Little is known about the signaling mechanisms that determine the highly regular patterning of the intestinal epithelium into crypts and villi. With the use of mouse models, we show that bone morphogenetic protein (BMP)-4 expression occurs exclusively in the intravillus mesenchyme. Villus epithelial cells respond to the BMP signal. Inhibition of BMP signaling by transgenic expression of noggin results in the formation of numerous ectopic crypt units perpendicular to the crypt-villus axis. These changes phenocopy the intestinal histopathology of patients with the cancer predisposition syndrome juvenile polyposis (JP), including the frequent occurrence of intraepithelial neoplasia. Many JP cases are known to harbor mutations in BMP pathway genes. These data indicate that intestinal BMP signaling represses de novo crypt formation and polyp growth.  相似文献

Phytophthora ramorum has been detected in official plant health surveys on Rhododendron, Viburnum and Camellia in ornamental nurseries in northern Spain since 2003. A collection of 94 isolates of P. ramorum was obtained from 2003 to 2008 from plants with symptoms at different geographical locations. Isolates were identified based on morphology and sequence of the rDNA ITS region. Mating type, genetic variation, sensitivity to phenylamide fungicides and aggressiveness of these isolates were determined. All isolates belonged to the A1 mating type, ruling out the possibility of genetic recombination. Seven microsatellite markers were used to study genetic diversity; three out of the seven microsatellite markers were polymorphic within the Spanish population of P. ramorum. This study confirms that all Spanish isolates of P. ramorum belonged to the EU1 lineage. Twelve intralineage genotypes were detected, five that are unique to Spain (EU1MG38, EU1MG41, EU1MG37, EU1MG39 and EU1MG40) and seven that are also present in at least one other European country (EU1MG1, EU1MG29, EU1MG22, EU1MG13, EU1MG2, EU1MG18 and EU1MG26). Genotypes EU1MG37, EU1MG39 and EU1MG40 were isolated from Rhododendron from one region; EU1MG38 and EU1MG41 were isolated from Camellia from two different regions. Isolates of genotype EU1MG38 were resistant to metalaxyl and mefenoxam. The level of genetic diversity within the Spanish population of P. ramorum is limited and indicates a relatively recent clonal expansion.  相似文献

Plantain virus X was first recognized by the ICTV as a species in the genus Potexvirus in 1982. However, because no sequence was available for plantain virus X (PlVX), abolishing the species was proposed to the Flexiviridae working group of the ICTV in 2015. This initiated efforts to sequence the original isolates from Plantago lanceolata samples. Here we report the full-genome sequencing of two original isolates of PlVX, which demonstrate that the virus is synonymous to Actinidia virus X, a species previously reported from kiwifruit (Actinidia sp.) and blackcurrant (Ribes nigrum). PlVX was previously noted to be widespread in the UK in P. lanceolata. This report additionally presents novel data on the distribution and diversity of PlVX, collected at the same site as the original UK isolates, and from three independent surveys, two in the Netherlands and one in Belgium. This study also includes two new host records for PlVX, Browallia americana and Capsicum annuum (sweet pepper), indicating the virus is more widespread and infects a broader range of hosts than previously reported. This stresses the importance of surveys of noncultivated species to gain insight into viral distribution and host range. This study also demonstrates the value of generating sequence data for isolates retained in virus collections. Additionally, it demonstrates the potential value in prepublication data sharing for giving context to virus detections such as the four independent studies here which, when combined, give greater clarity to the identity, diversity, distribution, and host range of plantain virus X.  相似文献

