Hazelnut is one of the most commonly consumed tree nuts, being largely used by the food industry in a wide variety of processed foods. However, it is a source of allergens capable of inducing mild to severe allergic reactions in sensitized individuals. Hence, the development of highly sensitive methodologies for hazelnut traceability is essential. In this work, we developed a novel technique for hazelnut detection based on a single-tube nested real-time PCR system. The system presents high specificity and sensitivity, enabling a relative limit of detection of 50 mg/kg of hazelnut in wheat material and an absolute limit of detection of 0.5 pg of hazelnut DNA (1 DNA copy). Its application to processed food samples was successfully achieved, detecting trace amounts of hazelnut in chocolate down to 60 mg/kg. These results highlight the adequacy of the technique for the specific detection and semiquantitation of hazelnut as potential hidden allergens in foods.  相似文献

A bioactivity-guided chemical study of aerial parts of Croton ciliatoglanduliferus Ort. led to the isolation for the first time of the flavonoids retusin (5-hydroxy-3,7,3',4'-tetramethoxyflavone) (1) and pachypodol (5,4'-dihydroxy-3,7,3'-trimethoxyflavone) (2) from the n-hexane extract. Compounds 1 and 2 were separated by preparative thin-layer chromatography. Compound 2 was the most active compound on ATP synthesis inhibition. The I50 value was 51 microM. Pachypodol behaves as a Hill reaction inhibitor. It inhibited the uncoupled electron flow on photosystem II partial reaction from water to dichlorophenol indophenol (DCPIP) and from water to sodium silicomolybdate. However, the uncoupled partial reaction from diphenylcarbazide to DCPIP and the uncoupled photosystem I from DCPIPred to MV were not inhibited by 2. These results were corroborated by fluorescence decay data. Therefore, pachypodol inhibits the water-splitting enzyme activity. Compound 1 with a 4'-methoxy group was a weak inhibitor, indicating that the 4' free -OH group is important for strong inhibition.  相似文献

A semiautomatic method for the determination of seven N-nitrosamines in beverages by gas chromatography with nitrogen-phosphorus detection is proposed. Beverage samples are aspirated into a solid-phase extraction module for preconcentration and cleanup. The influence of the experimental conditions was examined by using various sorbents among which LiChrolut EN was found to provide quantitative elution and the highest preconcentration factors of all. The proposed method is sensitive, with limits of detection between 7 and 33 ng/kg, and precise, with relative standard deviations from 4.3% to 6.0%. The recoveries of N-nitrosamines from beverage samples spiked with 0.5 or 1 microg/kg concentrations of these compounds ranged from 95% to 102%. The method was successfully applied to the determination of residues of the studied N-nitrosamines in beverages including beer, wine, liquor, whisky, cognac, rum, vodka, grape juice, cider, tonic water, and soft drinks. The analytes were only detected in beer samples, positives being confirmed by gas chromatography coupled with impact ionization mass spectrometry.  相似文献

In Brazilian Tropical Cerrado Region, reactions related to parental-material breakdown and oxisols genesis strongly affect the groundwater chemical characteristics. These reactions are responsible for the low water pH values, the soil charge balance, and the re-equilibrium of dissolved inorganic carbon species. Brasilia, the capital of Brazil, lies in the Distrito Federal, in the central part of the Brazilian Cerrado Region and has been experiencing an intense urban development during the last 15 years. Many town-house complexes have been built without proper water supply or wastewater disposal conditions. In this work, we discuss the adequacy of traditional ground waters geochemical classification models applied to the most exploited aquifers in DF. Forty groundwater samples were collected and pH, conductivity and total dissolved solids were determined in the field. Na, K, Ca, Mg, Fe, Al, Cu, Cd, Cr Mn e Zn were determined by Inductively Coupled Plasma Atomic Emission Spectrometry and Atomic Absorption Spectrophotometry. We identify inorganic indicators of human interference on the quality of these waters and also suggest a new mean of interpretation of dissolved inorganic elements based on statistic treatment of multivariate data. It is revealed a particular geochemical feature concerning to the alkali and the earth alkali metals concentrations, and a typical trace-element behaviour, closely related to the human activities and to the geological-bearing material characteristics. For all of the samples supposed to be altered by any kind of human activity, the physico-chemical parameters were lower than the standards established by the water quality legal system.  相似文献

A method for the simultaneous determination of aluminum (Al), arsenic (As), calcium (Ca), cadmium (Cd), chromium (Cr), copper (Cu), iron (Fe), magnesium (Mg), manganese (Mn), phosphorus (P), lead (Pb), sulfur (S), selenium (Se), vanadium (V), and zinc (Zn) in organic soil amendments using microwave-assisted acid digestion and inductively coupled plasma optical emission spectroscopy (ICP OES) is proposed. Concentrated or diluted acids mixtures (HNO3, HF, HBF4, and H3BO3) combined or not with H2O2 were systematically evaluated in order to achieve the best digestion procedure for masses of around 150 mg of samples. Principal component analysis (PCA) was applied in order to choose the best acid mixture for digestion (3 mL HNO3 + 1 mL HBF4 + 2 mL H2O). The determined concentrations were in accordance with certified values of CRM 029 at the 95% confidence level, according to the Student-t test. This acid mixture was successfully applied for the digestion of four organic soil amendment samples (organic fertilizers, substrates, and soil conditioners) and element determination.  相似文献

