In the present study, two methods for the genetic identification of the most important seaweed species used for human consumption were developed. Both are carried out through PCR amplification of an 18S rRNA gene fragment. The first one is based on the phylogenetic analysis of DNA sequences (FINS), while the second is based on length polymorphism and RFLP visualized by means of an ALF system. The main novelty of this work lies in the fact that it allows genetic identification of the main commercial species of seaweed. Moreover, the developed systems can be applied to all kinds of processed products, including those that have undergone intensive transformation, as for instance canned foods. These methodologies also permit the detection of species in complex matrixes where more than one algal species is present. The methods were validated using products manufactured in a pilot plant showing correct functioning. Finally, the methods were applied to 23 commercial samples including some that had been subjected to intensive thermal treatment, allowing the detection of those that were incorrectly labeled (30%). Therefore, these molecular tools can be used for clarifying questions related to the correct labeling and traceability of commercial products that include some seaweeds in their composition.  相似文献

In Brazil, citrus black spot (CBS) caused by Guignardia citricarpa is a major disease that has different symptoms on fruit. In this study, fruit of Citrus sinensis infected by G. citricarpa and showing the symptoms false melanosis, freckle spot and hard spot were cross-sectioned and analysed anatomically and histochemically by light microscopy. Immuno-histological assays were performed. All symptoms were accompanied by a thickening of the cuticle. False melanosis lesions did not contain pycnidia and remained restricted to the epicarp or to the first layers of the mesocarp. The stomata in this type of lesion showed phenolic compounds in the guard cells and in the sub-stomatal chamber. In some samples, the guard cells and their surrounding cells lysed, and a wound meristem began to form underneath them. Freckle spot and hard spot lesions had very similar histological alterations to the epicarp and mesocarp, but in our samples only hard spot lesions contained pycnidia. Both of these symptoms were accompanied by protein inclusions. Epidermal and sub-epidermal cells located in the oil-gland region were obliterated, causing alterations in these structures. All symptoms had regions that stained strongly for lipids and phenols.  相似文献

Endotheliotropic herpesvirus causes a fatal disease in young Asian elephants, but there are no methods for identifying latent carriers of the virus. During the postmortem study of one female African elephant and three male and two female Asian elephants, a lymph node located bilaterally caudoventral to the parotid gland, approximately 1.5 to 5 cm below the skin, was identified as suitable for transcutaneous ultrasound-guided biopsy. An ultrasonographic assessment and two biopsies were performed on 39 Asian elephants, and these lymph nodes were classified ultrasonographically as active, inactive or chronically active. The calculated mean (se) volume of 10 active lymph nodes was 17.4 (6.9) cm(3), and that of three chronically active lymph nodes was 10.6 (1.0) cm(3), whereas the mean volume of 17 inactive lymph nodes was 3.1 (0.6) cm(3). The presence of lymph node tissue in samples obtained by ultrasound-guided biopsy from three animals that were maintained under conditions that allowed for additional sampling was confirmed histologically. The dna extracted from the lymphoid tissue and the whole blood of all the elephants was negative for endotheliotropic herpesvirus by PCR.  相似文献

Shiga toxin (Stx)-producing Escherichia coli (STEC) strains isolated from animals and food in Argentina (n=44) and Brazil (n=20) were examined and compared in regard to their phenotypic and genotypic characteristics to evaluate their pathogenic potential. The clonal relatedness of STEC O157 isolates (n=22) was established by phage typing (PT) and pulsed-field gel electrophoresis (PFGE). All O157 strains studied carried eae and enterohemorrhagic E. coli (EHEC)-hly sequences. In Argentina, these strains occurred both in cattle and meat, and 50% of them carried stx2/stx2vh-a genes, whereas in Brazil the O157 strains were isolated from animals, and most harbored the stx2vh-a sequence. At least 13 different O:H serotypes were identified among the non-O157 strains studied, with serotype O113:H21 being found in both countries. All but one non-O157 strains did not carry eae gene, but EHEC-hlyA gene was found in 85.7% of them, and the stx2 genotype was also more prevalent in Argentina than in Brazil (P<0.01), where stx1 alone or in association was most common (68.8%). One STEC strain isolated from a calf in Brazil harbored the new variant referred to as stx2-NV206. PFGE analysis showed that STEC O157 strains were grouped in four clusters. One Brazilian strain was considered possibly related (> or =80%) to Argentinean strains of cluster I. Differences in the pathogenic potential, especially in regard to serotypes and stx genotypes, were observed among the STEC strains recovered from animals and food in both countries.  相似文献

