Seminal plasma (SP) contains several types of compounds derived from the epididymides and accessory glands. The aim of this study was to examine the protein composition of different ejaculate fractions. Trial I: fractionated ejaculates were collected from two normal and two subfertile stallions. Samples containing pre‐sperm fluid and the first sperm‐rich jets (HIGH‐1), the main sperm‐rich portion (HIGH‐2), the jets with low sperm concentrations (LOW), and a combined whole‐ejaculate (WE) sample was centrifuged, and the SP was filtered and frozen. A part of each SP sample was stored (5°C, 24 h) with spermatozoa from HIGH‐2 and skim milk extender. Sperm motility was evaluated after storage in extender mixed with the stallion’s own SP or SP from one of the other stallions (sperm from a normal stallion stored in SP from a subfertile stallion and vice versa). Protein composition was analysed using reverse‐phase liquid chromatography (RP‐HPLC), N‐terminal sequencing and mass spectrometry. The area‐under‐the‐curve (AUC) was used for quantitative comparison of proteins within fractions. Trial II: semen samples were collected from seven stallions. Fractions with the highest (HIGH) and lowest (LOW) sperm concentrations and WE samples were examined using SDS‐PAGE and densitometry. No significant differences emerged between fractions in the AUC‐values of the Horse Seminal Protein‐1 (HSP‐1) and HSP‐2 peaks, or the peak containing HSP‐3 and HSP‐4 (HSP‐3/4). Levels of HSP‐1, HSP‐2 and HSP‐3/4 were not significantly correlated with total sperm motility, progressive sperm motility or average path velocity after storage. Significant differences between ejaculate fractions in the amount of different protein groups present in SP were not found in Trial I; but in Trial II, the proteins in the 60–70 kDa range were more abundant in LOW than in HIGH and WE, indicating that this band contained proteins derived mainly from the seminal vesicles, which produce most of the SP in LOW.  相似文献

Diurnal raptors were surveyed in Central Sulawesi on foot in 16 sample areas, and by car along 2700 km road transects in August-September 2000. All 23 resident species were recorded and their frequency of occurrence assessed in broad habitat categories. Farmlands were unusually poor in raptors. Of their six associated species, the formerly common black kite (Milvus migrans) may be almost extirpated. Indiscriminate pesticide use is suspected to be a major factor. Wetlands and sea-coasts are limited in area and are also the most heavily disturbed natural habitats. All four piscivorous eagles were found to be rare or very local. Among them, only the grey-headed fishing-eagle (Ichthyophaga ichthyaetus) was found to be more frequent than was previously known. Excessive human fishing pressure may be the main threat, together with extensive wetland conversion into ricefields or fishponds. Forest raptors (13 species), including all Sulawesi endemic Falconiformes, were the most numerous and widespread birds of prey. Although they exhibited a fairly high tolerance to forest degradation and fragmentation (insular syndrome), their main threat is the high deforestation rate, especially the loss of the presumably more productive lowland forests.  相似文献

A 90‐day laboratory incubation study was carried out using six contrasting subtropical soils (calcareous, peat, saline, noncalcareous, terrace, and acid sulfate) from Bangladesh. A control treatment without nitrogen (N) application was compared with treatments where urea, ammonium sulfate (AS), and ammonium nitrate (AN) were applied at a rate of 100 mg N (kg soil)–1. To study the effect of N fertilizers on soil carbon (C) turnover, the CO2‐C flux was determined at nine sampling dates during the incubation, and the total loss of soil carbon (TC) was calculated. Nitrogen turnover was characterized by measuring net nitrogen mineralization (NNM) and net nitrification (NN). Simple and stepwise multiple regressions were calculated between CO2‐C flux, TC, NNM, and NN on the one hand and selected soil properties (organic C, total N, C : N ratio, CEC, pH, clay and sand content) on the other hand. In general, CO2‐C fluxes were clearly higher during the first 2 weeks of the incubation compared to the later phases. Soils with high pH and/or indigenous C displayed the highest CO2‐C flux. However, soils having low C levels (i.e., calcareous and terrace soils) displayed a large relative TC loss (up to 22.3%) and the added N–induced TC loss from these soils reached a maximum of 10.6%. Loss of TC differed depending on the N treatments (urea > AS > AN >> control). Significantly higher NNM was found in the acidic soils (terrace and acid sulfate). On average, NNM after urea application was higher than for AS and AN (80.3 vs. 71.9 and 70.9 N (kg soil)–1, respectively). However, specific interactions between N‐fertilizer form and soil type have to be taken into consideration. High pH soils displayed larger NN (75.9–98.1 mg N (kg soil)–1) than low pH soils. Averaged over the six soils, NN after application of urea and AS (83.3 and 82.2 mg N (kg soil)–1, respectively) was significantly higher than after application of AN (60.6 mg N (kg soil)–1). Significant relationships were found between total CO2 flux and certain soil properties (organic C, total N, CEC, clay and sand content). The most important soil property for NNM as well as NN was soil pH, showing a correlation coefficient of –0.33** and 0.45***, respectively. The results indicate that application of urea to acidic soils and AS to high‐pH soils could be an effective measure to improve the availability of added N for crop uptake.  相似文献