Cellulose nanofibers from durum wheat straw ( Triticum durum ) were produced and characterized to study their potential as reinforcement fibers in biocomposites. Cellulose was isolated from wheat straw by chemical treatment. Nanofibers were produced via an electrospinning method using trifluoroacetic acid (TFA) as the solvent. The nanofibers were 270 ± 97 nm in diameter. Analysis of the FT-IR spectra demonstrated that the chemical treatment of the wheat straw removed hemicellulose and lignin. XRD revealed that the crystallinity of the cellulose was reduced after electrospinning, but nanofibers remained highly crystalline. The glass transition temperature (T(g) value) of the fibers was 130 °C, higher than that of cellulose (122 °C), and the degradation temperature of the fibers was 236 °C. Residual TFA was not present in the nanofibers as assessed by the FT-IR technique.  相似文献

The quality of three types of treated sludges (sludge from the paper recycling industry, and sewage sludge treated by composting with pruning waste and thermally dried) was assessed in this work. The amendments were physicochemically characterized and evaluated for their microbiological quality and for contamination with pollutants. All three organic amendments showed an absence of pathogens, so they are microbially suitable for agricultural use. The pollutants studied in the amendments included eight heavy metals and 83 organic compounds. Heavy metals, pesticides, polycyclic aromatic hydrocarbons, octylphenol, bisphenol A, and parabens were found in the organic amendments. The amount and type of contaminants depended on the sample sources and differed between samples from a single origin. The presence of some of the pollutants found has not been previously reported in organic amendments. The germination assay showed that these amendments may be applied to soil in controlled doses. As far as we know, this is the first reported study that has evaluated a great variety of pollutants (heavy metals, pesticides, persistent and emerging organic pollutants) in sludge from the recycled paper industry; moreover, studies in other waste products in which such a broad range of pollutants has been evaluated have been scarce until now due to the complexity of these matrices.  相似文献

The effect of fermentation on antinutritional factors and also on total dietary fiber (TDF), insoluble (IDF) and soluble (SDF) dietary fiber fractions was studied in beans (Phaseolus vulgaris L.). The processes studied were two types of fermentation (lactic acid and natural), and a portion of the obtained flours were processed by autoclaving. The dietary fiber (DF) content and its components were determined using the enzymatic-gravimetric and enzymatic-chemical methods. The TDF content ranged from 24.5% dry matter (DM) in the raw to 25.2% DM in the processed beans. All the processing treatments significantly decreased the SDF content, and irrelevant changes were noticed in the IDF content of processed beans. Cellulose content of all samples was reduced by the processing treatments. Correspondingly, higher amounts of resistant starch was observed in the processed beans, except in the lactic acid fermented ones. However, the levels of pectic polysaccharides and Klason lignin were higher in the samples fermented by Lactobacillus plantarum. The action of microorganisms was determinant for the different degradation of the bean cell wall, disrupting the protein-carbohydrate integration, thus reducing the solubility of DF.  相似文献

Aliphatic and aromatic hydrocarbons, fatty acids and heavy metals (Pb, Cu, Ni, Zn, Fe and Cd) were analyzed in bulk deposition samples at two locations within the metropolitan area of Caracas, Venezuela. One of the sampling sites was located in the center (urban site, 1), while the other was in the outkirts of the city (rural site, 4). In general, a higher flux was observed for the deposition of pollutants at site 1 than at site 4. These fluxes showed relatively large short-term variations, while the long-term integrated deposition was found to be relatively constant throughout the sampling period. Wet deposition did not affect the flux of most of the pollutants studied, with the exception of Pb and the fatty acids of an anthropogenic origin.  相似文献

Neuroprotection in glaucoma using epoetin beta (EPOβ) has yielded promising results. Our team has developed chitosan-hyaluronic acid nanoparticles (CS/HA) designed to carry EPOβ into the ocular globe, improving the drug’s mucoadhesion and retention time on the ocular surface to increase its bioavailability. In the present in vivo study, we explored the possibility of delivering EPOβ to the eye through subconjunctival administration of chitosan-hyaluronic acid-EPOβ (CS/HA-EPOβ) nanoparticles. Healthy Wistar Hannover rats (n = 21) were split into 7 groups and underwent complete ophthalmological examinations, including electroretinography and microhematocrit evaluations before and after the subconjunctival administrations. CS/HA-EPOβ nanoparticles were administered to the right eye (OD), and the contralateral eye (OS) served as control. At selected timepoints, animals from each group (n = 3) were euthanized, and both eyes were enucleated for histological evaluation (immunofluorescence and HE). No adverse ocular signs, no changes in the microhematocrits (≈45%), and no deviations in the electroretinographies in both photopic and scotopic exams were observed after the administrations (p < 0.05). Intraocular pressure remained in the physiological range during the assays (11–22 mmHg). EPOβ was detected in the retina by immunofluorescence 12 h after the subconjunctival administration and remained detectable until day 21. We concluded that CS/HA nanoparticles could efficiently deliver EPOβ into the retina, and this alternative was considered biologically safe. This nanoformulation could be a promising tool for treating retinopathies, namely optic nerve degeneration associated with glaucoma.  相似文献