Invasive oomycete pathogens have been causing significant damage to native ecosystems worldwide for over a century. A recent well‐known example is Phytophthora ramorum, the causal agent of sudden oak death, which emerged in the 1990s in Europe and North America. In Europe, this pathogen is mainly restricted to woody ornamentals in nurseries and public greens, while severe outbreaks in the wild have only been reported in the UK. This study presents the results of the P. ramorum survey conducted in Swiss nurseries between 2003 and 2011. In all 120 nurseries subjected to the plant passport system, the main P. ramorum hosts were visually checked for above ground infections. Phytophthora species were isolated from tissue showing symptoms and identified on the basis of the morphological features of the cultures and sequencing of the ribosomal ITS region. Phytophthora was detected on 125 plants (66 Viburnum, 58 Rhododendron and one Pieris). Phytophthora ramorum was the most frequent species (59·2% of the plants), followed by P. plurivora, P. cactorum, P. citrophthora, P. cinnamomi, P. cactorum/P. hedraiandra, P. multivora and P. taxon PgChlamydo. The highest incidence of P. ramorum was observed on Viburnum × bodnantense. Microsatellite genotyping showed that the Swiss P. ramorum population is highly clonal and consists of seven genotypes (five previously reported in Europe, two new), all belonging to the European EU1 clonal lineage. It can therefore be assumed that P. ramorum entered Switzerland through nursery trade. Despite sanitation measures, repeated P. ramorum infections have been recorded in seven nurseries, suggesting either reintroduction or unsuccessful eradication efforts.  相似文献

Stilbenes have been shown to have cancer chemopreventive activity and to protect lipoproteins from oxidative damage. A method is described for their direct determination in different types of wine using high-performance liquid chromatography with ultraviolet detection. In a survey of 120 commercial wines from Portugal and France, the highest concentrations of stilbenes were found in red wines. The glucosides of resveratrol were present in higher concentrations than the free isomers. Isolation from wine and characterization of trans-astringin in a large quantity are described for the first time.  相似文献

We studied the effects of baiting on feral swine (Sus scrofa) movements and corresponding likelihood of disease spread under real and simulated culling pressure. Our objectives were to determine the proportion of feral swine that used the bait station site, and if baiting of feral swine altered areas of utilization, distances from location centroids to treatment location (control or bait station), and movement rates by survivors during culling activities. We hypothesized that the bait station would increase the sedentary nature of feral swine, thus reducing the potential for dispersal and hence disease dispersal. Our experiment was conducted between February and May 2011 on the Rob and Bessie Welder Wildlife Foundation (WWF) in San Patricio County, Texas. We trapped 83 feral swine and placed GPS collars on 21 animals. We established and maintained a centralized bait station on one side of the WWF from 13 March to 27 April. We conducted population-wide culling activities, including trapping, controlled shooting, drive shooting, and aerial gunning, from 3 to 27 April and removed 143 feral swine (4.6feral swine/km(2)). Areas of utilization did not differ between treatments (control or bait station). However, we found location centroids of bait station site feral swine to be closer to the treatment location than those of control site animals and daily movement rates of bait station site feral swine to be 39% greater than movement rates of control site animals. Based on our observation that only 62% of feral swine trapped in proximity to the bait station used it, we cannot recommend baiting as an alternative to fences for containing animals during culling activities. However, there is value in using bait stations to describe patterns of feral swine movements, facilitate observation, and improve efficacy when conducting removals.  相似文献

The northwest European heathland landscape with its characteristic communities of nutrient-poor and acidic soils has a high nature value, because of its locally high biodiversity and the distinct site conditions. In order to conserve and restore the heathlands, numerous rehabilitation projects have been performed, although with varying success. This is partly due to the fact that the key biogeochemical processes distinguishing the various vegetation types within the heathlands are not known in detail.Therefore, we performed a statistical survey on the main communities and their soil characteristics. In addition, we analyzed the data for key factors determining biodiversity in the heathland landscape.Data from previous studies and surveys was used to compile a dataset of 267 vegetation relevés (classified as EUNIS habitat types) with extensive soil measurements (22 parameters). A canonical discriminant analysis revealed that soil acidity explained most of the differences between the habitat types, while soil moisture content and soil fertility were less important. Acidity-related factors as Al3+, Al/Ca-ratio and pH were also strongly correlated to plant diversity in the majority of the habitat types, respectively, the species-rich Nardus grasslands, the Rhynchosporion communities and the species-rich Molinia meadows. In the dry heaths and over the total heathland landscape, plant diversity was negatively correlated with soil -concentrations. Only in wet heath, nutrient availability, in this case P, was the primary factor in explaining plant diversity.This study presents ranges for all major soil parameters for the studied well-developed heathland habitat types, thereby providing clear guidelines for conservation and restoration.  相似文献