The use of appropriate source of phosphorus (P) fertilizer at different planting densities has considerable impact on growth, grain yield as well as profitability of maize (Zea mays L). Field experiment was conducted in order to investigate the impact of P sources [(S0 = P not applied, S1 = SSP (single super phosphate) S2 = NP (nitrophos), and S3 = DAP (diammonium phosphate)] on maize growth analysis, yield and economic returns planted at different planting densities (D1 = 40,000, D2 = 60,000, D3 = 80,000, and D4 = 100,000 plants ha?1) at the New Developmental Agricultural Research Farm of Khyber Pakhtunkhwa Agricultural University, Peshawar, Pakistan, during summer 2006. This paper reports the profitability data with two objectives: 1) to compare agronomic efficiency and profitability of P-fertilizers, and 2) to know whether plant densities affect agronomic efficiency and profitability of P-fertilizers. Application of DAP and SSP resulted in higher partial factor productivity (PFP) (63.58 and 61.92 kg grains kg?1 P), agronomic efficiency (AE) (13.01 and 13.71 kg grains kg?1 P) and net returns (NR) (Rs. 16,289 and 16,204 ha?1), respectively, while NP stood at the bottom in the ranking with lower PFP (57.16 kg grains kg?1 P), AE (8.94 kg grains kg?1 P) and NR (Rs. 4,472 ha?1). Among the plant densities, D3 stood first with maximum PFP (69.60 kg grains kg?1 P), AE (18.21 kg grains kg?1 P) and NR (Rs. 21,461 ha?1) as compared to other plant densities. In conclusion, the findings suggest that growing maize at D3 applied with either SSP or DAP is more profitable in the wheat-maize cropping system in the study area.  相似文献

By using Particle-Induced X-ray Emission (PIXE) technique, the effect of arsenic (As) on the mineral contents and translocation in the xylem of rice (Oryza sativa L. cv. ‘Akihikari’) was studied. The results suggest that exogenous As increased the concentrations of phosphorus (P), calcium (Ca) magnesium (Mg), sulfur (S), and manganese (Mn) in xylem, while the concentrations of potassium (K) remained unchanged. The highest concentration of As to the rice roots did not have any clear effect on the translocation of P, Ca, S, and chlorine (Cl) in the xylem, indicating that the increasing concentrations of the minerals may be due to a condensation effect, resulting from the repression of water movement in xylem by As-toxicity. Among the metal micronutrients, As decreased the concentrations and translocations of iron (Fe), zinc (Zn), and copper (Cu).  相似文献

Development of aminopolycarboxylate chelants (APCs) having enhanced biodegradability is gaining increasing focus to replace the EDTA and its homologs with those used widely for the ex situ treatment of contaminated soils and are potential eco-threats. The paper reports the chelant-assisted extraction of the toxic metals (Cd, Cu, Pb, and Zn) from the metal-spiked European reference soils (Eurosoil 1 and Eurosoil 4) using biodegradable APCs, namely EDDS, GLDA, and HIDS. The effects of chelant-to-metal molar ratio, solution pH, and metal/chelant stability constants were evaluated, and compared with that of EDTA. The selectivity aptitude of the biodegradable chelants towards the toxic metals was assumed from the speciation calculations, and a proportionate correlation was observed at neutral pH. Pre- and post-extractive solid phase distributions of the target metals were defined using the sequential extraction procedure and dissolution of metals from the theoretically immobilized fraction was witnessed. The effect of competing species (Al, Ca, Fe, Mg, and Mn) concentrations was proven to be minimized with an excess of chelant in solution. The highlight of the outcomes is the superior decontamination ability of GLDA, a biodegradable APC, at minimum chelant concentration in solution and applicability at a wide range of pH environments.  相似文献

