glycoconjugate vaccines provide effective prophylaxis against bacterial infections. To date, however, no commercial vaccine has been available in which the key carbohydrate antigens are produced synthetically. We describe the large-scale synthesis, pharmaceutical development, and clinical evaluation of a conjugate vaccine composed of a synthetic capsular polysaccharide antigen of Haemophilus influenzae type b (Hib). The vaccine was evaluated in clinical trials in Cuba and showed long-term protective antibody titers that compared favorably to licensed products prepared with the Hib polysaccharide extracted from bacteria. This demonstrates that access to synthetic complex carbohydrate-based vaccines is feasible and provides a basis for further development of similar approaches for other human pathogens.  相似文献

Mercury in the aquatic biota and geologic materials in areas without anthropogenic sources has been stimulating the discussion about the possibility of natural Hg occurrence in the Amazon region. In this study the dispersion of Hg in different geologic materials as well as its relationship with high Hg levels, detected in some species of carnivorous fish consumed in the Rio Branco city, is evaluated. Bottom and suspended sediments, river water and fishes were sampled in the Acre and Purus Rivers and in some of their tributaries. Total mercury as well as physical and chemical characterization of the waters were done. Hg on top soil and upper horizons samples were also determined to evaluate the potential Hg accumulation in these materials. In addition the dominant minerals were determined by X-ray Diffraction analysis. Furthermore, ten samples of fossilized material were analyzed. The Hg analysis were carried out by CVAAS. The Hg mean levels in the bottom and suspensed sediments were 0.042 and 0.060 μg g-1, respectively. In the river water samples, the Hg levels were below the detection limit. The Hg mean values in the different soil samples were as follow: soil samples (0.077 μ g-1), lower (50–60 cm depth) horizons samples (0.117 μ g-1), ferruginous nodules within the clayey matrix (0.190 μ g-1) and fossilized material (0.379 μ g-1). In the fishes the Hg levels are high with mean of the 1.287 μ g-1 in the carnivorous species. These levels indicate a possible bioaccumulation of Hg mostly in the carnivorous fishes. Although low concentrations of Hg were found in the river waters, it is assumed here that the bioavailable Hg can is original large from the ferruginousnodules and fossilized materials.  相似文献

Ecological interactions of species and thus their spatial pat- terns may differ between homogeneous and heterogeneous forests. To account for this, techniques of point pattern analysis were implemented on mapped locations of tree individuals from two 1-ha tropicalforest plots in Vietnam. We analyzed the effect of environmental heterogeneity on tree distributions; spatial distribution patterns of dominant species; inter-specific associations; and conspecific associations between life stages. Our analyses showed that: environmental conditions were homo- geneous at plot 1 but heterogeneous at plot 2; in both plots, all six domi- nant species were aggregated at various scales up to 30 m, and tree spe- cies were aggregated at larger scales in the homogeneous site than in the heterogeneous site; attraction between pairs of species was remarkably higher at the homogeneous site while negative associations were more frequent in the heterogeneous site; some species, H. kurzii, T. ilicifolia (homogeneous plot) and D. sylvatica, S. wightianum (heterogeneous plot) showed a lack of early life-stage individuals near conspecific adults. Moreover, additional clustering of young individuals was independent from conspecific adults, except D. sylvatica in both sites. These findings are consistent with the Janzen-ConneU hypothesis. Overall, habitat het- erogeneity influences spatial patterns and inter-specific associations of the tree species and evidences of self-thinning are shown in most species.  相似文献

With the proliferation of private standards many significant decisions regarding public health risks, food safety, and environmental impacts are increasingly taking place in the backstage of the global agro-food system. Using an analytical framework grounded in political economy, we explain the rise of private standards and specific actors – notably supermarkets – in the restructuring of agro-food networks. We argue that the global, political-economic, capitalist transformation – globalization – is a transition from a Fordist regime to a regime of flexible accumulation (Harvey, 1989). We also argue that the standard making process of this new regulatory regime is increasingly moving from the front stage – where it is open to public debate and democratic decision-making bodies – to the backstage – where it is dominated by large supermarket procurement offices. We assert that transnational supermarket chains are increasingly controlling what food is grown where, how, and by whom. We also contend that the decision-making processes of transnational supermarket chains are typically “black-boxed.” The Euro-Retailer Produce Working Group (EUREP) is presented as a case of private governance by transnational supermarket chains. We conclude by examining the limitations and long-term efficacy of a system of private governance in the global agro-food system. In the area of Central Hesse we observed an unusual frequency of deaths of hares. The characteristic pathological alterations included general icterus, acute liver dystrophy, chromoproteinemic nephrosis, hyperemia in the tracheal mucosa and in the lungs, and splenic vascular congestion and necroses. Affected animals showed an altered behaviour with a diminished escape range and escape distance. Some animals had lost orientation completely. On the basis of sender questioning it appears probable that rape cultivation, as intensified in 1986--particularly newly generated 00-sorts--can be held responsible.  相似文献