The effect of trachyloban-19-oic acid isolated from Iostephane heterophylla (Asteraceae), was investigated on several photosynthetic activities in spinach thylakoids. The results indicated that this compound inhibited ATP synthesis and uncoupled electron transport, as well as basal and phosphorylating electron flow. Therefore, trachyloban-19-oic acid acts as Hill reaction inhibitor. This compound did not affect photosystem I activity but inhibited uncoupled photosystem II electron flow from H2O to DCPIP, and has not effect on electron flow from H2O to SiMo, indicating that the site of inhibition of this compound is at the level of QA-QB. Chlorophyll a fluorescence measurements confirm the behavior of this diterpene as QA-QB inhibitor, and in the other hand, this results indicate that a perturbation in the thylakoid membranes at the level of LHC II occurs.  相似文献

The aim of this study was to investigate how long-term feeding of ganders with diets containing different levels of phytoestrogens affects the morphometry of testes, plasma steroid levels, sperm parameters and reproductive performance. Male Bilgoraj geese were fed diets containing grass meal (with low phytoestrogen levels) and those containing alfalfa meal and soybean meal (with higher levels of phytoestrogens). Testes were obtained from ganders at the peak of mating and the ongoing laying period as well as at the beginning of photorefractoriness. The morphometric parameters were measured in frozen testicular sections. Blood was collected monthly from December to June, and plasma testosterone, androstenedione and corticosterone concentrations were determined. During the mating season, semen was collected and its quantity and quality were investigated. In the flocks (ganders with geese) reproductive output was noted. Seasonal changes were observed in the morphology of testes and in plasma testosterone, androstenedione and corticosterone concentrations in ganders. The feeding of ganders with dietary meals containing higher levels of phytoestrogens did not affect the plasma steroid contents or the majority of the morphometric parameters of testes, except for the height of the seminiferous epithelium, which was lowered. Moreover, diets with higher levels of phytoestrogens decreased the volume of ejaculates and the percentage of normal spermatozoa. However, these changes were not reflected in the fertilisation rates.  相似文献

Background: This study addresses the antitumoral properties of Penicillium purpurogenum isolated from a polluted lagoon in Northeastern Brazil. Methods: Ethyl Acetate Extracellular Extract (EAE) was used. The metabolites were studied using direct infusion mass spectrometry. The solid Ehrlich tumor model was used for antitumor activity. Female Swiss mice were divided into groups (n = 10/group) as follows: The negative control (CTL−), treated with a phosphate buffered solution; the positive control (CTL+), treated with cyclophosphamide (25 mg/kg); extract treatments at doses of 4, 20, and 100 mg/kg; animals without tumors or treatments (Sham); and animals without tumors treated with an intermediate dose (EAE20). All treatments were performed intraperitoneally, daily, for 15 days. Subsequently, the animals were euthanized, and the tumor, lymphoid organs, and serum were used for immunological, histological, and biochemical parameter evaluations. Results: The extract was rich in meroterpenoids. All doses significantly reduced tumor size, and the 20 and 100 mg/kg doses reduced tumor-associated inflammation and tumor necrosis. The extract also reduced the cellular infiltration of lymphoid organs and circulating TNF-α levels. The extract did not induce weight loss or renal and hepatic toxic changes. Conclusions: These results indicate that P. purpurogenum exhibits immunomodulatory and antitumor properties in vivo. Thus, fungal fermentation is a valid biotechnological approach to the production of antitumor agents.  相似文献

Citrus greasy spot (CGS), caused by Zasmidium citri, induces premature defoliation and yield loss in Citrus spp. The epidemiology of CGS is well understood in high humidity areas, but remains unaddressed in Brazil, despite differing climatic conditions and disease management practices. The spatiotemporal dynamics of CGS were characterized in the Recôncavo of Bahia (Brazil) at four hierarchical levels (quadrant, plant, grove, and region). A survey conducted in 19 municipalities found the disease to be present throughout the region with an incidence of 100% in groves and plants, and higher than 70% on leaves. Index of dispersion (D) values suggest the spatial pattern of units with symptoms lies between random and regular. This was confirmed by the parameters of the binary power law for plants and their quadrants (log[A] < 0 and b < 1). No consistent differences were observed in the disease incidence at different plant heights. We introduce a compartmental model synthesizing CGS epidemiology. The collected data allow such a model to be parameterized, albeit with some ambiguity over the proportion of new infections that result from inoculum produced within the grove versus external sources of infection. By extending the model to include two populations of growers—those who control and those who do not—coupled by airborne inoculum, we investigate likely performance of cultural controls accessible to citrus growers in northeastern Brazil. The results show that control via removal of fallen leaves can be very effective. However, successful control is likely to require area-wide strategies in which a large proportion of growers actively manage disease.  相似文献