Oocytes retrieval, in vitro maturation (IVM) and fertilization (IVF) efficiency are inevitable steps towards in vitro production of embryos. In the present study, these parameters were investigated in the ovaries of prepubertal (n = 31) and pubertal (n = 61) black Bengal goats obtained from a slaughterhouse. Nuclear maturation was evaluated upon aspiration and following IVM in TCM-199 (Earle''s salt with L-glutamine and sodium bicarbonate) for 27 h at 39℃ under 5% CO2 in humidified air. The oocytes retrieval and efficiency (mean ± SD) per prepubertal and pubertal goats were 5.2 ± 0.6 and 6.8 ± 0.6, and 77.3 ± 0.1% and 80.5 ± 0.6%, respectively. Anaphase I - telophase I stages differed significantly (7.3 ± 0.8 vs. 2.6 ± 0.2, p < 0.05) between the two groups of goats. After IVM, the percentages of metaphase II were significantly higher (66.3 vs. 60.3, p < 0.05) in pubertal goats than in their prepubertal counterparts. The percentages of normal in vitro fertilization (IVF) in Fert-Tyrode''s albumin lactate pyruvate of pubertal goat oocytes did not differ between Percoll and swim-up sperm separation methods (36.7 ± 0.9% vs. 32.7 ± 1.3%, p > 0.05). Furthermore, sperm capacitation by heparin alone or in combination with ionomycin did not lead to a significant increase in the normal fertilization rate (34.8 ± 1.7 vs. 32.2 ± 1.5%, respectively) in the oocytes of pubertal goats. In conclusion, the ovaries of pubertal black Bengal goats obtained from the slaughterhouse could be used for in vitro embryo production. However, further optimization of the IVM and IVF techniques are necessary for satisfactory in vitro embryo production.  相似文献

The bovine embryonic signal interferon‐τ (IFN‐τ) produced by the trophoblast is known to pass through the uterine fluid towards the endometrium and further into the maternal blood, where IFN‐τ induces specific expression of interferon‐stimulated gene expression (ISG), for example in peripheral leucocytes. In sheep, it was shown experimentally by administration of IFN‐τ that ISG is also detectable in the liver. The objective was to test whether ISG can be detected in liver biopsy specimens from Holstein–Friesian heifers during early pregnancy. Liver biopsies were taken on day 18 from pregnant and non‐pregnant heifers (n = 19), and the interferon‐stimulated protein 15 kDa (ISG‐15) and myxovirus‐resistance protein‐1 (MX‐1) gene expression was detected. The expression of both MX‐1 (p: 24.33 ± 7.40 vs np: 9.00 ± 4.02) and ISG‐15 (p: 43.73 ± 23.22 vs 7.83 ± 3.63) was higher in pregnant compared to non‐pregnant heifers (p < 0.05). In conclusion, pregnancy induced ISG‐15 and MX‐1 gene expression in the liver already at day 18 in cattle.  相似文献

We previously determined that five rather hydrophobic metabolites appeared in blood plasma after oral administration of cyanidin 3-O-beta-D-glucopyranoside, but a group of hydrophilic metabolites still remained unidentified. In the present study, 12 hydrophilic metabolites found were collected from urine and plasma samples by high-performance liquid chromatography (HPLC) and then analyzed by tandem MS spectrometry. From the MS spectra, four metabolites out of 12 were assigned as glucuronides of cyanidin 3-O-beta-D-glucopyranoside and six out of 12 were glucuronides of the primary metabolites of cyanidin 3-O-beta-D-glucopyranoside (O-methyl cyanidin 3-O-beta-D-glucopyranoside). Extended glucuronides of cyanidin 3-O-beta-D-glucopyranoside and O-methyl cyanidin 3-O-beta-D-glucopyranoside showed their maximum plasma concentrations at 15 and 60 min (or 30 min) after oral administration, respectively. Their maximum plasma concentrations ranged from 15 to 70 nM. From the profile of urinary-excreted anthocyanins after intravenous administration, it was deduced that extended glucuronides of cyanidin 3-O-beta-D-glucopyranoside and O-methyl cyanidin 3-O-beta-D-glucopyranoside were mainly produced in the liver rather than by intestinal flora. The area under the plasma concentration curve was 0.25 micromol min/L for extended glucuronides of cyanidin 3-O-beta-D-glucopyranoside and 0.14 micromol min/L for O-methyl cyanidin 3-O-beta-D-glucopyranoside, respectively, when evaluated as cyanidin 3-O-beta-D-glucopyranoside equivalent, indicating that extended glucuronidation is a critical pathway in cyanidin 3-O-beta-D-glucopyranoside metabolism in rats.  相似文献