In an era where the dominant implicitfarming policy has been ``get big or get out,'Titus Farms provides an exemplar to thecontrary. This paper explains the success storybehind a medium-sized 145-acre fruit andvegetable family farm in southeast Michigan.Using agroecosystems analysis, this case studydemonstrates a rigor and holism essential tofarming systems research and analysis. In-depthinterviews with the principal farm manager andowner were conducted during the fall and winterof 1999. Data, presented in the form ofvignettes, provide context and ascribe meaningto the rich and imaginative voice of the smallfamily farmer. It is concluded that thissuccess is directly attributed to the conceptof flexible diversification.It is through flexible diversification thatTitus Farms has imbued stability andsustainability seldom seen by similar familyfarms of this type, and for that reason alone,Titus Farms is a successful enterprize. This study was designed to observe the effects of conjugated linoleic acid (CLA) supplementation on lean content of pork carcass primal cuts (hams, loins, and bellies) and to determine the ability of total body electrical conductivity (TOBEC) to predict lean content. A total of 64 crossbred growing-finishing barrows were placed on a control (soybean oil) or CLA (0.75%) diet at an average weight of 40 kg. Pigs were penned in pairs according to diet and stress genotype (negative, carrier, and positive) and slaughtered at 115 kg. Stress genotype was included because of known variations in lean content. Hams (IMPS 401A; n = 64), loins (IMPS 410; n = 24), and bellies (IMPS 408; n = 63) were fabricated from carcasses at 24 h postmortem and scanned for electromagnetic (EM) absorption by a MQ-25 EM scanner. Each wholesale cut was scanned in triplicate at 2.5 MHz to yield a peak mean average (PMA) value then separated into lean, fat, bone, and skin components. Bellies were skinned prior to scanning then subjected to a belly bar firmness test before dissection. Supplementation with CLA had no effect (P > 0.05) on lean ham composition. Regression analysis was used for lean weight prediction using primal weight and PMA value as predictors. Lean content prediction of hams by TOBEC resulted in an R2 of 0.80. Loins from CLA-supplemented pigs exhibited increased lean weight (P < 0.05) and PMA values (P < 0.05) compared to controls. Lean prediction of loins by TOBEC resulted in an R2 of 0.66. Bellies from CLA-supplemented pigs had a higher percentage of moisture (P < 0.03) and protein (P < 0.01) and decreased percentage of lipid (P < 0.01). The R2 values from the regression analysis predicting protein, moisture, protein + moisture, and fat-free soft tissue composition of the skinless bellies were 0.67, 0.68, 0.71, and 0.78, respectively.  相似文献

Immune function (response to concanavalin A, cytokine production, and lymphocyte profiles) and blood chemistry variables were measured in growing-finishing pigs (Yorkshire/Landrace/Duroc dam × Hampshire sire) fed varying percentages of CLA (0, 0.12, 0.25, 0.50, and 1.0%). Blood was collected at 0, 14, 28, 42, and 56 d on feed (DOF). Total white blood cell (WBC) count increased (P < 0.01) linearly to 42 DOF. No differences (P = 0.53) were observed for WBC across CLA treatment. Nitric oxide was greater (P < 0.01) for the 1.0% CLA treatment compared with all other treatments. Flow cytometry using fluorescent labeled monoclonal antibodies to the CD4, CD8, double-positive CD4/CD8, and CD2 surface markers was used to determine lymphocyte subpopulations. Supplementation of CLA had no effect (P = 0.61) on lymphocyte subpopulation cell distribution. Most blood chemistry variables were within the normal metabolic range for pigs. A decrease was observed over DOF for P (P < 0.01) and K (P < 0.05). Additionally, Na and Cl concentrations increased (P < 0.05) from 14 to 28 DOF and decreased over the remainder of the trial. Electrolyte balance was not different (P = 0.38) across CLA treatments and was likely explained by no differences in feed intake among the CLA treatment groups. Blood lipid variables indicated that total cholesterol (P < 0.001), triglycerides (P < 0.001), high-density lipoproteins (P < 0.001), and low-density lipoproteins (P < 0.01) increased as the amount of CLA in the diet increased, but none of the results from these treatments exceeded the normal range of acceptability. These results suggested that CLA was safe when fed to growing-finishing pigs and had little effect on their immune function and blood chemistry variables.  相似文献
